Ultrasound-assisted extraction technology was utilized to extract total flavonoids from Chaenomeles speciosa (Sweet) Nakai, and response surface methodology was employed to optimize the extraction process. The anti-oxidant and lipoxygenase inhibitory activities were evaluated, along with an analysis of the type of inhibition. The results revealed that the optimal extraction conditions for total flavonoids from Chaenomeles speciosa (Sweet) Nakai were as follows: an ethanol concentration of 62%, a liquid-to-solid ratio of 15:1 mL/g, an ultrasonic temperature of 68°C, and an ultrasonic time of 40 min, resulting in a total flavonoid extraction rate of 10.18%. Antioxidant assays demonstrated that the Chaenomeles speciosa (Sweet) Nakai extract exhibited significant radical scavenging activities against 1,1-diphenyl-2-picrylhydrazyl radicals, 2,2’-azinobis (3-ethylbenzothiazoline-6-sulfonic acid ammonium salt) radicals, and hydroxyl radicals, with IC 50 values of 582 µg/mL, 538 µg/mL, and 1709 µg/mL, respectively. Furthermore, enzyme inhibition assays indicated that the Chaenomeles speciosa (Sweet) Nakai extract possesses notable inhibitory activity against lipoxygenase, with an IC 50 value of 2658 µg/mL. This inhibition is mediated through a mixed reversible inhibition mechanism.

This study aimed to enhance the valorization of Chaenomeles speciosa leaves as a sustainable source of bioactive phenolics. An innovative ionic liquid-based ultrasound-assisted extraction (IL-UAE) method was developed for extracting phenolic compounds. Among 10 structurally diverse ILs, [BMIM]Br demonstrated superior extraction performance. Using a combination of single-factor design and response surface methodology (RSM), the optimal parameters for IL-UAE were determined to be the [BMIM]Br concentration of 1.33 mol/L, ultrasonic power of 380 W, extraction time of 10 min, and liquid-to-solid ratio of 22 mL/g. Under these conditions, the yield of C. speciosa leaves total phenolics (CSL-TP) was 78.14 ± 0.35 mg/g, which was substantially higher than those obtained via conventional heat reflux and UAE. After extraction, the microstructures of C. speciosa leaves were examined using scanning electron microscopy (SEM), which confirmed the effectiveness of IL-UAE. Subsequently, NKA-II resin column chromatography was developed to effectively purify crude CSL-TP extracts, guided by leakage and elution curve evaluations, yielding phenolic extracts with a purity of 75.40 % ± 1.93 %. A UPLC–QqQ–MS/MS method was developed for the quantitative analysis of nine major phenolics in purified CSL-TP extracts. Furthermore, bioactivity assessments demonstrated that the purified CSL-TP extracts efficiently scavenged radicals and effectively inhibited the proliferation of HCT-116 and HT-29 cell lines. These results highlight the potential of C. speciosa leaves as a valuable resource for the pharmaceutical and food industries, paving the way for the development of innovative therapeutic products and functional foods.

Chaenomeles speciosa(Sweet) Nakai is a medicinal plant. Until date, there are no studies focusing on comparing the chemical profiles, antioxidant activity andα-glucosidase inhibitory activity of the dried fruits ofC. speciosafrom different production regions. In the study, we investigated the chemical components of dried fruits ofC. speciosafrom Yunnan, Chongqing, Zhejiang and Anhui provinces in China in relation to the antioxidant activity andα-glucosidase inhibitory activity.C. speciosafrom Yunnan had higher total flavonoid (47.92 ± 3.79 mg/g), total polyphenol (29.15 ± 0.29 mg/g) and polysaccharide (27.60 ± 1.56 mg/g) contents than plants from other production areas. Samples from Yunnan, Zhejiang and Anhui (all > 3200 mg/kg) had higher free amino acid contents than those from Chongqing (2286.66 mg/kg). Oleanolic acid and ursolic acid levels were highest in samples from Zhejiang (555.98 ± 20.88 μg/g) and Anhui (321.06 ± 14.64 μg/g), respectively.C. speciosafrom Chongqing had low total flavonoid, total polyphenol, polysaccharide, free amino acid, oleanolic acid and ursolic acid contents but high levels of palmitic acid (12.04 ± 0.02 mg/g) and stearic acid (2.23 ± 0.08 mg/g). Among four production areas, Yunnan represented the highest antioxidant activity andα-glucosidase inhibitory activity. In addition, correlation analysis revealed that total flavonoid, total polyphenol, polysaccharide and ursolic acid were the major components responsible for the antioxidant activity ofC. speciosa, while total flavonoid and polysaccharide were the main contributors forα-glucosidase inhibitory activity of the plant. These results would be helpful for evaluating the quality ofC. speciosain the different production areas. Keywords:Chaenomeles speciosa;chemical composition;antioxidant activity;α-glucosidase inhibitory activity

