Chlorin e6 conjugated chitosan as an efficient photoantimicrobial agent

Lin Yue,Meihong Zheng,Imran Mahmood Khan,Zhouping Wang

The development of antibacterial agents with high bacteria-binding capability and antibacterial efficiency is highly desirable. Herein, cationic polysaccharide chitosan (CS) was combined with photosensitizer Chlorin e6 (Ce6) to construct a novel photodynamic antibacterial agent (CS-Ce6 conjugates) for combating gram-positive bacteria Staphylococcus aureus (S. aureus) and gram-negative bacteria Escherichia coli (E. coli) . CS-Ce6 conjugates with different degrees of substitution (DS) were synthesized and characterized by a spectroscopic method and organic elemental analysis to understand the relationship between structure and antibacterial effect. CS-Ce6 conjugates revealed good reactive oxygen species (ROS) generation ability and photodynamic antibacterial effect. Meanwhile, they both were positively correlated with DS in the range of 4.81% ~ 11.56% resulting in stronger photodynamic antibacterial ability. These findings highlight that CS-Ce6 conjugates have the potential as an effective photodynamic bactericidal agent in the antibacterial field.

Glutathione-depleting polymer delivering chlorin e6 for enhancing photodynamic therapy

Shi-yin Wang,Guo Chen,Ji-feng Chen,Jin Wang,Shao-hui Deng,Du Cheng

The therapeutic effect of photodynamic therapy (PDT) is highly dependent on the intracellular production of reactive oxygen species (ROS). However, the ROS generated by photosensitizers can be consumed by the highly concentrated glutathione (GSH) in tumor cells, severely impairing the therapeutic effect of PDT. Herein, we synthesized a GSH-scavenging copolymer to deliver photosensitizer chlorin e6 (Ce6). The pyridyl disulfide groups, which have faster reactivity with the thiol groups of GSH than other disulfide groups, were grafted onto a hydrophobic block to encapsulate the Ce6. Under NIR irradiation, the Ce6 generated ROS to kill tumor cells, and the pyridyl disulfide groups depleted the GSH to prevent ROS consumption, which synergistically enhanced the therapeutic effect of PDT. In vitro and in vivo experiments confirmed the combinatory antitumor effect of Ce6-induced ROS generation and the pyridyl disulfide group-induced GSH depletion. Therefore, the pyridyl disulfide group-grafted amphiphilic copolymer provides a more efficient strategy for enhancing PDT and has promising potential for clinical application.

Photocatalytic degradation of ethylene by mesoporous nano silica loaded chlorin e6

Liangyu Jia,Weihao Jin,Weiwei Zhang,Jun Wang,Wanzhen Li,Yugui Tao

Green foods are usually harvested and stored before ripening, but excessive ethylene generated during storage can lead to the loss of vegetables and fruits. Photocatalytic oxidation provides a promising method to remove ethylene and extend the shelf life of green foods. In this study, a mesoporous nanosilica-loaded Ce6 composite nanomaterial (Ce6@SiO2) was designed and synthesized to adsorb ethylene into its pores and then degrade it under visible light illumination. The photosensitizer Ce6 in the mesopore produced large amounts of reactive oxygen species that degraded ethylene. The specific surface area was greatly increased by coating the material on the surface of a sodium alginate (SA) hydrogel membrane. This improved its degradation efficiency and facilitated the adsorption and degradation of ethylene during fruit storage. The ethylene removal capacity was studied by measuring the ethylene concentration and increases in the humidity in Tedlar gas bags. Practical applications were tested by observing color changes and surface decay of tomatoes stored with and without SA/Ce6@SiO2. The results showed the system may be used in practical commercial preservation systems.

Polyethylene glycol modified protamine-chlorin e6 conjugate nanoparticles for enhanced cancer photodynamic therapy

Hang Hu,Ting Yuan,Jiahao Qian,Xinya Lu,Xi Chen,Xiaoya Sun,Yuchen Lin,Yuxiang Tang,Qingbo Xu,Tianyu Zhu,Yu Hu,Defeng Xu

