Graphite carbon nitride (g-C 3 N 4 ) was regarded as a promising photodynamic antibacterial material due to its desirable bandgap and favorable visible light absorption. However, its photodynamic antibacterial effect was weakened by too rapid recombination of electron-hole and hypoxic environment of infection site. Herein, an oxygen self-supplied heterojunction was constructed via in-situ growing CuO 2 on g-C 3 N 4 (CuO 2 @g-C 3 N 4 ). For the heterojunction, on the one hand, it could response to the weak alkaline of infected site to decompose to CuO@g-C 3 N 4 , where CuO could form a built-in electric field with g-C 3 N 4 to promote the electron-hole separation. On the other hand, it could produce oxygen in the decomposition process, which achieved oxygen self-supply for photodynamic reaction. Subsequently, the heterojunction was introduced into poly-para-dioxane (PPDO) scaffold fabricated by laser additive manufacturing . The photoluminescence results confirmed that the electron-hole separation efficiency was significantly improved. The release kinetics results indicated that the scaffold could continuously release oxygen. As a consequence, the reactive oxygen species (ROS) generation ability of the scaffold was increased by approximately tenfold. The generated ROS effectively killed bacteria by disrupting membrane structure, triggering protein leakage and depleting glutathione . Eventually, the scaffold exhibited an antibacterial rate of 89.8% and 90.5% against S. aureus and E. coli , respectively.

Dopamine (DA) is a key neurotransmitter whose concentration affects various neurological disorders. Unlike previous methods that synthesize non-fluorescent polydopamine (NFL-PDA) under alkaline conditions, this study introduces a novel “turn-off” sensing method for DA using NFL-PDA synthesized through a unique reaction pathway. In our approach, CuO 2 nanodots, created via a simple reduction method, catalyze the formation of hydroxyl radicals (•OH) in acidic conditions, triggering the oxidative polymerization of DA into NFL-PDA. The reaction between DA and CuO 2 nanodots in acidic solution was examined to understand the process. UiO-66-NH 2 was then used to test NFL-PDA’s fluorescence quenching ability and to further investigate the DA determination mechanism. Results showed that fluorescence quenching was due to enhanced non-radiative energy transfer and Förster resonance energy transfer (FRET) between NFL-PDA and UiO-66-NH 2 . This led to the development of a simple “turn-off” fluorometric DA determination method with a linear range of 0.1–200 μmol/L and a limit of detection (LOD) of 0.0575 μmol/L. Although this method does not outperform existing NFL-PDA synthesis methods under alkaline conditions, it provides a new synthesis approach and application for sensing DA, contributing to the basic theory of chemical sensing. Additionally, DA determination in human serum samples was successfully achieved, with results consistent with high-performance liquid chromatography (HPLC). Graphical abstract

Bacterial infections considerably limit the success of scaffold transplantation. CuO 2 may generate hydroxyl radicals through a self-cascade catalytic process, which is effective for sterilization. However, reducing CuO 2 -derived Cu + ions to Cu° can lead to a decline in catalytic activity. To overcome these limitations, we anchored CuO 2 onto MoS 2 to establish stable Cu-S-Mo interfacial sites. The CuO 2 /MoS 2 nanozyme was fabricated using a low-temperature co-precipitation technique and introduced into poly- l -lactic acid scaffolds using selective laser sintering. The Cu-S-Mo interfacial sites showed a triad of functions that synergistically enhanced catalytic performance. These sites stabilized the Cu atoms, augmented catalytic activity by facilitating efficient electron transfer through the Mo 4+ /Mo 6+ redox couple, and improved the conduction of heat derived from the photothermal response of MoS 2 to the Cu catalyst. The synergistic action of chemodynamic and photothermal therapy, mediated by the Cu-S-Mo sites, resulted in remarkable antibacterial efficacy, with 92.3 % and 93.5 % inhibition rates against Staphylococcus aureus and Escherichia coli , respectively. Experimental evidence and simulations demonstrated that Cu-S-Mo structure stabilized the Cu atoms and improved electron transfer via interatomic charge transfer. These results highlight the potential of the CuO 2 @MoS 2 /PLLA scaffold in eliminating bacterial infections.

