Eleven polyphenols, classified as flavonoid glycosides, flavonoid aglycones, and phenolic acids, are important bioactive components in the capitula of Coreopsis tinctoria (CCT). Nevertheless, their full pharmacokinetic profiles have not been demonstrated simultaneously. Therefore, a liquid chromatography – tandem mass spectrometry (LC/MS/MS) method was developed in the present work and used it to study the pharmacokinetics of these 11 compounds. We performed LC/MS/MS with a gradient mobile phase composed of water containing 0.1 % formic acid and acetonitrile containing 0.1 % formic acid on a Proshell 120 SB C18 column (2.1 mm×100 mm, 2.7 μm). We achieved a good chromatographic peak shape, resolution, and mass signal response, and multiple reaction monitoring facilitated the simultaneous detection of 11 analytes. In addition, we validated the selectivity, correlation coefficient, precision, extraction recovery, matrix effects, and stability of the LC/MS/MS method to be acceptable for 11 analytes in rat plasma. Subsequently, rats were orally administered with 50 % ethanol eluent of CCT (ECCT). Nine of 11 polyphenols were absorbed quickly (except for QCD and TCA), and their plasma levels peaked within 40 min. The exposure and C max values of flavonoid glycosides and phenolic acids were lower than those of flavonoid aglycones. This is the first report to demonstrate the pharmacokinetics of 11 polyphenols in ECCT, which may play an important role in future studies of the bioactive components of ECCT and their bioactive mechanisms. α- Glucosidase is one of the main enzymes causing elevated blood glucose, and Coreopsis tinctoria extract can be used as a natural inhibitor of α -Glucosidase. Therefore, a new method was proposed for predicting the inhibitory activity on α- Glucosidase of Coreopsis tinctoria extract based on near infrared spectroscopy. The absorbance of the inhibitory system was measured by ultraviolet spectroscopy, which was used to study the inhibitory activity on a-glucosidase of Coreopsis tinctoria extract. The near infrared spectra of the solid samples were collected. By selecting spectral preprocessing and optimizing spectral bands, a rapid prediction model of the inhibitory activity was established by partial least squares regression. The root mean square error of cross-validation (RMSECV), correlation coefficient (R) value and the ratio of prediction to deviation (RPD) value were used as indicators of the evaluation model. The near infrared spectrum model was established by combining the best spectral preprocessing of the continuous wavelet transform (CWT) and the best spectral band. The root mean square error of cross-validation (RMSECV) of this model was 0.815%, the correlation coefficient (R) value was 0.942, and the ratio of prediction to deviation (RPD) was 3.0. The root mean square error of prediction (RMSEP) of the model by prediction set was 0.819%, the correlation coefficient (R) value was 0.950, and the RPD was 3.2. The model shows that the fitting relationship between the predicted inhibition value and the reference inhibition value of the near infrared spectral model is good. The results showed that there was a good correlation between near infrared spectroscopy and the inhibitory activity of Coreopsis tinctoria extract. Thus, the established model was robust and effective and could be used for rapid quantification of α -Glucosidase inhibitory activity. The prediction method is simple and rapid, and can be extended to study the inhibition of other medicinal plants.

Coreopsis tinctoria is being widely cultivated in Xinjiang of China, whose consumption is known to prevent diabetes and neurodegenerative diseases. To elucidate the bioactive ingredients responsible for these benefits, the alkaline soluble crude polysaccharide (CTB) was isolated from C. tinctoria . In vitro experiments showed that the inhibition of α -amylase and α -glucosidase by CTB was 13407-fold and 906-fold higher than that by positive control, respectively. Then, a novel arabinogalactan, CTBP-1, was isolated and purified from CTB. Structural analysis showed that CTBP-1 possessed a 1,6-linked β - d -Gal p and 1,5-linked α - l -Ara f backbone with branches substituted at the C-3 position of the 1,6-linked β - d -Gal p , and the side chains included 1,5-linked α - l -Ara f , T-linked β - d -Gal p and T-linked α - l -Ara f . The inhibitory effects of CTBP-1 on α -amylase and α -glucosidase were 2.7 and 17.9 times that of acarbose, respectively. CTBP-1 could avoid indigestion and similar side effects. In addition, CTBP-1 remarkably inhibited the release of nitric oxide (NO), tumor necrosis factor- α (TNF- α ) and interleukin-6 (IL-6) in lipopolysaccharide (LPS)-stimulated BV2 microglial cells. In summary, CTBP-1 is a novel arabinogalactan with great potential as a treatment for type 2 diabetes and Alzheimer’s disease.

