Background The development of effective and safe dietary supplements is essential for both the prevention and management of ulcerative colitis (UC), as its pathogenesis is intricate and difficult to completely resolve. Crataegus pinnatifida, a medicinal food with a long history of use, has broad medicinal value. Recent research has revealed promising insights into the role of polysaccharide derived from Crataegus pinnatifida on modulating short-chain fatty acids (SCFAs) to alleviate UC inflammation. However, the mechanisms by which CPP regulates the intestinal microbiota and key metabolites during the antagonistic phase of UC have yet to be elucidated. Objective This research elucidated the protective role of CPP in relation to UC, highlighted the mechanisms through which CPP operates, particularly regarding gut microbiota and metabolism, and offered a theoretical foundation for the potential use of CPP as a dietary supplement aimed at preventing UC. Methods The impact of CPP on acute UC induced by 3% DSS in mice was examined through the evaluation of the disease activity index, measurement of colon length, and observation of body weight changes. Enzyme-linked immunosorbent assay (ELISA) was used to measure inflammatory factor levels in both serum and colon, as well as to assess oxidative stress mediators. The intestinal histological damage, mucus layer damage and the level of tight junction protein were analyzed by histopathological staining and western blot (WB). The impact of gut microbiota on CPP in colitis was evaluated using 16S rRNA sequencing, microbiota depletion experiments, and fecal microbiota transplantation (FMT) studies. The key metabolic pathways and key metabolites affected by CPP in the treatment of UC were analyzed through untargeted metabolomics sequencing, ELISA, and WB assays. Results Prophylactic dietary supplementation with Crataegus pinnatifida polysaccharide (CPP) notably reduced thenotably reduced the fundamental clinical manifestations of UC induced by DSS, including DAI score, reduced colon length, and weight loss, as well as inflammation and oxidative stress. CPP promoted the expression of Caludin-1, ZO-1 and Occludin and promoted mucin secretion, which contributed to the mitigation of intestinal barrier damage caused by DSS. 16S sequencing results and metabolomics results revealed that CPP intervention upregulated the relative abundance of Lactobacillus, thereby reshaping the intestinal microbiota and activate the arginine biosynthesis pathway. The results of fecal microbiota transplantation and antibiotic clearance experiments indicated that the alleviating effect of CPP on UC was dependent on the intestinal flora and that this alleviating effect was transferred through fecal microbiota transplantation. Mechanistically, CPP indirectly promoted the expression of the rate-limiting enzyme argininosuccinate synthase (ASS1) in the intestinal Arginine biosynthesis pathway by reshaping the intestinal microbiota, thereby increasing intestinal Arginine level and alleviating the inflammatory response and oxidative stress induced by DSS and intestinal barrier damage. Conclusion Our research findings demonstrate that CPP is a plant-derived polysaccharide that alleviates UC by modulating the gut microbiota and enhancing arginine biosynthesis.

Background Intestinal epithelial barrier dysfunction is involved in the development and pathogenesis of intestinal diseases, such as irritable bowel syndrome, inflammatory bowel disease, and celiac disease. This study was performed to evaluate the ability of total flavonoid extract from hawthorn (TFH) to improve TNF-α-evoked intestinal epithelial barrier deficit. Material/Methods Caco-2 cells monolayers were exposed to TNF-α in different concentrations of TFH. Intestinal epithelial barrier function was evaluated using epithelial permeability and transepithelial electrical resistance (TER). Results Our findings showed that TFH alleviated the increase of paracellular permeability and the decline of transepithelial electrical resistance (TER) evoked by TNF-α. Additionally, 24-h pre-incubation with TFH inhibited TNF-α-evoked secretion of pro-inflammatory factors (IL-6, IL-8, MCP-1, and IL-1β). Furthermore, TFH inhibited TNF-α-evoked overexpression of pMLC and MLCK and alleviated breakdown of TJs protein (ZO-1 and occludin). The activations of Elk-1 and NFκBp65 were inhibited by TFH pre-incubation. Conclusions TFH can alleviate TNF-α-evoked intestinal epithelial barrier deficit via the NFκBp65-mediated MLCK-MLC signaling pathway.

