Clostridium autoethanogenum protein substitution and bile acids addition altered intestinal health and transcriptome profiles of hepatopancreas in Litopenaeus vannamei

Jia Xu,Chaozhong Zheng,Shuyan Chi,Shuang Zhang,Junming Cao,Beiping Tan,Shiwei Xie

Dietary Clostridium autoethanogenum protein (CAP) has the potential to replace fishmeal (FM), but excessive CAP substitutions will affect gut health. Bile acids (BAs) have multiple benefits, but little information is available on the effects of BAs on Pacific white shrimp ( Litopenaeus vannamei ) fed a CAP diet (CAP replacing FM). An 8-week feeding trial were used to investigate the impacts of CAP substitution and BAs addition on structural changes of intestine, apoptosis, inflammatory cytokine and NFκB pathway of intestine, and transcriptomic profiles of hepatopancreas in Pacific white shrimp. Compared to PC (control) treatment, the NC (CAP50, CAP replacing 50% FM) treatment significantly decreased the fold height (FH) and villus length (VL) of midgut, and impaired the mitochondrial cristae. Compared to NC group, the FH was increased in BA1 (CAP50 with 0.03% BAs) and BA2 (CAP50 with 0.06% BAs) groups, the muscle layer thickness (MT) was decreased in BA2 and BA3 (CAP50 with 0.09% BAs) groups, the VL was increased in BA1 group, and the mitochondrial cristae was normal and well-defined in BA1 and BA2 groups (all p  < 0.05). The NC increased the genes expression of apoptosis and NFκB signalling pathway, while the BA2 decreased the genes expression of apoptosis, inflammatory cytokine and NFκB signalling pathway. In transcriptomics analysis, a total of 171 differential expression genes (DEGs) were identified in NC group and 50 DEGs in BA2 group. There was 13 DEGs was observed both in NC and BA2 groups. The 5 KEGG pathways were statistically enriched in all treatments, including “Carbon metabolism”, “One carbon pool by folate”, “Glycine, serine and threonine metabolism”, “Amino sugar and nucleotide sugar metabolism”, and “PPAR signaling pathway”. In conclusion, CAP50 substitution impaired the intestinal health and 0.06% BAs addition improved it, both treatments altered the transcriptomics profiles of hepatopancreas in Pacific white shrimp.

Integrated metabolomics and proteomics analysis to provide insights into muscle atrophy of turbot Scophthalmus maximus by dietary Clostridium autoethanogenum protein

Shihui Pan,Nan Bai,Zezheng Qi,Chuwen Chen,Daisong Ma,Zhenlin Liang,Min Gu

Clostridium autoethanogenum protein (CAP) is a promising protein source for aquaculture. Previous studies proved that high levels of dietary CAP retarded the muscle development in turbot through mammalian target of rapamycin (mTOR)/amino acid response (AAR)/adenosine monophosphate-activated protein kinase (AMPK) pathways. However, how CAP regulating these pathways was still unclear and there was no information on the comprehensive analysis for the mechanisms of CAP on fish muscle. In the present study, the integration of metabolomic and proteomic was applied on the muscle of turbot (initial weight of 150 ± 3 g) fed with fishmeal-based diet or diet with 60 % of fishmeal protein replaced by CAP for 70 days. The present work confirmed that CAP decreased the growth, fillet yield and induced the muscle atrophy. Integrated analysis of metabolomics and proteomics showed that CAP negatively influenced the energy production by inhibiting the key enzymes of glycolysis and mitochondrial respiratory chain. Second, CAP inhibited mitogen-activated protein kinase (MAPK) and phosphatidylinositol-3-kinase (PI3K) pathways to down-regulate mTOR pathways and protein synthesis. Third, the amino acid contents in turbot muscle were imbalanced by CAP, which would be perceived by general control nonderepressible 1 in AAR signal pathways. Finally, the up-regulated AAR induced amino acids catabolism through purine-based combined deamination. In conclusion, the present work provided the universal knowledge on the working principle of CAP and laid the foundation for CAP improvement in the future.

