Effects of extremely low frequency electromagnetic field at 50 Hz on myofibrillar protein from grass carp (Ctenopharyngodon idellus) during chilled storage at 4 °C

Swati Mahato,Zhiwei Zhu,Da-Wen Sun

Biological interaction with extremely low-frequency electromagnetic fields (ELF-EMF) is complex and the 50 and 60 Hz frequencies are among a few ‘window’ frequency ranges reported with a stimulating effect. In the current study, ELF-EMF at frequencies from 40 to 70 Hz were examined for their possible significance in inducing biological effects during chilled storage of grass carp ( Ctenopharyngodon idellus ) at 4 °C for 4 and 6 h. The myofibrillar protein extracted from the grass carp was analyzed and the results indicate significant (p < 0.05) increases in Ca 2+ ATPase activity and pH, and a reduction in the total sulfhydryl content during the treatment of 50 and 60 Hz frequencies for both storage durations, in particular, the treatment of 50 Hz had pronounced effects with higher correlations. Therefore, the 50 Hz might be associated with the ‘window’ effect, enhancing myofibrillar protein quality during chilled storage.

Liver Damage in Ctenopharyngodon idellus Induced by Nanoplastics and Cadmium Exposure

Qifeng Gao,Jianbo Ma,Zixuan Li,Chunping Mao,Xiaodong Zhang,Chaonan Zhang

Nanoplastics and cadmium often occur together in freshwater, but their combined effects on fish health are not well understood. In this study, we exposed grass carp, an important food fish, to these pollutants alone or in combination for 72 h to examine liver damage. The mixture caused much more severe liver injury—including cell death, darkening, and tissue breakdown—than either pollutant alone. It also weakened the fish’s immune system and natural antioxidant defenses more strongly. By analyzing gene activity, we found that co-exposure broadly disturbed the liver’s normal fat and hormone processing. Our results show that the two pollutants together create complex harmful effects that are not simply the sum of their individual actions. Nanoplastics (NPs) and heavy metal cadmium (Cd) are common co-existing pollutants in freshwater environments, but their combined toxic effects on the liver of herbivorous economic fish remain unclear. In this study, grass carp ( Ctenopharyngodon idella ) were exposed to polystyrene nanoplastics (PS-NPs, 100 nm) and/or Cd to investigate their individual and combined effects on hepatic toxicity. The results revealed that co-exposure interactively suppressed interleukin-10 ( IL-10 ) expression and heme oxygenase-1 ( HO-1 ) antioxidant response, and induced more severe hepatic necrosis, melanization, and fibrinoid necrosis, with the highest integrated biomarker response index and extensive disruption of lipid and steroid metabolism pathways. This study clarified the toxicological interaction of NPs and Cd on the liver of grass carp, and provided a theoretical basis for understanding the combined toxicity of NPs and heavy metal pollution in extreme contamination scenarios or accidental pollution events.

Di-butyl phthalate induces apoptosis in Ctenopharyngodon idellus kidney cells through oxidative stress injury

Zijiang Yang,Beining Wu,Qiyi Chen,Jiarui Pan,Zongying Wang,Wei Wang

Di-butyl phthalate (DBP) is a major type of phthalate (PAE) contaminant widely used as a plasticizer. Its environmental presence poses threat to humans and aquatic organisms. In this study, Ctenopharyngodon idellus kidney (CIK) cell model was exposed to 100 μM DBP to investigate its effects. Apoptosis was assessed using acridine orange/ethidium bromide staining, flow cytometry, and fluorescein isothiocyanate/propidium iodide labeling. DBP exposure increased the percentage of apoptotic cells. Activities of antioxidant enzymes, superoxide dismutase, total antioxidant activity, glutathione peroxidase, and catalase were inhibited by DBP, whereas the levels of peroxide products were increased. Heat shock proteins were upregulated as a defense mechanism against DBP-induced stress. Further analysis revealed that the Pi3k/Akt pathway, which regulates physical processes to protect cell function, was suppressed by DBP exposure. Reverse transcription-quantitative PCR and western blotting revealed that DBP inhibited Pi3k/Akt signaling while apoptosis gene expression was increased. Notably, these effects of heat shock proteins and Pi3k/Akt, were reversed by N-acetylcysteine. In conclusion, DBP accelerates apoptosis of CIK cells by inhibiting the Pi3k/Akt pathway and anti-oxidative enzyme activities, promoting reactive oxygen species accumulation and enhancing peroxide product generation. These findings highlight the cytotoxic effects of DBP and underscore the need for further research. Our results provide a foundation for further study.

