Objective: A comprehensive monosaccharide quantifying method was developed in this article. Monosaccharides (including glucose, arabinose, galacturonic acid, galactose, mannose, rhamnose and xylose) were analyzed by RP-HPLC-DAD method. Meanwhile, mannitol and fructose were calculated
based on peak areas obtained by HPIEC-RID and corresponding data obtained by RP-HPLC-DAD. Monosaccharide compositions of CDSC were analyzed, nine kinds of monosacchardes (glucose, arabinose, galctose, galacturonic acid, mannose, rhamnose, xylose, fructose and mannitol) in were quantified.
Background: Cistanche deserticola Y. C. Ma, is a perennial holoparasite and mainly distributed in the desert region of northwestern China. It reported that C. deserticola soluble carbohydrate (CDSC) could prompt lymphocyte proliferation, modulate immunity, inhibit type 2 diabetes, anti-inflammation.
For these reasons, it is very appealing to researchers to have a convenient method for monosaccharides analysis of the CDSC. However, previous researches have neglected the faultiness in monosaccharide composition analysis. Therefore, in this study we will focus on establishing a quantifying
method of monosaccharides possibly found in C. deserticola. Methods: Soluble carbohydrates were hydrolyzed with trifluoroacetic acid, monosaccharides were labeled with 1-phenyl-3-methyl-5-pyrazolone (PMP), separated by HPLC system equipped with C18 column and detected with DAD (RP-HPLC-DAD).
RP-HPLC-DAD method was used for quantitative analysis of glucose, arabinose, galacturonic acid, galactose, mannose, rhamnose and xylose. Meanwhile, hydrolyzates were separated by HPIEC system equipped with aminex HPX-87H ion exclusion column and detected by refractive index detector (HPIEC-RID).
The quantity of mannitol and fructose can be calculated based on peak areas obtained by HPIEC-RID and corresponding data obtained by RP-HPLC-DAD. Results: Utilizing this method, monosaccharide compositions of CDSC with different molecular weight (CDCSA, CDCS-B, CDCS-C and CDCS-D) were
analyzed, nine kinds of monosacchardes (glucose, arabinose, galctose, galacturonic acid, mannose, rhamnose, xylose, fructose and mannitol) in were found. Conclusion: Fructose and mannitol, which are abundant present in C. deserticola, cannot be analysis by normally PMP modify method,
therefore, a comprehensive quantitative analysis method was developed in this research. Galacturonic acid was found out in macro-molecular polysaccharides from C.deserticola, which was not reported before. Galactose and galacturonic acid mainly exist in macro-molecular polysaccharide. Fructose
and mannitol mainly exist in mini-molecule or exist in free form.

The aim of this study was to optimize the ultrasound-assisted extraction (UAE) process of Phenylethanoid Glycosides (PhGs) from Cistanche deserticola and to elucidate the mechanism of bioactivity enhancement from a metabolomic perspective. A sequential optimization strategy was employed, combining single-factor experiments, Plackett-Burman design (PBD) to screen for significant variables, and Box-Behnken design (BBD) for response surface methodology (RSM). The optimal extraction parameters, which were the liquid-solid ratio of 14:1 v/w, the ultrasonic power of 215 W, and the ethanol concentration of 55%, which resulted in the yield of PhGs of 122.51±5.07 mg/g. Metabolomics analysis by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-ESI-MS/MS) revealed that the content of phenolic acids, flavonoids and terpenoids in the ultrasound-assisted ethanol extract (EEC) was significantly higher than that of the traditional solvent-extracted ethanol extract (ESC) ( p < 0.05), and the antioxidant capacity (DPPH IC 50 =57.79 μg/mL, ABTS IC 50 =51.56 μg/mL, FRAP = 0.74 mg FeSO 4 /g) were significantly better than that of ESC. Principal component analysis (PCA) confirmed a strong correlation between the enrichment of these bioactive constituents and the enhanced functional activities. This study reveals for the first time that ultrasound-assisted extraction enhances the antioxidant and anti-inflammatory potentials of C. deserticola extracts through selective enrichment of phenolic acids, flavonoids, and terpenoids compared to conventional extraction by metabolomics, which provides a theoretical basis for the highly efficient extraction of active ingredients of plant origin and the application of C. deserticola in the development of functional foods.

