Turmeric ( Curcuma longa ), a typical source with recognized anti-inflammatory activity, is one such medicine-food homology source, yet its anti-inflammatory mechanisms and specific component combinations remain unclear. In this study, a net fishing method combining bio-affinity ultrafiltration and ultra-high performance liquid chromatography-mass spectrometry (AUF-LC/MS) was employed and 13 potential COX-2 inhibitors were screened out from C. longa . 5 of them (C1, 17, 20, 22, 25) were accurately isolated and identified. Initially, their IC 50 values were measured (IC 50 of C1, 17, 20, 22 and 25 is 55.08, 48.26, 29.13, 111.28 and 150.48 μM, respectively), and their downregulation of COX-2 under safe concentrations (400, 40, 120, 50 and 400 μM for C1, 17, 20, 22 and 25, respectively) was confirmed on RAW 264.7 cells. Further, in transgenic zebrafish ( Danio rerio ), significant anti-inflammatory activity at safe concentrations (15, 3, 1.5, 1.5 and 3 μg/mL for C1, 17, 20, 22 and 25, respectively) were observed in a dose-dependent manner. More importantly, molecular docking analysis further revealed the mode of interaction between them and the key active site residues of COX-2. This study screened out and verified unreported COX-2 ligands, potentially accelerating the discovery of new bioactive compounds in other functional foods.

Objective Essential oil derived from the rhizome of Curcumin longa (CL-EO) has marked anti-inflammatory, antioxidant, antimicrobial, anticancer, and antiviral activities. However, its effect on photodamaged skin has not yet been evaluated. In this study, the anti-skin photoaging activity of CL-EO was determined by performing an ultraviolet B (UVB)-induced skin aging assay. Methods CL-EO was extracted by hydrodistillation and characterized using gas chromatography–mass spectrometry. The anti-skin aging effect was determined by topically applying 150 μl of CL-EO diluted to 1, 5, and 10% with ethanol to the dorsal area of UVB-irradiated nude mice every day except on Sunday for 8 weeks. Histological and immunohistochemical analyses were performed. Results In total, 56 compounds, which accounted for 94.36% of the contents of CL-EO, were detected. The major compounds in CL-EO were ar-turmerone (36.04%), curlone (8.78%), β-turmerone (7.05%), 8,9-dehydro-9-formyl-cycloisolongifolene (5.69%), β-sesquiphellandrene (5.39%), germacrone (4.51%), ar-curcumene (2.19%), α-himachalene (2.14%), and ledane (2.13%). Hematoxylin and eosin staining of tissue sections revealed that CL-EO decreased epidermal skin thickness. Immunohistochemistry showed that CL-EO inhibited interleukin-1β and tumor necrosis factor-α expression. Conclusions Thus, CL-EO can reduce cutaneous photoaging in a UVB-irradiated nude mouse model. Therefore, CL-EO could be used in the formulation of skin care and functional cosmetic products.

Helicobacter pylori ( H. pylori ) resistance due to antibiotics is becoming increasingly prevalent, and the search for healthy alternatives to drugs has become a popular research topic. Curcuma longa and Lactobacillus rhamnosus are not only inhibitory to H. pylori but also have good compatibility. It was found that acid-tolerant L. rhamnosus had a significant coaggregation effect on H. pylori , but the inhibitory effect was weak. The polyphenol or flavonoid content of Curcuma extract was significantly correlated with the antimicrobial effect, but the inhibition time would be shortened due to its decomposition or destabilization in the digestive system. Both were able to synergistically inhibit H. pylori adhesion and expression of certain genes ( SabA , UreE , UreI , and UreG ), with no significant synergistic effect in inhibiting urease. The combination of Curcuma extract with L. rhamnosus enhanced the inhibitory effect, solved the problem of drug resistance, and improved the short duration of inhibition of Curcuma extract. This study aimed to combine the antimicrobial effect of Curcuma extract with the function of L. rhamnosus in regulating the gastrointestinal flora and to investigate the synergistic inhibitory effect of the two on H. pylori . The findings provide a theoretical basis for subsequent cellular, animal, or clinical experiments.

