Dual-mode sensor based on the synergy of magnetic separation and functionalized probes for the ultrasensitive detection of Clostridium perfringens

Wenzhuo Wang,Wei Yuan,Debao Wang,Xutao Mai,Daoying Wang,Yongzhi Zhu,Fang Liu,Zhilan Sun

Clostridium perfringens is an important foodborne pathogen, which has caused serious public health problems worldwide. So, there is an urgent need for rapid and ultrasensitive detection of C. perfringens. In this paper, a dual-mode sensing platform using the synergy between fluorescent and electrochemical signals for Clostridium perfringens detection was proposed. An electrochemical aptasensor was constructed by a dual-amplification technology based on a DNA walker and hybridization chain reaction (HCR). When the C. perfringens genomic DNA was present, it specifically bonded with FAM-labeled aptamer which triggered the DNA walker on hairpin DNA (hDNA) tracks to start the synthesis of double-stranded DNA. HCR occurred subsequently and produced long-chain DNA to absorb more methylene blue (MB). In this cycle, the fluorescent signals of released FAM-labeled aptamer could also be detected. The synergistic effects of MB and FAM significantly improved the sensitivity and accuracy of the dual-mode sensor. As a result, the biosensor displayed an excellent analytical performance for C. perfringens at a concentration of 1 to 108 CFU g−1. A minimum concentration of 1 CFU g−1 and good accuracy were detected in real samples. The proposed ultrasensitive detection method for detecting C. perfringens in food showed great potential in controlling foodborne diseases.

A new method for rapid, portable, low-cost detection of Clostridium perfringens β2 toxin in animal fecal using smartphone-based electrochemical immunosensor

Beiying Yang,Xiu Zeng,Yu Ge,Weining Liu,Wenxue Hao,Chunyingzi Long,Linkang Wang,Qianyue Wu,Yangping Wen,Jinhua Zhang

Clostridium perfringens (Cp) β2 toxin poses a significant threat to the breeding industry because of its potential to cause enterotoxaemia in livestock. However, existing quantitative methods are very scarce. Thus, we developed a rapid, portable, low-cost quantitative detection method for Cp β2 toxin using a smartphone-based electrochemical immunosensor. Recombinant 32a-β2 toxin protein was successfully prepared as an antigen, as well as its monoclonal antibody. As an alternative to the traditional three-electrode system, a ternary nanocomposite-modified commercially integrated three-electrode chip was fabricated. A smartphone-based electrochemical immunosensor was constructed by combining a low-cost handheld potentiostat with a smartphone, and it was compared with a large-scale potentiostat. Under optimal conditions, the portable sensor for identifying Cp β2 strain expression displayed a linear relationship from 0.1 to 100 ng mL −1 , a low detection limit of 16.45 pg mL −1 , high sensitivity, a low cost, easy operation, outdoor rapid detection, and satisfactory practicability and reliability.

A novel strategy for rapid, portable, and sensitive detection of Clostridium perfringens cpb2 gene using smartphone-based electrochemical DNA biosensor based on screen-printed electrodes modified with nanocomposite

Wu Qianyue,Yang Beiying,Wang Yutang,Xiong Yanyan,Zhang Jinhua,Wen Yangping,Rajkovic Andreja

An accurate, sensitive, low-cost, portable, and easy-to-use method for the quantitative detection of Clostridium perfringens (Cp), a zoonotic pathogen widely found in nature and capable of spreading through contaminated food or environments, is essential for epidemiology, prevention, and diagnostics. Here, we have designed a smartphone-based electrochemical DNA biosensor, which utilizes carboxyl-functionalized multi-walled carbon nanotubes (COOH-MWCNTs) as the substrate, L-cysteine (L-Cys) as the linker, and a synergistic modification of the bioelectrode with gold nanoparticles (AuNPs) and polyamidoamine dendrimers (PAMAM). This design enables highly sensitive, low-cost, label-free, and portable detection of the cpb2 gene in Cp. The sensor not only enhances detection performance but also improves convenience and practicality. Differential pulse voltammetry (DPV) was used to monitor the electrochemical signal response to changes in target DNA concentration, enabling the tracking of the DNA hybridization process. Under optimized conditions, the biosensor exhibited a linear detection range from 10⁻ 14 to 10⁻⁸ M, with a detection limit of 1.5 fM. It demonstrated excellent selectivity for the cpb2 gene and was successfully applied to detect variations in cpb2 gene content in wastewater and fecal DNA samples. Compared with the traditional method, the detection time of this method is short, the operation of professional and technical personnel is not required, the instrument is small and portable, and the single detection cost is significantly reduced. This study provides a new strategy for the rapid, portable, and highly sensitive detection of bacterial toxin genes in livestock and aquaculture. Graphical

Fermented Maillard reaction product alleviates injurious effects in colon caused by Clostridium perfringens

