In Iranian, ethnopharmacological studies show that Cuminum cyminum is commonly used in pain relief. The most abundant component in C. cyminum seeds is cuminaldehyde (CUM). However, there are few articles that have evaluated the anti-biofilm activity against Staphylococcus aureus ( S. aureus ). The nobility of this study was to evaluate the anti-biofilm effect of CUM and to investigate its possible mechanism of action. Crystal violet staining assay showed that CUM decreased biofilm biomass. The morphological observation indicated that CUM restrained the adhesion of biofilm, and destroyed the integrated and cohesive structure of biofilm. MTT assay proved that the metabolic activity of adherent cells treated with CUM was reduced. In vitro assays exhibited that cell motility, cell surface hydrophobicity, and extracellular polymers production of S. aureus were also inhibited in the presence of CUM. Furthermore, quantitative real-time PCR analysis confirmed the ability of CUM to interfere with the expression of adhesion genes associated with biofilm formation such as agrA, icaA, sigB, ebpS, fnbA, fib and clfA . Molecular docking speculated that the interaction of CUM with SarA could inhibit SarA-DNA binding, and ultimately inhibit the formation of biofilm. The results of this trial provided scientific experimental data for the use of CUM as an anti-biofilm agent.

The previous study indicated that cuminaldehyde (CUM) could be used as an antibacterial agent in sauced beef to reduce the propagation of Staphylococcus aureus ( S. aureus ). This research took sauced beef treated with 0.4 μL/mL CUM as the research object. Transcriptomic and proteomic methods were used to comprehensively analyze the changes in genes and proteins of S. aureus under CUM stress. A total of 258 differentially expressed genes (DEGs, 178 up-regulated and 80 down-regulated) and 384 differentially expressed proteins (DEPs, 61 up-regulated and 323 down-regulated) were found. It was observed that CUM destroyed the cell wall and cell membrane by inhibiting the synthesis of peptidoglycan and fatty acid. Low energy consumption strategies were formed by reducing glycolysis and ribosome de novo synthesis. The levels of genes and proteins associated with the glycine, serine , threonine , methionine , cysteine, and branched-chain amino acids were dramatically changed, which impaired protein synthesis and reduced bacterial viability . In addition, the up-regulated DEGs and DEFs involved in DNA replication , recombination and single-stranded DNA-binding contributed to DNA repair . Moreover, ATP-binding cassettes (ABC) transporters were also perturbed, such as the uptake of betaine and iron were inhibited. Thus, this study revealed the response mechanism of S. aureus under the stress of CUM, and provided a theoretical basis for the application of CUM in meat products.

Cuminaldehyde (CA) has a strong antimicrobial activity against an array of pathogens. In this study, its growth inhibitory effect on a wild type strain (Pds01) and an imazalil-resistant strain (Pdw03) of Penicillium digitatum , the causative agent of postharvest green mold in citrus fruit was examined. CA strongly inhibited mycelial growth of both strains with minimum inhibitory concentration (MIC) of 0.3 mL L -1 and minimum fungicidal concentration (MFC) of 0.6 mL L -1 . In vivo assays revealed that wax + CA (WCA, 10 × MFC) treatment could significantly reduce green mold incidence in Ponkan fruit incubated with P. digitatum . After 5 d of incubation, the disease incidence of 10 × MFC WCA treated fruit was 65.0 ± 3.4 % for Pds01 and 50.0 ± 2.1 % for Pdw03 which were much lower than the corresponding values in the control samples. The antifungal activity was attributed to activation of massive oxidative stress , as evidenced by the accumulation of reactive oxygen species, increased lipid peroxidation , damage to plasma membrane permeability of hyphal cells, leakage of intracellular components and eventual cell death. The imazalil-resistant strain was more susceptible to the damaging effect of CA than the wild type strain. Collectively, our present results suggest that CA may potentially be a new antifungal drug molecule for the control of postharvest green mold in citrus fruit.

ObjectivesCuminaldehyde self-emulsified nanoemulsion (CuA-SEN) was prepared and optimised to improve its oral bioavailability and antihepatotoxicity.MethodsCuminaldehyde self-emulsified nanoemulsion was developed through the self-nanoemulsification method using Box–Behnken Design (BBD) tool while appropriate physicochemical indices were evaluated. The optimised CuA-SEN was characterised via droplet size (DS), morphology, polydispersity index (PDI), zeta potential (ZP), entrapment efficiency, in-vitro release, and pharmacokinetic studies while its antihepatotoxicity was evaluated.Key findingsCuminaldehyde self-emulsified nanoemulsion with acceptable characteristics (mean DS-48.83 ± 1.06 nm; PDI-0.232 ± 0.140; ZP-29.92 ± 1.66 mV; EE-91.51 ± 0.44%; and drug-loading capacity (DL)-9.77 ± 0.75%) was formulated. In-vitro drug release of CuA-SEN significantly increased with an oral relative bioavailability of 171.02%. Oral administration of CuA-SEN to CCl4-induced hepatotoxicity mice markedly increased the levels of superoxide dismutase, glutathione and catalase in serum. Also, CuA-SEN reduced the levels of tumour necrosis factor-alpha and interleukin-6 in both serum and liver tissues while aspartate aminotransferase, alanine aminotransferase and malonaldehyde levels were significantly decreased.ConclusionsThese findings showed that the improved bioavailability of cuminaldehyde via SEN provided an effective approach for enhancing antioxidation, anti-inflammation and antihepatotoxicity of the drug.

