Lead accumulation (adsorption and absorption) by the freshwater bivalve Corbicula fluminea in sediments contaminated by TiO2 nanoparticles

Xiulei Fan,Peifang Wang,Chao Wang,Bin Hu,Xun Wang

With the increasing production and applications of TiO 2 nanoparticles (NPs), their presence in aquatic environments, especially in sediments, will inevitably increase over time. Most studies investigating the influence of TiO 2 NPs on the bioaccumulation of co-existing contaminants have focused on the aqueous phase; however, few have examined the sediment phase, which contains more TiO 2 NPs and contaminants. We investigated the effects of TiO 2 NPs on Pb accumulation by Corbicula fluminea in sediments, and explored extracellular and intracellular Pb concentrations in the various soft tissues of the bivalve. Pb was spiked with 50 mg/kg in sediment and TiO 2 NPs/sediments ratios were within the range 0.2–3.0%. The results showed that TiO 2 NPs presented larger adsorption capacity and affinity to Pb ions than the sediments. In addition, the large adsorption capacity of TiO 2 NPs and the strong adsorption affinity to Pb ions caused part of the Pb ions released from sediments to aqueous phase were re-adsorbed by TiO 2 NPs in sediments. The concentration of TiO 2 NPs in C. fluminea tissues significantly increased with increasing TiO 2 NP content in sediments, following the order: gill > mantle > foot > visceral mass, which differed from the results found in the aqueous phase. In addition, the proportions of extracellular and intracellular Pb concentrations changed significantly in all the tissues as a result of TiO 2 NP contamination of sediments. TiO 2 NPs promote increased extracellular Pb in foot, mantle, and gill tissues, and increased intracellular Pb in the visceral mass. These results may be beneficial to more scientifically evaluate and predict the environmental risks of TiO 2 NPs to benthic organisms in sediments contaminated by heavy metals.

Combined toxicity of organophosphate flame retardants and cadmium to Corbicula fluminea in aquatic sediments

Dandan Li,Peifang Wang,Chao Wang,Xiulei Fan,Xun Wang,Bin Hu

Organophosphate flame retardants (OPFRs), as alternatives to polybrominated biphenyl ethers (PBDEs), are frequently detected in various environmental matrices. Owing to urbanization and industrial pollution, co-contamination of OPFRs and heavy metals is ubiquitous in the environment. The toxicity of OPFRs in aqueous phase is a significant concern, but uncertainty still exists regarding the co-toxicity to benthic organisms of OPFRs and metals in sediments. Hence, we explored the physiological response of Corbicula fluminea to OPFRs and Cd in sediments. The results indicated that the antioxidant system in the clams was stimulated in the presence of OPFRs and Cd, and the oxidative stress increased with increasing concentrations of OPFRs. In contrast, the cytochrome P450 (CYP450) content and acetylcholinesterase (AChE) activity were reduced by exposure to both OPFRs and Cd. The cytochrome P450 4 family (CYP4) mRNA expression and OPFR toxicity were lower than those in previously reported experiments conducted in the water phase. Moreover, the expression levels of metallothionein (MT) and AChE mRNA decreased when OPFRs and Cd were present together. The highest integrated biomarker response (IBR) index (IBR = 15.41) was observed in the presence of 45 mg kg −1 Cd + 200 mg kg −1 OPFRs, rather than the 45 mg kg −1 Cd + 400 mg kg −1 OPFRs treatment (IBR = 9.48). In addition, CYP450 and AChE in the digestive glands of C. fluminea exhibited significant correlations with the concentration of the OPFR/Cd mixture (p < 0.01) and could be effective biomarkers for OPFR and Cd co-contamination. The results potentially contribute to more realistic predictions and evaluations of the environmental risks posed by OPFRs in aquatic sediments contaminated with heavy metals, particularly with respect to the risk to benthic organisms.

