Formation of 8-hydroxylinalool in tea plant Camellia sinensis var. Assamica ‘Hainan dayezhong’

Ying Zhou,Wei He,Yunchuan He,Qiulin Chen,Yang Gao,Jiamei Geng,Zeng-Rong Zhu

Linalool and its derivatives contribute greatly to tea aroma. Here, 8-hydroxylinalool was found to be one of the major linalool-derived aroma compounds in Camellia sinensis var. assamica ‘Hainan dayezhong’, a tea plant grown in Hainan Province, China. Both ( Z )-8-hydroxylinalool and ( E )-8-hydroxylinalool were detected, and the E type was the main compound. Its content fluctuated in different months and was the highest in the buds compared with other tissues. CsCYP76B1 and CsCYP76T1, located in the endoplasmic reticulum, were identified to catalyze the formation of 8-hydroxylinalool from linalool in the tea plant. During withering of black tea manufacturing, the content of both ( Z )-8-hydroxylinalool and ( E )-8-hydroxylinalool significantly increased. Further study suggested that jasmonate induced gene expression of CsCYP76B1 and CsCYP76T1 , and the accumulated precursor linalool may also contribute to 8-hydroxylinalool accumulation. Thus, this study not only reveals 8-hydroxylinalool biosynthesis in tea plants but also sheds light on aroma formation in black tea.

A Novel Strain Burkholderia theae GS2Y Exhibits Strong Biocontrol Potential Against Fungal Diseases in Tea Plants (Camellia sinensis)

Yijie Dong,Xing Wang,Guang-Da Feng,Qing Yao,Honghui Zhu

Background: Tea plants (Camellia sinensis) are widely cultivated cash crops. However, fungal diseases lead to significant reductions in both the yield and quality of tea. Therefore, searching for economical, eco-friendly, and efficient biological control measures is crucial for protecting tea plants from pathogenic fungi. Methods: The confrontation assays were performed to identify the antagonistic bacteria against tea pathogenic fungi and evaluate the antifungal activity of these bacteria. Results: Here, three tea pathogenic fungi were identified:Colletotrichum siamenseHT-1,Diaporthe phaseolorumHT-3, andFusarium fujikuroiHT-4. Notably,D. phaseolorumwas the first to be reported in tea plants in China. Some tea pathogenic fungi showed a high relative abundance, suggesting a potential disease risk in tea plantations. Strain GS2Y, isolated from tea rhizosphere soil, exhibited strong antifungal activity against tea pathogenic fungi and represented a novel species within the genusBurkholderia, designated asBurkholderia theae. GS2Y could directly inhibit tea pathogenic fungi by disrupting the cellular structures and protect tea plants from fungal diseases caused byC. siamenseHT-1 andD. phaseolorumHT-3. Conclusions:B. theaeGS2Y might function as a potentially valuable resource for biocontrol agents, laying the foundation for the development of strategies to manage fungal diseases in tea plants.

Uptake, Translocation, Metabolism, and Distribution of Glyphosate in Nontarget Tea Plant (Camellia sinensis L.)

Mengmeng Tong,Wanjun Gao,Weiting Jiao,Jie Zhou,Yeyun Li,Lili He,Ruyan Hou

The uptake, translocation, metabolism, and distribution behavior of glyphosate in nontarget tea plant were investigated. The negative effects appeared to grown tea saplings when the nutrient solution contained glyphosate above 200 mg L–1. Glyphosate was highest in the roots of the tea plant, where it was also metabolized to aminomethyl phosphonic acid (AMPA). The glyphosate and AMPA in the roots were transported through the xylem or phloem to the stems and leaves. The amount of AMPA in the entire tea plant was less than 6.0% of the amount of glyphosate. The glyphosate level in fresh tea shoots was less than that in mature leaves at each day. These results indicated that free glyphosate in the soil can be continuously absorbed by, metabolized in, and transported from the roots of the tea tree into edible leaves, and therefore, free glyphosate residues in the soil should be controlled to produce teas free of glyphosate.

