The seed cake of Camellia oleifera Abel. (SCOC), a big residue produced after Camellia oil extraction, enriches with many bioactive components. However, it is often discarded directly due to the lack of cost-effective extraction. Herein, a three-phase partitioning (TPP) system is first time tried for the simultaneous extraction of three major ingredients i.e. residual oil, protein, and polysaccharide from SCOC. Results from the extraction yields show that the established method is more efficient or at least comparable to conventional techniques. The developed method takes only 1.5 h and can be carried out at mild temperature of 40 °C, making it a cost-effective method. The successful scaling up of the extraction system at a 30 L extraction tank confirms its scalability. Furthermore, compared with conventional extraction methods, the TPP extracts shows better product quality. These facts, along with simultaneous extraction features of the method, allow us to use TPP as a promising extraction technology of SCOC biomolecules.

Camellia oleifera Abel is a traditional and high-quality oil source in China, characterized as polyphenols. This study evaluated how extraction methods impacted the phenolic profiles and antioxidant activities of Camellia oils. The contribution rates of phenolic profiles to antioxidant activities were investigated. HPLC and UPLC-Triple-TOF/MS demonstrated that phenolic content significantly varied with extraction methods while phenolic profiles were not. Herein, 15, 15, 17, 14, and 14 compounds were identified in mechanical pressing (MP), solvent extraction-hexane (SE-H), solvent extraction-ethanol (SE-E), aqueous enzymatic extraction (AEE), aqueous extraction (AE) oils, respectively. Results presented that SE-E oils were rich in phenolic acids (caffeic acid and p-hydroxybenzaldehyde), while AE oils were abundant in flavonoids (rutin and quercetin). Besides, the antioxidant activities of AEE and AE oils were significantly higher than other oils, originating from flavonoids (rutin and quercetin) and non-phenolics (rhododendron acid). Furthermore, Camellia oils from five extraction methods were remarkably classified by principal component analysis based on phenolic categories and antioxidant activities. It was assumed that low-polar and high-polar phenolic profiles were simultaneously transferred into SE-E oils, while only low-polar phenolic profiles (mainly flavonoids) were extracted into AE or AEE oils. The antioxidative origin of Camellia oils was ascribed to rutin, quercetin, and rhododendron acid. Our results provided an in-depth understanding of Camellia oils on phenolic profiles and the relationship between extraction methods and antioxidant activities.

This study aimed to assess the effects of industrial-scale oil-pressing techniques (hydraulic pressing (HP), twin-screw pressing (TP)), and organic solvent extraction (OE) on the physicochemical properties and antioxidant activities of protein isolates derived from Camellia ( Camellia oleifera Abel.) seed cakes. The predominant protein fractions in camellia seed cake were albumin (32–37 mg/g) and glutelin (19–27 mg/g), and the twin-screw pressed cake had a lower total protein content than the others. Spectral analysis showed that the α-helix content increased at the expense of random coil content in proteins from HP and TP cakes, indicating that extrusion altered the structure of the protein. The camellia seed protein particle size from HP and TP cakes was significantly reduced compared to that from the OE cake. The proteins obtained by HP and TP had stronger antioxidant activities than those obtained by OE. This research presents the potential for further applications of camellia seed cake protein to be utilized as antioxidants and emulsifiers in the food industries.

We isolated and purified polysaccharide from the Camellia oleifera Abel. seed cake (CCP) and studied its hypoglycemic activity in vitro . The molecular weight of CCP was 4736 Da and the monosaccharide molar composition ratio was: Xylose :Glucuronic acid:Galactosamine:Mannose = 10.9:4.4:2.6:1.8. The CCP contained conjugated proteins and belonged to acidic polysaccharide, which can promote the consumption of the glucose in the medium by HepG2 cells within the range of 0.125–0.500 mg/mL. Relative consumption rate (RCR) of the glucose was the highest at 0.5 mg/mL, exceeded that of the metformin hydrochloride, as well as that of Tea Polysaccharide under the same concentration. A meaningful structure activity relationship (SAR) of hypoglycemia polysaccharides had established. This result suggests that CCP can be used as hypoglycemic medicine or health food.

