Hierarchical Cu7S4-Cu9S8 heterostructure hollow cubes for photothermal aerobic oxidation of amines

Haijing Gao,Yajie Chen,Huali Li,Fangfang Zhang,Guohui Tian

Copper-based chalcogenides have been paid intensive attention as alternate photothermal catalysts for solar energy conversion. Despite considerable progress of copper sulfides nanostructure fabrication, combining independent copper sulfides polymorphs with various stoichiometries by forming heterojunction is still challenging. Herein, unique hierarchical nanosheet-assembled Cu 7 S 4 -Cu 9 S 8 heterostructure hollow cubes with remarkable photothermal aerobic oxidation activity were prepared by a facile onepot chemical transformation route using Cu 2 O cubes as sacrificial template for the first time. In this reaction system, the introduction of proper sulfur source thioacetamide and moderate alkaline conditions plays a crucial role in phase transformation, and through tunning the reaction time, a series of Cu 7 S 4 , Cu 7 S 4 -Cu 9 S 8 , and Cu 9 S 8 can be selectively prepared. Furthermore, the Cu 7 S 4 -Cu 9 S 8 polymorphic heterostructure hollow cubes exhibit significantly enhanced photothermal aerobic oxidation performance of amines to the corresponding imines compared with the individual Cu 7 S 4 and Cu 9 S 8 in both N , N -dimethylformamide and acetonitrile solvent medium. Our results indicated that the strong near-infrared plasmonic absorption of Cu 7 S 4 -Cu 9 S 8 facilitated hot carrier transport between Cu 7 S 4 and Cu 9 S 8 , which subsequently promoted the catalytic reactions. Especially after uniform load of Au-Pt alloy nanoparticles, the photothermal aerobic oxidation activity was further enhanced due to their boosted synergistic local coupling enhancement effect between distinct components in the N , N -dimethylformamide solvent medium. Such enhanced photothermal reaction could effectively promote sunlight utilization in a wide range of photocatalytic reactions.

A hollow Cu9S8 theranostic nanoplatform based on a combination of increased active sites and photothermal performance in enhanced chemodynamic therapy

Yurui Wang,Lu An,Jiaomin Lin,Qiwei Tian,Shiping Yang

A theranostic nanoplatform, combined with tumor imaging and therapy, especially a highly reactive multimodal synergistic treatment, can effectively improve the efficacy of treatment and reduce costs. Copper sulfide can be used as a theranostic nanoplatform due to its strong photoacoustic, photothermal, and Fenton-like catalytic performance. However, its Fenton-like catalytic performance is limited by the availability of surface reaction sites. A hollow Cu 9 S 8 theranostic nanoplatform, with photoacoustic imaging and enhanced chemodynamic performance based on the combination of an increased number of active sites and photothermal performance was constructed in this study. The specific area of the hollow Cu 9 S 8 nanoparticles was 1.7 times greater than that of solid Cu 9 S 8 nanoparticle, which greatly increased the number of active sites where catalysis could occur in the Fenton reaction, thereby improving the chemodynamic therapy performance. In addition, the excellent photothermal properties of the Cu 9 S 8 nanoparticles accelerated the generation of hydroxyl radicals ( OH), which also enhanced the chemodynamic therapy performance. More importantly, the in vivo experiment demonstrated that the hollow Cu 9 S 8 theranostic nanoplatform exhibit good tumor photoacoustic imaging and enhanced chemodynamic therapy due to the combination of the increase in number of active sites and photothermal performance. Therefore, this work not only provides an excellent theranostic nanoplatform for tumor treatment, but also a new strategy for developing high performance chemodynamic agents.

A facile synthesis of CuFe2O4/Cu9S8/PPy ternary nanotubes as peroxidase mimics for the sensitive colorimetric detection of H2O2 and dopamine

Zezhou Yang,Fuqiu Ma,Yun Zhu,Sihui Chen,Ce Wang,Xiaofeng Lu

Synergistic effects play an important role in improving the catalytic activity for enzyme-like reactions. Compared to individual nanomaterials, a system consisting of multiple components usually exhibits enhanced catalytic activity as an enzyme mimic. Herein we describe the synthesis of CuFe2O4/Cu9S8/polypyrrole (PPy) ternary nanotubes as an efficient peroxidase mimic via a three-step approach involving an electrospinning process, annealing treatment and hydrothermal reaction. The remarkably enhanced catalytic activity of CuFe2O4/Cu9S8/PPy ternary nanotubes as peroxidase mimics over individual CuFe2O4 nanofibers, CuFe2O4/CuO composite nanofibers, CuFe2O4/CuS composite nanofibers, and PPy materials has been achieved, demonstrating the presence of a synergistic effect among the components. The steady-state kinetic experiment suggests a good catalytic efficiency of the CuFe2O4/Cu9S8/PPy ternary nanotubes. On the basis of high catalytic activity, a colorimetric platform for the sensitive detection of H2O2 and dopamine has been developed. This work not only offers a simple approach for the fabrication of a high performance peroxidase-like nanocatalyst, but also provides its promising potential applications in biosensors, medical diagnosis, and environmental monitoring.