This study was aiming at the differences in the aroma, flavor compounds, and bioactivities of essential oil of Coreopsis tinctoria flower (CTFEO) and essential oil of Chrysanthemum morifolium flower (CMFEO). Both essential oils were extracted by steam distillation. Their flavor compositions were determined by GC-MS and the antioxidant, antibacterial, and anti-inflammatory activities were also evaluated. It was found that CTFEO possessed a special medicinal herb odor, which was obviously stronger than those of CMFEO. A preferred condition for steam distillation was flower powder–water ratio of 7.5:100 and extraction for more than 3.0 h. Interestingly, the main flavor compounds of CTFEO were determined as D-limonene (34.54%), cis-carveol (5.49%), trans-α-bergamotene (4.44%), and α-pinene (3.90%), which were quite different from those of CMFEO. CTFEO showed better antioxidant activities with IC50 value of 121.4 ± 9.8 μg/mL than those of CMFEO. It also exhibited stronger antibacterial effects than those of CMFEO, with minimal inhibitory concentrations (MICs) ranging from 0.183 to 1.412 mg/mL against six strains of bacteria. Moreover, CTFEO was also found to exhibit strong anti-inflammatory effects that reduced the LPS-induced RAW 264.7 cell mRNA expression of the cytokines IL-1β, IL-6, and TNF-α at a concentration of 10.0 μg/mL. All these results suggest that the aroma, flavor compounds, and bioactivities of the two essential oils were quite different and they could be used as tea, drink, and food additive in the food industry for further development.

This work investigated the inhibitory effects of Coreopsis tinctorius essential oil (CTEO) and Coreopsis tinctorius microcapsules (CTM) on tyramine accumulation by Enterococcus faecium in pure culture and during the ripening of smoked horsemeat sausage, in order to provide a novel natural additive for the control of tyramine in smoked horsemeat sausage. To evaluate the expression of related-genes in the tyrosine decarboxylation (TDC) pathway, we used reverse transcription-quantitative polymerase chain reaction (RT-qPCR) to determine the expression of tyrosine decarboxylase (tyrDC) and tyrosine/tyramine antiporter (tyrP) genes. Furthermore, the pH, moisture content, microorganisms and tyramine accumulation of sausage were assessed. The results revealed that CTEO and CTM markedly ( P < 0.05) decreased tyramine accumulation by inhibiting E. faecium growth and the expression of tyrDC and tyrP genes. Furthermore, tyramine accumulation in sausage was evidently inhibited by CTEO and CTM, and the inhibitory effect of CTM was more significant ( P <0.05). Hence, CTM addition is a desirable approach to inhibit tyramine accumulation, and enhance the safety of smoked horsemeat sausages.

The antioxidant components of Coreopsis tinctoria have not been clearly identified; thus, here eleven constituents were isolated and identified from the capitula of Coreopsis tinctoria , in which taxifolin ( 7 ), 3,5-di-O-caffeoylquinic acid ( 9 ) and 4,5-di-O-caffeoylquinic acid ( 10 ) were identified for the first time. All eleven compounds possessed antioxidant activities based on DPPH assay and cellular antioxidant activity assay. Especially, okanin ( 11 ) and isookanin ( 6 ) exhibited excellent DPPH scavenging activities with the EC 50 of 6.2 and 10.6 µM, respectively, which were stronger than those of ascorbic acid and butylated hydroxytoluene (positive controls) with the EC 50 of 30.4 and 45.8 µM, respectively. Okanin and isookanin showed comparable cellular antioxidant activities to quercetin with the IC 50 of 11.0 and 14.4 µM, respectively. Additionally, the two most potent antioxidants, okanin and isookanin, were detectable and reached maximal concentration of 10.5 and 10.6 µM in plasma at 20 min after oral administration of 100 mg/kg ethanol extract of Coreopsis tinctoria capitula. In conclusion, our results indicate that Coreopsis tinctoria capitula might be developed as antioxidant sources.