Protective effects of calcium against cadmium-induced toxicity in juvenile grass carp (Ctenopharyngodon idellus)

Yang Xu,Yuting Gui,Dan Zhi,Jie Pi,Xinhua Liu,Jianguo Xiang,Deliang Li,Junhua Li

Cadmium (Cd) is one of the dominant metal pollutants present in the aquatic environment that affects ion homeostasis, oxidative stress (OS) and immune responses of aquatic organisms. Given the physicochemical similarities between Cd 2+ and calcium (Ca 2+ ) ions, their antagonism may facilitate the mitigation of Cd-induced toxicity. To better understand the role of Ca in protecting against Cd-induced toxicity in teleosts, juvenile grass carp were exposed to Cd (measured concentration 3 μg/L) and a gradient of Ca concentrations (measured concentration 1.5 mg/L, 2.5 mg/L, 3.0 mg/L, and 3.5 mg/L in the control (CTL) group, low calcium (LCA) group, medium calcium (MCA) group, and high calcium (HCA) group, respectively) for 30 days. Inductively coupled plasma mass spectrometry (ICP-MS) data analyses showed that simultaneous exposure to Ca impaired the accumulation of Cd in all tested tissues. Besides, Ca addition maintained the plasma ion (Na + , K + , Cl - ) homeostasis, alleviated Cd-induced oxidative stress (OS), and regulated the activities and transcriptional levels of ATPase. Furthermore, transcriptional heatmap analysis demonstrated that several indicator genes for OS and calcium signaling pathway were found to be significantly modulated by Ca addition. This work delineates a protective effect of Ca against Cd-induced toxicity in grass carp, providing new insight into the possible solutions to Cd pollution issues in aquaculture industry.

Bio-based active packaging: Gallic acid modified agarose coatings in grass carp (Ctenopharyngodon idellus) preservation

Liling Fu,Qiong Xiao,Yi Ru,Qinglin Hong,Huifen Weng,Yonghui Zhang,Jun Chen,Anfeng Xiao

Antioxidant and antimicrobial agarose coatings were developed by grafting gallic acid through the carbodiimide coupling method. Structural characterization revealed that the carboxyl group of gallic acid was successfully grafted onto the C6-OH of D-galactose in agarose, with the highest observed grafting ratio being 13.73 %. The grafting of gallic acid significantly increased the antioxidant and bacteriostatic activities of the agarose. As the grafting ratio of gallic acid-modified agarose (Ga Ag) increased from 0 to 13.73 %, the scavenging ratio of DPPH and the inhibition ratio of β -carotene bleaching were observed to increase from 0 % to 65.92 % and 6.89 % to 73.46 %, respectively. Ga Ag exhibited up to 100 % inhibition of Escherichia coli and Staphylococcus aureus . The physicochemical properties of gel strength , viscosity, gelling temperature and melting temperature decreased to 971.3 g/cm 2 , 17.9 mPa·s, 31.7 °C and 84.1 °C, respectively. The gel contact angle was increased from 22.1° to 73.6°. Fish preservation tests have demonstrated that it effectively inhibited bacterial growth, prevented fat oxidation , blocked light, reduced moisture loss, and enhanced the overall quality of grass carp ( Ctenopharyngodon idellus ) fillets during refrigeration, which was more effective than native agarose in extending the shelf life of fish. Therefore, Ga Ag holds promise as an aquatic product preservative.

