Application of Single-Walled Carbon Nanotubes/Au Nanosol Modified Electrode for the Electrochemical Determination of Esculetin in Cortex Fraxini

Yuanyuan Yao,Xiaomei Zhang,Na Li,Xuming Liang,Yangping Wen,Hui Zhang,Yilong Chen,Dajian Yang,Jingkun Xu

A novel simple, sensitive and selective electrochemical sensor was successfully prepared for the determination of esculetin in Cortex Fraxini based on the carboxylic acid-functionalized single-walled carbon nanotubes-Nafion–Au nanosol nanocomposite modified glassy carbon electrode (c-SWCNTs-NF–AuNs/GCE). Scanning electron microscopy, energy dispersive X-ray spectroscopy, electrochemical impedance spectroscopy and cyclic voltammetry were carried out to characterize the properties of c-SWCNTs-NF–AuNs nanocomposite. Owing to the synergistic effects of large surface area, superior electrical conductivity, and large amount of chemically active sites of c-SWCNTs, together with the good biocompatibility and high conductivity of AuNs, the c-SWCNTs-NF– AuNs/GCE exhibited a good electrocatalytic activity to esculetin with wide linear range of 0.004–55 μM and low detection limit of 0.12 nM. Additionally, the modified electrode was employed for analysis of esculetin in Cortex Fraxini with satisfactory results.

Effervescent salt and crown ether-assisted matrix solid-phase dispersion extraction of coumarins from Cortex fraxini

Yu-Han Hu,Qiu-Yan Wang,Li-Hong Ye,Juan Yang,Xin Dong,Hui Zheng,Jian Zhou,Jun Cao

A simple, rapid and environmentally friendly method named effervescence-assisted matrix solid-phase dispersion (EA-MSPD) was investigated for the extraction of four coumarins (esculin, esculetin, fraxin and fraxetin) from Crotex fraxini ( C. fraxini ). In this study, an effervescent tablet containing carbon dioxide sources (100 mg sodium bicarbonate and 200 mg sodium dihydrogenphosphate) and an adsorbent (25 mg benzo-15-crown-5) was prepared. The effervescent tablet was dissolved into an aqueous solution, and as a consequence of the effervescence reaction, carbon dioxide was generated, thus making the sample better a dispersion of the extraction solvent (100 mM sodium dodecyl sulfate). The variables that influenced the extraction efficiency included effervescent salts, extraction solvents and crown ether. These values were represented using ultra- high-performance liquid chromatography (UHPLC). Good linearity was exhibited by coefficients of determination all equal to 1, limits of detection ranging from 1.62 to 3.83 ng·mL −1 and limits of quantification ranging from 5.39 to 12.76 ng·mL −1 . The recoveries ranged from 91.37 to 100.29% with relative standard deviations of 0.57–4.58% under optimized conditions. In this work, the proposed EA-MSPD method was successfully applied to extract and identify four coumarins in C. fraxini . Compared with previously reported methods, the method examined herein was faster, greener and more sensitive.

An Ultra-Fast and Green LC-MS Method for Quantitative Analysis of Aesculin and Aesculetin in Cortex Fraxini

