Impact of surface tension of wastewater on biofilm formation of microalgae Chlorella sp.

Xinru Zhang,Hao Yuan,Zeyi Jiang,Dahao Lin,Xinxin Zhang

The organic matter and surfactants in wastewater may cause variations in the surface tension of wastewater (STW) ranging between ∼40 and ∼70 mJ·m −2 . This study focused on the influence of STW on microalgae biofilm formation. A theoretical analysis was first conducted, and then microalgae biofilm formation on hydrophilic and hydrophobic substrata in liquid and real wastewater with different surface tensions was studied. The results demonstrated that STW affected microalgae biofilm formation remarkably. When the surface tension of liquid medium ( γ lv ) was approximately equal to the average value of surface free energy of microalgae and substrata, biofilm formation reached the minimum. Microalgae biofilm formation on a hydrophilic surface first decreased (from ∼2200 to ∼1500 cells/mm 2 ) and then increased (from ∼1500 to 3100 cells/mm 2 ) with the decrease in γ lv (from ∼70 to ∼40 mJ·m −2 ), whereas biofilm on a hydrophobic surface continued to decrease (from ∼2500 to 1000 cells/mm 2 ).

Impact of Natural Microorganisms on the Removal of COD and the Cells Activity of the Chlorella sp. in Wastewater

Qingnan Sun,Xiaoping Zhang,Xin Zhang

In the treatment of wastewater containing only chemical oxygen demand (COD) byChlorellasp., the cell activity and proliferation ability ofChlorellasp. decreased with the culture time, which affected the removal of COD in wastewater. To solve these problems, theChlorellasp.–natural microorganism symbiosis system was prepared. The system was used to explore how natural microorganisms affect the cell activity and the proliferation ability ofChlorellasp. in wastewater. In the treatment of COD byChlorellasp., the removal rate of COD decreased from 45.47% to 28.88%, with a decrease in the cell activity and proliferation ability ofChlorellasp. In theChlorellasp.–natural microorganism symbiotic system, the removal rate of COD reached 45.75%. With the introduction of natural microorganisms, the circulation of CO2and O2betweenChlorellasp. and natural microorganisms promoted photosynthesis and respiration, which enhanced the cell activity ofChlorellasp. Under the condition that the dosage of natural microorganisms was between 1% and 6%, the concentration ofChlorellasp. was close to the logarithmic growth phase, which maintained the proliferation ability ofChlorellasp. At the same time, the natural microorganisms grew and proliferated in wastewater containing only COD through preying onChlorellasp.Keywords:Chlorellasp.;natural microorganisms;symbiosis system;cell activity;predation

Chlorella sp.-ameliorated undesirable microenvironment promotes diabetic wound healing

Hangyi Wu,Pei Yang,Aiqin Li,Xin Jin,Zhenhai Zhang,HuiXia Lv

Chronic diabetic wound remains a critical challenge suffering from the complicated negative microenvironments, such as high-glucose, excessive reactive oxygen species (ROS), hypoxia and malnutrition. Unfortunately, few strategies have been developed to ameliorate the multiple microenvironments simultaneously. In this study, Chlorella sp. (Chlorella) hydrogels were prepared against diabetic wounds. In vitro experiments demonstrated that living Chlorella could produce dissolved oxygen by photosynthesis, actively consume glucose and deplete ROS with the inherent antioxidants, during the daytime. At night, Chlorella was inactivated in situ by chlorine dioxide with human-body harmless concentration to utilize its abundant contents. It was verified in vitro that the inactivated-Chlorella could supply nutrition, relieve inflammation and terminate the oxygen-consumption of Chlorella-respiration. The advantages of living Chlorella and its contents were integrated ingeniously. The abovementioned functions were proven to accelerate cell proliferation, migration and angiogenesis in vitro . Then, streptozotocin-induced diabetic mice were employed for further validation. The in vivo outcomes confirmed that Chlorella could ameliorate the undesirable microenvironments, including hypoxia, high-glucose, excessive-ROS and chronic inflammation, thereby synergistically promoting tissue regeneration. Given the results above, Chlorella is considered as a tailor-made therapeutic strategy for diabetic wound healing.