Action of structural proteins in textural deterioration of grass carp (Ctenopharyngodon idellus) fillets during refrigerated storage

Jiandong Shen,Bijiang Zhong,Wenshui Xia,Yanshun Xu

Summary Textural deterioration is a severe problem in fish industry. In order to study the role of structural proteins in refrigerated grass carp, myofibrillar proteins and collagen degradation were investigated at different stages. Results revealed that connective tissues of fish muscle disintegrated from day 1, failing to hold muscle myotomes together, with a decrease of shear force. Meanwhile, collagen solubility increased from day 1 and degradation of nebulin was observed after 3 days, while there was no obvious change in the bands of myosin heavy chain and actin. Furthermore, Pearson coefficient analysis revealed that insoluble collagen (ISC) and nebulin were significantly correlated with shear force. Negative correlation was established between drip loss and contributing factors (ISC, titin and nebulin). All results suggested that textural loss of post-mortem grass carp fillets was mainly mediated by the degradation of collagen and to a lesser extent by myofibrillar proteins at the early storage.

Tea Polyphenols Affect Oxidative Modification and Solution Stability of Myofibrillar Protein from Grass Carp (Ctenopharyngodon idellus)

Li Xuepeng,Liu Cikun,Wang Jinxiang,Li Wenxie,Lin Boyan,Zhu Wenhui,Xu Yongxia,Yi Shumin,Mi Hongbo,Li Jianrong

This study investigated the effects of different concentrations (0, 5, 10, 20, 50, and 100 μmol/g protein) of tea polyphenols (TP) on the oxidative modification and the physicochemical, structural, and gelling properties of myofibrillar protein (MP) from grass carp ( Ctenopharyngodon idellus ) oxidized by a hydroxyl radical-generating system. The results showed that low concentrations (5 and 10 μmol/g protein) of TP could effectively inhibit the formation of carbonyl groups and dityrosine, the loss of sulfhydryl groups and α-helix conformation, and the change of the tertiary structure of MP caused by hydroxyl radicals. Moreover, the presence of TP in all concentrations decreased the surface hydrophobicity of MP. TP at 10 μmol/g protein had better effects on preventing the oxidation-induced cross-linking and aggregation of myosin heavy chain (MHC) and actin of MP, keeping the stability of MP solutions with lower turbiscan stability index (TSI) values, and improving gelling properties characterized by higher hardness and gel strength of MP gels. By contrast, excessive presence of TP (20, 50, and 100 μmol/g protein) showed pro-oxidative effects on oxidatively stressed MP, which was detrimental to the MP and contributed to the denaturation and irregular aggregation of MP, the loss of MP solution stability, and lower gelling capacity with poor texture and gel strength. The concentration-dependent effects of TP on MP depended on the extent of MP oxidative modification and MP-TP interactions, indicating that a proper amount of TP has the potential to protect MP from oxidation and to enhance the gelling capacity of surimi during processing.

Assessment of gelatinolytic proteinases in chilled grass carp (Ctenopharyngodon idellus) fillets: characterization and contribution to texture softening

