Screening Sugars Can Benefit the Parasitoid Cotesia chilonis (Hymenoptera: Braconidae) Without Benefiting Its Host, Chilo suppressalis (Lepidoptera: Crambidae)

Tian Jun-Ce,Chen Yang,Shelton Anthony M,Zheng Xu-Song,Xu Hong-Xing,Lu Zhong-Xian

Most adult parasitoids depend on carbohydrate-rich food as an energy source for longevity, fecundity, and mobility. Thus, providing sugars has been proposed as a technique to maximize the biological control efficacy of parasitoids. However, the sugars provided for parasitoids need to be carefully selected because herbivore hosts might also benefit. Here we explore the effects of 12 naturally occurring sugars on the longevity and fecundity of the rice pest, Chilo suppressalis, and the longevity of its parasitoid, Cotesia chilonis, as well as the effect of sugars on sugar consumption of C. chilonis. Results showed that none of the tested sugars significantly impacted the longevity of C. suppressalis, but fructose, glucose, maltose, and sucrose significantly increased its fecundity. The longevity of C. chilonis was significantly increased when fed fructose, glucose, sucrose, maltose, trehalose, and melezitose. Our data suggest that C. chilonis consumed larger quantities of glucose, fructose, and sucrose followed by maltose, melezitose, and trehalose and the longevity of C. chilonis was positively correlated to sugar consumption. Our data also suggest that the herbivore C. suppressalis and its parasitoid C. chilonis responded differently to trehalose and melezitose. Although additional studies are needed, our data suggest that these sugars could be considered as candidate components for sugar sprays to enhance the activity and efficacy of C. chilonis, but without benefiting its pest host.

Mechanisms for multiple resistances in field populations of rice stem borer, Chilo suppressalis (Lepidoptera: Crambidae) from Sichuan Province, China

Xia Zhao,Xiang Xu,Xue-gui Wang,Yong Yin,Ming-yang Li,Ya-qiong Wu,Ying-hong Liu,Qing-hua Cheng,Chang-wei Gong,Li-tao Shen

Chilo suppressalis Walker (Lepidoptera: Crambidae) is a widely destructive pest occurring in rice, particularly in the rice-growing regions of Asia. In recent years, C. suppressalis has developed resistance to several insecticides because of the extensive use of insecticides. The resistance levels to four insecticides were determined among populations from different regions of Sichuan Province, China, using a drop-method bioassay. Based on LC 50 values of a laboratory susceptible strain, all field populations showed moderate level of resistance to triazophos (23.9- to 83.5-fold) and were either susceptible or had a low level of resistance to abamectin (2.1- to 5.8-fold). All field-collected populations had a low or moderate level of resistance to chlorpyrifos (1.7- to 47.1-fold) and monosultap (2.7- to 13.5-fold). The synergism experiment indicated that the resistance of the XW19 to triazophos may be associated with cytochrome P450 monooxygenases (P450s), with the highest synergistic ratio (SR) of 3.05-fold and increased ratio (IR) of 2.28-fold for piperonylbutoxide (PBO). The P450 activity of the TJ19 population was the greatest among the six field populations. Moreover, the relative expression levels of four resistance-related P450 genes were detected with qRT-PCR, and the results indicated that CYP324A12 , CYP321F3 and CYP9A68 were overexpressed in the resistant population, especially in the XW19 population (by 1.2-, 3.4 -, and 18.0-fold, respectively). In addition, the relative expression levels of CYP9A68 among the CZ19 and TJ19 populations were also enhanced 10.5- and 24.9-fold, respectively. These results suggested that CYP324A12 , CYP321F3 and CYP9A68 may be related to the resistance development of C. suppressalis to triazophos.