Platelet membrane coated Cu9S8-SNAP for NIR-II photoacoustic imaging guided NO repairing acute kidney injury

Xiaojuan Hu,Danping Wu,Shijie Yao,Gongning Chen,Ping Li,Yang Chen,Wancheng Gong,Weijiao Fan,Xiaoyi Chen,Xiaozhou Mou,Yu Cai,Xianghong Yang

Acute kidney injury (AKI) is a prevalent critical condition in hospitalized patients with an elevated risk of death. Challenges in AKI management persist due to the absence of efficient early diagnostic modalities and the lack of effective therapeutic interventions, imposing significant burdens on medical resources for prolonged treatment. Here, we proposed a second near-infrared (NIR-II) photoacoustic imaging (PAI) guided triggered on-demand release of (nitric oxide) NO from the injury zone to monitor and treat AKI through the anti-inflammatory, anti-apoptotic, and oxidative stress-damage-reducing cell- or organ-protective effects of NO. We engineered hollow copper sulfide nanoparticles (Cu 9 S 8 NPs) loaded with platelet membrane-encapsulated NO-releasing precursors (S-Nitroso-N-acetylpenicillamine, SNAP), as a nanoplatform for early AKI diagnosis and precise spatiotemporal NO release Besides acting as a drug carrier, the inherent PAI compatible Cu 9 S 8 also enables targeted renal injury localization, exploiting the natural propensity of platelets to accumulate at vascular injury sites, facilitating real-time AKI monitoring in murine models. Experiments demonstrated that nitric oxide delivery nanoparticles can specifically target and accumulate in the kidney, reduce inflammation, and promote damage repair. Overall, this gas therapy strategy represents a new therapeutic pathway for the clinical AKI management.

Synergistic Photothermal and Photodynamic Therapy for Effective Implant-Related Bacterial Infection Elimination and Biofilm Disruption Using Cu9S8 Nanoparticles

Wanshun Wang,Xiaohang Cheng,Jiawei Liao,Zefeng Lin,Lingling Chen,Dandan Liu,Tao Zhang,Lihua Li,Yao Lu,Hong Xia

Implant-related bacterial infections are one of the most common but tricky problems in orthopedic clinics because the formation of biofilms inhibits the penetration of antibiotics to kill bacteria effectively; thus, a new strategy is urgently needed. Antibacterial nanomaterials [e.g., copper (Cu)-based nanoparticles (NPs)] combined with near-infrared (NIR) irradiation show enhanced antibacterial activity against clinical bacteria. However, their antibacterial efficiency toward implant-related infections and against biofilm formation remains unclear. Here, unique polyethylene glycol-modified Cu9S8 NPs with good biocompatibility were synthesized. We found that the Cu9S8 NPs exhibited high photothermal performance and could increase the generation of reactive oxygen species under NIR irradiation (808 nm, 1 W cm–2). The Cu9S8 NPs with NIR irradiation successfully destroyed the bacterial structure, resulting in the death of the clinically derived Staphylococcus aureus growing on titanium (Ti) plates. Moreover, this excellent antibacterial activity was indicated to have a synergistic effect with photothermal therapy (PTT) and photodynamic therapy (PDT) by comparison to Cu9S8 with heating treatment in a water bath with similar temperature changes compared to NIR + Cu9S8. Finally, the biofilm formation on the Ti plates was effectively disrupted by NIR + Cu9S8 treatment, while Cu9S8 with thermal treatment showed a mild impact. Hence, Cu9S8 NP-based PTT and PDT can provide a promising approach to eliminating implant-related bacteria and disrupting bacterial biofilms.

