Development of reliable in vitro long-term culture systems for hematopoietic cells of crayfish Cherax quadricarinatus and virus susceptibility assay

Yaqi Zhao,Ting Xue,Jinwu Wang,Huarong Guo

Long-term in vitro cultured crayfish cells can serve as a valuable tool for investigating various aspects of crayfish biology, encompassing cell physiology and viral mechanisms. Regrettably, they are difficult to maintain a highly active state and survive during long-term and continuous culture in vitro . In order to generate highly viable crayfish cell cultures, it is crucial to optimize dependable culture conditions and utilize a suitable cell medium. In this study, we have successfully developed reliable in vitro long-term culture systems for hematopoietic (HPT) cells of crayfish Cherax quadricarinatus by formulating an optimal crayfish cell growth medium (OCCM), including under two-dimensional (2D) and three-dimensional (3D) conditions. The novel medium was optimized by a mix of exogenous nutrients, crayfish plasma (CP) and shrimp serum-derived amino acids and saccharides within the Leibovitz's L-15 (L-15) medium. Our newly developed crayfish cell growth medium (CCM-28) enabled the culturing of HPT cells for over 142 days under 2D and over 60 days under 3D conditions, with consistent active cell proliferation observed over a 30-day incubation period. This proliferation was comparable to in vivo conditions, with a cell division rate exceeding 20%, suggesting that a high mitotic rate has been successfully achieved during long-term in vitro culture. Flow cytometry analysis further elucidated the mitosis-arrest of HPT cells cultured in vitro was predominantly observed in the G 0 /G 1 phase. Furthermore, the cell subculture techniques have been established using mechanical dissociation method (in 2D) and substrate exchange method (in 3D). Additionally, the susceptibility of these cell cultures to covert mortality nodavirus (CMNV) and white spot syndrome virus (WSSV) was confirmed by capturing cytopathic effects (CPE) and analyzing viral copy numbers. Species identification using the cytochrome oxidase subunit I ( COI ) gene established the origin of the cell cultures from C. quadricarinatus . In conclusion, the successful development of highly viable crayfish cells in long-term in vitro culture systems, as demonstrated in this study, will significantly advance the research on immortalization in crayfish cells and the study of mechanisms related to viruses.

Dietary astaxanthin alleviates microcystin-LR-induced hepatopancreatic digestive and absorptive dysfunction in red claw crayfish (Cherax quadricarinatus)

Yao Li,Beini Liu,Shuai Yuan,Kaixin Ren,Beixi Lu,Sirui Zhou,Shuquan Ding,Xilei Li

Astaxanthin is a functional feed additive with antioxidant and antistress properties in crustaceans. This study investigated the protective efficacy and underlying mechanisms of dietary astaxanthin against hepatopancreatic digestive and absorptive dysfunction caused by microcystin-LR (MC-LR) in red claw crayfish ( Cherax quadricarinatus ). Crayfish were fed diets containing 0, 3, 6, 9, or 12 mg/kg astaxanthin for 60 days. Growth performance was evaluated, after which crayfish from the control and 6 mg/kg groups were injected with saline or MC-LR (150 μg/kg body weight) and sampled after 24 h. MC-LR disrupted tubular architecture, reduced B cells, caused abnormal vacuolation in resorptive cells, and significantly decreased trypsin activity. Conversely, dietary astaxanthin preserved hepatopancreatic cell integrity and significantly elevated trypsin, lipase, and amylase activities during exposure. Transcriptomic analysis indicated that MC-LR suppressed genes involved in protein processing in endoplasmic reticulum (ER), proteasome, and protein digestion and absorption. Dietary astaxanthin restored the expression of key genes across these pathways, supporting protein folding, misfolded protein clearance, and nutrient transport. These findings demonstrate that dietary astaxanthin alleviates MC-LR-induced hepatopancreatic digestive and absorptive dysfunction by maintaining tissue structure, boosting digestive enzyme activities, and restoring protein homeostasis.

Multi-omics profiling of muscle tissue reveals metabolic signatures associated with size variation in redclaw crayfish (Cherax quadricarinatus)

Jinhua Xu,Zihan Wang,Lu Qiao,Zhe Xu,Zhen Yang,Jincheng Li,Xin Hu,Xiaodong Xu,Yingchun Mu

Muscle multi-omics reveal size-associated metabolic signatures. • Large crayfish show enhanced carbohydrate and purine metabolism. • Small crayfish exhibit constrained metabolism and potential oxidative stress. • Metabolic signatures may inform precision nutrition strategies. The rising market demand for large, premium grade redclaw crayfish ( Cherax quadricarinatus ) highlights the need for improved production efficiency, yet the molecular characteristics associated with body-size variation remain poorly understood. This study employs an integrated multi-omics approach (untargeted metabolomics, targeted taurine analysis, and transcriptomics) to characterize the metabolic signatures associated with crayfish size variation. Untargeted analysis of 18,204 metabolite features identified 229 differentially abundant metabolites, including a 28-fold higher level of sucrose in large individuals ( p  = 0.025). Targeted analysis confirmed that taurine, a key osmolyte and antioxidant, was significantly elevated in larger crayfish. Integrated metabolomic and transcriptomic analyses revealed metabolic features associated with carbohydrate and purine metabolism that correlated with body-size variation. In contrast, smaller crayfish exhibited elevated purine turnover (xanthine/uric acid) and reduced levels of key anabolic metabolites. These findings provide a multi-omics characterization of metabolic states associated with growth phenotypes and identify candidate biomarkers for future studies investigating the potential of nutritional strategies to improve crayfish production. Toxicological impacts of paddy field pesticides on red claw crayfish (Cherax quadricarinatus) and implications for integrated rice-crayfish farming