Cuproptosis shows great prospects in cancer treatments. However, insufficient intracellular copper amount, low-level redox homeostasis, and hypoxic tumor microenvironment severely restrict cuproptosis efficacy. Herein, hydrazided hyaluronan-templated decorated CuO 2 -doxorubicin (CuDT) nanodot clusters (NCs) are developed for efficient doxorubicin (DOX)-sensitized cuproptosis therapy in breast cancer via a three-pronged strategy. The CuDT NCs with an average size of 56.2 nm are fabricated from 3,3′-dithiobis(propionohydrazide)-conjugated hyaluronan, Cu 2+ , and DOX through a one-pot mineralization process. The CuDT nanoparticles exhibit pH-responsive H 2 O 2 , Cu 2+ , and DOX release profiles and catalytic activity. Upon entrance into tumor cells, CuO 2 -based exogenous H 2 O 2 supply and DOX-augmented endogenous H 2 O 2 generation jointly elevate intracellular H 2 O 2 level, which can further be transformed into hydroxyl radicals and O 2 through Fenton-like reaction to achieve oxidative stress amplification and hypoxia relief, respectively. Moreover, the CuDT NCs can efficiently deplete intracellular overexpressed glutathione via Cu 2+ /Cu + cycle and abundant disulfide bonds, further enhancing cellular oxidative stress. These results demonstrate that the novel CuDT NCs achieve DOX-sensitized cuproptosis in breast cancer cells through elevating copper level, amplifying oxidative stress and alleviating hypoxia, and display prominent in vivo antitumor efficacy. Such a three-pronged strategy of targetedly boosting cuproptosis in cancer cells represents a novel approach for antitumor treatments.

The development of metal-based redox nanozymes represents a new frontier in pollutant sensing. In this field, designing highly active nanozymes and precisely regulating their enzymatic activity are key challenges. In this work, we report the construction of a copper peroxide@polydopamine (CuO2@PDA) nanozyme with dual enzyme-like activity, mimicking the active centers of laccase and peroxidase. Here, CuO2 acts as the catalytic center, while PDA serves as a carrier to prevent CuO2 aggregation and promotes conversion of CuII/CuI active sites via the reduction effect of its surface catechol groups to complete the catalytic cycle. As expected, the obtained CuO2@PDA nanozyme exhibits significant laccase- and peroxidase-mimetic activities. Moreover, its dual enzymatic activity can be systematically switched by adjusting pH and temperature. Specifically, laccase activity dominates near neutral pH, while CuO2 decomposition into Cu ions and H2O2 at acidic pH triggers peroxidase activity. Similarly, CuO2@PDA exhibits temperature-dependent dual enzymatic activity with peroxidase activity prevailing at low temperatures and laccase activity at high temperatures. According to enzymatic performance and XPS results, a possible catalytic mechanism of the dual enzymatic activity of CuO2@PDA has been proposed. Then, based on the pH-dependent dual enzymatic activity of CuO2@PDA, we constructed a detection system for the isomers of organic pollutants, catechol (CC) and hydroquinone (HQ). The laccase-like activity of CuO2@PDA enables direct oxidation of CC into yellow o-benzoquinone, while HQ discolors the preoxidized substrate generated via the peroxidase-like activity of CuO2@PDA. Moreover, selective sensing for CC and HQ with high sensitivity was achieved in real water samples. This approach can guide the design of nanozymes with multienzymatic activity and unveil their potential uses in environmental pollutant discrimination.

Single-atom catalysts (SACs), characterized by atomically dispersed single metal atoms, provide superior antibacterial properties over common nanozyme materials due to their satisfactory catalytic activity. However, when used for the treatment of biofilm-infected wounds, SACs are still unable to effectively solve severe microbial infection due to the insufficient supply of reactive oxygen species (ROS). Herein, a new hybrid nanozymes consisting of Zn single-atom catalysts and copper peroxide (Zn SACs@CuO 2 ) were prepared to realize the upgrading of ROS generation and the combination of photothermal/chemodynamic (PTT/CDT) therapy for biofilm-infected wound. Specifically, CuO 2 exhibited not only excellent photothermal property, but also H 2 O 2 -producing and Fenton-like catalytic activities, which assisted Zn SACs to accelerate the ·OH conversion of endogenous and exogenous H 2 O 2 . Compared with single antimicrobial strategies of Zn SACs, Zn SACs@CuO 2 hybrid nanozymes exhibited stronger therapeutic effects of biofilm destruction, immune activation and inflammation improvement. In conclusion, this work developed a hybrid single atom nanozyme material with for efficient biofilm-infected wound healing.

