Anthracnose is a highly contagious and destructive plant disease caused by Colletotrichum gloeosporioides . The objective of this study was to investigate the antifungal activity and mechanism of ginsenoside Rk1 ( GRk1 ) against C. gloeosporioides. In vitro experiments demonstrated that GRk1 exhibited substantial antifungal activity, with EC 50 values of 64.58 mg L −1 , surpassing the plant-derived pesticide d -limonene . Additionally, GRk1 could effectively suppress the dissemination of anthracnose spots in postharvest fruit. By observing the microstructure of C. gloeosporioides , it was observed that the mycelium structure underwent notable changes, and autophagosomes engulfed the organelles in the experimental groups. Fluorescent staining revealed specific signals in mycelial autophagosomes, and Western Blot analysis confirmed the upregulation of the autophagy-related protein Atg8. These findings indicate that GRk1 may activate the autophagy process by promoting Atg8 expression. Proteomic analysis revealed that the expression of Sec20 and Mpv17 proteins was significantly downregulated, whose dysregulated expression is associated with the crosstalk between SNAREs and peroxisomes during vesicle trafficking and metabolic processes. Assessment of catalase (CAT) activity, hydrogen peroxide (H₂O₂) content, and superoxide anion (O₂ − ) levels further demonstrated that GRk1 could disrupt intracellular reactive oxygen species (ROS) homeostasis in mycelia. Moreover, the introduction of the ROS scavenger N -acetylcysteine (NAC) markedly inhibited autophagy, indicating that GRk1 promotes autophagy by inducing oxidative stress. Collectively, GRk1 inhibits pathogens via the ROS-autophagy axis. These findings suggested that GRk1 holds potential as a promising alternative to fungicides for the biological control of anthracnose.

Colletotrichum gloeosporioides , the primary fungal pathogen responsible for mango anthracnose, causes significant economic losses. Hence, we developed a xanthan gum/hydroxypropyl methylcellulose (XG/HPMC) film incorporating Dictyophora echinovolvata carbon quantum dots (DE-CQDs). These green-synthesized DE-CQDs are 2 nm spherical nanoparticles with good crystallinity and thermal stability, featuring functional groups such as -OH, -COOH, -NH₄ + , and -SO₃H, which enhance hydrophilicity, solubility, and charge properties. The negative charge of DE-CQDs, as evidenced by zeta potential measurements, enhances their dispersibility and inhibits aggregation, thereby contributing to the film's augmented UV-blocking and water vapor transmission characteristics. DE-CQDs inhibit C. gloeosporioides growth through a unique mechanism, inducing metacaspase-dependent apoptosis and causing ROS accumulation without DNA fragmentation. When incorporated into the XHC film, DE-CQDs improve UV-blocking by 30 % at 400 nm, enhance water vapor transmission, and increase water contact angle. The XHC film significantly reduces mango decay (decay index: 0.35 ± 0.05 to 0.21 ± 0.05) and boosts vitamin C retention (from 30 % to 61 % over 7 days), effectively extending shelf life and preserving nutritional quality. Among the XHC films, XHC6 shows the best overall performance. Thus, DE-CQDs serve as functional fillers in biodegradable, environmentally friendly packaging films, offering a novel solution for mango disease prevention and control.

Poplar anthracnose caused by Colletotrichum gloeosporioides is one of the major forest diseases worldwide, causing incalculable losses in forestry production every year. In this study, an endophytic bacterium with significant antagonistic activity against C. gloeosporioides was isolated from Populus canadensis leaves, identified and named Pseudomonas atacamensis GZ-3. The specific biological functions of strain GZ-3 were analyzed using whole genome sequencing. The volatile organic compounds (VOCs) produced by strain GZ-3 had good antifungal activity against C. gloeosporioides . These VOCs could effectively inhibit mycelial growth and conidial germination of C. gloeosporioides . In addition, metabolomics analysis revealed that VOCs could help poplar resist infection of C. gloeosporioides by inducing the synthesis of flavonoids in poplar. 3-methyl-1-butanol and 2-heptanone were the main antifungal compounds in VOCs. Therefore, Pseudomonas atacamensis GZ-3 had good potential for biocontrol. This study provided a new strategy for the biological control of poplar anthracnose.