Photodynamic therapy attracts great attention in cancer treatment due to its minimal invasiveness, good tolerance and specific cancer targeting. However, most of tumors are hypoxic and the tumors will become more hypoxic after photodynamic therapy, which might cause the poor prognosis by inducing tumor angiogenesis. In the present work, polyethylene glycol modified protamine-chlorin e6 conjugate (PEG-protamine-Ce6) was synthesized for enhanced cancer photodynamic therapy. PEG-protamine-Ce6 was synthesized by conjugating protamine with methoxypolyethylene glycol acetaldehyde and Ce6 sequentially. The in vitro studies show that PEG-protamine-Ce6 self-assembles into nanoparticles in aqueous solution with uniform size (∼200 nm), positive zeta potential and spherical morphology. The in vitro cell studies show that PEG-protamine-Ce6 nanoparticles exhibit enhanced cellular uptake amount, intracellular ROS level elevation upon laser irradiation, cell apoptosis and photocytotoxicity as compared to Ce6. The in vivo studies show that PEG-protamine-Ce6 nanoparticles exhibit enhanced antitumor effect as compared to Ce6 upon laser irradiation. PEG-protamine-Ce6 with laser irradiation not only inhibits tumor cell proliferation, but also inhibits tumor angiogenesis by suppression of VEGF expression, whereas Ce6 with laser irradiation promotes tumor angiogenesis by enhancing VEGF expression. PEG-protamine-Ce6 nanoparticles developed in this work offer a novel nanomedicine for enhancing cancer photodynamic therapy.

Highly Biocompatible Chlorin e6-Loaded Chitosan Nanoparticles for Improved Photodynamic Cancer Therapy

Yuan-Fu Ding,Shengke Li,Lijun Liang,Qiaoxian Huang,Lihui Yuwen,Wenjing Yang,Ruibing Wang,Lian-Hui Wang

The photosensitizer Chlorin e6 (Ce6) has been frequently employed for photodynamic therapy (PDT) of cancer; however, its nonspecific toxicity has limited its clinical applications. In this study, we prepared chitosan nanoparticles (CNPs), with a mean diameter of approximately 130 nm, by a nonsolvent-aided counterion complexation method in an aqueous solution, into which Ce6 could be physically entrapped during the preparation process. These CNPs and Ce6-loaded CNPs (CNPs-Ce6) were fully characterized by UV–vis, photoluminescence, and Fourier transform infrared spectroscopic analysis, as well as dynamic light scattering and transmission electron microscopy measurements. More importantly, the biocompatibility of the otherwise toxic Ce6 was significantly improved upon its loading into the CNPs, as demonstrated by both confocal laser scanning microscopy analysis and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays. Furthermore, the PDT efficiency of Ce6-loaded CNPs was dramatically enhanced, in comparison with that of the free Ce6, as shown by both MTT and flow cytometry assays. This discovery provides a novel strategy for improving the biocompatibility and therapeutic efficacy of PDT agents by using a natural, biocompatible polysaccharide carrier.

Foamed microemulsion nanodroplets loaded with chlorin e6 for epidermal-targeted treatment against psoriasis

Ma Xiaolu,Bian Qiong,Xu Yihua,Hu Jingyi,Hu Weitong,Wang Ruxuan,Zhang Yunting,Ye Yuxian,Sheng Xiaoxia,Zhang Tianyuan,Gao Jianqing

Psoriasis is a chronic skin disease characterized by the hyperproliferation of keratinocytes and an overactive autoimmune response. Photodynamic therapy (PDT) has been established as a promising intervention for alleviating psoriasis. However, the current transdermal delivery of photosensitizers is inefficient and imprecise. In this study, we developed a foamed microemulsion nanodroplets system containing chlorin e6 (Ce6 FM), exhibiting precise epidermal targeting and retention, which targeted the aberrantly proliferating epidermal cells at psoriatic skin lesions and avoided the damage to the normal cutaneous cells. Upon application in a psoriatic mouse model, Ce6 FM efficiently induced keratinocyte apoptosis by generating reactive oxygen species under laser. Furthermore, Ce6 FM-based PDT activated the cyclooxygenase-2-induced immunosuppressive pathway in keratinocytes, resulting in the amelioration of the autoimmune microenvironment in psoriatic skin. Additionally, Ce6 FM-based PDT did not induce skin damage or atrophy associated with non-targeted halometasone treatment. Overall, Ce6 FM-based PDT holds promise as an effective, safe and compliant strategy for psoriasis treatment.