Evaluation of the appropriate Clostridium autoethanogenum protein level in grass carp (Ctenopharyngodon idellus) diets by growth performance, health status, and intestinal microbiota

Liu Guoqing,Zhou Meng,Mao Xiangjie,Gu Dianchao,Chen Wangwang,Long Xianmei,Xie Shouqi,Tan Qingsong

The objective of this study was to evaluate the nutritional value of Clostridium autoethanogenum protein (CAP) as a sole protein source at different dietary protein levels by growth performance, health status, and intestinal microbiota of grass carp. Grass carp (4.56 ± 0.01 g) were randomly divided into 6 treatments in triplicate (30 fish per replicate) and fed with gradient protein level (24.45%, 27.31%, 31.82%, 35.79%, 38.64%, and 42.82%, respectively; named CAP1, CAP2, CAP3, CAP4, CAP5, and CAP6) diets for 8 weeks. Final body weight (FBW) and specific growth rate (SGR) were the significantly highest in the CAP4 group ( P  < 0.05). Based on SGR and feed efficiency (FE), the dietary protein requirement for juvenile grass carp was estimated to be 36.14–37.90%. CAP4 promoted hepatopancreatic health by reducing lipid deposition and structural lesions, and improved the anti-inflammatory ability by upregulating the relative expression level of il10 ( P  < 0.05). At the same time, the CAP4 improved the length of mucosal fold and the thickness of muscular layer. In addition, CAP4 can maintain the composition and metabolic function of intestinal microbiota and enhance the endocrine system and bacterial chemotaxis. However, it is worth noting that increasing CAP inclusion depressed hepatopancreatic antioxidant capacity. In conclusion, CAP is a promising novel protein source for grass carp, while careful consideration of its side effect at high inclusion level is deserved.

Dietary phosphorus supplementation in the diet of Pacific white shrimp (Litopenaeus vannamei) alleviated the adverse impacts caused by high Clostridium autoethanogenum protein

Chaozhong Zheng,Junming Cao,Shuyan Chi,Xiaohui Dong,Qihui Yang,Hongyu Liu,Shuang Zhang,Shiwei Xie,Beiping Tan

The study evaluated the effects of dietary phosphorus supplementation on the fishmeal replacement with Clostridium autoethanogenum protein (CAP) in the diet of L. vannamei . Four isonitrogenous and isolipid diets were formulated: the PC diet contains 25% fishmeal, the NC, P1 and P2 diets were replaced 40% fishmeal with CAP and supplemented with 0, 0.8 and 1.6% NaH 2 PO 4 respectively (equivalent to dietary phosphorus level of 0.96%, 1.12% and 1.27%). Sampling and V. parahaemolyticus challenge test were conducted after 50-day-feeding (initial shrimp weight 1.79 ± 0.02 g). The results showed that there were no significant differences in the growth performance of shrimp among the 4 groups. The expressions of dorsal in the gut were significantly lower in shrimp fed the P1 and P2 diets than shrimp fed the NC diet and the expression of peroxinectin in the gut was lower in shrimp fed the NC diet than others. The cumulative mortality of shrimp after V. parahaemolyticus challenge was significantly lower in shrimp fed the P2 diet than those fed the NC diet. After the challenge, genes expressions related to the prophenoloxidase activating system ( proPO , lgbp , ppaf ) were inhibited in the hepatopancreas of shrimp fed NC diet but activated in shrimp fed the P1 diet compared to those fed the PC diet. The AKP and T-AOC activities were higher in shrimp fed the P2 diet than those fed the other diets. The thickness of muscle layer of shrimp fed the P1 diet was thicker than that in the other groups, and significant stress damage happened in the midgut of the shrimp fed the NC diet. The abundance of Pseudoalteromonas , Haloferula and Ruegeria in shrimp fed the P1 diet was higher than those fed the other diets, while Vibrio in shrimp fed the P2 diet was higher than those fed the other diets. This indicated that a low fishmeal diet with dietary phosphorus level of 1.12% could improve the histology, enhance immune response, and increase the abundance of beneficial bacteria in the gut of shrimp. The low fishmeal diet with dietary phosphorus level of 1.27% could improve disease resistance and antioxidant capacity, but there was a possibility of damage to the gut histology as well as increasing abundance of Vibrio in the gut microbiota of shrimp.