The abnormal fluctuations of glutathione (GSH) in vivo often reflect disease progression and are closely linked to human metabolism and physiological functions. Highly sensitive and selective detection of GSH is crucial for guiding early diagnosis and treatment; however, achieving this remains a significant challenge. In this study, we developed an enzyme activity sensor platform for the efficient detection of GSH. This platform utilizes Au@CuS core–shell materials loaded with CuO 2 nanoparticles to create a composite nanosensor system. Under slightly acidic conditions, CuO 2 on the nanomaterial’s surface decomposes into H 2 O 2 and Cu 2+ ions. The generated H 2 O 2 then reacts with tetramethylbenzidine (TMB) in the presence of peroxidase-like CuS to yield oxidized tetramethylbiphenyl ( OX TMB), which generates a distinctive Raman signal. Upon addition of GSH to the system, the unique OX TMB signal diminishes due to GSH’s strong antioxidant capacity and the consequent consumption of OX TMB. This sensing method enables sensitive detection of GSH, with a detection limit as low as 1.2 × 10 -13 mol∙L -1 . This approach holds promise for providing researchers with rapid and precise in vitro analysis of GSH, serving as an indicator for early disease diagnosis and real-time evaluation of treatment efficacy.

Diabetic chronic wounds pose significant clinical challenges due to their characteristic features of impaired extracellular matrix (ECM) function, diminished angiogenesis, chronic inflammation, and increased susceptibility to infection. To tackle these challenges and provide a comprehensive therapeutic approach for diabetic wounds, the first coaxial electrospun nanocomposite membrane is developed that incorporates multifunctional copper peroxide nanoparticles ( n -CuO 2 ). The membrane's nanofiber possesses a unique “core/sheath” structure consisting of n -CuO 2 +PVP (Polyvinylpyrrolidone)/PCL (Polycaprolactone) composite sheath and a PCL core. When exposed to the wound's moist environment, PVP within the sheath gradually disintegrates, releasing the embedded n -CuO 2 . Under a weakly acidic microenvironment (typically diabetic and infected wounds), n -CuO 2 decomposes to release H 2 O 2 and Cu 2+ ions and subsequently produce ·OH through chemodynamic reactions. This enables the anti-bacterial activity mediated by reactive oxygen species (ROS), suppressing the inflammation while enhancing angiogenesis. At the same time, the dissolution of PVP unveils unique nano-grooved surface patterns on the nanofibers, providing desirable cell-guiding function required for accelerated skin regeneration. Through meticulous material selection and design, this study pioneers the development of functional nanocomposites for multi-modal wound therapy, which holds great promise in guiding the path to healing for diabetic wounds.

Melanoma is a malignancy of the skin that is resistant to conventional treatment, necessitating the development of effective and safe new therapies. The percutaneous microneedle (MN) system has garnered increasing interest as a viable treatment option due to its high efficacy, minimal invasiveness, painlessness, and secure benefits. In this investigation, a sensitive MN system with multiple functions was created to combat melanoma effectively. This MN system utilized polyvinylpyrrolidone (PVP) as microneedle substrates and biocompatibility panax notoginseng polysaccharide (PNPS) as microneedle tips, which encapsulated PVP-stabilized CuO2 nanoparticles as a therapeutic agent and disulfiram-containing F127 micelles to enhance the tumor treatment effect. The MN system had sufficient mechanical properties to pierce the skin, and the excellent water solubility of PNPS brought high-speed dissolution properties under the bio conditions, allowing the MNs to effectively penetrate the skin and deliver the CuO2 nanoparticles as well as the drug-loaded micelles to the melanoma site. CuO2 nanoparticles released by the MN system generated Cu2+ and H2O2 in the tumor acidic environment to achieve self-supply of hydrogen peroxide to chemodynamic therapy (CDT). In addition, Cu2+ was chelated with disulfiram to produce CuET, which killed tumor cells. And the MN system had excellent near-infrared (NIR) photothermal properties due to the loading of CuO2 nanoparticles and induced localized thermotherapy in the melanoma region to further inhibit tumor growth. Thus, the designed MN system accomplished effective tumor suppression and minimal side effects in vivo via combined therapy, offering patients a safe and effective option for melanoma treatment.