Perfluoropolyether Nanoemulsion Encapsulating Chlorin e6 for Sonodynamic and Photodynamic Therapy of Hypoxic Tumor

Liang Hong,Artem M. Pliss,Ye Zhan,Wenhan Zheng,Jun Xia,Liwei Liu,Junle Qu,Paras N. Prasad

Sonodynamic therapy (SDT) has emerged as an important modality for cancer treatment. SDT utilizes ultrasound excitation, which overcomes the limitations of light penetration in deep tumors, as encountered by photodynamic therapy (PDT) which uses optical excitations. A comparative study of these modalities using the same sensitizer drug can provide an assessment of their effects. However, the efficiency of SDT and PDT is low in a hypoxic tumor environment, which limits their applications. In this study, we report a hierarchical nanoformulation which contains a Food and Drug Administration (FDA) approved sensitizer chlorin, e6, and a uniquely stable high loading capacity oxygen carrier, perfluoropolyether. This oxygen carrier possesses no measurable cytotoxicity. It delivers oxygen to overcome hypoxia, and at the same time, boosts the efficiency of both SDT and PDT. Moreover, we comparatively analyzed the efficiency of SDT and PDT for tumor treatment throughout the depth of the tissue. Our study demonstrates that the strengths of PDT and SDT could be combined into a single multifunctional nanoplatform, which works well in the hypoxia environment and overcomes the limitations of each modality. The combination of deep tissue penetration by ultrasound and high spatial activation by light for selective treatment of single cells will significantly enhance the scope for therapeutic applications. Keywords:photodynamic;sonodynamic;hypoxia;nanoemulsion;delivery;tumor

Versatile Chlorin e6-based magnetic polydopamine nanoparticles for effectively capturing and killing MRSA

Chunbo Lu,Feifei Sun,Yinyin Liu,Yao Xiao,Yuanhao Qiu,Haibo Mu,Jinyou Duan

Bacterial infections are a growing global challenge for public health as antibiotic resistance could cause the failure of anti-infective treatment eventually. So, it is urgent to develop new potential antibacterial materials. Herein, a multifunctional chitosan (CS) functionalized magnetic Chlorin e6 (CS-MP-Ce6) was constructed to combat methicillin-resistant Staphylococcus aureus (MRSA) infection by integrating bacterial conjugation and enrichment, and near-infrared (NIR)-triggered photodynamic sterilization. CS-MP-Ce6 could efficiently capture bacteria due to positively charged property of CS, and Ce6 acted as an effective photodynamic killer to convert NIR light into local energy to enhance antibacterial activity. Specifically, after being trapped together with MRSA, CS-MP-Ce6 showed an excellent in vitro photodynamic sterilization ability. In vivo MRSA-induced abscess treatment studies showed faster healing when CS-MP-Ce6 was used as subcutaneous nano-localized energy sources with the assistance of external magnet to concentrate CS-MP-Ce6–bacteria conjugate. This work provides a promising framework for constructing a new system for efficiently combating MRSA.

Chlorin e6-modified iron oxide nanoparticles for photothermal-photodynamic ablation of glioblastoma cells

Hongqing Yao,Jian-Ying Zhou

Introduction: The effective treatment of glioblastoma still remains a great challenge. We herein report the development of chlorin e6 (Ce6)-conjugated iron oxide (Fe 3 O 4 -Ce6) nanoparticles for ablation of glioblastoma cells via combining photothermal therapy (PTT) with photodynamic therapy (PDT). Methods: Ce6 was conjugated to the synthesized Fe 3 O 4 nanoparticles to form Fe 3 O 4 -Ce6 nanoparticles displaying the optical property of Ce6. Results and discussion: Under 808 nm laser irradiation, Fe 3 O 4 -Ce6 nanoparticles generated heat and the temperature increase did not have obvious changes after five cycles of laser irradiation, suggesting their good photothermal effect and photothermal stability. In addition, 660 nm laser irradiation of Fe 3 O 4 -Ce6 nanoparticles produced singlet oxygen ( 1 O 2 ) to mediate PDT. The Fe 3 O 4 -Ce6 nanoparticles without laser irradiation showed a low cytotoxicity, but they would obviously kill C6 cancer cells after laser irradiation via the combinational effect of PTT and PDT. Fe 3 O 4 -Ce6 nanoparticles thus could be used as a nanotherapeutic agent for combinational ablation of glioblastoma cells.