The antibacterial mechanisms of cuminaldehyde ( p -Isopropylbenzaldehyde, CUM) against Staphylococcus aureus ATCC 6538 ( S. aureus ) were investigated from the aspects of membrane damage, and interaction with genomic DNA. The results demonstrated that the MIC of CUM against S. aureus was 0.8 μL/mL. Approximately 6 log CFU/mL of S. aureus could be killed after treatment with MIC of CUM after 8 h. Additionally, CUM increased the permeability of the cell membrane by inducing visible changes in cell morphology. The results of gel electrophoresis, spectroscopic analysis, and molecule docking indicated that CUM could bind to DNA through groove mode and change the secondary structure of DNA, and ultimately lead to the inactivation of cells. Compared with untreated samples, application of CUM (2 or 4 MIC) on sauced beef caused an obvious decrease in S. aureus counts and effectively inhibited the growth of S. aureus . Moreover, CUM at a concentration of 2 MIC had no significant effect on the taste and aroma score of sauced beef. Therefore, CUM could play a dual antibacterial effect by influencing cell membrane and genomic DNA, and controlling the growth of S. aureus in the food industry.

The inhibitory mechanism of CUM on Staphylococcus aureus exotoxins and invasive enzyme, and the effect of CUM on sauced beef were investigated. Treatment with sub-minimum inhibitory concentrations of CUM observed very little decrease in cell density after 24 h. Additionally, CUM reduced hemolytic activity by inhibiting the formation of heptamer, and inhibited the content of enterotoxin in culture medium and sauced beef. The transcription of genes encoding α-hemolysin, staphylococcal enterotoxin A and Panton-Valentine leukocidin were down-regulated after treatment with CUM. Compared with the control group, the addition of CUM caused a significant decrease in S. aureus counts, total volatile base nitrogen, acid value, and thiobarbituric acid reactive substances of sauced beef after 9 d. Furthermore, CUM improved the redness (a*) of sauced beef. These results confirmed that CUM could effectively restrain the synthesis of S. aureus toxin, and the addition of CUM to sauced beef was the potential way to inhibit the growth of food pathogens and maintain quality profiles.

With the continuous expansion of cultural scale and the deterioration of cultural environment recently, various viral diseases occur frequently in cultured shrimp industry. White spot disease (WSD) caused by white spot syndrome virus (WSSV) results in a high mortality as well as a huge economic loss of shrimp culture. As a kind of Chinese herbal medicine extract, the application of cuminaldehyde in prevention and treatment of WSSV was investigated in this study. The data showed that cuminaldehyde at 40 μmol/L could significantly inhibit WSSV replication in shrimp within the highest inhibition rate of 89% at 72 hpi. The survival rate of WSSV-infected shrimp was increased by >50% after continuous treatment of cuminaldehyde versus the virus group at 144 hpi. Obviously, the horizontal transmission of WSSV was inhibited by cuminaldehyde in a static cohabitation challenge model, further weakening the spread of the virus to reduce the mortality of the virus-infected shrimp. Meanwhile, cuminaldehyde increased resistance of shrimp to WSSV by activating the innate immune response, which was essential for prevention of WSSV. Thus, cuminaldehyde could potentially be used as a therapeutic agent with a great potential in culture of shrimp seedling and may also be suitable for applications in pond aquaculture settings against WSSV transmission.

The study investigated the influence of cuminaldehyde (CUM) on cell membrane and genomic DNA of Salmonella enterica serovar Typhimurium ( S . Typhimurium), and revealed its antibacterial mechanism. The minimum inhibitory concentration (MIC) of CUM for S . Typhimurium was determined to be 0.6 μL/mL. CUM could bind to bacterial membrane proteins and change its structure and conformation, leading to the leakage of intracellular substances and the destruction of cell integrity. DNA interaction analysis showed that CUM bound to genomic DNA through groove mode, affecting the normal functioning of S . Typhimurium. In addition, CUM at sub-MIC decreased bacterial motility, prevent cell attachment, restrain metabolic activity, and thus inhibiting S . Typhimurium biofilm formation. Furthermore, mild heat treatment combined with CUM at 1, 2 and 4 MIC could completely kill S . Typhimurium in powdered infant formula. Besides, the combination at 1 MIC of CUM had no obvious influence on the aroma of the powdered infant formula. The combined application of CUM has broad prospects in the control of food-borne pathogens.