The attenuation of atopic dermatitis (AD) by Camellia oil was investigated in Raw 264.7 macrophages and HaCaT keratinocytes cell lines in this paper. Results showed that phenolic-abundant Camellia oil (PACO) by the AEE method showed the lowest expression levels of pro-inflammatory cytokines, including TNF-α, IL-6 and IL-1β in LPS-induced Raw 264.7 cells, followed by the cold-pressed Camellia oil (CPCO), then the commercial refined Camellia oil (CRCO). This may be attributed to a higher content of polyphenols in PACO, particularly for typical phenolics such as caffeic acid, quercetin, kaempferol, kaempferol-3-O-[2-O-β-D-galactopyranosyl-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside (KGRG) and kaempferol-3-O-[2-O-β-D-xylopyranosyl-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside (KXR), which are associated with lower NO levels in model cells and higher antioxidant capacities in DPPH, ABTS, and FRAP experiments. In HaCaT keratinocytes, exposure to TNF-α/IFN-γ resulted in an 8-fold and 10-fold increase in CCL-17 and IL-33, compared with control , indicating skin inflammation occurred. Camellia oil treatment improved this inflammation, embodied by a significant restoration of skin inflammatory mediators, and this improvement is dose-dependent. High-dose PACO exhibited the best anti-skin inflammatory effects. The mRNA expression levels of MMP1 , MMP9 , MMP25 , VEGFC and TNFRSF6B were also significantly down-regulated after PACO treatment, indicating its potential for skin barrier protection. Moreover, 800 μg/mL PACO significantly alleviated the cell apoptosis rate and regulated the apoptosis-related biomarkers at gene expression and protein expression levels ( Bax , Bcl-2 and Caspase-3 ). Analysis results on the JC-1 ΔΨm and mitochondrial ROS indicated that mitochondrial damage was reversed after the Camellia oil treatment, which accounted for its alleviation effects on apoptosis related to inflammation. Overall, Camellia oil, rich in phenolics, exhibited great therapeutic potential for AD treatment.

One new and three known triterpenoid saponins were isolated and identified fromCamellia oleiferaseeds through IR, NMR, HR-ESI-MS and GC-MS spectroscopic methods, namely oleiferasaponin A3, oleiferasaponin A1, camelliasaponin B1, and camelliasaponin B2. The structure of oleiferasaponin A3was elucidated as 16α-hydroxy-21β-O-angeloyl-22α-O-cinnamoyl-23α-aldehyde-28-dihydroxymethylene-olean-12-ene-3β-O-[β-d-galactopyranosyl-(1→2)]-[β-d-xylopyranosyl-(1→2)-β-d-galactopyranosyl-(1→3)]-β-d-gluco-pyranosiduronic acid. Camelliasaponin B1and camelliasaponin B2exhibited potent cytotoxic activity on three human tumour cell lines (human lung tumour cells (A549), human liver tumour cells (HepG2), cervical tumour cells (Hela)). The hypoglycemic activity of oleiferasaponin A1was testified by protecting pancreatic β-cell lines from high-glucose damage. Keywords:Camellia oleifera;triterpenoid saponin;oleiferasaponin A3;cytotoxic activity;hypoglycemic activity

Liver fibrosis is a common part of the pathological development of many chronic liver diseases. As liver fibrosis progresses, it may lead to cirrhosis, portal hypertension, liver decompensation, liver tumours, and death. Camellia oil (Camellia oleifera Abel.) is widely used as an edible oil in China. It has a wide range of biological activities and is used as a traditional medicine to treat conditions such as burns and stomach pains. However, whether camellia oil can ameliorate liver fibrosis remains unclear. We constructed a liver fibrosis model induced by carbon tetrachloride (CCl4) and then confirmed the role of camellia oil in liver fibrosis by biochemical examination, histopathological morphology, immunohistochemistry, and western blot analysis. We found that camellia oil ameliorated histopathological lesions, improved liver function and antioxidant capacity, decreased the expression of TGF-β1 and α-SMA proteins, and downregulated the expression of the pro-apoptotic proteins in CCl4-induced liver fibrosis. Therefore, these results suggest that camellia oil attenuates CCl4-induced liver fibrosis in mice, and its mechanism may function via reduction of hepatocyte apoptosis to inhibit hepatic stellate cell (HSC) activation. Camellia oil may provide a potential new treatment for liver fibrosis as an auxiliary treatment by addition of the edible oil to the daily diet.

Summary We aimed here to investigate the ability of steam-exploded camellia seed protein (SECSP) in stabilising steam-exploded camellia seed oil-in-water emulsion in comparison with soy protein isolate (SPI). Emulsions with different SECSP and SPI contents (0.5, 1, 1.5 and 2 g 100 mL −1 ) were formulated and the effect of protein concentration on emulsion droplet size, zeta (ζ)-potential, physical stability and bioaccessibility was examined. The stability of emulsion increased with increasing protein concentration. SECSP emulsion was characterised by smaller droplet sizes, higher surface charge and better stability than SPI emulsion. Furthermore, SECSP emulsion exhibited good digestive stability. SECSP emulsion showed creaming only at protein concentrations above 0.5 g 100 mL −1 after 28 days of storage, while SPI emulsion showed stratification and flocculation during the same storage period. Hence, steam explosion pretreatment physical modification of camellia seed protein (CSP) provides a novel approach of utilising proteins for emulsions stabilisation, and digestive stability improvement under gastrointestinal conditions.

