Study on the extraction and stability of total flavonoids from Millettia speciosa Champ

Junhai Liu,Xiaosha Guo,Tengteng Yang,Xiaoli Wang,Yinku Liang

Millettia speciosa Champ, a member of the Leguminosae family and the genus Millettia , is a rich source of total flavonoids. Investigating the extraction methods and stability of total flavonoids from Millettia speciosa Champ using ultrasound-assisted extraction is crucial for enhancing its medicinal value and advancing the modernization of traditional Chinese medicine. In this study, the optimal extraction conditions were determined as follows, ethanol volume fraction of 64%, ultrasonic temperature of 60°C, solid-liquid ratio of 1:20 g/mL, ultrasonic time of 29 min, and ultrasonic power of 400 W. Under these conditions, the extract of total flavonoids from Millettia speciosa Champ was 6.485%. The stability of the total flavonoids was evaluated under various conditions. The results indicated that the total flavonoids should be stored in the dark. Metal ions such as K + , Na + , Ca 2+ , Mg 2+ , and Al 3+ had no significant impact on stability, whereas Cu 2+ , Zn 2+ , and Fe 3+ caused a notable loss of stability. The total flavonoids remained stable under neutral and weakly acidic conditions but were unstable under strongly acidic and alkaline conditions. Additives such as sodium benzoate, sodium chloride, glucose, and sucrose did not significantly affect stability. In contrast, ascorbic acid and citric acid had minimal effects on stability at low concentrations but significantly impacted stability at higher concentrations.

In Vitro Digestibility, Structural and Functional Properties of Millettia speciosa Champ. Seed Protein

Qing Yang,Shuxian Ding,Qinglong Wang,Li Xu,Xiaoxia Yan,Huan Tang,Langxing Yuan,Xiaoyan Chen,Zhunian Wang,Maoyuan Wang

As an underutilized industrial byproduct generated during bioactive compound extraction fromMillettia speciosaChamp. seeds, the residual protein fraction represents a promising sustainable resource for valorization.Millettia speciosaChamp. seed protein (MP) was extracted, and its fundamental physicochemical and functional properties were evaluated for potential applications in the food industry. Structural characterization revealed that MP had a molecular weight distribution with major components at 14.0 kDa and 116.0 kDa, with respective denaturation temperatures of 79.75 °C and 91.77 °C. The main structure of MP included different proportions of intramolecular α-helices and random coils in different pH microenvironments, based on circular dichroism spectroscopy. The MP displayed similar solubility profiles to the soy protein isolate (SP), but with lower solubility at slightly acidic pH, low solubility at pH 5.0, and comparable solubility above pH 8.0. Functional assessments showed that MP possessed emulsifying, foaming, water-binding, and fat-absorption capacities comparable to those of SPI, although the in vitro digestibility was relatively lower. These findings indicate that MP may serve as a safe and nutritious functional ingredient for health-oriented food products.

Polysaccharides from Millettia speciosa Champ. Ameliorates Excessive Exercise-Induced Fatigue by Regulating Gut Microbiota

Jinfeng Zhang,Wenkai Zhang,Longhui Yu,Qinru Yang,Wenjing Wang,Xinru Hu,Jingen Li,Guodong Zheng

This study aimed to explore the ameliorative effect of Millettia speciosa Champ. polysaccharides (MSP) on excessive exercise-induced fatigue (EF) mice. The physicochemical characterization and in vitro antioxidant activities were analyzed. The fatigue-related biochemical indexes were determined, and gut microbiota was analyzed by 16S rDNA sequencing. The results showed that MSP primarily consists of total carbohydrates, uronic acids, and proteins. It was mainly composed of glucose, arabinose, and galactose. Additionally, MSP showed good scavenging effects on free radicals. In the experiment of EF mice, MSP intervention effectively extended the pole-climbing and swimming time, significantly increased the levels of muscle and liver glycogen, blood glucose, adenosine triphosphate, glutathione peroxidase, and superoxide dismutase, while significantly decreased the levels of fatigue-related lactate dehydrogenase, creatine kinase, and urea nitrogen. Furthermore, MSP intervention reduced harmful bacteria (Helicobacter, Anaerotruncus, Erysipelatoclostridium, and Lachnospiraceae_FCS020_group), and enriched the beneficial bacteria (Lactobacillus, Alistipes, Ruminococcaceae, and Roseburia). Therefore, MSP may utilize the gut microbiota as a target to alleviate fatigue caused by excessive exercise.