Jiandong Shen,Qixing Jiang,Wei Zhang,Yanshun Xu,Wenshui Xia

BACKGROUND Texture softening is always a problem during chilling of grass carp fillets. To solve this problem and provide for better quality of flesh, understanding the mechanism of softening is necessary. Gelatinolytic proteinases are suspected to play an essential role in the disintegration of collagen in softening of fish flesh. In the present study, the types and contribution of gelatinolytic proteinases in chilled fillets were investigated. RESULTS Four active bands (G1, 250 kDa; G2, 68 kDa; G3, 66 kDa; G4, 29 kDa) of gelatinolytic proteinases were identified in grass carp fillets by gelatin zymography. The effect of inhibitors and metal ions revealed that G1 was possibly a serine proteinase, G2 and G3 were calcium-dependent metalloproteinases and G4 was a cysteine proteinase. The effect of the inhibitors phenylmethanesulfonyl fluoride (PMSF), l -3-carboxy- trans -2,3-epoxy-propionyl- l -leucine-4-guanidinobutylamide (E-64) and 1,10-phenanthroline (Phen) on chilled fillets revealed that gelatinolytic proteinase activities were significantly suppressed. Collagen solubility indicated that metalloproteinase and serine proteinase played critical roles in collagen breakdown during the first 3 days, and cysteine proteinase revealed its effect after 3 days. Meanwhile, during chilled storage for 11 days, the final values of shear force increased 19.68% and 24.33% in PMSF and E-64 treatments when compared to control fillets respectively, whereas the increase after Phen treatment was 49.89%. CONCLUSION Our study concluded that the disintegration of collagen in post-mortem softening of grass carp fillets was mainly mediated by metalloproteinase and to a lesser extent by serine proteinase and cysteine proteinase. © 2021 Society of Chemical Industry.

Concentrated mannan oligosaccharide supports grass carp (Ctenopharyngodon idellus) health during Ichthyophthirius multifiliis infection: Histopathological and immunological study

Fulong Li,Ming Xu,Baipeng Chen,Senyue Liu,Yanhao Cao,Yufan Zhang,Gang Lin,Ping Ouyang,Defang Chen,Yi Geng,Xiaoli Huang

Ichthyophthirius multifiliis is a highly pathogenic ciliate parasite that infects nearly all freshwater teleost fish and causes significant economic losses. However, none of the current treatments for ichthyophthiriasis are effective. Therefore, it is necessary to explore new effective, alternative, and environmentally friendly products for the control of I. multifiliis infection. Concentrated mannan oligosaccharide (cMOS) is widely used in aquaculture as a functional additive to enhance fish immunity and disease resistance. In this study, 1 ‰, 2 ‰, and 4 ‰ cMOS were included into the diet of grass carp ( Ctenopharyngodon idellus ) for 6 weeks while observing growth performance and immune response in order to evaluate the concentration and time of cMOS supplementation needed to support I. multifiliis infected grass carp. The results showed that feeding 2 ‰ and 4 ‰ cMOS significantly increased the body weight of grass carp ( P  < 0.05), and feeding 4 ‰ cMOS increased the total length and specific growth rate ( P  < 0.05). The histological results showed that grass carp fed with 4 ‰ cMOS for three weeks and 2 ‰ cMOS for four weeks had more mucous cells and T/NK cells in their gills ( P  < 0.05). Further qRT-PCR revealed higher expression of Muc-2 , TCR and IL-1β , as well as lower expression of IL-10 in cMOS fish ( P  < 0.05). After the I. multifiliis challenge, feeding 4 ‰ cMOS for 3 weeks and 2 ‰ cMOS for 4 weeks resulted in significantly less severity of I. multifiliis infection, lower mortality, and lessened pathological damage to gill tissue ( P  < 0.05). In addition, qRT-PCR revealed the expression of IL-1β , Muc-2 , MHC-II , IgM , and TCR in cMOS groups was significantly lower than that of the control-infection group, while the expression of IL-10 was significantly higher ( P  < 0.05). The results of this study contribute new knowledge for the formulation of commercial feed for grass carp and provide both theoretical support and practical guidance for research on the prevention and control of fish ichthyophthiriasis.