Anthracnose is a plant disease caused by Colletotrichum spp ., known for its widespread infectivity and extreme destructiveness. Colletotrichum gloeosporioides is a representative pathogen of anthracnose in China. Gypenosides GP4 - GP7 , derived from Gynostemma pentaphyllum (Thunb.) Makino, could significantly inhibit the growth of C. gloeosporioides mycelial, with EC 50 values of 96.98, 27.5, 38.48, and 61.59 mg L −1 . The inhibitory effect of these compounds surpassed the commonly used chemical pesticide chlorothalonant and plant-derived pesticide matrine. Among them, the most active compound GP5 also showed a significant inhibitory effect on spore germination and bud tube elongation of C. gloeosporioides . In addition, GP5 could effectively suppress the spread of anthracnose spots in postharvest fruit. Transmission electron microscopy and fluorescence microscopy demonstrated that GP5 primarily exerted its antifungal function by activating cellular autophagy. Additionally, proteomics analysis revealed that GP5 had an antifungal effect against C. gloeosporioides by enhancing cellular autophagy through upregulation of the expression of the autophagy-related protein Atg8. This study presents a novel approach for the control and management of anthracnose in C. gloeosporioides . Consequently, GP5 has the potential to be developed as a plant-derived fungicide for the biological control of anthracnose.

Pogostone, a major constituent of Pogostemon cablin, displays various biological properties. Here, we systematically assessed the antifungal and preservative activities of pogostone against Colletotrichum gloeosporioides in mango and explored its antifungal mechanism. Pogostone exhibited strong in vitro and in vivo inhibition against C. gloeosporioides, with EC50 values of 2.50 mg/L for hyphal growth and 3.23 mg/L for spore germination. Pogostone prolonged mango shelf life by reducing weight loss, maintaining firmness, and regulating TSS, TA, VC, malondialdehyde, and antioxidant enzyme activities. We also found that pogostone affected the microstructure of fungal hyphae, disrupting the integrity of the plasma membrane and ultimately causing leakage of cellular contents. Moreover, with increasing concentration and exposure time, the mode of cell death shifted from apoptosis to necrosis. In summary, this study showed that pogostone was a promising natural preservative against C. gloeosporioides in mangoes and enhanced our understanding of its antifungal mechanism.

Although much progress has been made in developing botanical fungicides to combat fungal diseases in crops, there remains a great need to improve the efficiency and long-term safety of these fungicides. This study proposes a novel strategy for designing citral-thiourea derivatives that feature such desirable properties. The motivation of the work herein was to enhance the antifungal activity of citral against C. gloeosprioides by exploiting the synergistic effect that arises from combining citral and thiourea compounds, thereby producing citral-thiourea derivatives that exhibit good long-term safety. The results revealed that the generated compounds e1, e3, e6, e18, and g showed remarkable antifungal activities against C. gloeosprioides, with corresponding EC50 values reaching 0.16, 1.66, 1.37, 4.76, and 4.60 mg/L, respectively, showing that the compounds significantly outperformed both the positive control kresoxim-methyl and the commercially available fungicide carbendazim. Furthermore, compound g showed stronger protective efficacy against C. gloeosprioides than carbendazim on mango fruit at 25 mg/L. Investigating the preliminary structure–activity relationship (SAR) of the compounds also revealed that the citral-thiourea derivatives exhibited higher antifungal activities against C. gloeosprioides compared to citral and thiourea compounds. This reinforcement of antifungal activity observed in the derivatives was found to be attributable to the two characteristics of low molecular size and the presence of a fluorine atom in the meta-position of the benzene ring. Beyond this, it was determined from QSAR that two molecular descriptors (the Kier–Hall index (order 3) and Tot dipole of the molecules) were negatively related to the antifungal activity of the citral-thiourea derivatives, while one other (the maximum resonance energy of a C–H bond) was positively related to their antifungal activity. More importantly, the citral-thiourea derivatives with high antifungal activities (i.e., compounds e1, e3, e6, e14, e15, e18, and g) exhibited negligible cytotoxicity to LO2 and HEK293T cell lines. The antifungal mechanism of the generated citral-thiourea derivatives was investigated by scanning electron microscopy (SEM) and relative conductivity. The derivatives were found to affect mycelial morphology and increase fungal cell membrane permeability, thereby resulting in the destruction of fungal cell membranes. These promising results provide novel insights into the study and potential application value of citral-thiourea derivatives as high-efficiency antifungal agents against C. gloeosprioides.