Monitoring and mechanisms of insecticide resistance in Chilo suppressalis (Lepidoptera: Crambidae), with special reference to diamides

Rong Yao,Dan-Dan Zhao,Shuai Zhang,Li-Qi Zhou,Xin Wang,Cong-Fen Gao,Shun-Fan Wu

BACKGROUND The rice stem borer, Chilo suppressalis Walker (Lepidoptera: Crambidae), is one of the most economically important pests of rice in Asia. Chemical control remains the most efficient primary means for controlling this pest. RESULTS Significant variations among field populations in their resistance to seven insecticides were observed. The populations exhibited LC 50 values that ranged between 0.605 and 108.088 mg AI L −1 for chlorantraniliprole and between 0.046 and 3.919 mg AI L −1 for flubendiamide. The YY14 population collected from Yuyao in Zhejiang Province in 2014 showed a moderate resistance level to the two diamides, i.e. up to 77.6-fold and 42.6-fold for chlorantraniliprole and flubendiamide respectively. Synergism tests and biochemical assays showed no obvious correlations between diamide resistance and three detoxifying enzymes. Sequence comparison of the ryanodine receptor gene between the YY14 resistant population and susceptible population revealed that a glycine to glutamic acid substitution (G4910E) was presented in the YY14 population. CONCLUSION G4910E mutation might be involved in the resistance evolution of C. suppressalis to the diamides. An appropriate insecticide resistance management programme should be established to maintain the effectiveness of the insecticides and to ensure sustainable management. © 2016 Society of Chemical Industry

CYP321F3 mediates metabolic resistance to methoxyfenozide in rice stem borer, Chilo suppressalis

Fang-Rui Guo,Shu-Chao Wang,Yan Liu,Shuai Wang,Jing-Mei Huang,Hao Sun,Lin-Feng He,Yuan Xie,Song-Tao Qiao,Feng-Xia Yang,Chris Bass,Cong-Fen Gao,Shun-Fan Wu

The development of insecticide resistance in insect populations is a major challenge to sustainable agriculture and food security worldwide. Methoxyfenozide, an insect growth regulator that acts as an agonist of 20-hydroxyecdysone (20E), has severely declined in its efficacy against the rice stem borer ( Chilo suppressalis ), a notorious pest of rice crops in East and Southeast Asia. To date, however, the genes involved in methoxyfenozide resistance in target pests remain unclear. We conducted a long-term (seven years from 2017 to 2023) and large geographical scale (8 provinces and 45 cities in China) resistance monitoring program for methoxyfenozide in C. suppressalis . Resistance was seen to arise rapidly in this species, with >100-fold resistance being detected in nearly all the field populations after 2018 . Piperonyl butoxide (PBO), an inhibitor of cytochrome P450 enzymes (P450s), significantly increased the sensitivity of resistant strains of C. suppressalis to methoxyfenozide, implicating P450s in resistance. Six P450 genes: CYP321F3, CYP6CV5, CYP9A68 , CYP6AB45, CYP324A12 and CYP6SN2 were identified as highly expressed in resistant C. suppressalis by transcriptome profiling. Of these, ectopic expression of CYP321F3 in Drosophila melanogaster resulted in a 7.0-fold increase in resistance to methoxyfenozide demonstrating its causal role in resistance. Collectively, these findings provide insight into the mechanisms mediating resistance to insect growth regulators and will inform the development of future pest and resistance management strategies.

Adaptive neofunctionalization of ancestral CYP6SN2vB following duplication contributes to abamectin resistance in Chilo suppressalis Walker

Shuai Wang,Yuan Xie,Chong Liu,Jin-Wei Liu,Fang-Rui Guo,Song-Tao Qiao,Yu-Ji Xing,Shun-Fan Wu,Chris Bass,Cong-Fen Gao

Abamectin serves as a key alternative for managing diamide-resistant Chilo suppressalis . However, resistance to abamectin itself presents a significant challenge to this pest control, and its genetic basis remains poorly understood. In this study, we report an increase in field resistance to abamectin in populations of C. suppressalis and provide evidence for a cytochrome P450-mediated metabolic resistance mechanism, supported by enzyme activity assays and synergist bioassays. Transcriptomic profiling analysis revealed six candidate P450 genes, among which the duplicated gene CYP6SN2 showed a strong correlation with resistance. Variant CYP6SN2vB was highly expressed in detoxification tissues and mainly produced isoforms with a short 3′-UTR. Functional validation in transgenic Drosophila melanogaster confirmed that overexpression of CYP6SN2vB conferred 2.4- and 1.9-fold resistance to abamectin and emamectin benzoate, whereas CYP6SN2vA had no detectable effect. Molecular docking further revealed that CYP6SN2vB could potentially hydroxylate abamectin via interactions near oxygen-binding motifs. In contrast, the inward-folded substrate recognition site 1 region of CYP6SN2vA compresses the ligand-binding cavity, precluding abamectin interaction. These findings suggest neofunctionalization of CYP6SN2vB following duplication and provide valuable insights into the molecular basis of abamectin resistance in C. suppressalis , with implications for resistance management.