Evidence that cadmium aggravate the toxicity of triphenyl phosphate in aquatic sediments to Corbicula fluminea

Dandan Li,Chen Xie,Ziwu Fan,Rui Ding,Xiaoyu Wang,Yipeng Liao

The ubiquitous co-existence of triphenyl phosphate (TPhP) and heavy metals in sediments raises significant biotoxicity concerns. However, uncertainty still exists regarding their combined toxicity to benthic organisms. Therefore, this research was conducted to elucidate the influences of cadmium (Cd) on TPhP toxicity to Corbicula fluminea ( C. fluminea ) in sediments. As a result, Cd promoted the accumulation of TPhP in C. fluminea and enhanced TPhP toxicity, manifested by damaged cell membranes and pronounced histological alterations. Molecular docking revealed that TPhP-Cd complexes exhibit greater binding affinity to cytochrome P4501A1 (CYP1A1) compared to TPhP alone. With the activity of CYP1A1 increasing, the biotransformation of TPhP was promoted in low-TPhP+Cd treatments (T5C0/T5C5/T5C35). Additionally, metabolites related to antioxidant defence and repair processes were reinforced to alleviate the toxicity of TPhP and Cd. However, excessive oxidative stress impaired the CYP1A1 activity in high-TPhP+Cd treatments (T35C0/T35C5/T35C35). Furthermore, metabolic pathway analysis revealed significant perturbations in the citrate cycle , alanine, aspartate and glutamate metabolism , purine metabolism, and pyrimidine metabolism . These disruptions weakened the repair capacity and aggravated apoptosis in digestive glands, potentially contributing to the synergistic toxicity of TPhP and Cd. The results highlight the ecological risks posed by TPhP in combination with heavy metals to benthic organisms.

Mechanistic Insights into Cadmium Sulfide Nanoparticles-Induced Digestive Gland Damage in Corbicula fluminea: Comparison with Cadmium Ions

Kena Zhang,Quanzhi Xiao,Jianbo Jia,Yan Gao,Yanfei Zhou,Xiao-Xia Zhou,Bing Yan

The extensive use of cadmium sulfide nanoparticles (CdS-NPs), along with their natural formation through the complex biogeochemical transformation of anthropogenic cadmium ions (Cd2+), poses substantial risks to ecosystems and human health. Despite this, the mechanisms underlying the toxicity of CdS-NPs remain unclear. A key question is whether their toxicity arises from the nanoparticulate form of cadmium (Cd) or from the release of Cd2+. To explore this, we exposed freshwater clams (Corbicula fluminea) to environmentally relevant concentrations (0.01–1 mg/L) of CdS-NPs or Cd2+ for 10 days. Hematoxylin and eosin (HE) staining revealed significant damage to the digestive gland in both cases. Although CdS-NPs released some Cd2+ (≤10.4%), transcriptomic and quantitative reverse transcription polymerase chain reaction (qRT-PCR) analyses indicated different toxicity mechanisms. CdS-NPs primarily induce ferroptosis, triggered by lysosomal dysfunction that releases Fe2+ into the cytoplasm, disrupting the cellular iron metabolism. In contrast, Cd2+ primarily induces an autophagic response, as evidenced by the upregulation of autophagy-related markers and activation of apoptosis pathways linked to mitochondrial membrane permeabilization. Overall, our findings suggest that the toxicity of CdS-NPs is not solely derived from Cd2+, highlighting the need to evaluate the risks posed by metal sulfide nanoparticles to benthic ecosystems.

TiO2 nanoparticles in sediments: Effect on the bioavailability of heavy metals in the freshwater bivalve Corbicula fluminea

Xiulei Fan,Chao Wang,Peifang Wang,Bin Hu,Xun Wang

Most studies investigating the influence of TiO 2 NPs on heavy metal bioavailability have focused on the aqueous phase; however, few have examined the sediments containing more nanoparticles. Here, we investigated the effects of TiO 2 NPs on heavy metal bioavailability in C. fluminea in sediments. The interactions between the TiO 2 NPs and metals in sediments, the influence of TiO 2 NPs on metals levels in aqueous phase and geochemical speciation were also explored. The results indicated the large adsorption capacity of TiO 2 NPs and the strong adsorption affinity to metals caused the metals adsorbed on nanoparticles, which decreased the metals concentrations in water phase. Changes in metal speciation caused by metals in EXC, CAR, and IMO partly transported from sediments to TiO 2 NPs during the aging of sediments. Heavy metals contents in C. fluminea tissues were in the order of gill   >   visceral mass   >   mantle   >   foot and increased with the increasing TiO 2 NPs contents in sediments. TiO 2 NPs enhanced the bioavailability of metals in the speciation of EXC, CAR, and IMO in sediments by the Trojan horse effects. The results can facilitate a more realistic evaluation of the environmental risks of TiO 2 NPs to benthic organisms in heavy metal-contaminated sediments.