Tumor hypoxia, limited H 2 O 2 levels, and abundant glutathione (GSH) in the tumor microenvironment (TME) pose challenges for high-efficiency reactive oxygen species (ROS)-mediated therapies. To address these limitations, an intelligent ROS-mediated nanoplatform, Ce6@ZIF-8/PDA/CuO 2 /HA (CZPCH), with photodynamic activity, self-supplied H 2 O 2 , Fenton-like reaction, GSH-depletion and photothermal activity was designed for efficient tumor treatment. In the acidic TME, CuO 2 in CZPCH deposits into Cu 2+ and H 2 O 2 , consuming GSH and triggering Fenton-like reactions to generate hydroxyl radical (•OH), enabling efficient chemodynamic therapy (CDT). Moreover, the presence of Ce6 and PDA enables CZPCH to exhibit excellent singlet oxygen ( 1 O 2 ) and temperature increase performance 660 nm and 808 nm laser irradiation, respectively, resulting in efficient photodynamic therapy (PDT) and photothermal therapy (PTT) efficacy. Interestingly, the temperature increase from PTT significantly enhances •OH and 1 O 2 generation, achieving high-efficiency ROS-based therapeutic efficacy. This synergistic CDT/PDT/PTT treatment demonstrates excellent anti-tumor efficacy and illuminates novel ROS-based therapeutic strategy design.

Nanoagents for chemodynamic therapy (CDT) hold a promising future in the field of antimicrobials, especially copper peroxide (CuO2) (CP) nanomaterials which have garnered significant attention due to their ability to self-supply H2O2. Nevertheless, the poor stability of CuO2 remains a critical challenge which restricts its practical application in the antibacterial field. In this study, an advanced nano-antimicrobial system HA-CP@Fe3O4 with enzyme-responsive properties is developed by coating hyaluronic acid (HA) on CuO2-loaded iron tetraoxide nanoparticles. The coating of HA not only stabilizes the CuO2 nanomaterials but also provides responsiveness towards the enzyme hyaluronidase, which is typically secreted by some bacteria. The outer layer of HA in HA-CP@Fe3O4 undergoes decomposition in the presence of hyaluronidase-secreting bacteria, resulting in the release of CuO2@Fe3O4. The released CuO2@Fe3O4 then self-supplies H2O2 and generates reactive oxygen species (ROS) within the infected microenvironment through Fenton and Russell effects, to ultimately achieve effective and precise antimicrobial activity. Simultaneously, the magnetic property provided by Fe3O4 allows the substance to be directed towards the infection site. Both in vitro and in vivo tests demonstrated that HA-CP@Fe3O4 exhibited excellent antimicrobial capabilities at low concentration (30 μg mL−1), exceptional biocompatibility and the ability to accelerate wound healing. The findings of this work offer a new and promising approach for targeted and precise CDT.

Marine biofouling is an important problem that disturbs the development of marine economy. Antifouling coating is an effective way to solve the problem. Here, we used free radical polymerization and condensation reaction to prepare a degradable zwitterionic polyurethane ( TA/CuO 2 -PU) n coating with poly(ε-caprolactone) (PCL) as the segments of the main chain and zwitterionic poly (sulfobetaine methacrylate) (PSBMA) as the side chains cross-linked with tannic acid (TA), a natural antibacterial agent, which could release a novel TA/CuO 2 nanoparticle antifouling agents. The TA/CuO 2 nanoparticles were synthesized and modified with tannic acid. The ultra-miniaturized nanostructures and the ability to produce reactive oxygen species (ROS) also endow the TA/CuO 2 nanoparticles with higher antimicrobial activity, which improve the utilization efficiency to reduce copper toxicity. In addition, the ( TA/CuO 2 -PU) n coating that was hydrophilic modified with zwitterionic PSBMA could block microbial adhesion. And slow biodegradation of the PU with PCL segment coating allowed stable release of the TA/CuO 2 nanoparticles and self-renewable surface. The degradation rate and antiadhesion properties of the ( TA/CuO 2 -PU) n coatings were increased with PSBMA content and the antifouling ability of the (TA/CuO 2 -PU) n coatings was increased with additive amount of TA/CuO 2 nanoparticles and degradation progress. These series of tests prove that we have successfully developed a self-renewable anti-fouling coating with both passive anti-adhesion and active release of biocides and this synergistic system may provide new ideas for the design and preparation of novel anti-fouling coatings.