The inhibitory effect of chlorogenic acid on lipid oxidation of grass carp (Ctenopharyngodon idellus) during chilled storage

Cao Qiongju,Du Hongying,Huang Yuan,Hu Yang,You Juan,Liu Ru,Xiong Shanbai,Manyande Anne

Grass carp ( Ctenopharyngodon idellus ) is a kind of freshwater fish which is rich in polyunsaturated fatty acids and easily exposed to lipid oxidation during refrigeration. The effect of chlorogenic acid (CGA) on lipid oxidation, protein oxidation, enzymatic activities, and color stability of grass carp muscle during chilled storage was investigated. The lipid oxidation was inhibited by CGA, as evidenced by lower thiobarbituric acid values, peroxide values, carbonyl valence, less free fatty acid content, and higher amount of unsaturated fatty acid compared to the control group. CGA also had a positive effect on the whiteness value and the stability of protein oxidation of fish samples. In addition, the inhibitory study of CGA on endogenous lipase and lipoxygenase activities of fish muscle can help to partly illustrate the mechanism that retains its freshness effect. The results indicate that CGA is a novel natural additive which can be used to inhibit lipid and protein oxidation and be applied in the storage of aquatic products or some similar fields.

Effects of glyphosate exposure on gut–liver axis: Metabolomic and mechanistic analysis in grass carp (Ctenopharyngodon idellus)

Biao Yan,Yumiao Sun,Kaiyu Fu,Yindan Zhang,Lei Lei,Jun Men,Yongyong Guo,Shengmin Wu,Jian Han,Bingsheng Zhou

Glyphosate, one of the most widely used herbicide worldwide, is potentially harmful to non-target aquatic organisms. However, the environmental health risks regarding impacts on metabolism homeostasis and underlying mechanisms remain unclear. Here we investigated bioaccumulation, metabolism disorders and mechanisms in grass carp after exposure to glyphosate. Higher accumulation of glyphosate and its major metabolite, aminomethylphosphonic acid, in the gut was detected. Intestinal inflammation, barrier damage and hepatic steatosis were caused by glyphosate exposure. Lipid metabolism disorder was confirmed by the decreased triglyceride, increased total cholesterol and lipoproteins in serum and decreased visceral fat. Metabolomics analysis found that glyphosate exposure significantly inhibited bile acids biosynthesis in liver with decreased total bile acids content, which was further supported by significant downregulations of cyp27a1 , cyp8b1 and fxr . Moreover, the dysbiosis of gut microbiota contributed to the inflammation in liver and gut by increasing lipopolysaccharide, as well as to the declined bile acids circulation by reducing secondary bile acids. These results indicated that exposure to environmental levels of glyphosate generated higher bioaccumulation in gut, where evoked enterohepatic injury, intestinal microbiota dysbiosis and disturbed homeostasis of bile acids metabolism; then the functional dysregulation of the gut-liver axis possibly resulted in ultimate lipid metabolism disorder. These findings highlight the metabolism health risks of glyphosate exposure to fish in aquatic environment.

Active packaging coatings based on agarose caffeate: Preparation, characterisation, and application in grass carp (Ctenopharyngodon idellus) preservation

Liling Fu,Yi Ru,Qinglin Hong,Yonghui Zhang,Huifen Weng,Qiuming Yang,Qiong Xiao,Anfeng Xiao

Caffeic acid was grafted to agarose by a nonradical synthesis method to improve the functional properties of agarose and its application value in food preservation. Structural characterization showed that the carboxyl group of caffeic acid was successfully grafted to the C6-OH of D -galactose in agarose, and the highest grafting ratio could reach 12.18 %. Incorporating caffeic acid significantly increased the antioxidant and bacteriostatic activities. With the increase in grafting ratio from 0 % to 12.18 %, the scavenging ratio of DPPH increased from 0 % to 50.09 %, and the inhibition ratio of β -carotene bleaching increased from 7.49 % to 72.21 %. The bacteriostatic ratio of agarose caffeate (Cf-Ag) against Escherichia coli and Staphylococcus aureus could reach 100 %. The fish preservation test indicated that Cf-Ag had positive effects on inhibiting bacterial growth and fat oxidation , reducing the water loss, and prolonging the overall quality of grass carp fillet during refrigeration, which was more effective than native agarose in extending the shelf life of fish.