Yujin Kim,Sejeong Kim,Heeyoung Lee,Nam-Su Oh,Min Suk Rhee,Yohan Yoon

Clostridium perfringens is an important bacterium among commensal bacteria in the gastrointestinal tract , which is known to possess potential pathogenicity during its inhabitation. In concordance with the concerns raised by its pathogenicity, this study aimed to elucidate if FMRP (fermented Maillard reaction products), the novel milk-derived material, can reduce the effect caused by the overabundance of C. perfringens in the gut. As a result, FMRP significantly reduced mRNA expression levels in the inflammatory cytokines (interleukin-10, interleukin-11, and interferon-γ) (p<0.05), and tumorigenic potential by activating the cell proliferation-suppressing factors (β-catenin, adenomatous polyposis coli , and E-cadherin) (p<0.05) and deactivation of the cell proliferation-inducing factor (cyclooxygenase-2) (p<0.05) in the colon. Moreover, the tight junction proteins (zonula occludens-1 and claudin-3) were recovered (p<0.05), and the antioxidant enzymes (catalase, superoxide dismutase, and glutathione peroxidase) were decreased by FMRP (p<0.05). However, biochemical analyses showed no significant difference. These results indicate that C. perfringens might cause inflammation, oxidative stress , disruption of tight junctions and abnormal cell proliferation to the colon, but FMRP may reduce inflammation and generation of reactive oxygen, and recover tight junction in the colon.

Pharmacokinetics and pharmacodynamics of isopropoxy benzene guanidine against Clostridium perfringens in an intestinal infection model

Lu Yixing,Yang Liuye,Zhang Wanying,Li Jie,Peng Xianfeng,Qin Zonghua,Zeng Zhenling,Zeng Dongping

This study aimed to evaluate the antibacterial activity of isopropoxy benzene guanidine (IBG) against C. perfringens based on pharmacokinetics/pharmacodynamics (PK/PD) modeling in broilers. The PK parameters of IBG in the plasma and ileal content of C. perfringens-infected broilers following oral administration at 2, 30, and 60 mg/kg body weight were investigated. in vivo PD studies were conducted over oral administration ranging from 2 to 60 mg/kg and repeated every 12 h for 3 days. The inhibitory Imax model was used for PK/PD modeling. Results showed that the MIC of IBG against C. perfringens was 0.5–32 mg/L. After oral administration of IBG, the peak concentration (Cmax), maximum concentration time (Tmax), and area under the concentration-time curve (AUC) in ileal content of broilers were 10.97–1,036.64 mg/L, 2.39–4.27 h, and 38.31–4,266.77 mg·h/L, respectively. After integrating the PK and PD data, the AUC0 − 24h/MIC ratios needed for the bacteriostasis, bactericidal activity, and bacterial eradication were 4.00, 240.74, and 476.98 h, respectively. For dosage calculation, a dosage regimen of 12.98 mg/kg repeated every 12 h for 3 days was be therapeutically effective in broilers against C. perfringens with MIC ≤ 2 mg/L. In addition, IBG showed potent activity against C. perfringens, which may be responsible for cell membrane destruction. These results can facilitate the evaluation of the use of IBG in the treatment of intestinal diseases in broilers caused by C. perfringens.

Three-mode ratiometric biosensor based on integrated DNA-driven magnetic beads for Clostridium perfringens detection

Wenzhuo Wang,Wei Yuan,Tingting Lin,Zhilan Sun,Fei Liu,Daoying Wang,Fang Liu

A three-mode biosensor incorporated ratiometric (electrochemical/colorimetric, electrochemical/photothermal) into its design was constructed using DNA-driven magnetic beads (DMBs) as a bridge to detect C. perfringens. It further enhances the accuracy of detection results while maintaining compatibility with applications in multiple scenarios. Briefly, the G-quadruplex was combined with aptamer and immobilized onto magnetic beads through amide-bond, resulting in the integration of DMBs. The DMBs and supernatant were separated by magnetic separation when the target was present. Subsequently, the DMBs were utilized to construct the electrochemical biosensor, whereas the supernatant was used to construct colorimetric and photothermal biosensors. The limits of detection the ratiometric biosensor were ultimately reduced to 0.26 and 0.27 lg CFU g −1 , respectively, in comparison to the single three-mode biosensor. Moreover, this biosensor had been applied in real-sample assays successfully. The establishment of this platform provides a new method for detecting pathogens in the fields of food safety and environmental monitoring.

Effects of dielectric barrier discharge cold plasma and EGCG on inactivation of Clostridium perfringens spores and quality changes of salt-baked chicken

Yue Wang,Yilin Lin,Xiaoqing Li,Shengpeng Liu,Xiuhua Li,Yigang Yu

Spore-forming microorganisms present a significant challenge to food safety and spoilage prevention. This study investigated the effects of dielectric barrier discharge cold plasma (CP) and epigallocatechin gallate (EGCG) on the inactivation of Clostridium perfringens spores and quality changes in salt-baked chicken during a 36-day storage. CP treatment achieved spore and total viable count reductions of 0.01 to 0.90 log CFU g −1 and 0.28 to 1.91 log CFU g −1 , respectively, depending on voltage and time. Spores treated with CP significantly lost their heat resistance under sublethal thermal conditions of 80 °C for 10 min. After 90 kV/600 s CP treatment, a minimum spore heat resistance ratio was attained 0.5% in salt-baked chicken matrix. Pyridine-2,6-dicarboxylic acid release and confocal laser scanning microscopy indicated that the CP-induced heat sensitization was associated with spore germination and structural damage. CP, EGCG, and EGCG + CP treatments extended the microbial shelf life, and suppressed the increase in total volatile basic-nitrogen values of salt-baked chicken during storage. Compared to the CP or EGCG treatments, the combined EGCG + CP application exhibited a better effect in delaying lipid oxidation. These outcomes provide scientific support for the application of CP as alternative non-thermal strategies for controlling C. perfringens spores and maintaining food quality in cooked meat products.