Constitutive overexpression of cytochrome P450 monooxygenase genes contributes to chlorantraniliprole resistance in Chilo suppressalis (Walker)

Lu Xu,Jun Zhao,Yang Sun,Dejin Xu,Guangchun Xu,Xiaolong Xu,Yueliang Zhang,Shuijin Huang,Zhaojun Han,Zhongyan Gu

BACKGROUND The rice striped stem borer (SSB), Chilo suppressalis (Walker), which is one of the most economically important phytophagous pests, has developed resistance to multiple insecticides. The resistance of SSB against chlorantraniliprole has been investigated in detail. However, the mechanism of its metabolic resistance has rarely been studied. RESULTS A field population from Wuhu City, China was used to establish chlorantraniliprole resistant and susceptible strains (WHR and WHS) by laboratory continuous selection. Enzyme activities data suggested the potential involvement of cytochrome P450 monooxygenase in WHR. CYP6CV5 , CYP9A68 , CYP321F3 , and CYP324A12 were significantly overexpressed in WHR (from 4.48 to 44.88-fold). These four P450 genes were expressed in the late developmental stages of WHR; however, they were almost absent during the egg stage. In addition, their expressions were much more sensitive to chlorantraniliprole induction in WHR than in WHS. Injection of individual and mixture dsRNAs reduced the expression of the four target genes (55.2–73.2% and 43.2–50.2%, respectively) and caused significant larvae mortality (55.1–65.1% and 88.2%, respectively). CONCLUSION Multiple overexpressed P450 genes were potentially associated with chlorantraniliprole resistance, as confirmed by the RNA interference (RNAi) assay. Our findings suggested that metabolic resistance to chlorantraniliprole might be mediated by P450s. © 2018 Society of Chemical Industry

Heavy metal exposure reduces larval gut microbiota diversity of the rice striped stem borer, Chilo suppressalis

Wang Jie,Huang Hexi,Yang Hailin,Wang Su,Li Mengnan,Zhu Zhengyang,Trumble John T.,Di Ning,Zang Liansheng

Cadmium (Cd), a widely distributed environmental pollutant in agroecosystems, causes negative effects on crops and herbivores through bottom-up processes. The gut microbial community of an insect can play a critical role in response to metal stress. To understand how microbiota affect the stress responses of organisms to heavy metals in agroecosystems, we initially used 16S rRNA sequencing to characterize the larval gut microbiota of Chilo suppressalis , an important agricultural pest, exposed to a diet containing Cd. The species richness, diversity, and composition of the gut microbial community was then analyzed. Results revealed that while the richness (Chao1 and ACE) of gut microbiota in larvae exposed to Cd was not significantly affected, diversity (Shannon and Simpson) was reduced due to changes in species distribution and relative abundance. Overall, the most abundant genus was Enterococcus , while the abundance of the genera Micrococcaceae and Faecalibaculum in the control significantly superior to that in Cd-exposed pests. Phylogenetic investigation of microbial communities by the reconstruction of unobserved states (PICRUSt) showed that the intestinal microorganisms appear to participate in 34 pathways, especially those used in environmental information processing and the metabolism of the organism. This study suggests that the gut microbiota of C. suppressalis are significantly impacted by Cd exposure and highlights the importance of the gut microbiome in host stress responses and negative effects of Cd pollution in agroecosystems.