Microalgae fuel cells enhanced biodegradation of imidacloprid by Chlorella sp.

Zhikang Deng,Jinyao Zhu,Lie Yang,Zulin Zhang,Bolin Li,Ling Xia,Li Wu

The widespread use of neonicotinoid pesticides has caused environmental threat that has attracted scientific concern. This study developed a microalgae fuel cells (MFCs) with Chlorella sp. as the cathode synergistically enhanced the removal of imidacloprid (IMI). The MFCs were operated at open-circuit and closed-circuit. Under closed-circuit condition (with an external load), 57.20–61.66% of IMI (10–50 mg/L) was removed within 12 days. However, only 39.66–42.66% was removed in the open-circuit (without an external load). Additionally, a voltage of 200 mV was also detected in the closed-circuit MFCs, indicating the green bioenergy was produced during the degradation of IMI. Cyclic voltammetry revealed that the cathodic electroactivity was higher with the addition of IMI. Biomass accumulation (indicated by chlorophyll and dry cell weight) were significantly enhanced by IMI. On the 12th day, there was no difference in MDA content between the different IMI concentration (10–50 mg/L), while a significantly upregulated superoxide dismutase (SOD) of each group was also found at the same time. Six intermediates were detected and five major degradation pathways were proposed. Based on the identified products and microalgae growth, the degradation products of IMI were less toxic /non-toxic than IMI. Considering the efficient removal of IMI and energy production, the MFCs could be a promising technique for controlling IMI contamination by effective detoxification.

Electro-flotation of Chlorella sp. assisted with flocculation by chitosan

Wenjun Zhou,Lili Gao,Wentao Cheng,Lin Chen,Junfeng Wang,Hui Wang,Wei Zhang,Tianzhong Liu

Harvest of algal biomass from dilute culture medium at low cost remains a major hurdle to industrial scale processing. Conventional harvesting methods are not only costly, but also affect any later downstream processes partly because of the metal ions contamination by flocculants . This work constructed an electro-flotation technique including two steps of flocculation by chitosan first and then sequential bubbling flotation by non-sacrificial graphite electrodes . Laboratory work has been carried out to investigate the influences of chitosan dosage, pH value, cell density of algal culture, stirring rate and mixing time on flocculation efficiency, and the voltage and current intensity imposed on electrodes and the space between electrodes on flotation efficiency. Moreover, a 1000   L pilot bench of electro-flotator constructed to harvest Chlorella sp. was tested. Around 90% of total recovery efficiency and 50 times concentration rate have been reached, and only 23.7   g chitosan and 0.43   kWh of electricity are required to harvest 1   kg biomass. Above results proved electro-flotation by graphite electrodes assisted with chitosan is a safe and cost effective approach for microalgae harvesting.

The potential of Co-cultivation of plant growth promoting bacteria (PGPB) and Chlorella sp. LZ-29 to enhance the biomass and protein content of Chlorella sp. LZ-29