Actinomycetes have emerged as significant biocontrol resources due to their rich array of bioactive natural products. While much research has historically focused on secondary metabolites isolated from their fermentation broth, there remains a dearth of reports on their volatile organic compounds (VOCs). Here, strain ML27, isolated from soil, was identified as Streptomyces albidoflavus based on morphological features, physiological, biochemical, and molecular characteristics ( 16S rRNA , atpD , recA , and rpoB gene sequences). VOCs from S. albidoflavus strain ML27 were effectively captured using solid-phase microextraction (SPME) and tentatively identified through gas chromatography–mass spectrometry (GC/MS). Among these compounds, 4-ethyl-1,2-dimethoxybenzene exhibited broad-spectrum antifungal activity and demonstrated efficacy in controlling citrus anthracnose, with a control efficacy of 86.67%. Furthermore, the inhibitory mechanism of 4-ethyl-1,2-dimethoxybenzene against Colletotrichum gloeosporioides was revealed. Results indicated that 4-ethyl-1,2-dimethoxybenzene induced swelling, deformity, and breakage in C . gloeosporioides mycelia, and significantly inhibited spore germination. Transcriptome analysis revealed that 4-ethyl-1,2-dimethoxybenzene inhibited the growth and development of C. gloeosporioides primarily by disrupting energy metabolism and the integrity of the cell wall and membrane. Based on these results, it is promising to develop 4-ethyl-1,2-dimethoxybenzene as a novel biopesticide for controlling citrus anthracnose.

The yeast Debaryomyces nepalensis was evaluated for effectiveness against Colletotrichum gloeosporioides in mangoes ( Mangifera indica L.) cv. ‘Tainong No. 1’, and possible mechanisms of action involved in the inhibition were identified. The results showed that D. nepalensis could survive in mango wounds and produce a hydrolytic enzyme (β-1,3-glucanase). Scanning electron microscopy showed that the yeast produced biofilms on the mango surface and yeast cells attached to each other and became embedded in the pathogen hyphal walls. In addition, D. nepalensis competed for nutrients (FeCl 3 ) and competed for space with the pathogen (p < 0.05). Morever, D. nepalensis enhanced defense-related enzymes of wounded mangoes, including peroxidase, β-1,3-glucanase, chitinase and phenylalanine ammonia-lyase. In addition, gas chromatographic-mass spectrometry (GC–MS) analysis of volatile compounds (VOCs) produced by the yeast showed that VOCs can play important roles over short distances in the suppression of C. gloeosporioides. The solid-phase microextraction (SPME) coupled with GC–MS technique analysis revealed phenylethyl alcohol (29.62% relative peak area, RA) as the dominant VOC mainly produced by D. nepalensis , and it had a significant inhibitory effect on fungal mycelia. According to the results obtained here, the combination of different mechanisms of action of the yeast increases their ability to control C. gloeosporioides. The use of antagonistic yeast to control C. gloeosporioides may contribute to the integrated management of disease caused by this pathogen.

