Background As a vital soil-borne pathogenic bacterium, Ralstonia solanacearum can cause wilt disease in multiple Solanaceae plants. Several phages, such as ϕPB2, could infect R . solanacearum acting as a potential biological control agent in soil. In addition, some nanoparticles, especially copper preparation, also showed high toxicity on R. solanacearum with low toxicity on plant. However, whether they can be administered in combination and how effective they are in inhibiting the plant disease caused by R . solanacearum is known very little. Results In this work, the characterization of CuONPs using scanning electron microscope, transmission electron microscope, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, and X-ray diffraction ascertained the presence of CuONPs which were nanometer particle of 83 nm. Then it was found that combined application of CuONPs with phage (ϕPB2) was superior to that of ϕPB2 or CuONPs alone in controlling tobacco bacterial wilt, with the CuONPs (250 mg/mL) and phage (10 6 PFU/mL) ratio being the best, at 79.1%. The combination of CuONPs and ϕPB2 also showed no obvious toxicity on tobacco growth than control like single application of CuONPs or ϕPB2. Furthermore, the transcriptome changes of R . solanacearum analysis indicated that the combination application and single allocation of CuONPs could inhibit “biofilm formation”, molecular function, biological processes, cellular components, metabolic process, and so on. In addition, the combination application showed higher inhibition of motility and biofilm, and better enhancement of cell membrane permeability, protein leakage, MDA concentration, and enzyme activity of their respiratory chain dehydrogenase than single application of CuONPs or phage (ϕPB2). Transcriptomes analysis also supported that the addition of ϕPB2 enhanced the toxicity of CuONPs by influencing the ABC transporters and quorum sensing, metabolic processes, and cellular biosynthetic processes of R. solanacearum . Conclusion In total, our work not only proposed a novel way to increase the bactericidal effect of nanomaterials by adding phage, but also discovered the influence, synergistic effects, and mechanisms, which is useful to design novel way to combat phytopathogenic bacteria in the complicated environment. Previous studies investigating the risk of engineered nanoparticles (NPs) to biological wastewater treatment have primarily tested NPs individually; however, limited data are available on the impact of NPs on the anaerobic ammonium oxidation (anammox) process. In this study, the toxicity of CuNPs on anammox granules was investigated individually and in combination with CuONPs or ZnONPs. Exposure to CuNPs at 5 mg g −1 suspended solids (SS) decreased the anammox activity to 47.1 ± 8.5%, increased the lactate dehydrogenase level to 110.5 ± 3.4% and increased the extracellular N 2 H 4 concentration by 16-fold but did not cause oxidative stress. The presence of CuONPs or ZnONPs at 5 mg g −1 SS did not significantly aggravate or alleviate the toxicity of the CuNPs; however, the introduction of EDTA or S 2− could attenuate the adverse effects of the CuNPs, CuONPs and ZnONPs on the anammox granules. EDTA captured Cu ions, whereas S 2− shielded and deactivated Cu ions and passivated CuNPs. Therefore, our results indicated that the toxicity of NPs was dependent on the amount of active metal reaching the anammox cells. Overall, the results of this study have filled knowledge gaps and provided insights into the combined toxicity of NPs on anammox biomass.

A method of green chemistry approach to synthesize copper oxide nanoparticles (CuONPs) was reported using extracts from the root, stem, and leaves of the Macleaya cordata (Wild.) R.BR as reducing and stabilizing agents. Copper acetate precursor (Cu(CH 3 COO) 2 ) was used for green synthesis of copper oxide nanoparticles through reduction and oxidation reactions. The synthesized CuONPs were characterized using UV-Vis, XRD, FTIR, SEM, TEM, zeta potential, and energy-dispersive X-ray spectroscopy (EDS). The results of these analyses provided valuable insights into the structure, shape, functional groups, and elemental composition of the synthesized CuONPs. The antibacterial activity of the green synthesized CuONPs was evaluated using an agar diffusion assay. The result shows that the inhibition effect was concentration-dependent, and at the highest concentration (5 mg/mL), the CuONPs exhibited strong antibacterial activity against Escherichia coli and Staphylococcus aureus , with inhibition zones of 21 mm and 23 mm, respectively. The photocatalytic degradation of methylene blue (MB) at a concentration of 10 mg/mL by CuONPs (10 mg/mL) was evaluated using a photochemical reaction apparatus under simulated natural light. The degradation rate was found to be 65% within 3 h and 93% after 24 h. The degradation rate of higher concentrations of MB (100 mg/mL) was 47.9%. These results indicate that the synthesized CuONPs have great potential for rapidly breaking down textile dyes and exhibit exceptional antibacterial properties. This suggests that these nanoparticles could play a significant role in the development of sustainable nanotechnology.

Macrophage-derived foam cells are crucial in the development of atherosclerosis, a multifaceted and progressive disorder characterized by lipid and fibrous accumulation in major arteries. Copper oxide nanoparticles (CuONPs) have found widespread applications but their potential role in atherosclerosis remains understudied. In this study, we investigated the impact of CuONPs on foam cell formation in RAW264.7 macrophages. Our results showed that CuONPs, at concentrations as low as 10 μg/ml, significantly exacerbated foam cell formation induced by oxidized low-density lipoprotein (ox-LDL). Exposure to CuONPs stimulated LDL release and elevated the expression of NLRP3 inflammasome components, including NLRP3, Caspase-1, and IL-1β. Transmission electron microscopy (TEM) revealed accumulation of CuONPs within macrophage lysosomes, leading to disrupted lysosomal function. CuONPs-treated cells exhibited autophagosome accumulation due to impaired lysosomal degradation, as confirmed by Western blot analysis showing abnormal expression of LAMP-1 and LAMP-2 proteins. Flow cytometry analysis further demonstrated decreased lysosomal acidity in CuONPs-exposed cells. Our findings reveal a novel mechanism whereby CuONPs activate the inflammasome, disrupt lysosomal function, and hinder cholesterol efflux, thereby exacerbating the formation of macrophage-derived foam cells. These results highlight the potential risks of CuONPs exposure and provide important insights into the role of environmental particulate matter in the development of atherosclerosis.