Crataegus pinnatifida is the dried fruit of Crataegus pinnatifida . Crataegus pinnatifida polysaccharides (CPPs) eluted with water as the mobile phase (CPPs-H-1) is one of the important active components. CPPs-H-1 has anti-oxidation and blood lipid regulation activities in vitro. Therefore, we investigated the effects of CPPs-H-1 on nonalcoholic fatty liver disease (NAFLD) in C57/BL6J mice. As a result, CPPs-H-1 inhibited the rapid growth of body weight and alleviated the liver injury caused by high-fat diets, significantly reducing the liver index and blood lipid levels in the model group, inhibiting the production of inflammation and cytokines, and inhibiting oxidative stress. CPPs-H-1 also enhanced intestinal barrier function. At the phylum level, CPPs-H-1 reduced the ratio of Firmicutes to Bacteroides (F/B) and regulated the content of short-chain fatty acids (SCFAs). The CPPs-H-1 may regulate intestinal flora composition, and improve intestinal barrier function, ultimately delaying the progression of NAFLD in mice. Also, CPPs-H-1 improved NAFLD progression by enhancing the Amp-activated protein kinase (AMPK)/peroxisome proliferator-activated receptor α (PPARα) signaling pathway to enhance fatty acid oxidation. All these results confirm that CPPs-H-1 plays a multi-pathway and multi-target role in the improvement of NAFLD.

Crataegus pinnatifida is widely used as functional food and traditional medicine in China for various applications. Polysaccharides may contribute to health benefits, such as modulating human gut microbiota (HGM). To investigate structure features of polysaccharides from this plant, fruits of C. pinnatifida were extracted with boiling water, yielding crude polysaccharides HAW. A novel polysaccharide, named HAW1-1, was pooled by DEAE Sepharose™ Fast Flow column and further purified by Sephacryl S-100 HR column. The molecular weight of HAW1-1 was 42.35 kDa. The carbohydrate content of HAW1-1 was 99.42% and no protein was detected. The monosaccharide analysis showed this polysaccharide was only composed of glucose. According to the partial acid hydrolysis, methylation analysis and NMR spectra, the backbone of HAW1-1 was consisted of 1,4-linked α-D-glucan and 1,4,6-linked α-D-glucan. The side chains were composed of 1,6-linked α-D-glucan and terminal-linked α-D-glucan. The bioactivity tests showed that HAW1-1 could promote the growth of three species of intestinal Bacteroides and produce short-chain fatty acids (SCFAs). The results suggested that HAW1-1 might be a potential prebiotic candidate to maintain intestinal homeostasis and improve human gut health.

Ultraviolet (UV) radiation is a primary factor contributing to photoaging, a form of premature skin aging characterized by the appearance of wrinkles, fine lines, uneven pigmentation, and reduced skin elasticity. Plant-derived polysaccharides exhibit notable antioxidant and anti-inflammatory properties, making them promising candidates for the management of skin photoaging. Nevertheless, the hydrophilic nature and large molecular size of polysaccharides make them less effective for topical application. This study aimed to develop a method to increase polysaccharide transdermal absorption, using Crataegus pinnatifida polysaccharide (CPP) as a model. Acetylation was employed to modify the CPP, yielding three derivatives with varying degrees of substitution (DS): 0.16 (Ace-CPP 1 ), 0.43 (Ace-CPP 2 ), and 0.56 (Ace-CPP 3 ). The in vitro antioxidant activity increased with increasing degree of substitution. A nanoemulgel formulation was developed, achieving approximately 72 % permeation of the native CPP. Furthermore, the acetylated CPP derivatives demonstrated enhanced permeation, exceeding 92 % within 4 h. In vivo studies revealed that the Ace-CPP 3 -based nanoemulgel significantly outperformed the native CPP in alleviating UVB-induced photoaging. This was evidenced by reduced oxidative stress, suppression of tissue inflammation, and promotion of collagen deposition. These findings underscore the potential of nanoemulgel formulation of acetylated CPP derivatives to advance applications in dermatology and cosmeceuticals.

