Structural elucidation and anti-osteoporosis activities of polysaccharides obtained from Curculigo orchioides

Xueqian Wang,Mengliu Zhang,Dawei Zhang,Xinluan Wang,Huijuan Cao,Qian Zhang,Chunyan Yan

Curculigo orchioides , is a traditional Chinese medicine, is used in strengthening tendons and bones. We evaluated the anti-osteoporosis activity of the crude polysaccharide (CO70) isolated from the rhizomes of C. orchioides in ovariectomized rats. CO70 showed excellent anti-osteoporosis activity comparable to that of 17 β -estradiol. To explore the constituents responsible for the anti-osteoporosis activity of CO70, a novel homogeneous heteropolysaccharide, COP70-3, was isolated and purified from CO70. COP70-3 has a main backbone chain of (1→5)-linked α -L-Ara f , (1,3→5)-linked α -L-Ara f , (1→6)-linked β -D-Gal p , (1→4)-linked β -D-Man p , (1,2→5)-linked α -L-Ara f , (1→3)-linked β -L-Rha p , (1, 3→6)-linked β -D-Man p , (1→3)-linked α -D-Gal p A, (1,3→6)-linked β -D-Gal p and (1→6)-linked α -D-Glc p residues. Furthermore, 1.87 nM COP70-3 obviously promoted the differentiation of MC3T3-E1 cells, while 0.94 and 1.87 nM COP70-3 significantly improved the osteogenic mineralization rate. These data indicate that COP70-3 has favorable anti-osteoporosis activity in vitro .

Structural characterization and anti-osteoporosis activity of two polysaccharides extracted from the rhizome of Curculigo orchioides

Yongbo Yu,Tianyu Li,Xueqian Wang,Mengliu Zhang,Qian Yu,Haiyun Chen,Dawei Zhang,Chunyan Yan

Curculigo orchioides is widely used to treat osteoporosis in China. In this study, we identified the active substances in the crude polysaccharide (CO50) from C. orchioides that had anti-osteoporosis activity in vivo. Two polysaccharides, COP50-1 and COP50-4, were purified from CO50. Based on structural analysis, COP50-1 was composed of α-D-Glcp-(1→, β-D-Galp-(1→, →4)-α-D-Glcp-(1→, →3,4)-α-D-Glcp-(1→, →4,6)-α-D-Glcp-(1→, →4,6)-β-D-Manp-(1→, whereas COP50-4 was composed of α-L-Araf-(1→, →2)-α-L-Rhap-(1→, β-D-Manp-(1→, α-D-Galp-(1→, →2,4)-α-L-Rhap-(1→, →2)-β-D-Manp-(1→, →4)-α-D-GlcAp-(1→, →3)-α-D-GalAp-(1→, →4,6)-α-D-Galp-(1→, →2,3,6)-β-D-Manp-(1→, →2,3,5)-α-L-Araf-(1→, →2,5)-α-L-Araf-(1→, →4)-α-D-Glcp-(1→ and →3)-α-D-Galp-(1→. Pharmacological assessment revealed that COP50-1 had no obvious osteogenic activity. However, COP50-4 (0.5 μM) significantly enhanced the differentiation and mineralization of osteoblasts in vitro. Moreover, the effect of COP50-4 was greater than that of 17β-estradiol. Therefore, COP50-4 may be an effective component of CO50 that has great potential for development as an alternative drug for the treatment of osteoporosis.

Ameliorative Effects of Curculigoside from Curculigo orchioides Gaertn on Learning and Memory in Aged Rats

Bao-Yuan Hou

This study was designed to evaluate the ameliorating effects of curculigoside fromCurculigo orchioidesGaertn on learning and memory in aged rats. In the present study, the ameliorating effects of curculigoside were determined through animal behaviour studies (including step-down test and Y-maze test), and the possible mechanisms were explored by evaluation of the activity of acetylcholinesterase (AchE) and determination of the expression of BACE1. Oral adminstration of the curculigoside (20, 40 mg/kg/day) for 14 days can significantly improve the latency and number of errors in aged rats based on the behaviour study results. In addition, the activity of AchE can be decreased by treatment of the curculigoside (10, 20, 40 mg/kg/day). Moreover, the expression of BACE1 can be down-regulated in the hippocampus of aged rats treated with curculigoside. The results of our present work have indicated that curculigoside can improve cognitive function in aged animals, possibly by decreasing the activity of AchE in the cerebra and inhibiting the expression of BACE1 in the hippocampus. In conclusion, our results suggested that curculigoside can be possible developed as a new drug for the treatment of Alzheimer’s disease in the future. Keywords:curculigoside;Curculigo orchioides;Alzheimer’s disease;behaviour study;ameliorative effects on learning and memory