Protocol: high-efficiency in-planta Agrobacterium-mediated transgenic hairy root induction of Camellia sinensis var. sinensis

Alagarsamy Karthikeyan,Shamala Lubobi Ferdinand,Wei Shu

Background Camellia sinensis var. sinensis is widely grown for tea beverages that possess significant health promoting effects. Studies on tea plant genetics and breeding are hindered due to its recalcitrance to Agrobacterium -mediated genetic transformation. Among the possible reasons, oxidation of phenolics released from explant tissues and bactericidal effects of tea polyphenols during the process of transformation play a role in the plant recalcitrance. The aim of the present study was to alleviate the harmful effects of phenolic compounds using in - planta transformation. Results Two-month old seedlings of tea cultivar “Nong Kangzao” were infected at the hypocotyl with wild type Agrobacterium rhizogenes and maintained in an environment of high humidity. 88.3% of infected plants developed hairy roots at the wounded site after 2 months of infection. Our data indicated that transgenic hairy root induction of tea can be achieved using A. rhizogenes following the optimized protocol. Conclusion With this method, composite tea plants containing wild-type shoots with transgenic roots can be generated for “in root” gene functional characterization and root-shoot interaction studies. Moreover, this method can be applied to improve the root system of composite tea plants for a better resistance to abiotic and biotic stresses.

Characterization of enzymes specifically producing chiral flavor compounds (R)- and (S)-1-phenylethanol from tea (Camellia sinensis) flowers

Ying Zhou,Qiyuan Peng,Ling Zhang,Sihua Cheng,Lanting Zeng,Fang Dong,Ziyin Yang

1-Phenylethanol is a chiral flavor compound that has enantiomers, ( R )- and ( S )-1-phenylethanol, with different flavor properties. Given that isolating these enantiomers from plants is low yielding and costly, enzymatic synthesis presents an alternative approach. However, the genes/enzymes that specifically produce ( R )- and ( S )-1-phenylethanol in plants are unknown. To identify these enzymes in tea ( Camellia sinensis ) flowers, 21 short chain dehydrogenase (SDR) genes were isolated from tea flowers, cloned, and functionally characterized. Several recombinant SDRs in Escherichia coli exhibited activity for converting acetophenone to ( S )-1-phenylethanol (CsSPESs, >99.0%), while only one SDR produced ( R )-1-phenylethanol (CsRPES, 98.6%). A pair of homologue enzymes (CsSPES and CsRPES) showed a strong preference for NADPH cofactor, with optimal enzymatic reaction conditions of 45–55 °C and pH 8.0. Identification of the tea flower-derived gene responsible for specific synthesis of ( R )- and ( S )-1-phenylethanolsuggests enzymatic synthesis of enantiopure 1-phenylethanol is possible using a plant-derived gene.

Genome-Wide Identification and Expression Analysis of the RADIALIS-like Gene Family in Camellia sinensis

Shaoying Wang,Beibei Wen,Yun Yang,Shanshan Long,Jianjun Liu,Meifeng Li

The RADIALIS-like (RL) proteins are v-myb avian myeloblastosis viral oncogene homolog (MYB)-related transcription factors (TFs), and are involved in many biological processes, including metabolism, development, and response to biotic and abiotic stresses. However, the studies on theRLgenes ofCamellia sinensisare not comprehensive enough. Therefore, we undertook this study and identified eightCsaRLsbased on the typical conserved domain SANT Associated domain (SANT) of RL. These genes have low molecular weights and theoretical pIvalues ranging from 5.67 to 9.76. Gene structure analysis revealed that sixCsaRLgenes comprise two exons and one intron, while the other two contain a single exon encompassing motifs 1 and 2, and part of motif 3. The phylogenetic analysis divided one hundred and fifty-eight RL proteins into five primary classes, in which CsaRLs clustered in Group V and were homologous with CssRLs of theShuchazaovariety. In addition, we selected different tissue parts to analyze the expression profile ofCsaRLs, and the results show that almost all genes displayed variable expression levels across tissues, withCsaRL1arelatively abundant in all tissues. qRT-PCR (real-time fluorescence quantitative PCR) was used to detect the relative expression levels of theCsaRLgenes under various abiotic stimuli, and it was found thatCsaRL1aexpression levels were substantially higher than other genes, with abscisic acid (ABA) causing the highest expression. The self-activation assay with yeast two-hybrid system showed that CsaRL1a has no transcriptional activity. According to protein functional interaction networks, CsaRL1a was well connected with WIN1-like, lysine histidine transporter-1-like, β-amylase 3 chloroplastic-like, carbonic anhydrase-2-like (CA2), and carbonic anhydrase dnaJC76 (DJC76). This study adds to our understanding of the RL family and lays the groundwork for further research into the function and regulatory mechanisms of theCsaRLsgene family inCamellia sinensis.Keywords:RADIALIS-like (RL);bioinformatics analysis;abiotic stress;self-activation;yeast two-hybrid system

Structural and digestive characters of a heteropolysaccharide fraction from tea (Camellia sinensis L.) flower