Carbohydrate microarray enabled rapid profiling of CDPs glycan epitopes. • Glucan-related epitopes were identified as the main detectable signals in CDPs. • Conventional structural analyses supported the microarray findings. To address the technical bottlenecks of conventional structural analysis for Cistanche deserticola polysaccharides (CDPs), including cumbersome operation, lengthy detection cycles and low throughput in structural screening, this study introduced carbohydrate microarray technology as a rapid epitope-profiling tool for the structural characterization of CDPs. With C. deserticola as the raw material, we prepared polysaccharide samples via six extraction methods and constructed a tailored carbohydrate microarray system. Specific carbohydrate-binding proteins (BS-400-2, BS-400-3, LM18, LM19, and LM21) were adopted to achieve high-throughput profiling of probe-recognizable polysaccharide epitope features. The reliability and applicability of this technology were evaluated and supported by determining polysaccharide yield, total sugar content, molecular weight ( M w) and monosaccharide composition using conventional analytical methods. Under the established carbohydrate microarray assay conditions, CDPs mainly exhibited BS-400-2-recognizable glucan-related epitope features, whereas obvious binding signals for the selected pectin- and mannan-related epitopes were not observed. Extraction processes markedly influenced the yield and structural characteristics of polysaccharides: ultrasonication-assisted alkaline extraction (UAAIE) achieved the highest polysaccharide yield (13.43%), while microwave-assisted extraction (MAE) produced polysaccharides with the highest total sugar content (85.72%) and M w (7.819 × 10 6 Da). The microarray results were generally consistent with those obtained by conventional methods, supporting the reliability of the carbohydrate microarray-based epitope profiling strategy. Furthermore, the binding performance and specificity of selected probes were examined using structurally defined homogeneous polysaccharides for system optimization, which supports the extended application of this method in other TCM polysaccharides. This rapid carbohydrate microarray system establishes a high-throughput and microscale approach for epitope-level characterization of CDPs, and lays an experimental foundation for the standardized application of carbohydrate microarrays in TCM polysaccharide research. The multi-target strategy can be further extended to other TCM polysaccharides, facilitating subsequent studies on their structure-activity relationships and high-efficiency bioactivity screening. Microbial fermentation is an effective method to enhance the bioavailability of herbs. This study utilized Lactiplantibacillus plantarum LZU-J-Q21 to ferment Cistanche deserticola and evaluated its metabolic properties and biological activity. Results showed that the contents of total acid and flavone, and the clearance rates of DPPH, ABTS and OH − in fermented Cistanche deserticola (FCD) were increased by 142.74 %, 56.45 %, 58.1 %, 62.3 %,51.2 %, compared with non-fermented Cistanche deserticola (NFCD). The metabolic profile of FCD had remarkable changes, especially elevated glucose and adenosine (97.31 % and 59.18 %). Further, FCD increased the weight-bearing swimming time of mice by 88.57 %, reduced fatigue markers BUN, BLA, and MDA (18.47 %, 12.92 %, and 15.16 %), and enhanced liver/muscle glycogen and SOD (28.99 %, 28.57 %, and 14.47 %). The investigation into its anti-fatigue mechanism suggested that FCD enhanced GS protein expression by activating PI3K/AKT/GSK3β signaling. These findings suggest that FCD enhances anti-fatigue effects by modifying its metabolic properties and biological activity.