This study explores the biochemical indices across three distinct experimental groups: the Curcuma longa L. fermentation Group (CF), the Curcuma longa L. and black tea fermentation Group (CT), and the black tea fermentation Group (KF). After fermentation, the detection of basic indexes revealed that the CT group had the highest total sugar content and gluconic acid levels, at 4.48 mg/mL and 3.20 mg/mL, respectively. Additionally, the KF group exhibited the highest levels of sucrose, polyphenols, and total flavonoids, measuring 28.67 mg/mL, 236.76 μg/mL, and 0.20 mg/mL, respectively. Ethanol content was no more than 0.16% vol in all groups. A qualitative analysis of the aforementioned biochemical parameters revealed the presence of 8 carbohydrates and analogs, 8 sesquiterpenoids, 6 flavonoids, 6 organic acids, 5 alkaloids, and 21 glycosides. We further identified two unique pathways pertinent to fermentation: glucuronic acid interconversion pathway and the cyanogenic amino acid metabolism pathway. Key metabolites influencing kombucha include α-ketoglutaric acid, D-xylitol, ribitol, dhurrin, and lotaustralin. Underscoring the significance of Curcuma longa L. and black tea fermentation. These results elucidate the differential expression of metabolites and their regulatory mechanisms.

Objective Retinal neovascularization is a leading cause of irreversible blindness worldwide. Chinese herbal medicines have unique therapeutic properties, such as antioxidant, anti-inflammatory, and neuroprotective effects. This study aims to investigate the therapeutic effects and underlying mechanisms of the Chinese medicinal herb Curcuma longa -derived extracellular vesicle-like particles (CL-EVLPs) in a mouse model of retinal neovascularization. Methods CL-EVLPs were extracted from fresh Curcuma longa rhizomes through differential centrifugation and sucrose density gradient centrifugation and characterized by transmission electron microscopy (TEM), and nanoparticle tracking analysis (NTA), and western blot analysis of EVLP-associated protein markers. CL-EVLPs were administered to oxygen-induced retinopathy (OIR) model mice via intravitreal injection. The therapeutic effects were evaluated using retinal flat mounts, fundus fluorescein angiography (FFA), and reactive oxygen species (ROS) staining. The underlying molecular mechanisms were explored through metabolomics, transcriptomics, and network pharmacology analyses, and the expression levels of relevant factors were quantified using real-time polymerase chain reaction (PCR) or western blotting. In vitro experiments were conducted using human retinal microvascular endothelial cells (hRMECs) and BV2 microglia. Results CL-EVLPs exhibited typical EV-like characteristics, including a saucer‑ or cup‑shaped morphology, a mean diameter of 140.7 nm, and enrichment of the EV‑related protein ARF1. CL-EVLPs effectively inhibited neovascularization in the OIR model, as evidenced by reduced pathological neovascular tufts and avascular zones and decreased vascular permeability and tortuosity. Furthermore, CL-EVLPs significantly decreased ROS production in the retinas of OIR mice. Mechanistically, CL-EVLPs inhibited HIF-1α/VEGF/ERK signaling and activated the NRF2/HO-1 antioxidant pathway. In vitro experiments confirmed that CL-EVLPs inhibited endothelial cell viability and alleviated oxidative stress in microglia. Integrated network pharmacology and metabolomics analyses proved that the cargo molecules of CL-EVLPs, such as curcumin and its derivatives, auraptene, and triptophenolide, exhibit regulatory antioxidant and anti-inflammatory effects and modulate multiple signaling pathways associated with retinal neovascular diseases. Conclusion CL-EVLPs effectively suppress retinal neovascularization by inhibiting the HIF-1α/VEGFA/ERK axis and activating the NRF2/HO-1 pathway. This study suggests that CL-EVLPs, as a multitarget therapeutic strategy, hold promising clinical translational potential for the treatment of retinal neovascular diseases. Turmeric has high commercial demand internationally, owing to its high nutrient and medical value. In this research, turmeric samples were subjected to several drying methods viz. infrared freeze drying (IRFD), freeze drying (FD), and pulse-spouted microwave-assisted freeze-drying (PSMFD). Over the range of operating conditions employed in this study, IRFD of infrared radiation was showed to shorten the drying time by 21.27% relative to FD, while the retention valves of ascorbic acid, curcumin, total phenols, and total flavonoids remains at 75.61%, 94.06%, 69.00%, and 80.09%, respectively over the range of experimental parameters. IRFD significantly (P < 0.05) reduced 25.19% of the energy consumption in comparison with FD. Furthermore, the interior of IRFD samples exhibited a honeycomb porous structure, and increased hardness, lower shrinkage, apparent density compared to PSMFD. Additionally, IRFD also performed better than PSMFD and FD under vacuum freezing conditions.