Eco-friendly chitosan base chlorantraniliprole nano-pesticides for effective control of Chilo suppressalis (Walker) through bidirectional transport

Ao Liang,Yunzheng Zhang,Xiang Xu,Hao Wang,Changwei Gong,Jie Hu,Xiangsong Li,Jizhi Yang,Anchun Peng,Xuegui Wang

Chilo suppressalis is a major pest that severely impacts rice production in China. However, the widespread use of insecticides has resulted in the development of resistance in C. suppressalis. The advancement of nanotechnology offers promising prospects for enhancing insecticide formulations and improving their efficacy. This study designed a pH-responsive release system composed of γ-PGA and chitosan (CS) loaded with chlorantraniliprole (CLAP). The synthesized CLAP-loaded nanoparticles had an average particle size of approximately 39.67 nm and a loading efficiency of 38.87%. Under a pH of 8.5, 64.4% of the pesticide was released within 120 hours. The CLAP@CS/γ-PGA formulation, after loading, exhibited a significant synergistic insecticidal effect, with bioassay results showing an 82.2% mortality rate of C. suppressalis six days post-treatment. Tests of metabolic genes and enzyme activities showed that CLAP@CS/γ-PGA rendered C. suppressalis more sensitive to insecticides by inhibiting the activities of P450 and by decreasing the expression of CYP9A68. CLAP@CS/γ-PGA also demonstrated favorable transport properties within C. suppressalis and rice plants, and due to the encapsulation by the nanoparticle carrier, it reduced toxicity to zebrafish. In summary, the system we investigated not only meets the needs of pest management but also enhances the utilization of pesticides.

Bt rice plants may protect neighbouring non-Bt rice plants against the striped stem borer, Chilo suppressalis

Yaoyu Jiao,Xiaoyun Hu,Yufa Peng,Kongming Wu,Jörg Romeis,Yunhe Li

The area planted with insect-resistant genetically engineered crops expressing Bacillus thuringiensis (Bt) genes has greatly increased in many areas of the world. Given the nearby presence of non-Bt crops (including those planted as refuges) and non-crop habitats, pests targeted by the Bt trait have a choice between Bt and non-Bt crops or weeds, and their host preference may greatly affect insect management and management of pest resistance to Bt proteins. In this study, we examined the oviposition preference of the target pest of Bt rice, Chilo suppressalis, for Bt versus non-Bt rice plants as influenced by previous damage caused by C. suppressalis larvae. The results showed that C. suppressalis females had no oviposition preference for undamaged Bt or non-Bt plants but were repelled by conspecific-damaged plants whether Bt or non-Bt. Consequently, C. suppressalis egg masses were more numerous on Bt plants than on neighbouring non-Bt plants both in greenhouse and in field experiments due to the significantly greater caterpillar damage on non-Bt plants. We also found evidence of poorer performance of C. suppressalis larvae on conspecific-damaged rice plants when compared with undamaged plants. GC-MS analyses showed that larval damage induced the release of volatiles that repelled mated C. suppressalis females in wind tunnel experiments. These findings suggest that Bt rice could act as a dead-end trap crop for C. suppressalis and thereby protect adjacent non-Bt rice plants. The results also indicate that the oviposition behaviour of target pest females should be considered in the development of Bt resistance management strategies.

Does cadmium cause cascading effects on the development and reproduction of the striped stem borer, Chilo suppressalis (Walker)?

Huang Hexi,Di Ning,Wang Jie,Wang Yuxing,Zhu Zhengyang,Lu Caige,Wang Su,Zang Liansheng

The heavy metal, cadmium (Cd), causing growth retardation and yield reduction on rice and impacting the fitness of organisms inhabiting on rice through bottom-up effects, has become a great challenge to rice production. However, the effect of Cd-exposure on the development of an economically important and destructive rice pest, Chilo suppressalis remains unexplored. By exposing the larvae of C. suppressalis to different Cd-exposed artificial diets (0, 0.2, 1.0, 2.5, 5.0, and 10.0 mg/kg), we found that Cd exposure did not affect the larval duration or pupation rate of C. suppressalis , but caused negative effects on pupal weight at high Cd levels (5.0 and 10.0 mg/kg) and on adult deformity rate from 2.5 and 5.0 mg/kg treatments. Although Cd significantly increased the female pupae ratio, C. suppressalis did not oviposit when Cd treatment was more than 2.5 mg/kg. Meanwhile, Cd transferred to pupae, females, exuviae of pupa and eggs of C. suppressalis from Cd treated larvae, and exhibited a dose-dependent response on Cd accumulation. Our results indicated that Cd had a negative effect on rice stem borer and can be transferred to eggs of C. suppressalis , but more work is needed to further assess the bottom-up effect on third tropic levels in Cd-polluted fields.