Zhilai Lu,Zhe Luo,Min Liao,Xiaomei Xie,Mingdi Fan,Yue Xiao,Yunqiang Zhu

Two plant growth promoting bacteria (PGPB) were isolated. • PGPB could significantly enhance the biomass and protein content of Chlorella. • PGPB could enhancing the nitrogen assimilation and cell division of Chlorella. • PGPB could inhibit the flow of carbon metabolism to fat. The co-cultivation of microalgae with bacteria that stimulate microalgae growth and metabolic changes is a promising strategy for improving microalgae yield and quality. In this study, two plant growth promoting bacteria (PGPB), Bacillus safensis BAI-6 (BAI-6) and Bacillus altitudinis FEN-9 (FEN-9), were isolated from a long-term open cultivation system of Chlorella sp. LZ-29. Both strains were confirmed to produce the growth-promoting phytohormone indole-3-acetic acid (IAA) and vitamin B12. The changes in biomass, protein, fat, polysaccharide, pigment and other active components of Chlorella sp. LZ-29 under co-cultivation with these strains were investigated. Compared with the axenic Chlorella culture, co-cultivation with BAI-6 or FEN-9 alone increased biomass by 46.57% and 58.75%, respectively, while the dual-strain co-culture achieved the highest biomass increase of 66.46%. Protein content increased by 7% with BAI-6, 2.56% with FEN-9, and 13.37% in the dual-strain co-culture. Conversely, fat content decreased by 3.05%, 3.67%, and 3.48%, respectively, while polysaccharide content also declined in all co-culture treatments ( p  < 0.05). The synergistic effect of BAI-6 and FEN-9 in dual co-culture resulted in significantly enhanced biomass and protein production ( p  < 0.01), likely due to increased secretion of IAA and vitamin B12, which promoted nitrogen assimilation and cell division, while inhibiting carbon flux towards lipid synthesis. These findings demonstrate that the dual-strain co-culture system has significant potential for enhancing both the yield and protein content of Chlorella sp. LZ-29.

Physiological responses of marine Chlorella sp. exposed to environmental levels of triphenyltin

Cao Zhihan,Li Ping,Ru Jinchuang,Cao Xuqian,Wang Xu,Liu Bin,Li Zhi-Hua

Triphenyltin (TPT) is a herbicide and antifouling agent that has been widely used. After TPT flows into water bodies, it will cause toxic effects on marine life. We evaluated the effect of environmental concentration level (0, 10, 100, and 200 ng/L) on the cell density, antioxidant capability, and photosynthesis-related genes in the marine Chlorella sp. The results showed that 10 and 100 ng/L TPT can promote the growth of marine Chlorella sp., 200 ng/L TPT can inhibit the growth of marine Chlorella sp., and the TPT toxicity was accumulative. The chlorophyll composition changed. The content of chlorophyll a in 100 ng/L and 200 ng/L groups was significantly higher than that in the control group ( p  < 0.05) in 13 days. The content of chlorophyll b in the 100 ng/L and 200 ng/L groups in 1 day and 13 days was significantly different from that in the control group ( p  < 0.05). The content of total chlorophyll in the 100 ng/L and 200 ng/L groups in 13 days was higher than that in the control group ( p  < 0.05). The 200 ng/L group began to suffer oxidative damage on the 12th day, and the pigment protein complex responded to oxidative damage through self-feedback regulation. On the 18th day, chld , cao , psy , rbcS , and rbcL genes were downregulated, and psbA gene was upregulated in the 10 ng/L and 100 ng/L groups, which may be a feedback regulation of self-oxidative damage. This paper analyzed toxicity of environmental levels of TPT to marine Chlorella sp., which provided new data support for the comprehensive evaluation of its marine ecological toxicity.

Metabolic Mechanism of Sulfadimethoxine Biodegradation by Chlorella sp. L38 and Phaeodactylum tricornutum MASCC-0025

Li Bing,Wu Di,Li Yan,Shi Yan,Wang Chenlin,Sun Jiasi,Song Chunfeng

Antibiotic resistance is one of the most important environmental challenges. Microalgae has been considered as a promising green media for environmental purification. In this work, sulfadimethoxine (SDM) biodegradation potential of Chlorella sp. L38 and Phaeodactylum tricornutum MASCC-0025 is investigated. Experimental results indicated that the tested freshwater and marine microalgae strains presented stress response to SDM addition. For Chlorella sp. L38, it has a good adaptability to SDM condition via antioxidant enzyme secretion (SOD, MDA, and CAT up to 23.27 U/mg, 21.99 μmol/g, and 0.31 nmol/min/mg) with removal rate around 88%. P. tricornutum MASCC-0025 exhibited 100% removal of 0.5 mg/L SDM. With increasing salinity (adding a certain amount of NaCl) of cultivation media, the removal rate of SDM by microalgae increased. Although its adaptive process was slower than Chlorella sp. L38, the salinity advantage would facilitate enzyme accumulation. It indicated that microalgae could be used to remove SDM from freshwater and marine environment via suitable microalgae strain screening.

