Dietary Clostridium autoethanogenum protein (CAP) has the potential to replace fishmeal (FM), but excessive CAP substitutions will affect gut health. Bile acids (BAs) have multiple benefits, but little information is available on the effects of BAs on Pacific white shrimp ( Litopenaeus vannamei ) fed a CAP diet (CAP replacing FM). An 8-week feeding trial were used to investigate the impacts of CAP substitution and BAs addition on structural changes of intestine, apoptosis, inflammatory cytokine and NFκB pathway of intestine, and transcriptomic profiles of hepatopancreas in Pacific white shrimp. Compared to PC (control) treatment, the NC (CAP50, CAP replacing 50% FM) treatment significantly decreased the fold height (FH) and villus length (VL) of midgut, and impaired the mitochondrial cristae. Compared to NC group, the FH was increased in BA1 (CAP50 with 0.03% BAs) and BA2 (CAP50 with 0.06% BAs) groups, the muscle layer thickness (MT) was decreased in BA2 and BA3 (CAP50 with 0.09% BAs) groups, the VL was increased in BA1 group, and the mitochondrial cristae was normal and well-defined in BA1 and BA2 groups (all p < 0.05). The NC increased the genes expression of apoptosis and NFκB signalling pathway, while the BA2 decreased the genes expression of apoptosis, inflammatory cytokine and NFκB signalling pathway. In transcriptomics analysis, a total of 171 differential expression genes (DEGs) were identified in NC group and 50 DEGs in BA2 group. There was 13 DEGs was observed both in NC and BA2 groups. The 5 KEGG pathways were statistically enriched in all treatments, including “Carbon metabolism”, “One carbon pool by folate”, “Glycine, serine and threonine metabolism”, “Amino sugar and nucleotide sugar metabolism”, and “PPAR signaling pathway”. In conclusion, CAP50 substitution impaired the intestinal health and 0.06% BAs addition improved it, both treatments altered the transcriptomics profiles of hepatopancreas in Pacific white shrimp.

The
objective of this study was to evaluate the nutritional value of Clostridium autoethanogenum protein (CAP) as a sole protein source at different dietary protein levels by growth performance, health status, and intestinal microbiota of grass carp. Grass carp (4.56 ± 0.01 g) were randomly divided into 6 treatments in triplicate (30 fish per replicate) and fed with gradient protein level (24.45%, 27.31%, 31.82%, 35.79%, 38.64%, and 42.82%, respectively; named CAP1, CAP2, CAP3, CAP4, CAP5, and CAP6) diets for 8 weeks. Final body weight (FBW) and specific growth rate (SGR) were the significantly highest in the CAP4 group ( P < 0.05). Based on SGR and feed efficiency (FE), the dietary protein requirement for juvenile grass carp was estimated to be 36.14–37.90%. CAP4 promoted hepatopancreatic health by reducing lipid deposition and structural lesions, and improved the anti-inflammatory ability by upregulating the relative expression level of il10 ( P < 0.05). At the same time, the CAP4 improved the length of mucosal fold and the thickness of muscular layer. In addition, CAP4 can maintain the composition and metabolic function of intestinal microbiota and enhance the endocrine system and bacterial chemotaxis. However, it is worth noting that increasing CAP inclusion depressed hepatopancreatic antioxidant capacity. In conclusion, CAP is a promising novel protein source for grass carp, while careful consideration of its side effect at high inclusion level is deserved.

Clostridium autoethanogenum protein (CAP) is a promising protein source for aquaculture. Previous studies proved that high levels of dietary CAP retarded the muscle development in turbot through mammalian target of rapamycin (mTOR)/amino acid response (AAR)/adenosine monophosphate-activated protein kinase (AMPK) pathways. However, how CAP regulating these pathways was still unclear and there was no information on the comprehensive analysis for the mechanisms of CAP on fish muscle. In the present study, the integration of metabolomic and proteomic was applied on the muscle of turbot (initial weight of 150 ± 3 g) fed with fishmeal-based diet or diet with 60 % of fishmeal protein replaced by CAP for 70 days. The present work confirmed that CAP decreased the growth, fillet yield and induced the muscle atrophy. Integrated analysis of metabolomics and proteomics showed that CAP negatively influenced the energy production by inhibiting the key enzymes of glycolysis and mitochondrial respiratory chain. Second, CAP inhibited mitogen-activated protein kinase (MAPK) and phosphatidylinositol-3-kinase (PI3K) pathways to down-regulate mTOR pathways and protein synthesis. Third, the amino acid contents in turbot muscle were imbalanced by CAP, which would be perceived by general control nonderepressible 1 in AAR signal pathways. Finally, the up-regulated AAR induced amino acids catabolism through purine-based combined deamination. In conclusion, the present work provided the universal knowledge on the working principle of CAP and laid the foundation for CAP improvement in the future.