Context Coreopsis tinctoria Nutt (Asteraceae), named snow chrysanthemum, is known to have a high level of polyphenols. However, the potential prebiotic effect on modulating intestinal microflora is still unclear. Objective The chemical composition, antioxidant properties of snow chrysanthemum polyphenols (SCPs) and their effects on human intestinal microbiota were investigated.Materials and methods SCPs were extracted using ultrasonic-assisted extraction, and further determined using UPLC-QE Orbitrap/MS. Five assays were used to investigate the antioxidant activities of SCPs. Subsequently, the effects of SCPs on intestinal microbiota in vitro were determined by high throughput sequencing and bioinformatics analysis. Results Marein, isookanin and cymaroside were the major phenolic compounds, which accounted for 42.17%, 19.53% and 12.25%, respectively. Marein exhibited higher scavenging capacities in DPPH (EC50 = 8.84 µg/mL) and super anion radical assay (EC50 = 282.1 µg/mL) compared to cymaroside and isookanin. The antioxidant capacity of cymaroside was weakest among the three phenolic compounds due to the highest EC50 values, especially for superoxide anion radical assay, EC50 > 800 µg/mL. The result of in vitro fermentation showed that the three phenolic compounds increased the relative abundances of Escherichia/Shigella, Enterococcus, Klebsiella, etc., and isookanin notably increased the relative abundance of Bifidobacterium and Lactobacillus.Discussion and conclusions SCPs exhibited antioxidant properties and potential prebiotic effects on modulating the gut microbiota composition. The findings indicated that SCPs consumption could exert prebiotic activity that is beneficial for human health.

Acute liver injury (ALI) is a syndrome characterized by rapid deterioration of liver function, rapid progression, and high mortality. In this study, a liver-targeted drug delivery system, galactosylated chitosan modified Coreopsis tinctoria flavonoid liposome (GC-CTF-Lip) was constructed. The targeted ligand was synthesized by the amidation reaction between chitosan and lactobionic acid, and its structure was identified. The modification conditions of GC on liposomes were optimized and characterization of liposomes was studied. The liver-targeting ability of GC-CTF-Lip was evaluated through in vivo and in vitro experiments. A mouse model of ALI induced by CCl4 was established to evaluate the hepatoprotective effect of GC-CTF-Lip. The results confirmed that lactobionic acid was successfully grafted onto chitosan. Finally, GC-CTF-Lip with particle size of 236.32 ± 0.60 nm and encapsulation efficiency of 68.67 ± 0.58% was obtained. GC-CTF-Lip improved the uptake efficiency of CTF in hepatocytes in vitro and exhibited excellent liver enrichment in vivo. Serum ALT/AST levels were significantly decreased, and the histological liver injury was alleviated. It exhibited a trend of liver enrichment and a hepatoprotective effect. This liver-targeted nanodrug delivery system can achieve active accumulation of drugs at the lesion site, providing a potential strategy for the prevention of ALI.

Coreopsis tinctoria Nutt., known for its vibrant flower colors, possesses significant ornamental and medicinal value, making it a highly promising medicinal flower variety. It is widely favored by consumers as a health tea, with flower color being a crucial quality factor. The growth indicators and pigment content during the flow-ering period were determined in three C. tinctoria cultivars exhibiting different petal colors: ‘JS’ (entirely golden yellow), ‘LS’ (golden yellow tops with reddish-brown bases), and ‘MS’ (entirely reddish-brown). Additionally, a comparative metabolomic analysis of carotenoid metabolites was conducted. The results showed that the three C. tinctoria cultivars exhibited distinct differences in flower phenotypes and growth characteristics. For example, plant height and stem diameter significantly differed among ‘JS’, ‘LS’, and ‘MS’ at various flowering stages. The total carotenoid content of ‘JS’, ‘LS’, and ‘MS’ was mainly concentrated in the early flowering stage (S2), full flowering stage (S3) and late flowering stage (S4), with significant differences observed. ‘LS’ and ‘JS’ reached their highest total carotenoid content in stages S3 and S4, respectively. Analysis revealed 58 carotenoid metabolites (seven carotenes and 51 xanthophylls) with abundances that varied significantly by flower color and developmental stage. Of these, 12 metabolites (two carotenes and 10 xanthophylls) were significantly enriched in the carotenoid pathway and exhibited notably high accumulation during the S2 and S3 stages, suggesting their important role in petal coloration variation. This study provides a theoretical basis for understanding the flower color mechanism of C. tinctoria and lays a foundation for the extraction and utilization of its active components.