Effects of environment-relevant concentrations of antibiotics on seawater Chlorella sp. biofilm in artificial mariculture effluent

Chong Yu,Chi Li,Ying Zhang,Xiang Du,Jing-Han Wang,Zhan-You Chi,Qian Zhang

Attached cultivation of microalgae using mariculture effluent could simultaneously realize cost-effective biomass production and effluent polishing, and the obtained microalgal biofilm could be readily applied to aquatic animals that feed directly on biofilms. However, the widespread existence of antibiotics in mariculture effluent may have inhibitory effects on microalgae. Existing studies regarding the influences of antibiotics on microalgae were overwhelmingly mostly conducted in suspended freshwater cultures, and the explored antibiotic concentrations were several orders of magnitudes higher than realist effluent, rendering limited guidance for practical applications. This study, therefore, studied three commonly observed antibiotics in mariculture effluent, i.e. sulfamethoxazole (SMX), tetracycline (TL), and clarithromycin (CLA), and explored their effects on attached seawater Chlorella sp. growth, nutrients removal, and antioxidative properties at environment-relevant concentrations (0.1–200 μg/L). Hormesis was shared among the three antibiotics, and highest biofilm density (14.6 g/m 2 ) was observed under 0.1 μg/L SMX. SMX displayed strongest chlorophyll impairment, while SMX, TL, and CLA respectively favored lipid (up to 42.4 %), protein (up to 39.6 %), and polysaccharides (up to 65.4 %) accumulation. Highest correlation between biofilm density and extracellular polymeric substances (EPS) contents as well as highest total nitrogen (TN) removal were observed under TL exposure, while total phosphorus (TP) removal among all groups met the strictest local discharge standard for mariculture effluent (TP < 0.2 mg/L). Antioxidative responses revealed least stress towards attached seawater Chlorella sp. under TL treatment, while best acclimation under SMX exposure. Potential coping mechanisms of this microalgae under each antibiotic exposure were proposed as well. This study expands the knowledge of microalgae biofilm under environment-relevant concentrations of antibiotics.

Effect of interactions between humic acid and cerium oxide nanoparticles on the toxicity to the Chlorella sp.

Yang Lei,Li Yucai,Li Xiaotong,Lu Haoqi,Wang Yuchao,Meng Hongyan,Ren Yongxiang,Lan Jun

With the wide application of nanomaterials, the concentration of nanomaterials in natural water continues to increase, which poses a severe threat to the water environment. However, the influence of organic matter and nanomaterials rich in natural water on the toxic effect of algae growth is still unclear. In this study, the effects of humic acid (HA) and nano-cerium oxide (nCeO 2 ) on the physiology and transcriptome of Chlorella sp. were analyzed, and the mechanism of the toxic effect of HA on Chlorella sp. under nCeO 2 stress was revealed. Under 20–200 mg/L nCeO 2 stress, the growth of Chlorella cells was inhibited and the highest inhibition rate reached 52% within 200 mg/L nCeO 2 . The Fv/Fm and ETRmax values of Chlorella sp. decreased from 0.490 and 24.45 (20 mg/L nCeO 2 ) to 0.488 and 23.4 (100 mg/L nCeO 2 ), respectively. Under the stimulation of nCeO 2 , the level of reactive oxygen species in algal cells was increased, accompanied by lipid peroxidation and membrane damage. However, the addition of HA at concentrations of 5–10 mg/L effectively alleviated the toxic effect of nCeO 2 on Chlorella sp. Transcriptome analysis showed that 10 mg/L HA could alleviate the cellular stress at 100 mg/L nCeO 2 on Chlorella sp. by regulating genes related to photosynthesis and metabolism pathways. Moreover, the downregulation of genes (e.g., Lhca1, Lhcb1, AOC3, and AOC2) indicated that HA reduced the level of oxidative stress in Chlorella sp. These findings offer novel insights of evaluating the ecotoxicity nCeO 2 and HA in natural water environment and their impact on Chlorella sp. Graphical abstract