Xiaodong Wang,Wenhao Wang,Qinggui Lei,Zhengming Qian,Wenbin Deng

This study aims to develop a fast and eco-friendly liquid chromatography–mass spectrometry (LC-MS) method for the determination of aesculin and aesculetin in Cortex Fraxini. Ultrapure water was used as the solvent during the microwave-assisted extraction process to prepare the Cortex Fraxini sample. This extraction method reduces the cost of the harmful solvent (only ultrapure water was used) and microwave extraction time (1 min). The LC separation was conducted using an Agilent InfinityLab Poroshell 120 EC-C18column (2.1 mm × 30 mm, 2.7 µm) with a mobile phase consisting of 0.1% formic acid and acetonitrile (90:10,v/v) at a flow rate of 0.6 mL/min. Isocratic elution was employed, and the analytes were detected by MS. Through careful optimization and selection of LC-MS conditions, the analysis time was reduced to 1 min, demonstrating the method’s efficiency. The developed method was validated and exhibited excellent specificity, linearity, limit, precision, accuracy, and stability in quantifying aesculin and aesculetin in the Cortex Fraxini samples. The analysis result revealed the presence of aesculin (ranging from 3.55 to 18.8 mg/g) and aesculetin (ranging from 1.01 to 16.2 mg/g) in all ten batches of Cortex Fraxini samples. Compared to the reported LC methods, this approach substantially reduces the total analysis time and requires a minuscule volume of organic solvents. An “Analytical Eco-Scale” assessment was used to evaluate the different assay methods of Cortex Fraxini. The current LC-MS method scored an impressive 90; it was better than the other four reports’ LC methods. Thus, the developed LC-MS method is rapid and green, which is helpful for the quality evaluation of Cortex Fraxini.Keywords:LC-MS;Cortex Fraxini;aesculin;aesculetin;green;fast

Advanced applications of spectral technology combined with chemometrics in rapid quantitative analysis and holistic quality evaluation of Cortex Fraxini

Mengyao Chen,Chunling Yin,Leqian Hu,Xinyao Lin,Liuchuang Zhao,Zhimin Liu,Peng Li

Quality of Cortex Fraxini is closely related to its clinical efficacy and drug safety. Traditional quality assessment methods are either non-objective or time-consuming. It is imperative to establish a rapid method for quality detection and evaluation of Cortex Fraxini . In this study, fourier transform infrared spectroscopy (FT-IR) and near-infrared spectroscopy (NIR) combined with chemometrics were used for rapid quantitative analysis of the main active components in Cortex Fraxini . Partial least squares regression (PLSR) combined with variable select by competitive adaptive reweighted sampling (CARS) algorithm gave rise to good predictions of the content of the three active components with R 2 values greater than 0.98. Furthermore, the quality evaluation results of Cortex Fraxini cannot be simply derived from data on the content of active ingredients. This study used technique for order preference by similarity to an ideal solution (TOPSIS) method and consistency analysis to evaluate the quality of Cortex Fraxini from five geographical origins. Two methods respectively realized the quality superiority ranking and quality consistency grading of Cortex Fraxini samples. The evaluation results showed that Cortex Fraxini derived from SN had the best performance in quality and was consistent with the standard of genuine regional herbs. This study introduces an effective technique for swift identification of the primary active compounds in Cortex Fraxini , as well as for an overall assessment of its quality. The combination of spectroscopic techniques and chemometrics provides a rapid and efficient method for the quality analysis of Cortex Fraxini .

Optimization of the ultrasound-assisted extraction for phenolic compounds content and antioxidant activity of Cortex fraxini using response surface methodology

Huang Dan,Wang Jing,Li Fanghua,Xie Min,Qu Qingli,Wang Yimin,Sun Wenjuan,Wu Caie,Xu Wenhui,Xiong Ranhua,Ding Yichun,Yang Anquan,Huang Chaobo

In this study, for the first time, optimized phenolic compounds and antioxidant properties were derived from the extraction process of Cortex fraxini (CFR) using response surface methodology. Four factors and three levels of Box-Behnken (BBD) were used to study the extraction time (X 1 ), extraction temperature (X 2 ), ethanol concentration (X 3 ), and solvent-material ratio (X 4 ). The analysis of variance (ANOVA) revealed that temperature and solvent-material ratio had a relatively large effect on the response. The optimal combination of phenolic compounds and antioxidant activity was obtained with the optimal conditions of X 1  = 1.629 h, X 2  = 53.367 ℃, X 3  = 60.971% (v/v), and X 4  = 45.582 mL/g under ultrasonic assisted extraction (UAE). The experimental parameters were consistent with the predicted values. Caffeic acid, erucic acid, ferulic acid and p-hydroxycinnamic acid and other 19 phenolic acids were characterized by high performance liquid chromatography-mass spectrometry (HPLC–MS). The results of this study indicate that optimization of the extraction process is essential for the accurately determined measurement of phenolic components and antioxidants in CFR for its industrial applications.