Dan Chen,Ao Wang,Jialiang Lv,Chao Tang,Chang-hai Jin,Jun Liu,Xiaoxiong Zeng,Li Wang

Tea ( Camellia sinensis L.) flower polysaccharides (TFPS) have various health-promoting functions. In the present work, the structure of a purified TFPS fraction, namely TFPS-1-3p, and its in vitro digestive properties were investigated. The results demonstrated that TFPS-1-3p was a typical heteropolysaccharide consisting of rhamnose (Rha), arabinose (Ara), galactose (Gal) and galacturonic acid (GalA) with a molecular weight of 47.77 kDa. The backbone of TFPS-1-3p contained → 4)-α- d -Gal p A(-6-OMe)-(1 → 4)-α-Gal p A-(1 → and → 4)-α- d -Gal p A(-6-OMe)-(1 → 2,4)-α- l -Rha p -(1 → linkages. The branch linkages in TFPS-1-3p contained → 6)-β- d -Gal p -(1→, →3,6)-β- d -Gal p -(1→, →5)-α- l -Ara f -(1 → and → 3,5)-α- l -Ara f -(1 →. Subsequently, TFPS-1-3p could not be degraded under simulated human gastrointestinal conditions but could be of use to human fecal microbes, thereby lowering the pH and increasing the production of short-chain fatty acids (SCFAs) of the gut microenvironment and altering the composition of the gut microbiota. The relative abundance of Fusobacterium_mortiferum Megasphaera_elsdenii_DSM_20460, Bacteroides thetaiotaomicron , Bacteroides plebeius and Collinsella aerofaciens increased significantly, potentially contributing to the degradation of TFPS-1-3p.

Studies on the Biochemical Formation Pathway of the Amino Acid l-Theanine in Tea (Camellia sinensis) and Other Plants

Sihua Cheng,Xiumin Fu,Xiaoqin Wang,Yinyin Liao,Lanting Zeng,Fang Dong,Ziyin Yang

Tea (Camellia sinensis) is the most widely consumed beverage aside from water. The flavor of tea is conferred by certain metabolites, especially l-theanine, in C. sinensis. To determine why more l-theanine accumulates in C. sinensis than in other plants, we compare l-theanine contents between C. sinensis and other plant species (Camellia nitidissima, Camellia japonica, Zea mays, Arabidopsis thaliana, and Solanum lycopersicum) and use a stable isotope labeling approach to elucidate its biosynthetic route. We quantify relevant intermediates and metabolites by mass spectrometry. l-Glutamic acid, a precursor of l-theanine, is present in most plants, while ethylamine, another precursor of l-theanine, specifically accumulates in Camellia species, especially C. sinensis. Most plants contain the enzyme/gene catalyzing the conversion of ethylamine and l-glutamic acid to l-theanine. After supplementation with [2H5]ethylamine, all the plants produce [2H5]l-theanine, which suggests that ethylamine availability is the reason for the difference in l-theanine accumulation between C. sinensis and other plants.

Metabolic Changes of Caffeine in Tea Plant (Camellia sinensis (L.) O. Kuntze) as Defense Response to Colletotrichum fructicola

Yu-Chun Wang,Wen-Jun Qian,Na-Na Li,Xin-Yuan Hao,Lu Wang,Bin Xiao,Xin-Chao Wang,Ya-Jun Yang

Tea plant (Camellia sinensis) is one of the most economically valuable crops in the world. Anthracnose can affect the growth of leaves and cause serious yield losses of tea. Tea plants are rich in secondary metabolites; however, their roles in resistance to anthracnose are unclear. Herein we compared the contents of total phenolics, catechins, and caffeine in two cultivars with different resistances to anthracnose during Colletotrichum fructicola infection. (−)-Epigallocatechin-3-gallate (EGCG), (+)-catechin (C), caffeine, and critical regulatory genes were induced in C. fructicola-resistant tissues. In vitro antifungal tests showed that caffeine more strongly inhibited mycelial growth than tea polyphenols and catechins. Both electron microscopy and bioactivity analysis results showed that caffeine can affect mycelial cell walls and plasma membranes. Through promoter sequences analysis, a number of stress response-related cis-acting elements were identified in S-adenosylmethionine synthetase and tea caffeine synthase. These results demonstrated that (−)-EGCG, (+)-C, and caffeine may be involved in the resistance of tea plants to anthracnose.