Background Studies have shown that phenylethanoid glycosides (PhGs) have multiple pharmacological effects such as anti-inflammatory, hepatoprotective or neuroprotective functions, whereas their anti-tumor effects are rarely studied. Tubuloside B (Tub B) is a PhG isolated from Cistanche deserticola , a traditional Chinese medicine. To date, there is a lack of comprehensive research regarding the biological activity of Tub B. Purpose The subject of the current study was to investigate the anti-hepatocellular carcinoma (HCC) cell activity and the underlying mechanism of Tub B. Methods We evaluated the in vitro anti-migratory effect of Tub B by scratch and transwell assays. RNA-seq was employed to identify the differential genes by Tub B. Besides, the functional mechanism of Tub B was investigated by distinct molecular biology techniques including immunofluorescent staining, quantitative PCR, as well as western blot analysis. Subsequently, we utilized Hep3B cells for in vivo metastasis assays through spleen injection and evaluated the anti-migratory effect of Tub B in hepatocellular carcinoma (HCC). Results Tub B exhibited in vitro and in vivo inhibition of HCC cell migration. Tub B decreased the expression of transcriptional target genes downstream of the Hippo pathway, including CTGF, CYR61, and N-cadherin as determined by RNA-seq. Furthermore, mechanistic studies confirmed that Tub B increased phosphorylation of YAP at S127, which contributes to YAP cytoplasmic localization. Additionally, overexpression of YAP abrogated Tub B-induced inhibition of HCC migration and the mRNA levels of CTGF, CYR61, and N-cadherin . Conclusions Taken together, these results illustrated that Tub B demonstrated great potential in inhibiting migration of HCC, and a portion of its impact can be attributed to the modulation of the Hippo-YAP pathway.

The primary objective of this study is to investigate the potential mechanism behind the protective effect of Cistanche deserticola polysaccharides (CP) against alcoholic liver disease (ALD). Multiple chromography techniques were employed to characterize CP from polysaccharide, the molecular weight distribution of polysaccharides, monosaccharide composition, isomeric hydrogen and isomeric carbon, in order to clarify the material basis of CP. To create the ALD mouse model, we utilized the well-established Lieber-DeCarli alcoholic liquid feed method. Findings from the study revealed that CP administration resulted in significant improvements in intestinal permeability, upregulation of barrier proteins expression , and reduced levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in mouse liver and serum. Additionally, CP treatment reduced the presence of inflammatory cytokines both in serum and liver while enhancing the activity of antioxidant enzymes in the liver. Furthermore, CP effectively reduced alcohol-induced oxidative damage by downregulating Keap1 protein levels in the liver, leading to increased expression of Nrf2 protein . The 16S rDNA sequencing results revealed that CP significantly restored the intestinal microbiota composition in ALD mice. These findings establish a strong association between gut microbiota and liver injury indicators, highlighting the potential of CP in preventing and treating ALD by modulating the gut-liver axis.

Cistanche deserticola (Orobanchaceae) has been widely used in China for food and medicinal purposes. Chemical differentiation between the raw and steamed C. deserticola was performed by high-performance liquid chromatography coupled with diode array detection and mass spectrometry. Eight chemicals (echinacoside, cistanoside A, acteoside, cistanoside C, 2′-acetylacteoside, isoacteoside, isocistanoside C, and tubuloside B) were obtained from raw and steamed C. deserticola, and the kinetics in the steaming process were investigated in detail. When the steaming time increased, the concentrations of echinacoside, cistanoside A, acteoside, cistanoside C, and 2′-acetylacteoside decreased, while the levels of isoacteoside, isocistanoside C, and tubuloside B increased. Furthermore, two compounds, 5-hydroxymaltol and 5-hydroxymethylfurfural, were found only in the steamed form. In addition, hierarchical clustering analysis and principal component analysis were performed to evaluate the classify the chemical concentrations among steamed C. deserticola. The results showed that steaming had a significant influence on the chemical constitution. The study provide chemical analysis of raw and steamed C. deserticola, including possible transformation pathways, and should allow better understanding of the steaming process of this herb.