Imaging-Guided and Light-Triggered Chemo-/Photodynamic/Photothermal Therapy Based on Gd (III) Chelated Mesoporous Silica Hybrid Spheres

Dan Yang,Guixin Yang,Shili Gai,Fei He,Ruichan Lv,Yunlu Dai,Piaoping Yang

Exploring a combined anticancer therapeutic strategy to overcome the limitations of a single mode and pursue higher therapeutic efficiency is highly promising in both fundamental and clinical investigations. Herein, a theranostic nanoplatform based on mesoporous silica, which is functionalized by hybrid nanosphere photosensitizer Chlorin e6 (Ce6), photothermal agent carbon dots (CDs), and imaging agent Gd (III) ions has been rationally designed and fabricated. A thermo/pH-coupling sensitive polymer (P(NIPAm-co-MAA)) coated on a composite acted as a key “gatekeeper” to control drug release at the appropriate time and location. Upon light irradiation, two-mode synergistic therapeutic effect of photodynamic and photothermal therapy can be achieved by photoactive Ce6 and CDs. Meanwhile, the CDs loaded in the channels of mesoporous silica hybrid spheres can also play a role in handling the “gatekeeper” polymer to control the drug release process. Combined with the thermo/pH-sensitive drug release-induced controllable chemotherapy, this platform shows synergistic therapeutic efficacy better than any single/dual therapy, which is confirmed with evidence from in vivo and in vitro assays. Considering the chelated Gd3+ simultaneously introduced magnetic resonance imaging (MRI) and computed tomography (CT) properties, this multifunctional platform should have excellent potential in the imaging-guided cancer therapy field.

Nanobody–chlorin e6 conjugate for Nectin-4-mediated tumor targeting and enhanced photodynamic therapy

Haodian Wan,Tao Ye,Yawei Bi,Jiayu Zhang,Wei Wu,Aochi Liu,Li Sun,Jie Lin,Ningli Chai,Manjie Zhang,Jian Ma

Photodynamic therapy (PDT) holds promise for tumor ablation, but the clinical translation of photosensitizers, such as chlorin e6 (Ce6), is limited by poor aqueous solubility, low bioavailability, and insufficient tumor selectivity. To address these limitations, we constructed a nanobody–photosensitizer conjugate (NPC) by covalently linking Ce6 to a variable heavy-chain domain targeting Nectin-4, a clinically relevant tumor biomarker. Cell-based assays demonstrated that NPC exhibited markedly enhanced aqueous solubility, efficient cellular uptake, and strong Nectin-4-dependent binding compared with free Ce6. Upon light activation, NPC generated abundant reactive oxygen species (ROS), triggered lysosomal disruption, and induced apoptosis and immunogenic cell death. In xenograft mouse models, NPC displayed prolonged circulation, rapid and selective tumor-specific accumulation, and potent PDT to inhibit tumor growth, whereas free Ce6 displayed weak tumor retention and limited therapeutic benefit. Importantly, NPC treatment caused no measurable systemic toxicity or body weight loss during repeated administration. Collectively, these findings demonstrate that nanobody conjugation can overcome key pharmacological limitations of conventional photosensitizers by improving solubility, tumor targeting, and therapeutic index. NPC, thus, represents a promising next-generation PDT agent with potential for tumor-selective and clinically translatable phototherapy.

The conjugates of carbon nanodots and chlorin e6 for enhancing cellular internalization and photodynamic therapy of cancers

Jing Wang,Xiongwei Wang,Shimiao Wang,Zheng Huang,Jun Liu

Chlorin e6 (Ce6), a large heterocyclic aromatic molecule, is a promising photosensitizer for photodynamic therapy (PDT). We propose an efficient nano-photosensitizer delivery system based on covalent interactions between Ce6 and polyethylenimine (PEI) coated carbon nanodots (CDots). We observed  >50% Ce6 drug loading content for PEI, due to this compound's unique 'proton sponge effect.' We found that the covalently incorporated Ce6 molecules retained their functional properties for near-infrared (NIR) fluorescence imaging and PDT. The chemical characteristics of CDot-PEI-Ce6 and Ce6 were evaluated using different analytical methods, including transmission electron microscopy and UV–Visible absorption spectra. Time-correlated single photon counting (TCSPC) and fluorescence spectra were used to demonstrate that Ce6 successfully conjugated to the CDots. The high cellular uptake of CDots-PEI-Ce6 was confirmed using flow cytometry and confocal laser scanning microscopy. According to the MTT assay, the CDots-PEI-Ce6 exhibited low dark toxicity and efficient PDT efficacy to HeLa cancer cells. These results indicate that CDot-PEI-Ce6 conjugates are potential photosensitizer delivery systems for PDT.