Platelet Membrane Coated Cu9S8-SNAP for Targeting NIR-II Mild Photothermal Enhanced Chemodynamic/Gas Therapy of Triple-Negative Breast Cancer

Danping Wu,Xiaoyi Chen,Shijie Yao,Yichen He,Gongning Chen,Xiaojuan Hu,Yang Chen,Zhenye Lv,Jing Yu,Ketao Jin,Yu Cai,Xiaozhou Mou

Triple-negative breast cancer (TNBC) is a highly aggressive and uncommon subtype of breast cancer with a poor prognosis. It is crucial to prioritise the creation of a nanotherapeutic method that is highly selective and actively targeting TNBC. This study explores a new nanosystem, Cu 9 S 8 -SNAP@PM (C-S@P), composed of Cu 9 S 8 -SNAP coated with a platelet membrane (PM). The purpose of this nanosystem is to cure TNBC using multimodal therapy. The utilisation of PM-coated nanoparticles (NPs) enables active targeting, leading to the efficient accumulation of C-S@P within the tumour. The Cu 9 S 8 component within these NPs serves the potential to exert photothermal therapy (PTT) and chemodynamic therapy (CDT). Simultaneously, the S-Nitroso-N-Acetylvanicillamine (SNAP) component enables nitric oxide (NO) gas therapy (GT). Furthermore, when exposed to NIR-II laser light, Cu 9 S 8 not only increases the temperature of the tumour area for PTT, but also boosts CDT and stimulates the release of NO through thermal reactions to improve the effectiveness of GT. Both in vitro and in vivo experimental results validate that C-S@P exhibits minimal side effects and represents a multifunctional nano-drug targeted at tumors for efficient treatment. This approach promises significant potential for TNBC therapy and broader applications in oncology.

CuS–Cu9S8 nanoparticles as an efficient catalyst: A facile method via anion controlled

Mengfan Liang,Yan Zhu,Shuhong Sun

In this paper, a facile, surfactant-free and mild solution chemistry route was employed to prepare copper sulfide (CuS–Cu 9 S 8 ) nanoparticles (NPs) by using anion-controlled agents with a different electronegativity. Counterions CH 3 COO − were found to be suitable for regular hexagonal morphology formation, whereas in the presence of NO − 3 NO3− , irregular structure is obtained. By changing the anion of the copper precursor, the ratio of formation of Cu 9 S 8 and CuS can be changed, and the band gap energy ( Eg ) of as-prepared NPs can be controlled in the presence of anion-controlled agents. Moreover, we demonstrate here the catalytic performance of the as-prepared samples for the degradation of methylene blue (MB) both under the simulated natural light and in dark, and the result shows that the CuS–Cu 9 S 8 NPs exhibited better catalytic activity under visible light ( > 5 0 >50 %) than in darkness (45%), and CuS–Cu 9 S 8 NPs with higher electronegativity of anion-controlled exhibited the higher catalytic performance. In addition, a simple and direct diagram was applied to illustrate the mechanism of the degradation of MB.

Non-Small Cell Lung Cancer Treatment via Cu9S8-Mediated NIR-II Photothermal Therapy

Huifang Hao,Zhe Yu,Cheng Cao,Yingao Jiao,Yu Cao,Xinyuan Cui,Yan Gao,Dengyao Gao,Shengsheng Cui,Chenyu Wang,Yanlei Liu

Non-small cell lung cancer (NSCLC) treatment is often compromised by acquired resistance and, in the context of photothermal therapy (PTT), by tumor thermotolerance─a protective stress response mediated by the molecular chaperone heat shock protein 90 (HSP90). To address this dual challenge, we developed a multifunctional nanoplatform (Cu9S8@Lum NPs) that synergistically integrates NIR-II PTT with HSP90 inhibition. This nanosystem is based on a Cu9S8 nanocore with high photothermal conversion efficiency, loaded with the HSP90 inhibitor Luminespib and designed for pH-responsive drug release. This spatiotemporally coordinated strategy enables concurrent tumor hyperthermia and molecular interference with the heat-shock defense mechanism. In vitro, the nanoparticles effectively suppressed HSP90 expression, exacerbated cellular oxidative stress, and induced mitochondrial apoptosis, resulting in markedly enhanced cytotoxicity compared to PTT alone. In vivo, the nanoplatform demonstrated effective tumor accumulation and, under mild NIR-II irradiation, achieved significant tumor growth inhibition in a syngeneic model. This work presents a mechanism-informed nanomaterial strategy to overcome thermotolerance in NSCLC, advancing the development of synergistic cancer nanotherapeutics.

