The green extraction and separation of natural produce is a significant part in pharmaceutical industry which has received extensive attention. Comparing with the extraction of hydrophilic products, hydrophobic natural products are further challenging. An extraction means depended in high efficiency hydrophobic deep eutectic solvent (HDES, TCP) was screened by designing ternary HDES. Utilizing the established method, the hydrophobic compound betulin was successfully extracted and recovered from Celtis sinensis leaves. The results showed that HDESs were further productive in extracting hydrophobic compounds than hydrophilic DES. The extractions of TPC were statistically optimized by response surface methodology and central composite design. Under the optimum conditions, the extraction rate of betulin was increased by 125.00% compared with the initial yield. At the same time, it was proved that among the six resins, D-101 macroporous resin had the highest recovery rate of 71.12% for betulin, which was the most effective enrichment and purification method. To sum up, HDES has obvious advantages in the extraction of hydrophobic natural products due to its mild extraction conditions, high yield, and environmental friendliness. At the same time, limitless feasibilities of HDES compositions and their associations furnish a plentiful selection space for the extraction about hydrophobic compounds.

In recent years, deep eutectic solvent (DES) has attracted comprehensive attention on the extraction of natural products, and is regarded as an alternative to traditional organic solvents for the environmental advantages. Twenty-six DESs were compared for their extraction yield of total flavonoids and isovitexin (the main flavonoid in Celtis sinensis) from Celtis sinensis. The results show that the extraction yields of total flavonoids by betaine/glycolic acid (DES8), ethylamine hydrochloride/1,2-propanediol (DES12) and tetrapropylammonium bromide/lactic acid (DES17) are the highest, while the extraction yields of isovitexin by ethylene glycol/malonic acid (DES23), ethylene glycol/glycolic acid (DES24) and 1,2-propanediol/glycolic acid (DES26) are the highest. The extraction conditions using the above six DESs were further optimized systematically. Under optimum conditions, the extraction rates of total flavonoids and isovitexin can be increased up to 95.39 and 10.58 mg g−1, respectively, which were significantly higher than that of methanol extraction. In order to exclude the effect of DESs on the bioactivity of Celtis sinensis extract, the macroporous resin D-101 was used to purify the total flavonoids from DESs extract, and the recovery rates of flavonoids from the above six kinds of DESs were all over 80%. Next, the anti-inflammatory activity of DES extracts was compared using a lymphocyte transformation experiment. The result showed that the inhibition rate of the DES24 extract on the proliferation of Con A-activated T cells was up to 72% with an IC50 value of 124.8 μg mL−1. None of the DESs extracted exhibited cytotoxicity on normal T cells. The mechanism of the anti-inflammatory activity against Con A-activated T cells may be that DES24 flavonoids extract induced the apoptosis of inflammatory T cells, and activated the expression of pro-apoptotic protein. Taken together, DES has showed significant advantages on the extraction of natural products for the relatively mild extraction condition, high yield and environmental-friendliness.

This study used deep eutectic solvents (DES) to extract the active components from Celtis sinensis leaves, with the residue comprising at least 90% of the total biomass. The main components included cellulose, hemicellulose, and lignin, representing a renewable lignocellulose resource. In recent years, DES has attracted considerable attention in the field of biomass pretreatment due to its straightforward preparation methods, cost-efficiency, and ease of recovery. Therefore, DES may be an effective method for lignin separation. This study analyzed the compositional changes in different types of DES-extracted residues and compared the hydrophilic and hydrophobic DES extraction methods for the enzymatic hydrolysis of cellulose in Celtis leaves. The results showed that hydrophilic DES was more competitive during decomposition. Pretreatment reduced the lignin from 37.55 to 22.18%, while the cellulose levels displayed a minimal decline from 19.94 to 16.45%. Therefore, DES effectively removed lignin and hemicellulose from the leaves, while retaining most of the cellulose in the specified conditions. Furthermore, scanning electron microscopy (SEM) was used to characterize the surface morphology changes in the original leaves and residues pre-treated with different solvents. The influence of chemical structural changes on the subsequent enzymatic hydrolysis was investigated further via thermogravimetric (TG) analysis, Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD). Compared to hydrophobic DES, hydrophilic DES effectively removed amorphous components such as hemicellulose and lignin from the extracted residue, which improved cellulase hydrolytic efficiency. The results confirmed that the crystal structure of cellulose in the extraction residue displayed no changes, while the relative crystallinity increased. The rate and degree of the subsequent enzymatic glycation were enhanced, while the hydrolytic efficiency of hydrophilic DES increased 2.13-fold (40.19 ~ 85.86%).

Deep eutectic solvents (DES) have received extensive attention in the extraction of natural products in the last few years. They are considered a substitute for conventional organic solvents because of their environmental advantages. To efficiently extract bioactive polyphenols and triterpenoids from the medicinal plant Celtis sinensis, fifteen DESs were prepared. The resulting DES1 (choline chloride-levulinic acid) showed the maximum extraction yield of total polyphenols, while DES9 (choline chloride-1,2-propylene glycol) had the maximum yield of total triterpenes. Further, the extraction conditions of DES1 and DES9 were methodically optimized. After optimizing the extraction conditions of DES1 and DES9, the extraction yield of total polyphenols was enhanced from 82.05 mg g−1 to 101.38 mg g−1 and that of total triterpenes from 57.87 mg g−1 to 72.72 mg g−1, both significantly superior to those of traditional water- and methanol-based extraction approaches. We designed a new DES by mixing DES1 and DES9 in the ratio of 7.25 : 2.75 to perform extraction of both polyphenols and triterpenes with high efficiency. The new DES solvent simultaneously increased the extraction yields of polyphenols and triterpenes to 118.46 mg g−1 and 88.51 mg g−1, respectively, after response surface methodology. More importantly, the extract obtained by the mixed solvent of DES1 and DES9 displayed significant antioxidant activity superior to those of extracts individually produced by DES1 and DES9. Our results indicate that a DES-based approach has promising potential in extracting bioactive substances from medicinal arbor species with high efficiency and eco-friendliness.
