2′-Epi-uscharin from the Latex of Calotropis gigantea with HIF-1 Inhibitory Activity

Parhira Supawadee,Zhu Guo-Yuan,Jiang Ren-Wang,Liu Liang,Bai Li-Ping,Jiang Zhi-Hong

Two stereoisomeric cardenolides, uscharin (1) and a new compound, 2′- epi -uscharin (2), were isolated from the latex of Calotropis gigantea (Asclepiadaceae). Their structures were fully elucidated based on their spectroscopic data, X-ray crystallographic data and chemical evidences. Both epimers (1 and 2) exhibited strong inhibitory effects on HIF-1 activity with different magnitudes. Compound 1 showed much more potent activity than 2 and digoxin, a well-known HIF-1 inhibitor. Discrepancy in potencies between 1 and 2 revealed the contribution of a β-configuration of 2′ hydroxyl moiety for HIF-1 inhibitory activity. This is a first report of the activity of HIF-1 inhibition of thiazoline ring-containing cardenolides.

Open hollow structured Calotropis gigantea fiber activated persulfate for decomposition of perfluorooctanoic acid at room temperature

Bihui Niu,Min Yu,Changduo Sun,Luhan Wang,Kun Zang,Xiaowen Hu,Lei Zhou,Yian Zheng

Perfluorooctanoic acid (PFOA) is an anthropogenic persistent organic compound that resistant to traditional oxidative decomposition, bringing thus huge pressure to the environment. Here, the open hollow structured Calotropis gigantea fiber (CGF) was developed using a facile pyrolysis and subsequent magnetization method, and the resulting magnetic carbonized fiber (MC-CGF) was applied as an efficient catalyst for PFOA decomposition at room temperature in persulfate-based oxidation process. The experimental results indicated that with the addition of MC-CGF, the removal efficiency of PFOA could reach to 75.76% at 30℃ and pH 2.0. By calculation, the apparent activation energy was reduced from 62.76 kJ/mol to 22.49 kJ/mol, demonstrating that in the presence of MC-CGF, a lower reaction temperature and a shorter time were enough to reach the higher efficiency for PFOA decomposition. Mechanism research showed that during the decomposition process, PFOA underwent a first decarboxylation process and two possible decomposition pathways should also be taken into discussion. Taking pakchoi as the plant model, the process toxicity was finally evaluated, and the changes in chlorophyll content stated that the decomposition process of PFOA by MC-CGF assisted persulfate system was relatively harmless and environmental-friendly.

Calotropis gigantea fiber confined ZIF-67 derived Co, N in-situ assembled hollow carbon fiber to activate peroxymonosulfate for degradation of perfluorooctanoic acid

Wenli Yao,Fenghui Qin,Ziwei Zhao,Zhijun Ma,Chunyang Hu,Lei Zhou,Yian Zheng

Perfluorinated compounds have serious impacts on ecosystems and threats to human health, and exploring technologies to remove them is urgently needed. Using Calotropis gigantea fiber (CGF) as the biological template, the precursor of ZIF-67 was firstly in-situ anchored in the inner/outer walls of the fiber and then pyrolyzed to yield the composite catalyst labelled as Co/C@C. In combination with peroxymonosulfate (PMS), the advanced oxidation system, i.e. Co/C@C/PMS, was constructed for the catalytic removal of perfluorooctanoic acid (PFOA) from water. The experimental results showed that under the optimized conditions of PMS dosage of 1.0 g/L, catalyst dosage of 0.5 g/L and pH 3.0, PFOA achieved the degradation efficiency of 72.2% within 4 h at ambient temperature. Combined with free radical quenching, electron paramagnetic resonance, electrochemistry and nitroblue tetrazolium dichloride-based UV-Vis analysis, the reactive oxygen species and diverse pathways were identified, including SO 4 • − SO4•− , O 2 • − O2•− , O 2 1 O21 and electron transfer process in the Co/C@C/PMS system. The coexisting anions ( HCO 3 − HCO3− , Cl − Cl− , H 2 PO 4 − H2PO4− and NO 3 − NO3− ) and real wastewaters showed no significant effects on PFOA degradation, demonstrating that the Co/C@C/PMS system has good resistance to external ions and different media. After four cycles, the degradation efficiency of PFOA was still 62.8%, indicating that Co/C@C has high stability and good reusability. The degradation products of PFOA were mainly short-chain perfluorinated carboxylic acids (C4-C7) and hydrogen-substituted perfluorinated compounds. Based on quantitative structure-activity relationship analysis, their ecotoxicity was finally evaluated via ECOSAR method. In short, natural CGF derived Co/C@C has great potential for the reduction of emerging PFOA to achieve a sustainable water environment for humans.