A glucose rich heteroglycan named HAW1-2 was isolated and purified from fruit of Crataegus pinnatifida by hot-water extraction, ethanol precipitation, DEAE-cellulose anion exchange and gel permeation chromatography. HAW1-2 was proved as a homogenous polysaccharide with a molecular weight ( Mw ) of 8.94 kDa. Monosaccharide composition results indicated that HAW1-2 was composed of arabinose, galactose and glucose. Methylation analysis and NMR spectrum showed that HAW1-2 contained →4)-β- d -Glc p -(1→, →4)-β- d -Gal p -(1→, α-L-Ara f -(1→, →5)-α-L-Ara f -(1→, β- d -Glc p -(1→ and →4,6)-β- d -Glc p -(1→. Bioactivity test showed that HAW1-2 could significantly promote the growth of Bacteroides thetaiotamicron ( BT ), Bacteroides ovatus ( BO ) and Bifidobacterium longum ( BL ), which showed competition with the Bacteroides spp. under co-culture. Interestingly, Bacteroides spp. generated more acetic acids and propionic acids while BL only generated acetic acids. These results imply that polysaccharide HAW1-2 may be useful for human by modulating intestinal bacteria and producing short chain fatty acids.

In this study, twenty-three commercial yeasts were inoculated into hawthorn ( Crataegus pinnatifida , Bunge (Rosaceae)) extracted juice to explore their fermentation characteristics, β-glucosidase activity, ethyl carbamate production, volatile compounds of resulted wines, along with the relationship of β-glucosidase activity on ethyl carbamate and volatile compounds. β-Glucosidases were yeast and culture medium dependence. However, their activities in wines were merely 9.50 %–30.7 % of that in YM (yeast extract, malt extract) medium. Ethyl carbamate, which was also yeast dependence, was correlated with β-glucosidase (0.426*) whereas less affected by physicochemical indices of hawthron wines. Volatile compounds were not correlated with β-glucosidase activity whereas related to the physicochemical indices. α-Terpineol, which was originally existed in the juice, was increased by the fermentation, while (-) linalool oxide, rose oxide, linalool, terpinen-4-ol, citronellol and nerolidol were newly produced during fermentation. FC9 was the most optimal yeast for hawthorn wine making because of its excellent ethanol production, higher β-glucosidase activity, moderate amounts of volatile compounds, best sensory score and lower EC (5.48 μg/L).

As important functional food ingredients with excellent biological activities, Crataegus pinnatifida polysaccharides (CPPs) are widely applied in the food industry. However, their traditional extraction methods often encounter issues such as thermal degradation and shear damage, which result in the instability of polysaccharide structure and the reduction of biological activity. In this study, a magnetic field-assisted extraction (MFAE) method of CPPs was developed to solve the problems of thermal degradation, shear damage, and decreased bioactivity in conventional polysaccharide extraction. Compared with hot water, ultrasound-assisted, and microwave-assisted extraction, MFAE significantly improved the extraction yield and total soluble sugar content of CPPs. Through AFM, SEM, antioxidant and immunomodulatory identification, magnetic field-assisted extracted CPPs maintained stable triple-helix conformation and unique microstructure, exhibited excellent antioxidant activity, and showed no cytotoxicity toward RAW264.7 macrophages while exerting immunomodulatory effects. This demonstrates that MFAE can effectively protect the structural integrity and biological activity of hawthorn polysaccharides while achieving efficient extraction. The optimal MFAE parameters optimized by response surface methodology were as follows: solid-liquid ratio 1:30.81, magnetic field intensity 93.20 mT, and extraction temperature 92.37 °C, with a predicted extraction yield of 20.90%. This study provides a novel strategy for the high-yield and structure-preserving extraction of bioactive polysaccharides, and offers theoretical basis and technical reference for their development in the field of functional foods.

