Currently, pulmonary complications such as lung infections during the perioperative period are still the main cause of prolonged hospitalization and death in patients with lung injury due to the lack of effective drugs. Clusterzyme, a kind of artificial enzyme with a high enzyme-like activity and safety profile, exhibits good effects on reducing oxidative stress and immunomodulation. Here, we present the functionalized patches that is administered on the lung airways and rescues the injured organ via clusterzymes. The long-term antioxidant capacity of the patches significantly ameliorated lipopolysaccharide-induced lung function impairment with a significant reduction in lung goblet cell metaplasia and oxidative stress. The inflammatory factors such as cytokines interleukin-1β, interleukin-6, and tumor necrosis factor-α levels decreased by 50%, while the mtDNA copy number increased by 50% and ATP production increased by 100%. Mice lung function was significantly improved, suggesting that the patches can rescue lung injury by modulating oxidative stress and immune responses as well as protecting the mitochondria, providing an avenue for effective intervention of lung injury.

The simultaneous detection of multiple bioanalytes with similar structures has been a long-standing challenge for biological research and disease diagnosis. Bioreceptor-inspired sensor arrays provide an attractive and competitive solution for addressing this challenge, but applying this technique to biosensing in practical application scenarios is complicated by many factors such as the lack of robust probes, interference from the biological matrix, and difficulty for on-site analysis. In this work, by taking advantage of the intrinsic fluorescence and enzyme-mimic properties of gold nanoclusters (AuNCs), we report the design of an innovative ratiometric sensor array toward enhanced fluorescent visual biosensing. With biologically important phosphates as an example, we show that the present fluorescent clusterzyme-based ratiometric sensor array could effectively discriminate and detect eight types of phosphates. In particular, AuNCs with three different emission colors (blue, green, and red) and good peroxidase-mimic properties were employed as the sensing units, and the presence of phosphates affected both the intrinsic fluorescence and the enzymatic activity of these AuNCs, yielding distinct optical responses in a ratiometric manner. Moreover, a portable sensor array was established by further integrating these fluorescent clusterzymes into a hydrogel matrix, which could visually identify different phosphates based on their distinct fluorescence color changes. Consequently, the point-of-care diagnosis of urinary tract infections at different levels was achieved by analyzing urinary microbial ATP via the present strategy. This study illustrates the great potential of clusterzyme-based fluorescent sensor arrays as a promising biosensing platform for disease diagnosis.

The accurate identification and sensitive quantification of heavy metal ions are of great significance, considering that pose a serious threat to environment and human health. Most array-based sensing platforms, to date, utilize nanozymes as sensing elements, but few studies have explored the application of the peroxidase-like activity of clusterzymes in identification of multiple analytes. Herein, for the first time, we developed a clusterzyme sensor array utilizing gold nanoclusters (AuNCs) as sensing elements for five heavy metal ions identification including Hg 2+ , Pb 2+ , Cu 2+ , Cd 2+ and Co 2+ . The heavy metal ions can differentially regulate the peroxidase-like activity of AuNCs, and that can be converted into colorimetric signals with 3,3′,5,5′-tetramethylbenzidine (TMB) as the chromogenic substrate. Subsequently, the generated composite responses can be interpreted by combining pattern recognition algorithms. The developed clusterzyme sensor array can identify five heavy metal ions at concentrations as low as 0.5 μM and their multi-component mixtures. Especially, we demonstrated the successful identification of multiple heavy metal ions in tap water and traditional Chinese medicine, with an accuracy of 100% in blind test. This study provided a simple and effective method for identification and quantification of heavy metal ions, rendering a promising technique for environmental monitoring and drug safety assurance.

The treatment of diabetic wounds is hindered by multiple factors, including Staphylococcus aureus (S. aureus) biofilms (the predominant pathogen in diabetic wound infections), dysregulated immune responses, and insufficient angiogenesis. The complex interactions among these local pathological factors have been insufficiently addressed in the development of existing therapeutic approaches. This study reports a hybrid biocatalytic platform consisting of abundant iridium (Ir) clusters supported on ceria (Ir-CeO2). This architectural configuration enables pH-responsive reactive oxygen species (ROS) catalysis, allowing the biocatalyst to exert concurrent antimicrobial and anti-inflammatory effects, which collectively promote healing of diabetic wounds. Ir-CeO2 displays dual pH-dependent ROS modulation: ROS generation under acidic conditions and ROS scavenging under neutral pH. In S. aureus-infected environments, the biocatalyst exerts potent antibacterial effects via catalytic ROS production. Following pathogen clearance, Ir-CeO2 continues to play a bifunctional role by alleviating oxidative stress in inflamed wounds, promoting the polarization of macrophages toward the M2 phenotype-a critical event for accelerating wound healing. Collectively, these results highlight the multifaceted therapeutic efficacy of Ir-CeO2, which seamlessly integrates potent antibacterial activity against S. aureus infections with anti-inflammatory and pro-angiogenic functions. This synergistic profile establishes Ir-CeO2 as a highly promising agent for the holistic management of complicated diabetic ulcers, thereby addressing key shortcomings of existing treatment modalities.