BACKGROUND Acrylamide (ACR), an environmental neurotoxicant prevalent in thermally processed foods, contributes to brain injury via oxidative stress and neuroinflammation. The cumulative effects of long-term low-dose exposure are particularly alarming. As a traditional food ingredient, the inflorescence of Coreopsis tinctoria Nutt. is rich in polyphenolic compounds that exhibit significant anti-inflammatory and antioxidant properties. This study systematically evaluated the neuroprotective potential of Coreopsis tinctoria Nutt. polyphenols (CTNP) against chronic low-dose ACR exposure in mice. RESULTS Behavioral analyses demonstrated that CTNP (0.25–1.00 g kg −1 ) significantly ameliorated ACR-induced gait abnormalities and restored voluntary activity. CTNP mitigated neuronal misalignment and loss of synaptic density in the hippocampal CA1/CA3 regions and markedly reduced serum levels of brain injury markers MBP and GFAP by 30.6% and 41.7%, respectively. Mechanistic investigations revealed that CTNP attenuated oxidative damage via activation of the Nrf2/HO-1 pathway, leading to decreased brain reactive oxygen species and malondialdehyde levels while enhancing catalase, superoxide dismutase and glutathione peroxidase activities. Furthermore, CTNP suppressed MAPK pathway-mediated neuroinflammation, reducing pro-inflammatory factors such as COX-2, TNF- α and IL-1 β . CONCLUSION These findings demonstrate that CTNP, as diet-derived bioactives, mitigate ACR neurotoxicity through coordinated modulation of Nrf2/HO-1 antioxidant and MAPK anti-inflammatory pathways, supporting their potential as a functional food component for preventing environmental toxicant-associated neural damage. © 2026 Society of Chemical Industry.

Coreopsis tinctoria (snow chrysanthemum) has been reported to exert antihyperlipidemic effects. The present study aimed to identify the active compounds of Coreopsis tinctoria and to investigate the molecular mechanisms underlying its effects on lipid dysregulation by measuring lipid levels, reactive oxygen species, lipid peroxidation and fatty acid synthesis. The present results demonstrated that snow chrysanthemum aqueous extracts significantly reduced serum lipid levels and oxidative stress in vivo. The main compounds that were isolated were identified as flavanomarein (compound 1) and eriodictyol 7‑O‑β‑D glucopyranoside (compound 2). Compounds 1 and 2 demonstrated potent antioxidative properties, including free radical scavenging activity, inhibition of lipid peroxidation, as well as lipid‑lowering effects in human HepG2 hepatocellular carcinoma cells treated with free fatty acids (FFAs). Compound 2 was revealed to suppress the elevation of triglyceride levels and inhibit lipid peroxidation following FFA treatment. In addition, it was demonstrated to significantly reduce intracellular levels of reactive oxygen species and improve the mitochondrial membrane potential and adenosine triphosphate levels, thus protecting mitochondrial function in FFA‑treated HepG2 cells. Furthermore, compound 2 markedly suppressed the protein expression levels of disulfide‑isomerase A3 precursor and fatty acid synthase, thus suppressing FFA‑induced lipogenesis in HepG2 cells. In conclusion, the present study identified compound 2 as one of the main active compounds in Coreopsis tinctoria responsible for its lipid‑lowering effects. Compound 2 was revealed to possess antihyperlipidemic properties, exerted via reducing oxidative stress, protecting mitochondrial function and suppressing lipogenesis.