Optimization of ultrasonic-assisted supramolecular solvent microextraction of coumarins from Cortex fraxini using response surface methodology combined with artificial neural network-genetic algorithm

Dingli Ye,Junqiang Hao,Rongxu Zhang,Yangyang Zhou,Shurong Chen,Weijian Zhang,Lei Zhao,Jiahan Xie,Zhibing Wang

A simple, fast, and efficient ultrasonic-assisted supramolecular solvent microextraction combined with high performance liquid chromatography method was developed for the determination of coumarins in Cortex fraxini , including esculin, esculetin and fraxetin. In this study, a novel supramolecular solvent was prepared with 1-octanol, tetrahydrofuran and water for the first time, and its composition, viscosity, density, structure, and micromorphology were characterized. The prepared supramolecular solvent exhibited vesicular structures and had the characteristics of low viscosity. Through single-factor experiments, response surface methodology and artificial neural network-genetic algorithm, the optimal extraction conditions were obtained as follows: NaCl concentration of 1 mol mL −1 , pH value of 10, solid-liquid ratio of 10:1, vortex time of 30 s, ultrasonic power of 100 W, ultrasonic temperature of 60 °C, ultrasonic time of 15 min, centrifugation speed of 5000 rpm, and centrifugation time of 1 min. The results demonstrated that the artificial neural network model exhibited maximum R -values of 0.98703, 0.97440, 0.99836, and 0.95447 for training, testing, validation, and all dataset, respectively. The minimum mean square errors were 0.75, 10.15, 1.99, and 2.63, respectively. This indicated that the predicted values were almost consistent with the actual values. Under the optimal conditions, the total extraction yields of target analytes reached 2.80 %. The calibration curves for each analyte exhibited excellent linearity within the linear range ( r > 0.9993). The limits of detection and quantification ranged from 4.87 to 6.55 ng mL −1 and 16.24 to 21.84 ng mL −1 , respectively. The recoveries ranged from 98.71 % to 111.01 % with relative standard deviations of less than 3.6 %. The present method had the advantages of short extraction time (15 min) and less solvent consumption (0.5 mL). The prepared supramolecular solvent was proved to have great potential in extracting coumarins from medicinal plants.

An approach of ionic liquids/lithium salts based microwave irradiation pretreatment followed by ultrasound-microwave synergistic extraction for two coumarins preparation from Cortex fraxini

Zaizhi Liu,Huiyan Gu,Lei Yang

Ionic liquids/lithium salts solvent system was successfully introduced into the separation technique for the preparation of two coumarins (aesculin and aesculetin) from Cortex fraxini. Ionic liquids/lithium salts based microwave irradiation pretreatment followed by ultrasound-microwave synergy extraction (ILSMP-UMSE) procedure was developed and optimized for the sufficient extraction of these two analytes. Several variables which can potentially influence the extraction yields, including pretreatment time and temperature, [C 4 mim]Br concentration, LiAc content, ultrasound-microwave synergy extraction (UMSE) time, liquid–solid ratio, and UMSE power were optimized by Plackett–Burman design. Among seven variables, UMSE time, liquid–solid ratio, and UMSE power were the statistically significant variables and these three factors were further optimized by Box–Behnken design to predict optimal extraction conditions and find out operability ranges with maximum extraction yields. Under optimum operating conditions, ILSMP-UMSE showed higher extraction yields of two target compounds than those obtained by reference extraction solvents. Method validation studies also evidenced that ILSMP-UMSE is credible for the preparation of two coumarins from Cortex fraxini. This study is indicative of the proposed procedure that has huge application prospects for the preparation of natural products from plant materials.