Effect of DNA methylation on gene transcription is associated with the distribution of methylation sites across the genome in osteoarthritis

Peng Yi,Xiongfeng Xu,Jiawei Yao,Bo Qiu

Genetics and epigenetics are important subjects in the field of osteoarthritis (OA) research. DNA methylation may affect gene transcription, but the specific mechanisms have remained to be fully elucidated. In the present study, the ChAMP methylation analysis package was used to identify differentially methylated genes (DMGs) from the dataset GSE63695 from the Gene Expression Omnibus (GEO) database. The distribution of differentially methylated sites (DMS) and the total array sites across the genome were analyzed by enrichment analysis. Subsequently, two mRNA expression profiling datasets, GSE114007 and GSE113825, were obtained from the GEO database and common differentially expressed genes (DEGs) were identified using the Limma package. Key genes were screened by analyzing the distribution of DMS across the genome consisting of DEGs and DMGs. A total of 1,662 and 1,986 DEGs were identified between OA and normal human cartilage from the GSE113825 and GSE114007 dataset, respectively. A further screening revealed 292 genes with common differences between the two datasets. A total of 574 DMS containing 394 DMGs were observed between OA and normal cartilage. Integrative analysis revealed a corresponding subset of 15 genes. Of these, 6 genes were verified by reverse transcription‑quantitative PCR, confirming that the mRNA expression of 5 genes (MAP1B, FNDC1, ANLN, SCNN1A and STC2) in OA cartilage was consistent with the mRNA expression from the analysis of the datasets. Upon treatment with the DNA methylation inhibitor 5‑aza‑2'‑deoxycytidine, the mRNA levels of FNDC1 and SCNN1A were decreased, and no significant alteration in the mRNA levels of MAP1B, ANLN, KCNN4 and STC2 was observed. The incidence of differential methylation varied in subregions of the genome and the effects on transcription were associated with the distribution of DEGs across the genome. The regulation of this appears more complex than initially postulated. Combining the data on epigenetic differences of OA with the genome or transcriptome data for analysis may improve the understanding of the pathophysiological processes of OA. FNDC1 and SCNN1A may potentially be valuable biomarkers for OA.

Optimization of ultrasound, microwave and Soxhlet extraction of flavonoids from Millettia speciosa Champ. and evaluation of antioxidant activities in vitro

Zhao Zhenyu,Liu Pinghuai,Wang Shenglin,Ma Shasha

Ultrasonic-assisted extraction (UAE) and microwave-assisted extraction (MAE) were developed to extract flavonoids antioxidants from the root of Millettia speciosa ( Millettia speciosa Champ.). The optimal experimental parameters for antioxidant extraction from M. speciosa Champ. were measured using single-factor experimentation combined with response surface methodology (RSM). Final extraction rate (FER) of flavonoids assay was used to quantify antioxidant compounds. Then, 2,2ˊ-diphenyl-1-picryhydrazyl (DPPH) scavenging capacity, 2,2ˊ-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) scavenging capacity and ferric reducing antioxidant power (FRAP) were used to determine the antioxidant capacities of extract. Besides, UAE and MAE were compared with conventional Soxhlet extraction (CSE) and M. speciosa Champ. powders were observed by scanning electronic microscopy to analyze the damage of cell structure caused by different extraction methods. The results showed that UAE was the optimal extraction method to extract flavonoids compounds from M. speciosa Champ. and the optimized UAE conditions of flavonoids was as follows: ethanol, 80%, temperature, 40 °C and ultrasonic power, 500 W. This paper provided useful information to utilize the resource of Chinese herbal medicine and manifest the great antioxidant activities of M. speciosa Champ., and the optimized UAE conditions of flavonoids from M. speciosa Champ. could be used in industrial production.

Therapeutic effects of Ainsliaea fragrans champ. and its active compounds on cervicitis: An integrated approach combining metabolomics, network pharmacology, and experimental validation

Zhengwen Zhang,Zhaojun Wang,Yushun Cui,Min Yao,Jiajian Lu,Mingzhen He,Yulin Feng,JunMao Li