A creative dual-template molecularly imprinted polymer (Fe 3 O 4 @MCM-41-MIPs) with good magnetic properties and specific recognition of echinacoside and acteoside was prepared by surface imprinting. Echinacoside and acteoside served as dual templates, 4-vinylpyridine (4-VP) was used as a functional monomer , the cross-linker was ethylene glycol dimethacrylate (EGDMA), and the selected initiator was azobisisobutyronitrile (AIBN). The prepared Fe 3 O 4 @MCM-41-MIPs were applied to the selective separation and enrichment of echinacoside and acteoside from Cistanche deserticola Y. C. Ma ( C. deserticola ). The resulting Fe 3 O 4 @MCM-41-MIPs were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR), laser particle size scanning and vibration sample magnetometry (VSM). The results of the isotherm adsorption experiment showed that the maximum adsorption capacities of Fe 3 O 4 @MCM-41-MIPs for echinacoside and acteoside were 6.02 and 1.63 mg·g -1 , respectively, at 298 K. The pseudo-second-order reaction mechanism showed that the equilibrium adsorption capacities of Fe 3 O 4 @MCM-41-MIPs for echinacoside and acteoside were 4.81 and 0.93 mg·g -1 , respectively. The characterization of Fe 3 O 4 @MCM-41-MIPs showed that the imprinted layer with dual-template molecules was wrapped on the outside of the carrier, and the obtained Fe 3 O 4 @MCM-41-MIPs showed good saturation magnetization , desirable capacity for echinacoside and acteoside adsorption, and satisfactory reuse (echinacoside and acteoside maintained relative adsorption rates of 97.5% and 95.8% after five cycles). After separation, the purities of echinacoside and acteoside were 33.50% and 27.00%, respectively. In addition, in the selective adsorption experiment, the synthesized Fe 3 O 4 @MCM-41-MIPs showed better selectivity for echinacoside and acteoside than their structural analogs. Therefore, this method proved to be sustainable, rapid and highly efficient for the separation and enrichment of echinacoside and acteoside.

Ethnopharmacological relevance Cistanche deserticola Ma (Orobanchaceae), a widely used botanical drug in Traditional Chinese Medicine, is traditionally utilized to moisten the intestines and relieve constipation, particularly for senile and deficiency-induced constipation. Aim of the study To evaluate the restorative effects of C. deserticola (CD) aqueous extract on functional constipation (FC) and elucidate its multi-omic mechanisms mediated by the gut microbiota-short-chain fatty acid (SCFA)-host axis. Materials and methods The CD extract was phytochemically characterized via UPLC-Q-TOF-MS/MS. Loperamide induced FC mice were treated with physiologically relevant doses of CD extract (369 and 615 mg/kg/day). Efficacy was assessed via phenotypic, biochemical, and histopathological evaluations. Mechanisms were explored integrating 16S rRNA sequencing, targeted SCFA metabolomics, and colonic transcriptomics. Fecal microbiota transplantation (FMT) was conducted to verify microbiota dependency. Key signaling networks were validated using Western blotting and immunofluorescence. Results CD administration significantly accelerated intestinal transit, repaired mucosal barriers, restored gastrointestinal hormones, and mitigated oxidative and inflammatory stress. Microbiome profiling demonstrated CD reversed dysbiosis by enriching beneficial SCFA-producers (e.g., Lachnospiraceae NK4A136 group), correspondingly elevating colonic propionate and butyrate. FMT confirmed these therapeutic effects were microbiota-driven. Transcriptomic and molecular validations revealed that CD-mediated microecological restoration reactivated the Integrin αL/p-FAK/Filamin C mechanotransduction cascade and PDGFC/SGK1 signaling pathways, thereby promoting focal adhesion and structural mucosal repair. Conclusions CD alleviates FC by concurrently remodeling the gut microbiota, enhancing SCFA production, and reactivating colonic mechanotransductive tissue repair networks. This provides a robust scientific rationale for its traditional ethnopharmacological use in constipation management.

As a well-known Chinese herb medicine, the Cistanche deserticola has been used for the treatment of kidney deficiency syndrome in China for thousands of years. Both the raw product of Cistanche deserticola slices (RCD) and its Wine Steam-Processed Product (WSCD) are used clinically for different effects. In this study, the influences of steaming process with wine (SPW) from Cistanche deserticola on chemical compositions and biological effects were investigated. The principal component analysis (PCA) and quantitative analysis were used to study the differences of the chemical compositions. The effects of nourishing kidney were also investigated to compare the differences between the RCD and the WSCD. The PCA results indicated that the obvious separation was achieved in the RCD and WSCD. The results of quantitative analysis showed that the WSCD has higher amounts of total polysaccharides, total PhGs, isoacteoside, and osmanthuside B than RCD, while the content of 2′-acetylacteoside and acteoside decreased after the SPW. The comparison of RCD and WSCD on biological activities showed that both could restore the level of sex hormone in the model of kidney-yang deficiency and improve the antioxidant effect. The WSCD were much better in increasing the viscera weight of kidney and seminal vesicle. The results indicated that SPW changed its chemical components and enhanced its biological activities.