CUM encapsulated by β-CDP enhances water solubility and stability, improving its effects. • CUM-β-CDP shows lower MIC on E. coli (40 mg/ml) and S. aureus (5 mg/ml) than pure CUM. • CUM-β-CDP effectively inhibits HepG2 cell proliferation, showcasing potent anti-cancer ability. A pH-responsive delivery system based on supramolecular vesicles was fabricated via a novel host–guest inclusion complex between water-soluble pillar[5]arene (WP5) and cuminaldehyde (CUM). This CUM-WP5 inclusion complex was studied using 1 H NMR, UV-vis, and FT-IR spectroscopy. The resulting inclusion complex could self-assemble into well-defined supramolecular vesicles via host–guest interactions and exhibited significant pH-responsive behavior, which was investigated using dynamic light scattering, transmission electron microscopy, and UV-vis spectroscopy. The preservation time of CUM-WP5 and the molar ratio between host and guest molecules were both significant for the aggregated morphology of the supramolecular vesicles. Furthermore, the CUM complexes demonstrated good antibacterial activity, inhibiting the proliferation of Agrobacterium tumefaciens GV3101 and EHA105. These vesicles therefore present a promising platform for drug delivery and antibacterial applications. Cuminaldehyde demonstrates strong antimicrobial properties against several disease-causing organisms. This study investigated its antifungal effectiveness against an imazalil-resistant strain of Penicillium italicum , the pathogen responsible for postharvest blue mold disease in citrus fruits. Cuminaldehyde significantly inhibited the mycelial growth of P. italicum , with both the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) estimated at 0.5 mL/L. In vivo assays revealed that treatment with 4 × MFC cuminaldehyde in Tween-80 significantly reduced blue mold incidence in Ponkan mandarins inoculated with P. italicum . After 6 days of incubation, the disease incidence in fruits treated with 4 × MFC cuminaldehyde was approximately 72%, compared to 80% in the Prochloraz-treated fruits and 100% in the control group. The antifungal activity was attributed to the activation of oxidative stress, as evidenced by the accumulation of reactive oxygen species, damage to plasma membrane permeability in hyphal cells, and leakage of intracellular components. Additionally, treatment with 4 × MFC cuminaldehyde preserved the quality of the inoculated Ponkan mandarins by day 6. Overall, these findings indicate that cuminaldehyde is a potent antifungal alternative to current chemical fungicides used for controlling postharvest blue mold disease in Ponkan mandarins.

The contamination of food with Aspergillus flavus has caused serious economic losses and posed a great threat to human and animal health. Cumin , a condiment and flavoring in many dishes, was showed to have great potential to prevent A. flavus contamination in the present study. By using cumin essential oil (CEO), the occurrence of A. flavus contamination on peanuts was prevented during the storage period even up to 9 months. We further identified cuminaldehyde as the predominant component in CEO with great inhibitory activity to both the growth of A. flavus and aflatoxin B 1 (AFB 1 ) biosynthesis. The data showed that the minimum inhibitory concentration (MIC) of cuminaldehyde to conidial growth was 0.64 μL/mL, and the mycelia exposed to 0.80 μL/mL cuminaldehyde reduced AFB 1 production by more than 99% in comparison to the control. RNA-Seq profiles showed that some genes, involving the synthesis pathway of ergosterol and AFB 1 , were significantly down-regulated in the transcription by cuminaldehyde. Further investigation confirmed a significantly reduced cellular ergosterol content in the presence of cuminaldehyde, and the qRT-RCR results were also in agreement with that by RNA-Seq, indicating down-regulated expressions of those genes involving ergosterol and AFB 1 synthesis. Taken together, our study strongly supported cuminaldehyde as a potential agent for the prevention of A. flavus contamination.

Cuminaldehyde (CA), a major bioactive constituent of cumin essential oil, was incorporated into gelatin/konjac glucomannan (Gel/KGM) composite films at different levels, expressed relative to the polymer content (mL/g polymer), to investigate its effects on film structure, physicochemical properties, and antioxidant activity. Structural characteristics of the films were examined using Fourier transform infrared spectroscopy (FTIR), Raman spectroscopy, X-ray diffraction (XRD), and scanning electron microscopy (SEM). Mechanical properties, surface wettability, optical properties, water sensitivity, and antioxidant activity were also determined. Spectroscopic analyses indicated changes in the molecular interactions of the Gel/KGM matrix following CA incorporation, while XRD and SEM analyses showed reduced structural order and a more heterogeneous microstructure at higher CA levels. Representative contact-angle observations showed an initial angle of 40.2° for the Control and 58.0° for CA-10, suggesting a tendency toward reduced surface wettability after CA incorporation. The addition of CA also improved UV-shielding properties and enhanced antioxidant responses, although the magnitude and trend of the antioxidant effect depended on the assay. In contrast, higher CA levels significantly reduced tensile strength and increased water solubility, whereas the tensile strength of films containing 1 mL CA did not differ significantly from that of the control (p> 0.05). These results demonstrate that the structural, physicochemical, and functional responses of Gel/KGM composite films varied with the nominal CA loading. Among the tested formulations, CA-3 represented a possible compromise between structural integrity and functional performance. Further studies using real food matrices are required to validate its practical applicability under actual food-packaging conditions.