Differences in susceptibility to chlorantraniliprole between Chilo suppressalis (Lepidoptera: Crambidae) and two dominant parasitic wasps collected from Sichuan Province, China

Ming-yang Li,Chang-wei Gong,Yun-zheng Zhang,Xia Zhao,Yan Jia,Jian Pu,Xue-mei Liu,Xiang Xu,Xue-gui Wang

Chilo suppressalis Walker (Lepidoptera: Crambidae) is one of the most destructive pests occurring in the rice-growing regions of Asia. Parasitoids , mainly egg parasitoids , have been of interest for several years even with practical used cases. Therefore, the potential impact of insecticides on natural enemies needs great attention. In this study, chlorantraniliprole was evaluated for its impact on C. suppressalis and two dominant parasitic wasps . Bioassays showed that chlorantraniliprole had negligible toxicity to Eriborus terebrans but was significantly toxic to Chelonus munakatae ; the mortality exceeded 50% when the concentration reached 46.83 ng/cm 2 . Enzyme assays suggested that the significantly different carboxylesterase activity may be involved in the high-level detoxification metabolism of E. terebrans . According to the results of enzyme gene correlation analysis, P450s may be the dominant factor in the detoxification metabolism of C. munakatae . In addition, the ryanodine receptor C-terminus of C. suppressalis (CsRyR), C. munakatae (CmRyR) and E. terebrans (EtRyR) were successfully cloned. Different amino acids at resistance mutation I4758 M between susceptible C. suppressalis (I) and parasitic wasps (M) may be related to susceptibility differences. Simulated docking showed that CsRyR and CmRyR can interact with chlorantraniliprole but not EtRyR. More interaction forces were formed between CsRyR and chlorantraniliprole than CmRyR. Furthermore, a Pi-Pi T-shape formed between 73PHE in CsRyR and the benzene ring in chlorantraniliprole. These results indicated that both detoxification metabolism and the target site could mediate the susceptibility difference between C. suppressalis and its parasitic wasps.

Infestation of Rice Striped Stem Borer (Chilo suppressalis) Larvae Induces Emission of Volatile Organic Compounds in Rice and Repels Female Adult Oviposition

Chen Shen,Shan Yu,Xinyang Tan,Guanghua Luo,Zhengping Yu,Jiafei Ju,Lei Yang,Yuxuan Huang,Shuai Li,Rui Ji,Chunqing Zhao,Jichao Fang

Plants regulate the biosynthesis and emission of metabolic compounds to manage herbivorous stresses. In this study, as a destructive pest, the pre-infestation of rice striped stem borer (SSB,Chilo suppressalis) larvae on rice (Oryza sativa) reduced the subsequent SSB female adult oviposition preference. Widely targeted volatilomics and transcriptome sequencing were used to identify released volatile metabolic profiles and differentially expressed genes in SSB-infested and uninfested rice plants. SSB infestation significantly altered the accumulation of 71 volatile organic compounds (VOCs), including 13 terpenoids. A total of 7897 significantly differentially expressed genes were identified, and genes involved in the terpenoid and phenylpropanoid metabolic pathways were highly enriched. Correlation analysis revealed that DEGs in terpenoid metabolism-related pathways were likely involved in the regulation of VOC biosynthesis in SSB-infested rice plants. Furthermore, two terpenoids, (−)-carvone and cedrol, were selected to analyse the behaviour of SSB and predators. Y-tube olfactometer tests demonstrated that both (−)-carvone and cedrol could repel SSB adults at higher concentrations; (−)-carvone could simultaneously attract the natural enemies of SSB,Cotesia chilonisandTrichogramma japonicum, and cedrol could only attractT. japonicumat lower concentrations. These findings provide a better understanding of the response of rice plants to SSB and contribute to the development of new strategies to control herbivorous pests.