Effects of dietary tributyrin supplementation in low fish meal diet containing high-Clostridium autoethanogenum protein on the growth performance, antioxidative capacity, lipid metabolism and intestinal microbiota of largemouth bass (Micropterus salmoides)

Kai Luo,Yuhui Shi,Kunlin Yang,Qiaoqing Xu,Jiang Zhou,Weihua Gao,Chenglong Wu,Zhuoduo Wang

This study aimed to investigate the impact of dietary tributyrin (TB) in the low fish meal (high- Clostridium autoethanogenum protein, CAP) diet on the growth, antioxidation, lipid metabolism and intestinal microbiota of largemouth bass ( Micropterus salmoides ). For eight weeks, largemouth bass (15.05 ± 0.58 g) were fed on eight isonitrogenous and isolipid diets with graded levels of dietary TB (0 g/kg, 0.2 g/kg, 0.4 g/kg, 0.8 g/kg, 1.6 g/kg, 3.2 g/kg, 4.0 g/kg and 8.0 g/kg). The findings showed that the weight gain rate (WGR) and specific growth rate (SGR) were increased by 1.6 g/kg of dietary TB. The protein efficiency and deposition ratios were linearly affected by dietary TB. The broken-line model based on the WGR revealed that 1.617 g/kg of dietary TB was sufficient for optimal growth. The digestive enzyme activities and villus length of the intestine were increased by dietary TB (1.6 g/kg). Total antioxidant capacity (T-AOC), superoxide dismutase (SOD) and catalase (CAT) activities were significantly improved, and the malondialdehyde (MDA) and protein carbonyl (PC) contents were significantly decreased after dietary TB. The expressions of nuclear factor erythroid 2-related factor 2 ( nrf2 ) and kelch like ech associated protein 1 ( keap1 ) in the liver and intestine were significantly increased and decreased by 1.6 g/kg of dietary TB, respectively. The mRNA levels of copper-zinc superoxide dismutase ( cuznsod ), cat , heme oxygenase-1 ( ho-1 ) and glutathione peroxidase ( gpx ) were significantly upregulated. Dietary TB could significantly affect the total protein (TP) and albumin (ALB) content in the serum. The aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were the lowest at 1.6 g/kg of TB group. Besides, the hepatocyte vacuolation and nuclear translocation were alleviated after supplementation of 0.2‰− 3.0‰ of dietary TB. The total cholesterol (T-CHO), high density lipoprotein cholesterol (HDLC) and low density lipoprotein cholesterol (LDLC) were significantly increased by dietary TB. The triglycerides (TG) content was decreased by dietary TB. The mRNA levels of sterol regulatory element-binding protein 1-c ( srebp1-c ), acetyl-coa carboxylase alpha ( accα ), fatty acid synthase ( fasn ) and accβ in the liver were significantly downregulated, and peroxisome proliferator-activated receptor alpha ( pparα ) and carnitine palmitoyltransferase 1 ( cpt-1 ) were upregulated by 1.6 g/kg of dietary TB. Dietary TB had effects on the intestine microbes of largemouth bass. The Cetobacterium was present in the highest percentage and increased from 41% (0 g/kg TB) to 59% (1.6 g/kg TB). The Mycoplasmataceae was the second most predominant genus and decreased from 90% in the 0 g/kg of TB group to 10% in the 1.6 g/kg of TB group. In summary, the dietary TB could be supplemented in the CAP-based diet for better growth and antioxidation, lipid metabolism and intestinal microbiota of largemouth bass.

Engineering energy-robust Clostridium autoethanogenum via integrated nanotechnology and transcription-guided metabolic reprogramming

Yida Zhai,Limei Chen,Fuguo Liu,Fei Li,Liucheng Long,Menghan Chi,Jinju Ma,Wuxi Chen,Yang Yang,Yu Duan,Guofu Chen,Demao Li

Microbial conversion of CO and CO 2 offers a promising route to C1 gas valorization, but acetogenic syngas fermentation remains constrained by bioenergetic bottlenecks. To address this, a biohybrid system integrating bovine serum albumin-stabilized gold nanoparticles with Clostridium autoethanogenum was constructed, increasing intracellular ATP levels, modulating redox balance, and promoting cell growth under illumination. Transcriptomic analysis of this system systematically identified key energy complexes and positive regulators associated with enhanced cellular energy metabolism. Based on these elements, a Push–Pull–Block strategy was implemented to generate the target strain CAB181, which increased OD 600 -normalized production of ethanol, acetate, and total acids and alcohols by 46.5%, 35.1%, and 34.2%, respectively. Multiomics analysis confirmed molecular-level metabolic reprogramming in CAB181. This work provides an effective strategy to improve the bioenergetic robustness and fermentation performance of C. autoethanogenum -based cell factories, and offers a reference framework for future engineering of other acetogens.