Glucose as a mitigator of cadmium-induced toxicity in chlorella sp.: Insights from cell division and photosynthesis

Bocheng Su,Jianke Huang,Enci Ye,Xiwen Xue,Jindong Zhao,Zi Ye

Glucose increased Chlorella biomass by counteracting Cd inhibition. • Cd 2+ disrupts cell wall degradation and photosynthetic systems. • Glucose co-treatment restored the cell wall degradation and chlorophyll homeostasis. Cadmium (Cd) pollution is a growing environmental challenge worldwide, impairing cellular integrity and transcriptional regulation across species and undermining ecosystem stability. This study investigates the effects of Cd 2 + stress on Chlorella pyrenoidosa in terms of growth, cell division, and photosynthetic function, and evaluates the alleviating role of soluble organic carbon supplementation, using glucose as a representative source. Results indicate that Cd 2+ exposure markedly suppresses algal growth, disrupts photosynthetic systems, reduces chlorophyll content, diminishes key photosynthetic metrics, and interferes with cell wall degradation. Biomass and specific growth rate declined from 0.6 g L −1 and 0.49 d −1 in controls to 0.38 g L −1 and 0.42 d −1 under Cd 2+ stress, but recovered to 0.95 g L −1 and 0.61 d −1 with glucose addition. Glucose supplementation substantially counteracted these inhibitory effects, partially reviving photosynthetic activity through enhanced QA-to-QB electron transfer and stabilization of porphyrin metabolism. Transcriptomic profiling revealed that Cd 2+ downregulated genes involved in cellulose degradation while upregulating those associated with lignin biosynthesis; glucose treatment restored the expression of lignin-related genes to near-control levels. Collectively, our findings demonstrate that glucose alleviates Cd 2+ toxicity in C. pyrenoidosa by restoring photosynthetic competence and cell division capacity, thereby elucidating key metabolic pathways underlying heavy-metal detoxification. Evaluating the Single and Combined Effects of BMDM and PS Microplastics on Chlorella sp.: Physiological and Transcriptomic Insights

Jin Liu,Yankun Zhang,Fengyuan Chen,Dandan Duan,Xiaoping Diao

In the environment, the coexistence of microplastics (MPs) with other pollutants may either enhance or reduce the toxicity of MPs themselves or the co-occurring pollutants toward microalgae. This phenomenon is particularly notable when MPs interact with emerging pollutants, such as ultraviolet absorbers. This study investigates the single and combined exposure effects of ultraviolet absorber (Butyl methoxydibenzoylmethane, BMDM, 50 μg/L) and MPs (Polystyrene, PS, 10 mg/L, d = 1 μm) onChlorellasp. with a stress duration of 7 days. The results showed that cell density, chlorophyll a (Chla) concentration, and physical properties of cell surface integrity were higher in the combined stress group compared to the BMDM single stress group. Furthermore, transcriptome sequencing analysis revealed that the number of differentially expressed genes (DEGs) in the combined exposure group (885 DEGs) was lower than in the single exposure groups (BMDM: 1870 DEGs and PS: 9109 DEGs). Transcriptomic profiling indicated that individual stressors of BMDM and PS disrupted 113 and 123 pathways, respectively, predominantly associated with protein synthesis and energy metabolism. Conversely, combined exposure significantly enriched 86 pathways, including ribosome function and oxidative phosphorylation, thereby manifesting an antagonistic effect. This study provides new insights into the effects of BMDM and PS onChlorellasp. and offers valuable information for the risk assessment of multiple pollutants.
