Correlation between Chemical Composition and Antifungal Activity of Clausena lansium Essential Oil against Candida spp.

Xiaowen He,Lantong Zhang,Jinping Chen,Jinlei Sui,Guohui Yi,Jinyan Wu,Yinzheng Ma

Essential oils (EOs) have been shown to have a diversity of beneficial human health effects.Clausenais a large and highly diverse genus of plants with medicinal and cosmetic significance. The aim of this study was to analyze the composition ofClausena lansiumEOs and to investigate their potential antifungal effects. The chemical compositions ofClausena lansiumEOs obtained by hydrodistillation were analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 101 compounds were identified among the diverse extracts ofC. lansium. EOs of leaves and pericarps from different cultivars (Hainan local wampee and chicken heart wampee) collected in Hainan (China) were classified into four clusters based on their compositions. These clusters showed different antifungal activities against fiveCandidaspecies (C. albicans,C. tropicalis,C. glabrata,C. kruseiandC. parapsilosis) using the disc diffusion method.Clausena lansiumEOs of pericarps displayed noteworthy antifungal activitives against all the testedCandidastrains with inhibition zone diameters in the range of 11.1–23.1 mm. EOs of leaves showed relatively low antifungal activities with inhibition zone diameters in the range of 6.5–22.2 mm. The rank order of antifungal activities among the four EO clusters was as follows: Cluster IV> Cluster III > Cluster I ≥ Cluster II. These results represent the first report about the correlation between chemical composition ofC. lansiumEOs and antifungal activity. Higher contents of β-phellandrene, β-sesquiphellandrene and β-bisabolene in EOs of pericarps were likely responsible for the high antifungal activity of Cluster IV EOs. Taken together, our results demonstrate the chemical diversity ofClausena lansiumEOs and their potential as novel antifungal agents for candidiasis caused byCandidaspp. Furthermore, the obtained results showing a wide spectrum of antifungal activities provide scientific evidence for the traditional use of these plants. Keywords:correlation;Clausena lansium;essential oil;antifungal activity;Candidaspp.

Characterization and prebiotic properties of pectin polysaccharide from Clausena lansium (Lour.) Skeels fruit

Can Song,Feihong Huang,Linyu Liu,Quan Zhou,Dan Zhang,Qi Fang,Hui Lei,Hong Niu

Pectins have proven to be advantageous for human health as they regulate beneficial microbial communities and enhance immunity. The fruit of Clausena lansium (Lour.) Skeels (Wampee), also referred to as “treasure in fruit”, is rich in pectin polysaccharides . In this study, a homogalacturonan-type pectin (CCP2) with a molecular weight of 8.9 × 10 4  Da and degree of esterification of 42.86% was isolated from Wampee fruit. The gut microbiota regulation and phagocytosis-enhancing properties of CCP2 were examined in vivo and in vitro, respectively. Oral administration of CCP2 dramatically decreased the abundance of Bacteroidetes and increased the abundance of Firmicutes in intestinal bacteria in mice. The content of short-chain fatty acids in the feces also significantly improved. Moreover, CCP2 exhibited excellent phagocytosis-enhancing activities on RAW 264.7 macrophages. These results suggested that CCP2 could be a potential gut microbiota regulator and phagocytosis-enhancer, which could be used in food products to promote health through beneficial manipulation of gut microbiota.

Bioassay-guided isolation of antifungal amides against Sclerotinia sclerotiorum from the seeds of Clausena lansium