Biomass-derived activated carbon (BAC) materials are ideal electrode candidates for supercapacitors due to their eco-friendliness, cost-effectiveness, and robust physicochemical stability. The development of porous carbons with high specific surface area and energy density necessitates optimized biomass selection and advanced carbonization-activation strategies. Herein, Chaenomeles cathayensis seeds were employed as carbon precursors to synthesize oxygen self-doped porous activated carbons (CSAC) through sequential carbonization and chemical activation using K2CO3, KOH, or NaOH. Distinct pore-forming mechanisms were observed: K2CO3 preferentially generates micropores, NaOH favors mesopore formation, while KOH creates hierarchical micro-mesoporous architectures. Such hierarchical porosity synergistically facilitates rapid electrolyte ion diffusion through mesopores and provides abundant adsorption sites via micropores. Consequently, CSAC-KOH demonstrates superior electrochemical performance, achieving a specific capacitance of 378 F g-1 at 0.5 A g-1, superior rate capability (249 F g-1 at 20 A g-1), and exceptional cycling stability (99.5% retention after 5000 cycles). The symmetric supercapacitor (SC) fabricated with CSAC electrodes and KOH electrolyte exhibits specific capacitance values of 65.4 F g-1 (0.5 A g-1) and 40 F g-1 (5 A g-1). Meanwhile, the Na₂SO₄-based SC achieves enhanced energy density (11.3 Wh kg-1 at 490 W kg-1) owing to its expanded voltage window (1.4 V), demonstrating the critical role of electrolyte selection in optimizing energy storage performance. This work not only advances the understanding of activator-biomass interactions but also provides a sustainable pathway for valorizing medicinal plant wastes into high-performance energy storage materials.

Wrinkle Floweringquince ( Chaenomeles speciose (Sweet) Nakai) seed, as an unexploited forestry residue, contains considerable amount of bioactive carbohydrates with potential functionality, which was not widely concerned. The aim of this study is to determine the basic characterizations (molecular weight and functional group), specific components (carbohydrate, protein and uronic acid contents), and functional properties of Chaenomeles speciosa seed gum (CSG). Results indicated that carbohydrate (63.80%), protein (13.69%) and uronic acid (10.30%) contents were achieved. The CSG (average molecular weight, 9.85 × 10 6 u) consists rhamnose, arabinose, xylose and glucose in a molar percentage of 29.77꞉10.54꞉18.55꞉15.84, respectively. The Fourier transform infrared (FT-IR) analysis revealed hydroxyl, carboxyl and methyl groups and α -glycosidic linkages are founded in the CSG. The CSG was surface active and its ability to decrease surface tension was comparable to commercial gums. Moreover, the CSG solutions showed pseudoplastic flow behavior under dynamic shear rate at high concentrations. The GSC also presented good emulsifying and foaming properties, indicating the potential of the GSC as bioresource stabilizer and thickener in industry.