Ru Liang,Qianxue Shao,Yongming Ruan,Min Wang,Jiarong Cui,Jiaojiao Xia,Yanfei Zhu,Ziyao Liu,Zengpei Lin,Hongxing Xu,Zhongxian Lu,Pingyang Zhu

The integrated "rice-aquaculture" farming system is increasingly promoted in China as a sustainable agricultural model offering "dual benefits of grain and aquaculture". However, the safety of pesticides to co-cultured aquatic species, such as the red claw crayfish ( Cherax quadricarinatus ), requires thorough investigation. This study assessed the acute and chronic toxicological effects of six widely used chemical pesticides in paddy fields—abamectin, trifloxystrobin, triflumezopyrim, glufosinate-ammonium, zinc thiazole, and pretilachlor—on C. quadricarinatus under laboratory conditions. Acute toxicity tests revealed that abamectin and trifloxystrobin were highly toxic to C. quadricarinatus . Pretilachlor exhibited moderate toxicity. In contrast, triflumezopyrim, glufosinate-ammonium, and zinc thiazole demonstrated low acute toxicity. Subsequent chronic toxicity assessments (7-day exposure followed by a 14-day recovery) indicated that sub-lethal concentrations of triflumezopyrim, glufosinate-ammonium, and zinc thiazole induced neurotoxicity in C. quadricarinatus . Additionally, triflumezopyrim exposure led to oxidative stress, as evidenced by altered antioxidant enzyme activities and increased lipid peroxidation and DNA damage. Most notably, these sub-lethal effects-including significant changes in growth parameters (SGR, HSI), neurotoxicity (AChE inhibition), oxidative stress (SOD, CAT, MDA), and genotoxicity (8-OHdG) were largely reversible, consistently returning to control levels after a 14-day recovery in clean water. This demonstrated resilience provides a strong scientific basis for the periodic and safe application of these low-toxicity agents in integrated rice-crayfish system, while the highly toxic abamectin, trifloxystrobin, and moderate toxic pretilachlor pose significant risks and their use should be limited. This research provides a scientific basis for pesticide selection to support the sustainable development of integrated rice-red claw crayfish farming systems, contributing to the protection of aquatic ecosystems while ensuring the economic benefits of agricultural production.

Effects of dietary octacosanol supplementation on growth, feed utilization, and physiological and molecular responses of red claw crayfish (Cherax quadricarinatus)

Alhoshy Mayada,Shehata Akram Ismael,Habib Yusuf Jibril,Wang Xuexi,Wang Yilei,Zhang Ziping

Crayfish species like Cherax quadricarinatus are crucial nutrients that contribute to the world economy but face challenges in production due to dietary supplements. Octacosanol offers diverse health benefits, from immunity boosts to anti-inflammatory and anti-tumor effects. The current investigation assessed the ovarian changes, immune reaction, availability of antioxidants, gene response pattern, and entire organ performance of the red claw crayfish after supplementation with different levels of octacosanol. Two hundred twenty-five C. quadricarinatus with an initial weight of 53 ± 1.11 (g/crayfish) were randomly stocked into fiberglass tanks (15 crayfish/tank) and fed each day with three diverse dietary octacosanols. Different levels of octacosanol (4, 8, 12, and 16 mg/kg) were tested in triplicates over 60 days, denoted as D1, D2, D3, and D4, respectively, excluding the control group. The study revealed that 8 mg/kg octacosanol effectively enhances growth and feed utilization in C. quadricarinatus , while also improving ovarian performance by increasing hepatosomatic index (HSI), gonadosomatic index (GSI), and condition factor (CF). Additionally, it decreased levels of MDA and NO in crayfish. Supplementation with 16 mg/kg octacosanol boosted antioxidant capacity by enhancing activities of T-AOC and SOD, as well as influencing nonspecific immune enzymes in the hepatopancreas at doses of D3 (12 mg/kg) and D4 (16 mg/kg). Furthermore, it positively impacted GLU and protein profiles while reducing levels of T-BIL, T-CHOL, TG, LDL-c, and VLDL-c in crayfish. These dietary interventions also regulated gene expression associated with reproduction, lipid metabolism, antioxidants, and immune response in various organs of C. quadricarinatus . Octacosanol demonstrated potential as a safe and effective feed additive. According to quadratic regression analysis, nutritional supplementation of octacosanol at 9 mg/kg diet is considered the recommended dosage in the red claw crayfish diet.