Effect of ibuprofen on the behavior, histopathology, antioxidant system, DNA damage, and apoptosis of Asian freshwater clams (Corbicula fluminea)

Yun-xia Hou,Xiang-sheng Hong,Shi-yin Tang,Wang Liu,Zhi-tong Li,Juan Wang,Sai-hong Yan,Jian-hui Qin,Jin-miao Zha

The accumulation of ibuprofen (IBU) in freshwater sediments poses significant risks to benthic bivalves, yet its chronic ecological impacts remain insufficiently studied. Here, Asian clams ( Corbicula fluminea ) were exposed to 20, 200, and 2000 µg/L IBU for 28 days. The viscera were identified as the primary target organ for IBU toxicity, with bioconcentration factors (BCFs) ranging from 3.03–6.05 across treatments and a 1.3- to 3.3-fold increase in in digestive gland vacuolation damage. Chronic IBU exposure also induced neurotoxicity, evidenced by acetylcholinesterase inhibition, and potentially associated behavioral disturbances. Filtration and burrowing rates decreased significantly at 200 and 2000 µg/L, while escape rates increased, reflecting a trade-off between avoidance and burrowing under stress. At 20 µg/L, escape fluctuations were most pronounced, suggesting sub-threshold behavioral modulation. The trade-off between escape and burrowing highlights the adaptive yet costly responses of bivalves to pharmaceutical stress. Transcriptomic profiling demonstrated marked enrichment of apoptotic and pro-inflammatory pathways. Notably, while antioxidant defenses remained unaltered in clams, IBU exposure consistently upregulated COX-2 activity, with concomitant induction of genotoxic effects evidenced by DNA strand breakage and activation of caspase-9 mediated intrinsic apoptotic pathways. This work provides critical insights for managing pharmaceutical contamination in freshwater ecosystems and protecting benthic biodiversity and ecosystem functions.

Integration of physiological, transcriptomic and metabolomic analyses reveals acute carbonate alkalinity stress responses in the gill of Asian clam (Corbicula fluminea)

Ting Zhang,Junlei Ma,Yidong Liang,Yicheng Li,Jinshuo Liang,Yanfeng Zhou,Dongpo Xu

Aquaculture is an effective approach to improve the utilization of saline-alkaline water. Corbicula fluminea is an important economically freshwater bivalve species in China, capable of adapting to certain levels of salinity and alkalinity. However, the mechanisms underlying its acute response to alkalinity remain unexplored. In this study, we investigated the effects of alkalinity (0, 10 and 20 mmol/L) on the gill tissue of C. fluminea (54 individuals, 22.18 ± 1.9 mm in shell length and 4.41 ± 0.66 g), employing integrated physiological, transcriptomic, and metabolomic analyses to elucidate its early responses. Histological and physiological results showed that alkalinity induced damage to gill tissue and significant physiological stress, evidenced by marked changes in indicators related to osmoregulation, oxidative stress, and immune response. Transcriptomic analysis identified many differentially expressed genes (DEGs) related to ion transport, oxidative stress, apoptosis, immune response, and metabolism. Metabolomic analysis revealed that differential metabolites (DMs) were predominantly lipids and lipid-like molecules. Integrated transcriptomic and metabolomic analysis revealed that both DMs and DEGs were enriched in pathways related to apoptosis, carbohydrate metabolism, and lipid metabolism. Overall, these results indicated the responses of C. fluminea to acute alkalinity stress involving in ion transport, apoptosis, immnue, and metabolism processes. These research results will accumulate data on the acute stress responses of freshwater bivalves to alkalinity, and provide a theoretical foundation for the saline-alkaline aquaculture of C. fluminea .