Ethnopharmacological relevance Ainsliaea fragrans Champ. (AFC) , a traditional Chinese medicinal herb with a well-documented history in the management of gynecological disorders, serves as the principal component of several Chinese patent medicines that have demonstrated significant clinical efficacy against cervicitis, endometritis, and pelvic inflammatory disease. However, its specific mechanism of action against cervicitis has not been fully elucidated. Purpose This study aims to determine the therapeutic efficacy and underlying mechanisms of AFC in treating cervicitis, and identifying its key active components. Materials and methods First, an in vivo model of cervicitis was established using a phenolic mucilage solution, while an in vitro LPS induced RAW264.7 cell inflammation model was employed. The therapeutic effects of AFC were evaluated through HE staining, uterine index measurements, RT-qPCR and ELISA. The chemical constituents of AFC were identified using ultra-performance liquid chromatography coupled with UPLC-Q-TOF/MS. Subsequently, cellular metabolomics and network pharmacology were utilized to predict the potential targets, mechanisms, and active components of AFC in treating cervicitis. Finally, the underlying mechanisms and active ingredients were validated using methods such as RT-qPCR, Western blot, and molecular docking. Results Both in vivo and in vitro experiments demonstrated that AFC ameliorated phenol mucilage-induced cervicitis in rats and significantly suppressed the expression of inflammatory factors including IL-6, TNF-α, and IL-1β. UPLC-Q-TOF-MS/MS analysis identified 93 chemical constituents in AFC. Cellular metabolomics revealed 79 differential metabolites, including PGE2 and COX-2, among others. Integrated network pharmacology analysis identified potential targets including IDO1 and COX-2, and enriched signaling pathways such as TLR4 and PI3K/AKT-mTOR. Among the nine candidate compounds screened, luteolin, apigenin, and isorhamnetin ranked as the top three by degree value were hypothesized to be the primary active constituents of AFC against cervicitis. Subsequent experimental validation through RT-qPCR, Western blot, and molecular docking demonstrated that AFC treats cervicitis by modulating the PGE2/COX-2/IDO1-FLG in coordination with the TLR4 and PI3K/AKT-mTOR pathways, with luteolin, apigenin, and isorhamnetin confirmed as the key active compounds. Conclusion AFC exerts its therapeutic effects against cervicitis through coordinated regulation of the PGE2/COX-2/IDO1-FLG in conjunction with the TLR4 and PI3K/AKT-mTOR signaling pathways.

Optimized brewing of Millettia speciosa Champ. leaf golden-flower fungus fermented tea and its in vitro lipid-lowering and glycemic-lowering activity

Li Qing,Wang Ke,Chen Xiaoling,Hu Jinmei,He Xin,Zhou Lin

Given the global rise in obesity and metabolic syndrome, plant-based functional beverages that regulate glycemia and lipidemia have attracted increasing interest. This study focused on Millettia speciosa Champ. leaf Golden-flower fungus fermented tea (MLGFT), a novel fermented tea developed from underutilized leaf biomass. The brewing process was optimized via single-factor and response surface methodology, yielding optimal parameters of 85 °C, 7.3 min, and 146 mL of water, which maximized total phenolic content (124 µg/mL). The optimized tea infusion (OTI) exhibited potent in vitro enzyme inhibition: (93.45 ± 0.98) % against α-glucosidase, (45.52 ± 1.11) % against α-amylase, and (81.47 ± 0.90) % against pancreatic lipase. In cellular models, the freeze-dried extract (ME) dose-dependently enhanced glucose consumption in insulin-resistant HepG2 cells, with effects comparable to metformin at 100 mg/L. In a model of lipid accumulation, ME at 100 mg/L significantly improved the lipid profile, reducing TC, TG, and LDL-C by 63.02%, 43.50%, and 71.79%, respectively, while increasing HDL-C by 46.15%. UPLC-HRMS analysis revealed that the extract is rich in phenolics, particularly epicatechin gallate. These findings demonstrate that MLGFT exhibits notable dual hypoglycemic and hypolipidemic effects, supporting its use as a promising functional beverage for metabolic health management.

Millettia speciosa Champ cellulose-based hydrogel as a novel delivery system for Lactobacillus paracasei: Its relationship to structure, encapsulation and controlled release

Meng-Fan Li,Hua-Ling Cui,Wen-Yong Lou

We report for the first time the usage of Millettia speciosa Champ cellulose (MSCC) and carboxymethylcellulose (MSCCMC) for the fabrication of 3D-network hydrogel as delivery system for probiotics. The structural features, swelling behavior and pH-responsiveness of MSCC-MSCCMC hydrogels and their encapsulation and controlled-release behavior for Lactobacillus paracasei BY2 ( L. paracasei BY2) were mainly studied. Structural analyses demonstrated that MSCC-MSCCMC hydrogels with porous and network structures were successfully synthesized through the crosslinking of –OH groups between MSCC and MSCCMC molecules. An increasing concentration of MSCCMC significantly improved the pH-responsiveness and swelling ability of the MSCC-MSCCMC hydrogel toward neutral solvent. Besides, the encapsulation efficiency (50.38–88.91 %) and release (42.88–92.86 %) of L. paracasei BY2 were positively correlated with the concentration of MSCCMC. The higher the encapsulation efficiency was, the higher the release in the target intestine. However, due to the existence of bile salts , controlled-release behavior decreased the survivor rate and physiological state (degrading cholesterol) of encapsulating L. paracasei BY2. Even so, the number of viable cells encapsulated by hydrogels still reached the minimum effective concentration in the target intestine. This study provides an available reference for the practical application of hydrogels fabricated from the cellulose of the Millettia speciosa Champ plant for probiotic delivery.