Tea ( Camellia oleifera Abel) seed oil (TSO) has antioxidant and pharmacological properties. In this study, TSO was obtained from tea seeds by subcritical n- butane extraction (SBE), which is an environmentally friendly method. The oil yield, quality characteristics, and chemical composition of the extracted TSO were compared with those of oils obtained by supercritical carbon dioxide extraction (SCDE) and conventional cold pressing (CP). The TSO yield from SBE (51.35 %) was similar to that from SCDE (50.30 %), but higher than that from CP (31.82 %). TSO from SBE had better oxidative stability than that from SCDE and a lower toxic aldehyde content (0.41 mg/L) than that from SCDE (0.71 mg/L) and CP (2.67 mg/L). Moreover, TSO from SBE had relatively high tocopherols (26.20 mg/100 g) and phytosterols (842.25 mg/100 g) contents. Based on these findings, SBE is a promising method for obtaining high-quality TSO enriched with tocopherols and phytosterols.

In this study, a novel aqueous enzymatic extraction (AEE) was developed to prepare camellia oil enriched with polyphenols, and the potential mechanism of in situ phenolics enrichment was explored. Camellia seeds were first subjected to AEE in a 25 % isopropanol solution containing 0.15 M glucose for 4 h, followed by the introduction of α -glycosidase β -glycosidase, or pectinase, and further hydrolysis was performed for 1 to 4 h. A higher free oil yield (FOR, 86.98 %) was obtained. Meanwhile, the highest total phenolic content (TPC) in oil (95.72 ± 1.53 mg GAE/Kg oil) was detected in the 4-h β -glycosidase-treated group, approximately 4-fold that of the control group (20.00 ± 1.02 mg GAE/Kg oil). HPLC analysis of the hydrolysate revealed that typical glycosylated polyphenols declined in the aqueous phase, while the content of less polar polyphenols, such as kaempferol-3-O-sambubioside, rutin, and quercetin, in the oil phase increased. These findings suggest that hydrophilic KGRG and KXR were hydrolyzed into relatively hydrophobic deglycosylated phenolics, which migrated into the oil phase, accounting for the enrichment of phenolics in camellia oil by the novel AEE. Furthermore, the stability of phenolics-abundant camellia oil was evaluated with respect to physicochemical properties, phenolics content, and lipid accompaniments content under various conditions. Results showed that the phenolics and lipid accompaniments remained relatively stable under a nitrogen atmosphere and at 4 °C refrigeration. Hence, the in-situ phenolics enrichment technique developed has the potential to prepare functional camellia oil with high biological activity and stability on a large scale.

Cellulose nanocrystals (CNCs) extracted from the waste shell of Camellia oleifera Abel ( C. oleifera ) are gaining attention as valuable materials. In this study, CNCs were extracted from the agricultural waste shell of C. oleifera through phosphoric acid and sulfuric acid hydrolysis , respectively. Firstly, we optimized the alkaline treatment process for cellulose isolation by using response surface methodology. Furthermore, the properties of CNCs were investigated by neutralizing them with NaOH and NH 3 ·H 2 O, and by dialysis in water. In addition, the characterization methods including FT-IR, TGA , AFM and TEM were used to analysis the properties of the synthesized CNCs. Finally, CNCs were studied for their application in essential oil-based Pickering emulsions. CNCs obtained from sulfuric acid showed the smallest particle size and good dispersibility. Moreover, the release profiles of essential oils in the emulsions were followed by Peppa's kinetic release model. The antibacterial activity of the emulsions against E. coli and S. aureus showed that CNCs-stabilized emulsions enhanced the antibacterial activity of essential oils. Therefore, neutralization treatments may enhance the properties of CNCs, and CNCs stabilized Pickering emulsions can enhance antibacterial activity of essential oil. This study provides insight into the potential application of CNCs derived from C. oleifera waste shells.

Camellia saponins are important by-products of Camellia Oleifer Abel. processing. In this study, an eco-friendly method based on natural deep eutectic solvents (NaDESs, proline and glycerol at a molar ratio of 2:5) was established to extract saponin s from C.oleifera cakes. The content of saponin (702.22 ± 1.28 mg/g) obtained using NaDES was higher than those extracted using water or methanol. UPLC-Q-TOF MS analysis of chemical structure showed that the difference in the extraction technique alter individual saponins. A widely targeted metabolomic approach and KEGG metabolic pathway analysis showed that the upregulated metabolites in the NaDES-based extract mainly included flavonoids, alkaloids, and phenolic acids; and they were involved in arginine and proline metabolism, metabolic pathways, phenylpropanoid biosynthesis, biosynthesis of secondary metabolites, and flavonoid biosynthesis. The present study proposes a selective substitute for use in the extraction of camellia saponins with composition analysis.