Anti-depressant-like effect of curculigoside isolated from Curculigo orchioides Gaertn root

Jing Wang,Xiao-Li Zhao,Lan Gao

Purpose: To investigate the anti-depressant-like activity of curculigoside from Curculigo orchioides Gaertn and its underlying mechanism(s).Methods: Antidepressant-like activity was determined in mice through forced swimming test (FST), tail suspending test (TST), and open field test (OFT). Mechanism of action was investigated by measuring levels of dopamine (DA), norepinephrine (NE), and 5-hydroxytryptamine (5-HT) in chronic mild stress (CMS) rats using high-performance liquid chromatography-electron capture detector (HPLC-ECD). Western blotting was used to investigate the effect of curculigoside on the expression of brain-derived neurotrophic factor (BDNF) protein in rats. Results: In FST and TST, treatment of mice with curculigoside (10, 20, 40 mg/kg, p.o.) significantly reduced immobility time, which was, however, unaffected by locomotor activity when assessed in the OFT. The treatment led to significant increases in DA, NE and 5-HT, and up-regulation of BDNF protein expression in the hippocampus of the CMS rats. Conclusion: These results demonstrate that curculigoside exerts significant anti-depressant-like activity on mice, the mechanism of which involves increase in the levels of DA, NE, 5-HT, and up-regulation of BDNF expression. Thus, curculigoside can be considered a potential drug candidate for the treatment of depressive disorders. Keywords: Curculigo orchioides Gaertn, Curculigoside, Antidepressant-like activity, Behavioral studies

An O-acetyl-glucomannan from the rhizomes of Curculigo orchioides: Structural characterization and anti-osteoporosis activity in vitro

Xueqian Wang,Mengliu Zhang,Dawei Zhang,Shumei Wang,Chunyan Yan

Osteoporosis is characterized by a reduction in bone mass and bone mineral density, which weakens the bone. Due to the side effects associated with drugs that are currently used to treat this disease, an increasing number of studies have focused on the research of effective ingredients derived from natural products. In particular, polysaccharides extracted from Chinese herbal medicines have received increasing attention. In this study, we isolated a homogeneous polysaccharide (COP90-1) from the dried rhizomes of Curculigo orchioides , a famous traditional Chinese medicine that is widely used in China, and determined its structure using the combined methods of chemical and spectral analyses. In addition, its effects on the proliferation and differentiation of primary mouse osteoblasts were assessed. The results showed that COP90-1 can effectively promote the proliferation and differentiation of primary osteoblasts in vitro . Additional studies are warranted to study the effects of this compound in vivo .

Optimization of enzyme-based ultrasonic/microwave-assisted extraction and evaluation of antioxidant activity of orcinol glucoside from the rhizomes of Curculigo orchioides Gaertn.

Liu Xin,Hu Yan,Wei Dongfeng

The orthogonal design was used to optimize enzyme-based ultrasonic/microwave-assisted extraction (EUMAE) of orcinol glucoside (OG)-enriched extract from the rhizomes of Curculigo orchioides Gaertn. (RCO). The results indicated that the highest extraction yield of OG by EUMAE could reach 92.13 % using 70 % ethanol as solvent under the optimal conditions (the ratio of raw material to solvent was 1:8, extraction time was 15 min, extraction temperature was 50 °C, amount of enzyme was 3 mL, and microwave irradiation power was 400 W), which was more efficient than that by the conventional and classical extraction methods. Finally, the crude extract was then purified by macroporous adsorption resin, the OG content in the purified extract increased to 91.70 %. Compared to the conventional and classical methods, EUMAE was superior in terms of higher yields, less energy consumption, and better selectivity of the extracts controlled by the extraction parameters. Furthermore, it was obvious that OG from RCO had significant effect on the antioxidant activity.
