MXene Loaded With Cu(2-x)Se Nanozyme for Nanocatalytic Tumor Therapy

Mengtian Lu,Tianye Zhang,Yue Yang,Xin Lin,Jin Huang,Yuan Sun,Tiedong Sun

Graphical The study innovatively combines etched/stripped monolayer Ti 3 C 2 MXene with Cu (2- x ) Se nanoparticles in a one-pot method. The synthesized Ti 3 C 2 /Cu (2- x ) Se nanocomposite has catalase (CAT) and peroxidase (POD) -like activities combined with the excellent photothermal properties of MXene, which enhances the catalytic performance of Cu (2- x ) Se under near infrared (NIR) irradiation, and realizes the synergic photothermal catalytic treatment. Traditional tumor treatments (surgery, radiotherapy, chemotherapy, etc.) have certain limitations and can have serious negative effects, such as difficulty in cutting out tumors, damage to normal tissues, and complications. Ordinary nanozymes have low catalytic activity and require higher doses for treatment, which can increase in vivo toxicity and side effects. To address these limitations, we developed a Ti 3 C 2 MXene-based nanocomposite (Ti 3 C 2 /Cu (2- x ) Se) integrating Cu (2- x ) Se nanozymes with dual enzyme-mimicking activities (catalase and peroxidase) and MXene's photothermal properties. The Cu (2- x ) Se component catalyzes the decomposition of tumor-overexpressed H 2 O 2 into O 2 and cytotoxic ·OH, alleviating hypoxia while inducing oxidative stress. Simultaneously, MXene's high surface area and photothermal capability enhance nanozyme stability, biocompatibility, and catalytic efficiency under near-infrared irradiation. Notably, the photothermal effect amplifies enzymatic activity, enabling synergistic nanocatalytic-photothermal therapy. This synergy not only degrades glutathione to suppress tumor antioxidant defenses but also achieves targeted tumor ablation with reduced dosage requirements. Our work highlights a rational design of MXene-based nanozymes for enhanced multimodal tumor therapy, offering a paradigm for nanocomposite-driven disease treatment.

Rapid photothermal assay for ultrasensitive point-of-care detection of tumor markers based on a filter membrane

Liu Shendong,Chen Xinlian,Zhao Hong,Lin Tianran,Hou Li,Zhao Shulin

An ultrasensitive photothermal assay was designed for point-of-care testing (POCT) of tumor markers based on a filter membrane. Firstly, Cu 2-x Se was successfully encapsulated in liposome spheres with biotin on the surface and connected to carcinoembryonic antigen (CEA) aptamer with 3'end modified biotin by streptavidin. Secondly, the CEA antibody was successfully modified on the surface of the nitrocellulose membrane through simple incubation. Finally, the assay process was completed using a disposable syringe, and the temperature was recorded using a handheld infrared temperature detector. In the range 0–50 ng mL −1 , the temperature change of the nitrocellulose membrane has a strong linear relationship with CEA concentration, and the detection limit is 0.097 ng mL −1 . It is worth noting that the entire testing process can be easily performed in 10 min, much shorter than traditional clinical methods. In addition, this method was successfully applied to the quantitative determination of CEA levels in human serum samples with a recovery of 96.2–103.3%. This rapid assay can be performed by “one suction and one push” through a disposable syringe, which is simple to operate, and the excellent sensitivity reveals the great potential of the proposed strategy in the POCT of tumor biomarkers. Temperature-Sensitive Polymer-Driven Nanomotors for Enhanced Tumor Penetration and Photothermal Therapy

Beng Ma,Ying Yu,Jiayi Li,Yunqi Zhang,Bo Sun,Anqi Ji,Kexing Song,Linlin Shi,Hao Hu,Shegan Gao,Haoyan Cheng

Self-propelled nanomotors possess strong propulsion and penetration abilities, which can increase the efficiency of cellular uptake of nanoparticles and enhance their cytotoxicity against tumor cells, opening a new path for treating major diseases. In this study, the concept of driving nanomotors by alternately stretching and contracting a temperature-sensitive polymer (TS-P) chain is proposed. The TS-Ps are successfully linked to one side of Cu 2-x Se@Au (CS@Au) nanoparticles to form a Janus structure, which is designated as Cu 2-x Se@Au-polymer (CS@Au-P) nanomotors. Under near-infrared (NIR) light irradiation, Cu 2-x Se nanoparticles generate photothermal effects that change the system temperature, triggering the alternation of the TS-P structure to generate a mechanical force that propels the motion of CS@Au-P nanomotors. The nanomotor significantly improved the cellular uptake of nanoparticles and enhanced their penetration and accumulation in tumor. Furthermore, the exceptional photothermal conversion efficiency of CS@Au-P nanomotors suggests their potential as nanomaterials for photothermal therapy (PTT). The prepared material exhibited good biocompatibility and anti-tumor effects both in vivo and in vitro, providing new research insights into the design and application of nanomotors in tumor therapy.

Ameliorating Mitochondrial Dysfunction of Neurons by Biomimetic Targeting Nanoparticles Mediated Mitochondrial Biogenesis to Boost the Therapy of Parkinson's Disease

Qing Zheng,Hanghang Liu,Hao Zhang,Yaobao Han,Jiaxin Yuan,Tingting Wang,Yifan Gao,Zhen Li

Mitochondrial dysfunction of neurons is the core pathogenesis of incurable Parkinson's disease (PD). It is crucial to ameliorate the mitochondrial dysfunction of neurons for boosting the therapy of PD. Herein, the remarkable promotion of mitochondrial biogenesis to ameliorate mitochondrial dysfunction of neurons and improve the treatment of PD by using mitochondria-targeted biomimetic nanoparticles, which are Cu 2- x Se-based nanoparticles functionalized with curcumin and wrapped with DSPE-PEG 2000 -TPP-modified macrophage membrane (denoted as CSCCT NPs), is reported. These nanoparticles can efficiently target mitochondria of damaged neurons in an inflammatory environment, and mediate the signaling pathway of NAD + /SIRT1/PGC-1 α /PPAR γ /NRF1/TFAM to alleviate 1-methyl-4-phenylpyridinium (MPP + )-induced neuronal toxicity. They can reduce the mitochondrial reactive oxygen species, restore mitochondrial membrane potential (MMP), protect the integrity of mitochondrial respiratory chain, and ameliorate mitochondrial dysfunction via promoting mitochondrial biogenesis, which synergistically improve the motor disorders and anxiety behavior of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice. This study demonstrates that targeting mitochondrial biogenesis to ameliorate mitochondrial dysfunction has a great potential in the treatment of PD and mitochondria-related diseases.