He Yan,Zi Xiong,Na Xie,Shanzhi Liu,Lianlian Zhang,Fang Xu,Wenhui Guo,Juntao Feng

Clausena lansium ( Lour.) Skeels is an attractive shrub or small tree, a member of the Rutaceae family. The seeds, leaves and roots of this plant have been used as herbal medicines for a long time. In this study, seven amides were isolated from the seeds of Clausena lansium . The antifungal assay indicated that lansiumamide B ( 4 ) and lansiumamide C ( 2 ) displayed the highest antifungal activity against Sclerotinia sclerotiorum with EC 50 values of 4.95 and 13.24 μg/mL, which were both lower than that of carbendazim (EC 50  = 0.64 μg/mL). The structure-activity relationships of these amides were investigated using assays to measure their in vitro antifungal activities against Sclerotinia sclerotiorum . The effect of the most potent compound lansiumamide B on morphology and physiology of S. sclerotiorum were also evaluated. Lansiumamide B caused cell rupture and mycelial abnormalities of S. sclerotiorum , and its curative efficacy (75.17%) against S. sclerotiorum infection was better than that of carbendazim (56.57%). This study demonstrated that amides isolated from C. lansium possessed the potential to be exploited as botanical fungicides for commercial application or can be used as templates for designing new fungicides with novel action modes.

The influence of the chemical composition of essential oils of Clausena lansium seeds on the growth of Candida strains

Ma Yinzheng,Wang Yuanxiao,Zhou Xia,Yang Heng,Zhang Huixin,Chen Wenhan,Zhang Haiying,Zhang Yunxia,He Xiaowen

Clausena lansium (Lour.) Skeels seeds have been shown to have diverse beneficial medical value due to their unique active components. This study analysed the composition of essential oils (EOs) of C. lansium seeds and investigated their potential antifungal effects against Candida strains . A total of forty-six components were identified in all samples by gas chromatography-mass spectrometry (GC–MS). The main components were sabinene, β -phellandrene and 4-terpineol. Thirteen EOs of C. lansium seeds were classified into three clusters based on their components. Cluster analysis showed that the difference between the tropics and subtropics was the greatest. These EOs and the three main chemicals showed different antifungal activities against five Candida species ( C. albicans , C. tropicalis , C. glabrata , C. krusei and C. parapsilosis ). The antifungal activity against C. glabrata and C. krusei was higher than that against other Candida strains. EOs of C. lansium seeds displayed noteworthy antifungal activity against both sensitive and fluconazole-resistant strains, with inhibition zone diameters in the range of 9.4–23.4 mm. Comprehensive analysis illustrated the importance of sabinene, β -phellandrene and 4-terpineol to antifungal activity, and there may be some synergistic effects with other components. These results represent the first report about the correlation between the chemical composition of EOs of C. lansium seeds and antifungal activity. Taken together, the results obtained provide scientific evidence for the traditional use of C. lansium seeds waste.

Antimicrobial and anti-biofilm activity of essential oils extracted from Clausena lansium (Lour.) Skeels and their main constituents against Streptococcus mutans

Zibei Wu,Xufeng Zhang,Lili Wang,Jinlei Sui,Mingqi Yu,Bingzhangke Bao,Jianing Qian,Guohui Yi,Yinzheng Ma,Xiaowen He

Streptococcus mutans is one of the most significant pathogens leading to dental caries by producing highly cariogenic biofilms within the oral cavity. However, the increasing antimicrobial resistance brings great difficulties to the prevention and treatment of dental caries. Therefore, it is very important to develop new and effective antimicrobial agents from natural sources. Clausena lansium (Lour.) Skeels, a plant commonly cultivated in Southern China, is rich in various active constituents. In this study, the main constituents of the essential oils extracted from Clausena lansium leaves (EOL), seeds (EOS), peels (EOP) were precisely analyzed by gas chromatography–mass spectrometry. The antimicrobial and anti-biofilm activities of EOL, EOS, and EOP, along with their main chemical constituents, against Streptococcus mutans were comprehensively evaluated. The primary constituent in EOL was β-caryophyllene, with a content of 112.29 mg/mL. EOS was found to be abundant in sabinene and 4-terpineol, with constituents of 147.21 mg/mL and 54.36 mg/mL, respectively. In EOP, the most predominant constituent was β-phellandrene, with a content of 173.95 mg/mL. The minimum inhibitory concentrations of EOL, EOS, and EOP were 0.12, 2.00, and 1.00 mg/mL, respectively. The minimum concentrations of inhibiting at least 90% biofilm formation of EOL, EOS, and EOP was 0.12 mg/mL, 2.00, and 2.00 mg/mL, respectively. Intriguingly, the antimicrobial and anti-biofilm efficacies of the main constituents were significantly lower than those of the essential oils. By observing the dynamic changes of the biofilm formation of S. mutans treated with essential oils, it was found that the process of biofilm formation was greatly delayed. In some cases, there was even no biofilm formation. The results of real-time quantitative PCR indicated that the expression levels of brpA, gtfB, gtfC, gtfD, and luxS genes were down-regulated after treatment with the essential oils. This study reinforces the potential of Clausena lansium as a promising source for developing new antimicrobial and anti-biofilm agents. They could inhibit the proliferation and virulence of S. mutans, thereby contributing to the control of dental caries.