Chaenomeles speciosa (CS) and Crataegus pinnatifida (CP) are two common Chinese fruits with health benefits. The effect of in vitro digestion on the chemical composition, antioxidant activity and α -glucosidase inhibitory activity of these two fruits, as well as the accessibilities of their active compounds were studied. The inhibitory effects on starch digestion of CS, CP and the 5 identified compounds also were measured. It was observed that the chemical compositions of CS and CP were changed after simulated gastric and intestinal digestions. Their antioxidant and α -glucosidase inhibitory activities both decreased, the chemical composition and bioactivities of CS were changed more after intestinal digestion than after gastric digestion. On the other hand, the chemical composition and bioactivities of CP were changed more after gastric digestion than after intestinal digestion. Bioaccessiblities of compounds in CS and CP varied. Specifically, there were 31, 36 and 68% increases in oleanolic acid, protocatechuic acid and ursolic acid concentrations, respectively, after gastric and intestinal digestions in CS and its bioaccessibilities were more than doubled. The high bioaccessibility observed for protocatechuic acid, oleanolic acid and ursolic acid suggested the bioactive potential of CS. CS, CP, gallic acid , chlorogenic acid , catechin and oleanolic acid all inhibited in vitro starch digestion. Therefore, it may help understand the changes of the chemical composition, antioxidant activity and α -glucosidase inhibitory activity of CS and CP during digestion. It also supported that CS and CP are good potential candidates for controlling postprandial hyperglycemia in diabetics.

Crataegus pinnatifida is a common food in China, Europe and North America. In order to confirm polysaccharide was the material basis for C. pinnatifida to exert immune regulation. A polysaccharide (CPP) with a molecular weight of 13.58 kDa was isolated from C. pinnatifida . The structure of CPP was determined to be a backbone composed of → 3,5)-α- l -Ara f -(1→, with two branches consisting of → 4)-α- d -Gal p -(1 → and → 5)-α- l -Ara f -(1→, with α- l -Ara f and α- d -Man p as the terminal unit. CPP (10 ∼ 500 μg/mL) could promote the secretion of nitric oxide, interleukin-2, interleukin-6 and tumor necrosis factor-α in vitro . CPP could significantly restore the body weight of immunosuppressive mice and improve the immune organ index and interleukin-2, interleukin-6, and tumor necrosis factor-α secretion. In addition, CPP increased the abundance of Bacteroidetes and Verrucomicrobia and decreased the abundance of Proteobacteria at the phylum level. So CPP can regulate the gut microbiota and play an important role in immune regulation.

Hawthorn (Crataegus spp.) is a functional food with long medical usage and excellent health-promoting effects. In this study, comparative metabolomics analysis between the yellow hawthorn berry (Crataegus pinnatifida) and the red-skinned ones was first carried out with the peel and flesh separated to fully understand the polyphenol composition and metabolite variations. A total of 453 polyphenols was characterized, among which phenolic acids and flavonoids were abundant, and were closely relevant to the antioxidant capacity of hawthorn fruits. Polyphenol profile showed accession-specific accumulation in peel or flesh of different colored hawthorn. The unique yellow hawthorn was found to have few anthocyanins but show enhanced flux to synthesize flavones and flavonols, especially flavone C-glycosides and acylated flavonol glycosides. The specific acylation decoration includes acetylation, p-coumaroylation, and malonylation. In addition, the yellow hawthorn showed excellent α-glucosidase inhibitory effect, which might be associated with the high concentration of 8 polyphenols including 5 phenolic acids, 2 flavone C-glycosides, and an acylated flavonol glycoside, namely quercetin-7-O-(6''-malonyl)glucoside. Such acylated flavonol showed the highest correlation ship with the inhibition effect of hawthorn fruits on α-glucosidase, and was predicted to had the lowest binding energy with the enzyme by molecular docking analysis, indicating its great potential as a strong α-glucosidase inhibitors and an important antidiabetic ingredient in yellow hawthorn. Thus, acylated flavonol glycosides and C-glycosyl flavones might be chemotaxonomic markers differentiating varieties and bioactivities of yellow hawthorn from the traditional red-skinned ones. These findings complement the existing knowledge on the metabolite composition and nutritional properties of hawthorn fruits.