Microbiomic and metabonomic analysis provide new insights into the enhanced intestinal health in large-size largemouth bass (Micropterus salmoides) when fed novel proteins: Novel proteins are promising foods for future aquaculture

Lukuan Li,Yu Wang,Zhen Zhang,Chunfang Wang

The novel proteins, i.e., Clostridium autoethanogenum (CAP), Tenebrio molitor (TM), Chlorella vulgaris (ChM), and cottonseed protein concentrate (CSM), are promising food for the future aquaculture due to the limited fishmeal (FM) resources. However, in-depth investigations on the intestinal barriers of fish when fed novel proteins are sparse, and the association between intestinal flora and physiological biomarkers is still unknown. Five isonitrogenous and isolipidic diets comprising various proteins were developed, with fishmeal serving as the control and novel proteins as the experimental diets. Largemouth bass ( Micropterus salmoides ) with an initial body weight of 211.9 ± 1.2 g were used as experimental model and fed with these five diets for eight weeks. The findings from the current study indicated that the intestinal histological structure as well as immunochemistry barrier performance of the ChM, CAP, and TM groups were superior to those of the CSM and FM groups, despite the fact that the villi height of FM was the greatest of all groups. Similarly, the intestinal inflammatory response and the permeability were elevated in CSM while decreased in ChM, CAP, and TM group. Concerning the intestinal short fatty acids (SCFAs), the levels in the CAP as well as ChM group were greater than the other proteins. From the data analysis of 16S rRNA gene sequencing, the abundance of opportunistic pathogens including (Ralstonia , Acinetobacter , Aeromonas , and Protebacteria) were higher in CSM group while attenuated in ChM, CAP and TM group. In addition, the abundance of nine types of intestinal-barriers-repairing probiotics were elevated in all novel proteins compared with the fishmeal group. These altered microbiota were highly correlated with intestinal inflammatory response, antioxidant status, immunochemistry barrier, physical barrier and SCFAs indexes by Mantel test and Pearson's correlation analysis. Collectively, the ranking of the beneficial functions among these five proteins was ChM > CAP>TM > FM > CSM according to Z -score and this study provides in-depth data manifesting the effects of novel proteins on the gut health status of large-size largemouth bass.

Multi-omics approach to study the dual effects of novel proteins on the intestinal health of juvenile largemouth bass (Micropterus salmoides) under an alternate feeding strategy

Li Lukuan,Wang Yu,Huang Yanqing,Wang Chunfang

Introduction: In an effort to minimize the usage of fishmeal in aquaculture, novel protein diets, including Tenebrio molitor, cottonseed protein concentrate, Clostridium autoethanogenum, and Chlorella vulgaris were evaluated for their potential to replace fishmeal. Nevertheless, comprehensive examinations on the gut health of aquatic animals under an alternate feeding strategy when fed novel protein diets are vacant. Methods: Five isonitrogenous and isolipidic diets containing various proteins were manufactured, with a diet consisting of whole fishmeal serving as the control and diets containing novel proteins serving as the experimental diets. Largemouth bass (Micropterus salmoides) with an initial body weight of 4.73 ± 0.04g employed as an experimental animal and given these five diets for the first 29 days followed by a fishmeal diet for the next 29 days. Results: The results of this study demonstrated that the growth performance of novel protein diets in the second stage was better than in the first stage, even though only the C. vulgaris diet increased antioxidant capacity and the cottonseed protein concentrate diet decreased it. Concerning the intestinal barriers, the C. autoethanogenum diet lowered intestinal permeability and plasma IL-1β/TNF-α. In addition, the contents of intestinal immunological factors, namely LYS and sIgA-like, were greater in C. vulgaris than in fishmeal. From the data analysis of microbiome and metabolome, the levels of short chain fatty acids (SCFAs), anaerobic bacteria, Lactococcus, and Firmicutes were significantly higher in the C. autoethanogenum diet than in the whole fishmeal diet, while the abundance of Pseudomonas, aerobic bacteria, Streptococcus, and Proteobacteria was lowest. However, no extremely large differences in microbiota or short chain fatty acids were observed between the other novel protein diets and the whole fishmeal diet. In addition, the microbiota were strongly connected with intestinal SCFAs, lipase activity, and tight junctions, as shown by the Mantel test and Pearson’s correlation.Discussion: Taken together, according to Z-score, the ranking of advantageous functions among these protein diets was C. autoethanogenum diet > C. vulgaris diet > whole fishmeal diet > cottonseed protein concentrate > T. molitor diet. This study provides comprehensive data illustrating a mixed blessing effect of novel protein diets on the gut health of juvenile largemouth bass under an alternate feeding strategy.