Inhibitory potential of proanthocyanidins from the fruit pulp of Clausena lansium (Lour.) Skeels against α-glucosidase and non-enzymatic glycation: Activity and mechanism

Chong Ou-Yang,Weiming Chai,Xiaohui Xu,Shuang Song,Qiming Wei,Qian Huang,Zhengrong Zou

Inhibiting the activity of α-glucosidase and glycosylation of protein is an important way to treat diabetes mellitus and its complications. In this study, we investigated the anti-α-glucosidase activity, anti-glycation potential and structure of proanthocyanidins from fruit pulp of Clausena lansium (Lour.) Skeels. C. lansium fruit pulp proanthocyanidins showed a remarkable inhibition against α-glucosidase activity with IC 50 vaule of 0.26 ± 0.01 μg/mL in a competitive manner, and quenched the fluorescence of α-glucosidase by forming proanthocyanidin-α-glucosidase complex. Furthermore, compared to positive agent aminoguanidine (AG), the proanthocyanidins were the more significant inhibitors of non-enzymatic glycation by strongly inhibiting the formation of α-dicarbonyl compounds and advanced glycation end products. In addition, the structure of the proanthocyanidins was characterized in detail. These compounds were mainly composed of prodelphinidins, and their gallates. The main extender units, gallocatechin epigallocatechin and their gallates, were critical factor of the strong anti-α-glucosidase and anti-glycation activity. Therefore, this study authenticated a efficient α-glucosidase inhibitors and antiglycation agents, which would contribute to the development of anti-diabetic drug.

Chemical Composition of Cuticle and Barrier Properties to Transpiration in the Fruit of Clausena lansium (Lour.) Skeels

Huang Hua,Wang Ling,Qiu Diyang,Lu Yusheng

The plant cuticle, as a lipid membrane covering aerial plant surfaces, functions primarily against uncontrolled water loss. Herein, the cuticle chemical composition and the transpiration of wampee fruit (Clausena lansium (Lour.) Skeels) at the green, turning, and yellow stages in cultivars of “Jixin” and “Tianhuangpi” were comprehensively studied. The coverage of wax and cutin monomers per unit of fruit surface area at the green stage was lower in “Jixin” than in “Tianhuangpi” and increased gradually during development. Cutin monomers accumulated ranging from 22.5 μg cm−2 (green) to 52.5 μg cm−2 (turning) in “Jixin” and from 36.5 μg cm−2 (green) to 81.7 μg cm−2 (yellow) in “Tianhuangpi.” The total composition of waxes ranged between 6.0 μg cm−2 (green) and 11.1 μg cm−2 (turning) in “Jixin,” while they increased from 7.4 μg cm−2 (green) to 16.7 μg cm−2 (yellow) in “Tianhuangpi.” Cutin monomers were dominated by ω-, mid-dihydroxy fatty acids (over 40%), followed by multiple monomers of α,ω-dicarboxylic acids with or without added groups, α-monocarboxylic acids with or without ω- or mid-chain hydroxy or mid-epoxy groups, primary alcohols, and phenolics. The very-long-chain (VLC) aliphatic pattern of cuticular waxes was prominently composed of n-alkanes (ranging from 21.4% to 39.3% of total wax content), fatty acids, primary alcohols, and aldehydes. The cyclic waxes were dominated by triterpenoids (between 23.9 and 51.2%), sterols, and phenolics. Water loss in wampee fruit exhibited linear changes over time, indicating an overall monofunctional barrier to transpiration. Permeance for water in wampee fruit was higher at the green stage than at the yellow stage in both “Jixin” and “Tianhuangpi,” which showed a negative correlation with the changes of VLC n-alkanes. The results showed the cuticular chemicals, including cutin monomers and waxes, in wampee fruit and further indicated the potential contributions of the cuticular chemical composition to the physiological functions in fruits.