Multi-functional Calotropis gigantea fabric using self-assembly silk fibroin, chitosan and nano-silver microspheres with oxygen low-temperature plasma treatment

Zeyu Zhao,Jia Yan,Tao Wang,Yanxue Ma,Maobin Xie,Xuan Mu,Xiaoqin Wang,Zhaozhu Zheng,Yuling Li,Gang Li

The present paper describes a novel multi-functional Calotropis gigantea (CG) fabric with durable antibacterial property and comfortable wearability for various applications. First, antibacterial microspheres (AMs) were prepared by self-assembly of silk fibroin (SF), chitosan (CS) and nano-silver microspheres (NSMs). Oxygen low-temperature plasma (OLTP) treatment was used to improve the adhesion between AMs and CG yarn. The AMs modified CG fabric has soft hand feeling, high moisture regain (10.37 ± 1.87%), good breathability, strong wrinkle resistance, and durable antibacterial properties. The antibacterial rate of the modified CG fabric against Escherichia coli ( E. Coil ) and Staphylococcus aureus ( S. Aureus ) is as high as 99.9 ± 0.1%, and it still has an antibacterial activity after washing 20 times (90.24 ± 0.65% and 80.25 ± 1.14%, respectively). The in vivo biocompatibility test showed clear signs of angiogenesis at the implantation site in the rats. Thus, this study offers the foundation for the development of functional CG fiber-based biomedical textiles.

Cardenolides from Calotropis gigantea as potent inhibitors of hypoxia-inducible factor-1 transcriptional activity

Supawadee Parhira,Guo-Yuan Zhu,Ming Chen,Li-Ping Bai,Zhi-Hong Jiang

Ethnopharmacological relevance Calotropis gigantea (L.) Dryand (Apocynaceae) is a medicinal plant native to southern China, India and Southeast Asia. It has been traditionally used for the treatment of several diseases including cancers in these countries. Aim of the study This study aimed to isolate bioactive cardenolides from C. gigantea , to screen their hypoxia-inducible factor (HIF-) 1 inhibitory activity, and to analyze the structure-activity relationship (SAR). Materials and methods Isolation and purification of cardenolides from the latex and the fruits of C. gigantea were performed by using a series of separation techniques. Their structures were fully characterized by elucidating their NMR and HRMS data. The HIF-1 inhibitory activities of cardenolides were evaluated by using a T47D cell-based dual-luciferase reporter assay. The potent cardenolides were selected to further evaluate their dose-response manner. Cytotoxic effects of selected cardenolides were also examined against breast cancer cell line (MCF-7) and normal mammary epithelial cell line (MCF-10A) by MTT assay. Results Among twenty isolated cardenolides, compounds 1 , 3 , 4 , 6–8 , 14 and 17 exhibited stronger HIF-1 inhibitory activities than that of digoxin, a well-known HIF-1 inhibitor ( P <0.001). These eight cardenolides inhibited HIF-1 transcriptional activity in a dose-dependent manner with IC 50 values in nanomolar potency (21.8–64.9   nM). An analysis of SAR revealed the great contributions of a β -configuration of the substituents at positions of C-2′ and C-3′, an aldehydic moiety on C-19, and the dioxane moiety between the aglycone and sugar parts of cardenolides to the HIF-1 inhibitory activity. In contrast, a hydroxyl group at any positions of C-15, C-16 and C-4′ of cardenolides showed negative effects on suppressing HIF-1 transcriptional activity. In addition, these eight cardenolides also exhibited potent cytotoxic effects against human breast cancer cell MCF-7 (IC 50 values ranged from 30.5 to 68.8   nM), but less toxic effects to human normal mammary epithelial cell MCF-10A (IC 50 values >20   µM). Conclusions This is the first report of a comprehensive study of SAR on cardenolides from C. gigantea as HIF-1 inhibitors. Eight cardenolides ( 1 , 3 , 4 , 6–8 , 14 and 17 ) showed both potent HIF-1 inhibitory activity and strong cytotoxic effect against MCF-7 cancer cells in nanomolar level. The findings of these cardenolides provided important insights into the development of these potent HIF-1 inhibitors as anticancer drug .