Regulating copper homeostasis of tumor cells to promote cuproptosis for enhancing breast cancer immunotherapy

Guan Meng,Cheng Kai,Xie Xiao-Ting,Li Yong,Ma Meng-Wen,Zhang Bin,Chen Si,Chen Wei,Liu Bo,Fan Jin-Xuan,Zhao Yuan-Di

Cuproptosis is an emerging mode of programmed cell death for tumor suppression but sometimes gets resisted by tumor cells resist under specific mechanisms. Inhibiting copper transporter ATPase (ATP7A) was found to disrupt copper ion homeostasis, thereby enhancing the effect of cuproptosis and eventually inhibiting tumor invasion and metastasis. In this study, we develop a multifunctional nanoplatfrom based on Cu 9 S 8 (CAPSH), designed to enhance cuproptosis in tumor cells by specifically targeting ATP7A interference, while combining thermodynamic therapy with immune effects. The release of copper ions from CAPSH and the copper homeostasis interference by siRNA cooperatively increases the concentration of copper ions in tumor cells, which induces effectively cuproptosis and activates immune responses for suppressing development and metastasis of tumor. This nanoplatform simultaneously regulates cuproptosis from both principles of onset and development, facilitating the application of cuproptosis in tumor therapy.

Near-infrared-II photocharging nanozyme for enhanced tumor immunotherapy

Anshuo Li,Shuzhen Chu,Meng Yuan,Jinhui Zhang,Hengrui Liu,Yuhui Zhu,Jingyi Xu,Xinquan Jiang,Weili Xue

In tumor therapy, copper (Cu)-based nanozymes with peroxidase-like activity play a crucial role in converting hydrogen peroxide into hydroxyl radicals ( OH). This process induces immunogenic cell death, which in turn activates the body’s immune response, enhancing the efficacy of tumor immunotherapy. Nonetheless, the efficiency of this reaction is curtailed due to the oxidation of Cu(I) to Cu(II), leading to the self-depletion of the nanozyme’s activity and an insufficient yield of OH for effective immunotherapeutic activation. To surmount this challenge, our research introduces a photocharging self-doped semiconductor nanozyme, copper sulfide (Cu 9 S 8 ). The photocharging effect enables the nanozyme to convert internal Cu(II) back to Cu(I) through charge transfer induced by near-infrared (NIR)-II photothermal energy, thereby effectively maintaining the enzyme-like activity of the nanozyme. Additionally, Cu 9 S 8 is enhanced with a calcium sulfide (CaS) coating. This coating reacts in the acidic microenvironment of tumors to generate hydrogen sulfide (H 2 S) gas, which in turn suppresses the catalase activity inherent in tumor cells, ensuring a plentiful supply of H 2 O 2 for the nanozyme’s operation. This dual strategy of amplifying enzyme-like activity and substrate availability culminates in the generation of ample OH within tumor cells, leading to significant immunogenic cell death and thereby realizing potent immunotherapy.

Camouflaging multifunctional nanoparticles with bacterial outer membrane for augmented chemodynamic/photothermal/starvation/chemo multimodal synergistic therapy of orthotopic glioblastoma

Yichen He,Yi Pan,Xin Zhao,Luyi Ye,Longcai Liu,Wenliang Wang,Meng Li,Dapeng Chen,Yu Cai,Xiaozhou Mou

Innovative therapeutic strategy and effective drug delivery are significant challenges in current glioblastoma multiforme (GBM) therapy. The development of efficient combination therapy based on chemodynamic therapy (CDT) has attracted widespread attention in recent years. Herein, a lipopolysaccharide-free bacterial outer membrane vesicles (dOMV) camouflaged biomimetic nanoreactor was constructed for quadruple synergistic therapy of orthotopic GBM. Benefiting from the immune escaping property and blood–brain barrier (BBB) penetration capacity of dOMV, nanoreactors can target cancer cells and accumulate in the tumor site. Firstly, for quadruple synergistic therapy, Glucose oxidase (GOx) can rapidly consume intratumoral glucose to generate H 2 O 2 and gluconic acid for starvation therapy. Secondly, the abundant H 2 O 2 produced by the enzyme-catalyzed reaction can continuously supply Cu 9 S 8 -mediated Fenton-like reaction to enhance CDT. Meanwhile, tumor hypoxic microenvironment exacerbated by GOx-induced oxygen depletion activated prodrug AQ4N for chemotherapy. Finally, the second near-infrared (NIR-II) absorbed Cu 9 S 8 increased the temperature in local tumor site upon NIR-II laser irradiation, which can achieve photothermal-enhanced CDT, elevate GOx activity, and accelerate drug release. In vitro and in vivo results demonstrated that the camouflaging multifunctional nanoparticles with bacterial outer membrane ( [email protected] 9 S 8 @dOMV) was a promising multifunctional therapeutic nanomedicine platform for highly efficient treatment of GBM, which may be extended to as a treatment option for clinical cancer theranostics.