RNA-Seq-Based Transcriptomics and GC–MS Quantitative Analysis Reveal Antifungal Mechanisms of Essential Oil of Clausena lansium (Lour.) Skeels Seeds against Candida albicans

Yinzheng Ma,Jinlei Sui,Yan Wang,Wanying Sun,Guohui Yi,Jinyan Wu,Shi Qiu,Lili Wang,Aihua Zhang,Xiaowen He

Infections caused byCandida albicans(C. albicans) and increasing resistance to commonly used drugs lead to a variety of mucosal diseases and systemic infectious diseases. We previously confirmed that the essential oil ofClausena lansium(Lour.) Skeels seeds (CSEO) had antifungal activity againstC. albicans, but the detailed mechanism between the chemical components and antifungal activity is unclear. In this study, a quantitative analysis of five volatile components of CSEO, including sabinene,α-phellandrene,β-phellandrene, 4-terpineol, andβ-caryophyllene, was carried out using the gas chromatography–mass spectrometry (GC–MS) method. Both the broth dilution and kinetic growth methods proved that the antifungal activity of CSEO against fluconazole-resistantC. albicanswas better than that of its main components (sabinene and 4-terpineol). To further investigate the inhibitory mechanism, the transcriptional responses ofC. albicansto CSEO, sabinene, and 4-terpineol treatment were determined based on RNA-seq. The Venn diagram and clustering analysis pattern of differential expression genes showed the mechanism of CSEO and 4-terpineol’s anti-C. albicansactivity might be similar from the perspective of the genes. Functional enrichment analysis suggested that CSEO regulated adherence-, hyphae-, and biofilm-formation-related genes, which may be CSEO’s active mechanism of inhibiting the growth of fluconazole-resistantC. albicans. Overall, we preliminarily revealed the molecular mechanism between the chemical components and the antifungal activity of CSEO againstC. albicans. This study provides new insights to overcome the azole resistance ofC. albicansand promote the development and application ofC. lansium(Lour.) Skeels seeds. Keywords:Clausena lansium(Lour.) Skeels seeds;essential oil;Candida albicans;drug resistance;gas chromatography–mass spectrometry;transcriptomics analysis

Phenolic profile and α-glucosidase inhibitory potential of wampee (Clausena lansium (Lour.) Skeels) peel and pulp: In vitro digestion/in silico evaluations

Hua Chen,Yousheng Shi,Lu Wang,Xiaoping Hu,Xue Lin

To investigate the changes in phenolics, flavonoids, and their bio-activities of wampee ( Clausena lansium (Lour.) Skeels) during digestion, the peel and pulp were subjected to simulated in vitro digestion, encompassing oral, gastric, small intestine, and large intestine digestion stages. The peel exhibited a total release of 91.93 mg GAE/g DW of phenolics and 61.86 mg RE/g DW of flavonoids, whereas the pulp displayed a release of 27.83 mg GAE/g DW of phenolics and 8.94 mg RE/g DW of flavonoids. Notably, the phenolics and flavonoids were mostly released during the oral digestion stage for peel, while they were mostly released during the small intestine digestion stage for pulp. The results of the targeted flavonoids analysis indicated that rutin and l-epicatechin were the two most widely released compounds in each digestion step. Moreover, myricetin has been identified as the best inhibitor against α -glucosidase, probably because it formed the most H-bonds, 8, with 6 catalytic residues, which was the highest number. Furthermore, the soluble substances released from the peel exhibited significantly higher antioxidant activities and inhibitory activity against α -glucosidase ( p  < 0.05) compared to those from the pulp. Positive correlations were observed between the total phenolic content or total flavonoid content and the antioxidant activities (r > 0.73 (peel), > 0.61 (pulp)), as well as α -glucosidase inhibitory activity (r <  − 0.48 (peel), < −0.64 (pulp)) of peel and pulp. In conclusion, these findings provide valuable insights into the digestive characteristics and health benefits of both wampee peel and pulp.