Chaenomeles speciosa(Sweet) Nakai (CF), a traditional food in East Asia and a recent addition to clinical dietary recommendations, has demonstrated potential for managing hyperuricemia. However, its bioactive components and therapeutic mechanisms remain largely unexplored. In this study, we used an integrative approach incorporating serum pharmacochemistry, metabolomics, bioinformatics, molecular docking, and in vitro/vivo validation to investigate CF’s effects and mechanisms in hyperuricemia. In hyperuricemic mice, CF significantly reduced serum uric acid, creatinine, and blood urea nitrogen (BUN) levels, improved kidney histopathology, and restored redox balance by increasing antioxidant enzyme activities (SOD and GSH-Px) while lowering malondialdehyde (MDA) levels. Metabolomic analysis revealed that CF modulated pathways associated with oxidative stress, including purine metabolism, arachidonic acid metabolism, and α-linolenic acid metabolism, to reverse hyperuricemia-associated metabolic perturbations. Correlation analysis between differential metabolites and serum-absorbed constituents identified androsin, cynaroside, and salicin as potential bioactive compounds. These compounds showed high predicted binding affinities to COX-1, PGE2, and XOD in molecular docking, and these interactions were validated by in vitro assays, where the compounds effectively suppressed inflammatory cytokine production and inhibited XOD activity. Overall, CF exerts anti-hyperuricemic and renoprotective effects through coordinated regulation of purine metabolism, inflammation, and oxidative stress, supporting its potential as a functional food or complementary therapy for hyperuricemia-related conditions.

Purification and structural characterization of a novel polysaccharide fraction from Chaenomeles speciosa seeds were investigated. After hot water extraction and ethanol precipitation, the crude polysaccharide was sequentially purified with Cellulose DEAE-52 and gel-filtration chromatography, and a highly purified polysaccharide fraction (F3) was obtained. The structure of F3 was characterized by high-performance gel permeation chromatography (HPGPC), high performance liquid chromatography (HPLC), ultraviolet-visible (UV), Fourier transform infrared (FT-IR) and nuclear magnetic resonance (NMR) spectrum, together with methylation, scanning electron microscopy (SEM), atomic force microscope (AFM), and Congo-red test analysis. The results indicated that F3 was a homogeneous polysaccharide fraction with a molecular weight of 8.65 × 10 6 Da, and it was composed of Rha, GlcA, Gal, and Ara in a molar ratio of 6.34:5.73:47.14:40.13. The backbone of F3 was consisted of →3,6)-Gal p -(1→, and the side chains of F3 were composed of Ara f -(1→, →4)-Glc p A-(1→, →4)-Gal p -(1→ and →3)-Rha p -(1→. The hypoglycemic assays demonstrated F3 had good α-amylase and α-glucosidase inhibition activities, and their IC 50 values were 6.24 mg/mL and 4.59 mg/mL respectively. Thus, the polysaccharide from Chaenomeles speciose could be applied as a potential natural source in retarding postprandial hyperglycemia effects.

This study investigated the effects of lactic acid bacteria fermentation on the bioactive components and functional properties of Chaenomeles sinensis (Thouin) Koehne ( C. sinensis ). Fermentation conditions were optimized as follows: ratio of Lactiplantibacillus plantarum and Lactobacillus acidophilus 2:1, inoculum size 0.46% ( w / v ), fermentation time 6 days and sugar addition 7.6 g, resulting in an 80% increase in proanthocyanidin content (1.84 mg/mL). Furthermore, significant increases in total phenolics, total flavonoids, and superoxide dismutase activity were observed under the optimized conditions. Untargeted metabolomics revealed the differential metabolites with antioxidant activity were significantly enriched in citrate cycle, propanoate metabolism, inositol phosphate metabolism, pentose and glucuronate interconversions and starch and sucrose metabolism. C. sinensis fermentation broth demonstrated strengthened free radical scavenging ability and effectively attenuated oxidative stress in HepG2 cells by reducing reactive oxygen species and malondialdehyde levels while enhancing antioxidant enzyme activities, supporting its potential application as functional food ingredients with antioxidant activity.