Introduction Cistanche deserticola Ma (CD), an edible and medicinal plant native to Xinjiang, Inner Mongolia, and Gansu in China, is rich in bioactive polysaccharides known for their health-promoting properties. The polysaccharides of C. deserticola (CDPs) have been shown to possess a range of beneficial activities, including immunomodulatory, anti-aging, antioxidant, and anti-osteoporosis effects. Objective This study seeks to identify the optimal conditions for extracting CDPs using hot water. Additionally, it aims to evaluate their chemical properties, antioxidant activity, hypoglycemic effects, and cytotoxicity. The findings will provide a theoretical foundation for the potential use of CDPs in functional foods and pharmaceuticals. Methodology The study employed response surface methodology to optimize the hot water extraction conditions for CDPs. The extracted CDPs were characterized using a range of chemical, spectroscopic, and instrumental analyses. Furthermore, their antioxidant activity, hypoglycemic effects, and cytotoxicity were evaluated through relevant assays to assess their potential health benefits. Results Under optimal conditions, the yield of CDPs was 45.85% ± 1.91%. CDPs were identified as acidic heteropolysaccharides with a wide molecular weight distribution, ranging from 0.3 to 128.2 kDa. They were composed primarily of glucose (51.21%), arabinose (32.86%), galactose (17.88%), and smaller amounts of galacturonic acid (4.66%), rhamnose (1.85%), mannose (1.32%), glucosamine hydrochloride (1.08%), and xylose (0.56%). Antioxidant assays demonstrated that CDPs exhibited significant free radical scavenging activity, metal ion chelation, and reducing power. Additionally, CDPs inhibited α-glucosidase and α-amylase in vitro through a mixed-type mechanism, as well as static fluorescence quenching. Cytotoxicity assays showed that CDPs were nontoxic to L02 and AML12 cells. Conclusion This study offers a theoretical foundation for the potential use of CDPs in functional foods and pharmaceuticals and provides valuable insights for the development of new antioxidant and hypoglycemic agents from natural sources.

Cistanches Herba is a famous traditional Chinese medicine that has been in use for treating kidney deficiency, impotence, female infertility, morbid leucorrhea, profuse metrorrhagia, and senile constipation. With the exception of studies available for a few single active ingredients such as echinacoside, acteoside, and poliumoside, comprehensive and systematic studies on in vivo metabolism of Cistanches Herba are lacking despite its widespread clinical application. There is no comparative study yet on the metabolites resulting from the traditional usage of Cistanche deserticola and C. tubulosa water extract – two species that are recorded in Chinese Pharmacopoeia. This further restricts research on the therapeutic effect of Cistanches Herba to a great extent. In this study, a robust and unbiased UPLC-Q-TOF-MS combined pattern recognition analysis (orthogonal partial least squared discriminant analysis, OPLS-DA) was employed to rapidly screen prototype components and metabolites of C. deserticola and C. tubulosa water extract in rat urine, feces, and serum. A total of 71 metabolites from C. deserticola including 25 prototype components and 46 metabolites, and 45 metabolites from C. tubulosa including 18 prototype components and 27 metabolites were tentatively identified. Out of these, 10 metabolites were characterized for the first time in these two species. Results of this study indicate that phenylethanoid glycosides (PhGs) are mainly metabolized into degradation products in the gastrointestinal tract of rats. The chemical components cistanoside B, C, D, and E exist only in C. deserticola and release methylated hydroxytyrosol (HT) following metabolism. This factor contributes to the difference between metabolites of C. deserticola and C. tubulosa water extract in rats and is responsible for the differential therapeutic effect that these two species of Cistanches Herba have on the same diseases.