In this study, we used virus-mediated gene silencing technology and found that the HSP17.4 gene-silenced cultivar Sweet Charlie plants were more susceptible to Colletotrichum gloeosporioides than the wild-type Sweet Charlie, and the level of infection was even higher than that of the susceptible cultivar Benihopp. The results of differential quantitative proteomics showed that after infection with the pathogen, the expression of the downstream response genes NPR1 , TGA , and PR-1 of the salicylic acid (SA) signalling pathway was fully up-regulated in the wild-type Sweet Charlie, and the expression of the core transcription factor MYC2 of the jasmonic acid (JA) pathway was significantly down-regulated. The expression of the proteins encoded by these genes did not change significantly in the HSP17.4 -silenced Sweet Charlie, indicating that the expression of HSP17.4 activated the up-regulation of downstream signals of SA and inhibited the JA signal pathway. The experiments that used SA, methyl jasmonate, and their inhibitors to treat plants provide additional evidence that the antagonism between SA and JA regulates the resistance of strawberry plants to C . gloeosporioides .

Colletotrichum leaf disease (CLD), mainly caused by the Colletotrichum gloeosporioides species complex (CGSC), is a serious disease of rubber tree in most rubber-growing countries. Few fungicides are available for rubber farmers to manage this disease. To control CLD effectively, it is important to test the baseline sensitivity and efficacy of fungicides against CGSC isolates. Penthiopyrad is a new succinate dehydrogenase inhibitor (SDHI) that has been registered for use in many countries. In this study, baseline sensitivity to penthiopyrad was established by mycelial growth methods using 91 CGSC isolates collected from rubber trees in China. The EC 50 values of penthiopyrad ranged from 0.243 to 3.377 mg/L with a mean EC 50 value of 1.685 mg/L. Penthiopyrad greatly decreased the mycelial biomass and increased cell membrane permeability of CGSC isolates. The mycelia were severely distorted, and the number of top branches increased with penthiopyrad treatment. Penthiopyrad inhibited succinate dehydrogenase activity, causing a decrease in the ATP content of CGSC isolates, and significantly inhibited conidia germination and appressorium formation on rubber tree leaves. The protective and curative efficacy of 100 mg/L penthiopyrad reached up to 98.53% and 66.79% against CLD, respectively. Our results demonstrated the high inhibitory activity of penthiopyrad against CGSC isolates and its potential for CLD management on rubber tree.

Our previous study found that 1-(4-bromophenyl)-5-phenyl-1H-1,2,3-triazole (BPT) exhibited very low toxicity and higher biology activity. In the present study, BPT was used to investigate its mechanism against Colletotrichum gloeosporioides in mango by chemical proteomics and chemicobiological methods. The experimental results of some phenotypic traits and cytotoxicity indicated that there are some changes in permeability of the cell membranes and BPT has fungistatic activity with lower cytotoxicity. Next, a biotin probe named BPTN with a higher activity (EC 50 value of 11.08 mg/L) than that of BPT(EC 50 value of 14.13 mg/L) was designed and synthesized by the chemical proteomics approach. The major differentially expressed proteins between BPT or BPTN and Colletotrichum gloeosporioides were identified and analyzed by Matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOF-MS) and bioinformatics. The results indicated that there are 25 differentially expressed proteins in Colletotrichum gloeosporioides under BPT intervention, and 29 target proteins in Colletotrichum gloeosporioides captured by BPTN. For BPTN-anthracnose system, the protein No. 8 belonged to cytochrome P450 proteins (CYP) family can be taken as a target protein of BPTN, which confirmed by bioinformatics analysis. Further, a classic fungal target protein of triazoles, sterol 14α-demethylase (CYP51) was used to verify the interaction in the presence of BPT by various spectroscopic and molecular docking methods. The results showed that BPT exhibited strong interactions with CYP51 and it was predominantly driven by hydrophobic forces and hydrogen bonds. The present study has significant theoretical and practical implications for the development of novel 1,2,3-triazole antifungal agents.