A comparative UPLC-Q-Orbitrap-MS untargeted metabolomics investigation of different parts of Clausena lansium (Lour.) Skeels

Ruiyi Fan,Cheng Peng,Xinxin Zhang,Diyang Qiu,Genlin Mao,Yusheng Lu,Jiwu Zeng

In this study, the non-targeted large-scale plant metabolomics (UPLC-Q-Orbitrap-MS) was performed for the comparison of chemical profiling of the leaves, barks, flowers, peels, pulps, and seeds of Clausena lansium (Lour.) Skeels (called “wampee”). A total of 364 metabolites were identified, and 62 potential biomarkers were selected by the multivariate statistical analysis. Hierarchical cluster analysis suggested that the selected biomarkers were significant differential metabolites among various parts of wampee. Metabolic pathway analysis showed a significant enrichment of the “Flavone and flavonol synthesis” and “Isoquinoline alkaloid biosynthesis” pathway. This study provides important information for the isolation and identification of functional components from different tissues of wampee and the metabolic biosynthesis pathway elucidation in detail.

Anti-Inflammatory Potential of Wampee (Clausena lansium (Lour.) Skeels) Polyphenol Extract in Ulcerative Colitis: Gut Microbiota and TLR4-p38 MAPK/NF-κB Signaling Axis Regulation

Kaijie Shang,Zhiheng Zhao,Hua Chen,Xiaonan Bian,Xianquan Zhong,Xiaoping Hu,Xue Lin,Lu Wang

The consumption of wampee has traditionally been utilized to alleviate gastrointestinal inflammation and associated disorders; however, its exact mechanism has remained unknown. The aim of this study was to elucidate the therapeutic efficacy and underlying mechanism of wampee polyphenol extract (WPE) in dextran sulfate sodium (DSS)-induced ulcerative colitis (UC). The findings revealed that WPE alleviated diverse symptoms of UC, regulated various inflammatory cytokines, and effectively protected the colon tissue structure and barrier integrity, thereby inhibiting LPS translocation. Moreover, WPE restored the richness and diversity of gut microbiota and optimized its structure at the phylum and genus levels, causing a notable improvement in short- chain fatty acid (SCFA) metabolism, particularly acetic acid, propionic acid, and butyric acid. Consequently, WPE was demonstrated to effectively suppress the LPS-induced TLR4-p38 MAPK/NF-κB signaling pathway by modulating gut microbiota and SCFA metabolism. These findings provided a theoretical basis for the use of wampee as a potential functional natural food for UC.

Formulation and functional performance of edible chitosan films incorporating Clausena lansium leaf essential oil

Li Jie,Perera Vindya,Cao Shihao,Zhang Mengluo,Ding Yuheng,Tang Minmin,Zhu Tingting

Edible composite films were developed by incorporating Clausena lansium leaf essential oil (CLEO) into chitosan (CS) matrix to enhance the functionality of food-contact films. The optimal formulation (2.0% CS, 1.5% glycerol and 0.4% CLEO) was determined using principal component analysis and response surface methodology. The optimized films exhibited favorable physicochemical, mechanical, and barrier properties. Fourier transform infrared spectroscopy and SEM verified successful CLEO incorporation into the polymer matrix without compromising structural integrity. Furthermore, CLEO significantly enhanced antioxidant and antibacterial activities, with DPPH and ABTS⁺ scavenging rates of 49.15 ± 1.25 and 52.25 ± 3.33%, respectively, and inhibition rates of 72.84 to 98.63% against tested pathogenic bacteria. Preliminary biological evaluation indicated good cytocompatibility, with cell viability above 90%; however, this does not constitute a comprehensive food safety assessment. Overall, CLEO represents a promising natural functional additive for chitosan-based edible films, although further validation in real food systems is required to confirm practical preservation efficacy.