Chaenomeles speciosa (CS) and Crataegus pinnatifida (CP) are two common Chinese fruits with health benefits. The effect of in vitro digestion on the chemical composition, antioxidant activity and α -glucosidase inhibitory activity of these two fruits, as well as the accessibilities of their active compounds were studied. The inhibitory effects on starch digestion of CS, CP and the 5 identified compounds also were measured. It was observed that the chemical compositions of CS and CP were changed after simulated gastric and intestinal digestions. Their antioxidant and α -glucosidase inhibitory activities both decreased, the chemical composition and bioactivities of CS were changed more after intestinal digestion than after gastric digestion. On the other hand, the chemical composition and bioactivities of CP were changed more after gastric digestion than after intestinal digestion. Bioaccessiblities of compounds in CS and CP varied. Specifically, there were 31, 36 and 68% increases in oleanolic acid, protocatechuic acid and ursolic acid concentrations, respectively, after gastric and intestinal digestions in CS and its bioaccessibilities were more than doubled. The high bioaccessibility observed for protocatechuic acid, oleanolic acid and ursolic acid suggested the bioactive potential of CS. CS, CP, gallic acid , chlorogenic acid , catechin and oleanolic acid all inhibited in vitro starch digestion. Therefore, it may help understand the changes of the chemical composition, antioxidant activity and α -glucosidase inhibitory activity of CS and CP during digestion. It also supported that CS and CP are good potential candidates for controlling postprandial hyperglycemia in diabetics.

Chaenomeles speciosa (Sweet) Nakai fruit is a good source of phenolics with many health benefits. In this work, the enrichment of C. speciosa fruit total phenolics (CSFTP) using macroporous resins was studied. NKA-Ⅱ resin was selected for enriching CSFTP due to its highest adsorption/desorption quantity. Adsorption characteristics of CSFTP on NKA-Ⅱ resin exhibited a good fit with the Langmuir isotherm model and pseudo-second order kinetics model. This adsorption was spontaneous, exothermic, and entropy-decreasing through a physisorption mechanism. The breakthrough-elution curves were studied to optimize CSFTP enrichment conditions. One-step enrichment increased CSFTP content in the extracts from 26.51 % to 78.63 %, with a recovery of 81.03 %. A UPLC-QqQ-MS/MS method in multiple reaction monitoring (MRM) mode was established and validated for the simultaneous quantification of seven phenolic compounds. This study demonstrates the feasibility of industrial enrichment of CSFTP using NKA-Ⅱ resin and proposes a reliable method for quality control of CSFTP-rich products.

Background Previous studies have demonstrated that numerous medicine and food homology (MFH) possess the potential to regulate purine metabolism disorders, promote uric acid excretion, and alleviate hyperuricemia symptoms. Examples include CS (Chaenomeles speciosa (Sweet) Nakai), SR (Smilax glabra Roxb.) and PL (Pueraria montana var. lobata). Methods Metabolomics was employed to analyze the compositional changes in medicinal and edible extracts before and after fermentation. Network pharmacology and molecular docking studies were further utilized to elucidate the interactions between these differential metabolites and the core targets of hyperuricemia. In vitro enzyme activity assays were conducted to confirm the therapeutic effects. Results A total of 283, 248, and 18 differential metabolites were identified in CS,SR and PL samples, respectively. Among these, 54 significantly upregulated differential metabolites were selected for screening. Based on these metabolites, 53 HUA-related targets were identified for CS, SR and PL. Functional enrichment analysis revealed their roles in inflammatory stress and uric acid production pathways, particularly the MAPK signaling pathway and purine metabolism regulated by XDH. Additionally, other targets in the purine metabolism pathway, such as ADA, PNP, AMPD3, and IMPDH2, were co-regulated. Enzyme activity assays indicate that fermented MFH more effectively inhibits XOD, thereby regulating the conversion of xanthine and hypoxanthine into uric acid. Molecular docking revealed two significantly upregulated compounds in CS; and five in PL; and four in SR. exhibit strong binding to XOD.Conclusion These findings provide theoretical support for FMFH as a potential effective component in preventing and treating hyperuricemia. Our research demonstrates that FMFH targets multiple pathways associated with hyperuricemia, offering a promising approach for preventing this condition.
