This study presents the development of a magnetic relaxation switching (MRS) immunosensor for the sensitive detection of chlorpyrifos (CPF) with a signal amplification strategy using Pd/Cu-TCPP(Fe) hybrid nanosheets and polydopamine (PDA). Pd/Cu-TCPP(Fe) nanosheets, which exhibit high peroxidase-like activity and excellent storage stability, making them suitable replacements for natural enzymes in biosensors, are further functionalized with goat anti-mouse IgG (Ab 2 ) to construct an immunosensor. The mechanism relies on the competition between free CPF and the immobilized bovine serum albumin-CPF conjugates for antibody binding, which modulates the aggregation of magnetic nanoparticles (MNPs) that are related to the transverse relaxation time. The optimized immunosensor shows a linear detection range from 0.5 ng/mL to 100 ng/mL and a limit of detection (LOD) of 0.24 ng/mL, outperforming traditional enzyme-linked immunosorbent assay (ELISA) methods by achieving an LOD that is about 9 times lower. Real sample analysis demonstrates the applicability of the Pd/Cu-TCPP(Fe)-PDA-MRS immunosensor for detecting CPF residues in tomato and water samples, with results well consistent with those obtained using gas chromatography. This work highlights the potential of nanomaterials in enhancing the performance of MRS immunosensors for pesticide residue analysis in environmental and food safety monitoring.

The development of biosensors capable of assessing umami intensity has elicited significant attention. However, the detection range of these biosensors is constrained by the sensing components and strategies used. In this study, we introduce a novel competitive, ultra-high-sensitivity electrochemical impedance biosensor by utilizing composite nanomaterials and T1R1 as a composite signal probe. Pd/Cu-TCPP(Fe) had a substantial surface area, effectively enhancing the loading capacity of the T1R1 and thus augmenting the biosensor's recognition precision. Furthermore, the Pd/Cu-TCPP(Fe) elevated peroxidase-like activity catalyzed the formation of insoluble precipitates of 4-CN, resulting in cascaded amplification of the impedance signal. The remarkable catalytic activity of the composite signal probe endowed the biosensor with exceptional analytical performance, featuring a detection limit of 0.86 pg/mL and a linear detection range spanning from 10 to 10000 pg/mL. Successful application of the biosensor for umami substance detection in fish was also demonstrated, signifying its substantial potential in food-flavor evaluation.

Metal-organic frameworks(MOFs) have been widely regarded as promising carriers for enzyme immobilization owing to their advantages in improving loading and regulating interaction with enzymes. However, they are still suffering from the problems of slow mass transfer and compromising activity. In this paper, the active two-dimensional(2D) MOF of Cu-TCPP(Fe)[TCPP=tetrakis(4-carboxy-phenyl)porphyrin], which possesses the biomimetic architecture of peroxidase, was adopted to anchor cytochrome(Cyt c) for the enhancement of catalytic activity. The atomic/nanometer thickness and micrometer lateral dimension of 2D MOFs can ensure the full exposure of immobilized enzymes and a shorter diffusion distance for the reactant molecules. Besides, the active carrier can provide synergistic catalysis and activity compensation during the reaction. When tested in the decomposition reaction of H 2 O 2 , Cyt c/Cu-TCPP(Fe) exhibited nearly twice catalytic activity and an accelerated catalytic rate compared to free Cyt c.

Background The detection and quantification of glucose in human biofluids are critical for the early diagnosis and management of metabolic disorders such as hypoglycemia and diabetes. Non-invasive wearable sensors for sweat glucose monitoring have emerged as promising alternatives to traditional blood glucose testing due to their ease of use, rapid response, and portability. However, the development of stable, cost-effective, and enzyme-free wearable platforms capable of reliable sweat glucose detection remains a significant challenge. Current enzyme-based sensors often suffer from poor stability under ambient conditions, complex fabrication processes, and high costs, limiting their widespread applicability in resource-constrained settings. Results Au NPs/Cu-TCPP(Fe) nanozymes exhibiting dual peroxidase-like and glucose oxidase-like catalytic activities were synthesized and characterized. These nanozymes catalyze a cascade reaction: glucose oxidation generates H 2 O 2 , which is then decomposed to produce hydroxyl radicals that oxidize 3,3′,5,5′-tetramethylbenzidine (TMB), yielding a blue-colored product for colorimetric detection. A flexible, multi-layer hydrophilic-hydrophobic array patch was fabricated by integrating the nanozymes and TMB into a paper-based matrix. This design enables efficient in situ sweat collection, concentration, and colorimetric analysis. The patch exhibits a linear detection range of 20–200 μM with a detection limit of 12.72 μM, suitable for physiological sweat glucose levels. Selectivity tests confirmed negligible interference from common sweat components. A smartphone application was developed to enable real-time, quantitative analysis of colorimetric signals. Clinical validation on multiple volunteers across different skin locations (chest, back, forearm, forehead) showed strong correlation with reference methods (HPLC-MS, commercial glucose detection kit), with relative errors <10 %. Significance This study presents a low-cost and enzyme-free sweat glucose detection platform based on Au NPs/Cu-TCPP(Fe) nanozymes. The integration of smartphone technology enables point-of-care testing, making the platform particularly suitable for personalized health monitoring. The successful validation in human subjects demonstrates its potential for real-world applications in diabetes management and early disease detection. This work bridges the gap between nanozyme catalysis and wearable sensor technology, providing a versatile platform for non-invasive biomarker detection.

Ferroptosis is a recently discovered route of regulated cell death that offers the opportunities for the treatment of chemotherapy-resistant tumor indications, but its efficacy can be affected by the glutathione peroxidase 4 (GPX4) and ferroptosis suppressor protein 1 (FSP1) antioxidant mechanisms, posing significant challenges for its clinical translation. In this study, we report a Cu-tetra(4-carboxyphenyl)porphyrin chloride(Fe(III)) (Cu-TCPP(Fe)) metal organic framework (MOF)-based nanosystem for the efficient incorporation of Au nanoparticles (NPs) and RSL3, which can demonstrate enzyme-like activities to universally suppress the antiferroptotic pathways in tumor cells for amplifying ferroptotic damage. Herein, Cu-TCPP(Fe) MOF nanosheets were integrated with Au NPs via in situ nucleation and loaded with RSL3 via π–π stacking, which were eventually modified with polyethylene glycol (PEG) and iRGD for tumor-targeted drug delivery. Specifically, the Au NPs can demonstrate glucose oxidase-like activities for efficient glucose depletion, thus disrupting the pentose phosphate pathway to impede reduced glutathione (GSH) biosynthesis and prevent the recycling of coenzyme Q10 (CoQ10) to CoQ10H2, while Cu species can oxidize GSH into oxidized glutathione (GSSG). These nanocatalytic activities can lead to the simultaneous inhibition of the GPX4/GSH and FSP1/CoQ10H2 pathways and cooperate with the GPX4-deactivating function of RSL3 to cause pronounced ferroptotic damage, thereby providing a strong rationale for the application of ferroptosis therapy in the clinic.