Comprehensive analysis of environmental factors on the quality of tea (Camellia sinensis var. sinensis) fresh leaves

Wei Ran,Qinghui Li,Xinlong Hu,De Zhang,Zhi Yu,Yuqiong Chen,Mingle Wang,Dejiang Ni

“High mountain, cloud and mist produce high-quality tea”; however, the specific mechanism has not been fully understood. To explore the influence of environmental factors on the quality of tea fresh leaves, we firstly selected the same variety of tea plant growing at different altitudes in Xuan'en County, China. And then, based on the analyses of the physical and chemical properties of the soil where they grew, we comprehensively analyzed the composition and content of volatile and non-volatile substances in these tea fresh leaves harvested from 27 sampling sites. The relative contents of linalool and its oxides I and II, phenylethanol and nerol in paddy soil were higher than those in red soil and yellow soil, but the contents of acids and esters were significantly lower than those in other soils ( P < 0.05). Benzoic acid content and palmitic acid content are relatively high in red soil and yellow soil, respectively. The amino acid content of tea fresh leaves produced in paddy soil is significantly lower than that in red soil and yellow soil, while the contents of catechins and flavonol glycosides were significantly higher than that in other soils ( P < 0.05). The relative contents of EGCG, ECG and C in paddy soil were significantly higher than those in yellow soil and red soil. The content of alkaloids in red soil is high. With the increase of altitude, the content of amino acids gradually increased, and the catechins content showed a trend of first decreasing and then increasing; the phenolic acid content changes in accordance with catechins with the elevation; the flavonoids content (alcohols) glycosides did not change obviously in each altitude range. These results are helpful for us to fully understand the influence of environmental factors on the quality of fresh tea leaves.

Elucidation of the Regular Emission Mechanism of Volatile β-Ocimene with Anti-insect Function from Tea Plants (Camellia sinensis) Exposed to Herbivore Attack

Guotai Jian,Yongxia Jia,Jianlong Li,Xiaochen Zhou,Yinyin Liao,Guangyi Dai,Ying Zhou,Jinchi Tang,Lanting Zeng

Herbivore-induced plant volatiles (HIPVs) play an important role in insect resistance. As a common HIPV in tea plants (Camellia sinensis), β-ocimene has shown anti-insect function in other plants. However, whether β-ocimene in tea plants also provides insect resistance, and its mechanism of synthesis and emission are unknown. In this study, β-ocimene was confirmed to interfere with tea geometrid growth via signaling. Light was identified as the key factor controlling regular emission of β-ocimene induced by the wounding from tea geometrids. β-Ocimene synthase (CsBOS1) was located in plastids and catalyzed β-ocimene formation in overexpressed tobacco. CsBOS1 expression in tea leaves attacked by tea geometrids showed a day-low and night-high variation pattern, while CsABCG expression involved in volatile emission showed the opposite pattern. These two genes might regulate the regular β-ocimene emission from tea plants induced by tea geometrid attack. This study advances the understanding on HIPV emission and signaling in tea plants.

Characterizing relationship between chemicals and in vitro bioactivities of teas made by six typical processing methods using a single Camellia sinensis cultivar, Meizhan

Guanhua Xie,Jingna Yan,Anxia Lu,Jirui Kun,Bei Wang,Chengda Song,Huarong Tong,Qing Meng

Processing method is considered as a major factor that affects biotransformation of phytochemicals in tea and leads to diverse flavor and bioactivity of tea. In the present work, six typical tea manufacturing processings were employed to compare the effect on chemical composition of teas through using leaves of the single tea cultivar – – Camellia sinensis var. Meizhan. And in vitro antioxidant activity, inhibition against α-glucosidase and three lipid metabolism enzymes of these teas were also investigated, the relationships among them were analyzed further. As fresh leaves were processed into six categories of teas, the content of total catechins (TCs) has decreased in varying degrees while theaflavins (TFs) has increased. The antioxidant capacity composite index (ACCI) from high to low were green tea, yellow tea, oolong tea, white tea, dark tea, and black tea with the range from 98.44 to 58.38, which dominated by the content of TCs. Furthermore, all categories of teas possessed an inhibition effect on the pancreatic lipase (PL), 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-COA reductase), lecithin cholesterol acyltransferase (LCAT), and α-glucosidase. The inhibition rate of PL and α-glucosidase appears to be positively influenced by TFs content (r =0.863, r =0.857, p < 0.05) while that of LCAT showed significant positive correlations with the content of tea polyphonels (TPs) (r = 0.902, p < 0.01). These results provide a better understanding of the relationships between processing method and chemical components of tea. It is suggested that various tea categories possess potential healthy effects which could serve as promising nutritional supplements.