This study aimed to investigate the impact of dietary tributyrin (TB) in the low fish meal (high- Clostridium autoethanogenum protein, CAP) diet on the growth, antioxidation, lipid metabolism and intestinal microbiota of largemouth bass ( Micropterus salmoides ). For eight weeks, largemouth bass (15.05 ± 0.58 g) were fed on eight isonitrogenous and isolipid diets with graded levels of dietary TB (0 g/kg, 0.2 g/kg, 0.4 g/kg, 0.8 g/kg, 1.6 g/kg, 3.2 g/kg, 4.0 g/kg and 8.0 g/kg). The findings showed that the weight gain rate (WGR) and specific growth rate (SGR) were increased by 1.6 g/kg of dietary TB. The protein efficiency and deposition ratios were linearly affected by dietary TB. The broken-line model based on the WGR revealed that 1.617 g/kg of dietary TB was sufficient for optimal growth. The digestive enzyme activities and villus length of the intestine were increased by dietary TB (1.6 g/kg). Total antioxidant capacity (T-AOC), superoxide dismutase (SOD) and catalase (CAT) activities were significantly improved, and the malondialdehyde (MDA) and protein carbonyl (PC) contents were significantly decreased after dietary TB. The expressions of nuclear factor erythroid 2-related factor 2 ( nrf2 ) and kelch like ech associated protein 1 ( keap1 ) in the liver and intestine were significantly increased and decreased by 1.6 g/kg of dietary TB, respectively. The mRNA levels of copper-zinc superoxide dismutase ( cuznsod ), cat , heme oxygenase-1 ( ho-1 ) and glutathione peroxidase ( gpx ) were significantly upregulated. Dietary TB could significantly affect the total protein (TP) and albumin (ALB) content in the serum. The aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were the lowest at 1.6 g/kg of TB group. Besides, the hepatocyte vacuolation and nuclear translocation were alleviated after supplementation of 0.2‰− 3.0‰ of dietary TB. The total cholesterol (T-CHO), high density lipoprotein cholesterol (HDLC) and low density lipoprotein cholesterol (LDLC) were significantly increased by dietary TB. The triglycerides (TG) content was decreased by dietary TB. The mRNA levels of sterol regulatory element-binding protein 1-c ( srebp1-c ), acetyl-coa carboxylase alpha ( accα ), fatty acid synthase ( fasn ) and accβ in the liver were significantly downregulated, and peroxisome proliferator-activated receptor alpha ( pparα ) and carnitine palmitoyltransferase 1 ( cpt-1 ) were upregulated by 1.6 g/kg of dietary TB. Dietary TB had effects on the intestine microbes of largemouth bass. The Cetobacterium was present in the highest percentage and increased from 41% (0 g/kg TB) to 59% (1.6 g/kg TB). The Mycoplasmataceae was the second most predominant genus and decreased from 90% in the 0 g/kg of TB group to 10% in the 1.6 g/kg of TB group. In summary, the dietary TB could be supplemented in the CAP-based diet for better growth and antioxidation, lipid metabolism and intestinal microbiota of largemouth bass.

The study evaluated the effects of dietary phosphorus supplementation on the fishmeal replacement with Clostridium autoethanogenum protein (CAP) in the diet of L. vannamei . Four isonitrogenous and isolipid diets were formulated: the PC diet contains 25% fishmeal, the NC, P1 and P2 diets were replaced 40% fishmeal with CAP and supplemented with 0, 0.8 and 1.6% NaH 2 PO 4 respectively (equivalent to dietary phosphorus level of 0.96%, 1.12% and 1.27%). Sampling and V. parahaemolyticus challenge test were conducted after 50-day-feeding (initial shrimp weight 1.79 ± 0.02 g). The results showed that there were no significant differences in the growth performance of shrimp among the 4 groups. The expressions of dorsal in the gut were significantly lower in shrimp fed the P1 and P2 diets than shrimp fed the NC diet and the expression of peroxinectin in the gut was lower in shrimp fed the NC diet than others. The cumulative mortality of shrimp after V. parahaemolyticus challenge was significantly lower in shrimp fed the P2 diet than those fed the NC diet. After the challenge, genes expressions related to the prophenoloxidase activating system ( proPO , lgbp , ppaf ) were inhibited in the hepatopancreas of shrimp fed NC diet but activated in shrimp fed the P1 diet compared to those fed the PC diet. The AKP and T-AOC activities were higher in shrimp fed the P2 diet than those fed the other diets. The thickness of muscle layer of shrimp fed the P1 diet was thicker than that in the other groups, and significant stress damage happened in the midgut of the shrimp fed the NC diet. The abundance of Pseudoalteromonas , Haloferula and Ruegeria in shrimp fed the P1 diet was higher than those fed the other diets, while Vibrio in shrimp fed the P2 diet was higher than those fed the other diets. This indicated that a low fishmeal diet with dietary phosphorus level of 1.12% could improve the histology, enhance immune response, and increase the abundance of beneficial bacteria in the gut of shrimp. The low fishmeal diet with dietary phosphorus level of 1.27% could improve disease resistance and antioxidant capacity, but there was a possibility of damage to the gut histology as well as increasing abundance of Vibrio in the gut microbiota of shrimp.
