Co-culture of Clostridium acetobutylicum and Saccharomyces cerevisiae was used to enhance solvents (acetone-butanol-ethanol, ABE) production from cassava without amylolytic enzymes addition. Under the simultaneous inoculation condition of C. acetobutylicum and S. cerevisiae with the inoculation level of 5% (v/v) for both strains, 21.74 g/L solvents was produced from 75 g/L cassava with the yield of 0.36 g solvents/g glucose. The solvents titer and yield obtained in the co-culture process increased by 14.3% and 9.1%, respectively, compared to those achieved in ABE fermentation. For further improvements in total solvents titer and yield from cassava, ABE fermentation and co-culture process from higher cassava concentration media were conducted. 42.56 g/L total solvents was achieved from 150 g/L cassava by co-culture with the yield of 0.46 g solvents/g glucose. The solvents titer and yield obtained in the co-culture process increased by 141.0% and 43.8%, respectively, compared to those obtained from ABE fermentation. The results showed great potential for the use of co-culture of C. acetobutylicum and S. cerevisiae for solvents production from starchy substrate.

Summary Immobilized fermentation has several advantages over traditional suspended fermentation, including simple and continuous operation, improved fermentation performance and reduced cost. Carrier is the most adjustable element among three elements of immobilized fermentation, including carrier, bacteria and environment. In this study, we characterized carrier roughness and surface properties of four types of natural fibres, including linen, cotton, bamboo fibre and silk, to assess their effects on cell immobilization, fermentation performance and stability. Linen with higher specific surface area and roughness could adsorb more bacteria during immobilized fermentation, thereby improving fermentation performance; thus, linen was selected as a suitable carrier and was applied for acetone–butanol–ethanol (ABE) fermentation. To further improve fermentation performance, we also found that microbes of Clostridium acetobutylicum were negatively charged surfaces during fermentation. Therefore, we then modified linen with polyetherimide (PEI) and steric acid (SA) to increase surface positive charge and improve surface property. During ABE fermentation, the adhesion between modified linen and bacteria was increased, adsorption was increased about twofold compared with that of unmodified linen, and butanol productivity was increased 8.16% and 6.80% with PEI- and SA-modified linen as carriers respectively.

Butanol is an attractive second-generation biofuel as well as an important chemical. Production of butanol from lignocellulosic feedstock via consolidated bioprocessing has attracted increasing interest in recent years. However, this has been limited by the inefficient degradation of extracellular polysaccharides in butanol-producing strains. In this study, production of butanol directly from hemicellulose was achieved simply through overexpression of an indigenous xylanase in Clostridium acetobutylicum. The xylanase (XynB) encoded by CA_P0053 was highly soluble and fully secreted from C. acetobutylicum. The extracellular xylanase activity was increased 88-fold, and 4.03 g/L biobutanol was obtained from hemicellulose, which has been the highest yield yet reported from either hemicellulose or xylan. This study proved that the solventogenic C. acetobutylicum had great potential for consolidated production of biofuels. Understanding and applying its secretion mechanism will be of great significance in the future.

The main route for biobutanol production is anaerobic clostridial fermentation using corn and sugarcane as substrates. The high-price of raw materials in above processes largely limits its application as an advanced biofuel. The renewable lignocellulosic biomass is an ideal feedstock to produce butanol. However, the structure of lignocellulose is complicated which needs pretreatment to facilitate enzymatic efficiency and substrate fermentability. Herein, the green ternary deep eutectic solvent (DES) composed of choline chloride, succinic acid, and glycerol was synthesized. To improve DES pretreatment performance and reduce the viscosity of reaction system, water was added as co-solvent. Results indicate that xylan removal and glucan recovery were 61.7% and 93.6% when pretreating reed straw (RS) by DES/H 2 O (80:20, wt%) at 120 °C for 4 h. In this case, glucose yield reached 87.9% by cellulase after 72 h enzymolysis, which is 6.2-fold to untreated RS. Afterward, saccharification yield of treated reed was further elevated to 93.1% assisted by Tween 80 (100 mg/g-substrate). The reed hydrolysate was then applied as substrate for biobutanol production by Clostridium acetobutylicum ATCC 824, and approximately 6.5 g/L butanol and 8.8 g/L total solvents was obtained from the real hydrolysate. Finally, the enzymatic residues of reed were prepared to porous hydrochar materials via hydrothermal carbonization at 220 °C for 8 h. Adding 2.0 g/L hydrochar was verified for enhancing butanol titer to 11.5 g/L from synthetic medium containing typical pretreatment-derived inhibitors. In summary, this study provides an efficient butanol bioproduction route integrated with the complete valorization of renewable lignocellulosic biomass.