Curcuminoids originated from the plant Curcuma longa L. show high medical values. Herein, ionic liquids (ILs) were synthesized using fatty acids and alcohol amines to extract curcuminoids. Through single-factor experiments and response surface methodology (RSM) optimization, the extraction amount of curcuminoids by [TEA][FA-C 7 ] aqueous solution reached 34.89 mg/g at a liquid-solid ratio of 16.36 mg/g and an extraction temperature of 36 °C, surpassing that of methanol. The extraction capacity of [TEA][FA-C 7 ] to curcuminoids was 299.33 mg/g. Furthermore, the separation of extracts and recovery of fatty acids were studied. Quantum chemical calculations and molecular dynamics simulations were performed to investigate the extraction mechanism profoundly. This study not only proposed a novel green solvent for the extraction of curcuminoids from the plant Curcuma longa L. but also provided the possibility of applying fatty acid-based ILs to extract natural medicine efficiently.

Integrated tandem ultrafiltration membrane technology (UMT) was used to separate Curcuma longa polysaccharides (CLPs) into four grades: CLP-100, CLP-50, CLP-10, and CLP-5. CLP-10 was a purified polysaccharide consisting of d -glucose, L -arabinose, L -rhamnose, and D -galactose. Its backbone comprised →2)- α - L -Rha p -(1 → 2,4)- α - L -Rha p -(1 → 4)- α - D -Glc p -(1→. CLP-100 and CLP-50 showed significant protective activity against H 2 O 2 -induced oxidative stress in human umbilical vein endothelial cells (HUVECs) through the VEGFA/AKT/SOD2 pathway. The structure-activity relationship of four CLPs was further analyzed. Molecular weight ( Mw ) distribution, monosaccharide composition, and morphology were the main factors affecting the activity. In addition, a preliminary investigation into the separation mechanisms by which UMT separated the polysaccharides was conducted, which revealed potential influences of intra-molecular aggregation and inter-molecular repulsion between the polysaccharides and membrane materials. These findings provide a profound elucidation of the complex relationship between UMT and factors such as Mw , conformation, particle size, charge number, and apparent viscosity during the separation process.

The combination of microwave irradiation, ultrasonication treatment, and enzyme-assisted extraction approach was applied to study the extraction efficiency and antioxidative activity of curcumin from turmeric ( Curcuma longa L.) using Response Surface Optimization (RSM). The effects of the power and treatment time of microwave irradiation and ultrasonication treatment, the types of enzymes, the amounts, and interaction time of pectinase on the extraction efficiency were studied. RSM was used to select the optimum extraction conditions by implementing Box–Behnken design. The extraction ratio and antioxidant activity of extracted species are 2.89% and 83.95% under the optimal conditions, respectively, which are close to the predicted values, and are much higher than the single extraction approach. Our results show that a versatile approach for biological compounds extraction from agricultural and natural products was successfully developed, and the bioactivity of extracted species could be kept well by using our proposed combination extraction methods.

Diosgenin is an important intermediate of steroid hormone drugs, green production of diosgenin with high yield is a key research issue. In this study, a convenient, efficient, and eco-friendly approach was developed to produce diosgenin from saponins in D. zingiberensis C. H. Wright (Curcuma longa L) using a magnetic solid acid. Fe 3 O 4 @SiO 2 -Pr-S-SO 3 H is a heterogeneous catalyst and was obtained by loading 3-(mercaptopropyl) trimethoxysilane on the surface of Fe 3 O 4 @SiO 2 , followed by sulfonation with chlorosulfonic acid. The magnetic solid acid catalyst was fully characterized by Fourier Transform-Infrared (FT-IR) spectra, thermo gravimetric analysis (TGA), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), and X-ray photoelectron spectroscopy (XPS), scanning electron microscope(SEM), transmission electron microscope (TEM), NH 3 -TPD, elemental mapping analysis. The effects of single factors, such as the solid acid mass, alcoholysis temperature, extraction time, and solvent volume on the diosgenin yield were analyzed. The extraction parameters for diosgenin were optimized by response surface methodology (RSM). The optimized values for the solid acid mass, solvent volume, extraction time, and temperature were 0.22 g, 7.8 mL, 7.4 h, and 100 °C, respectively. Under the optimized conditions, the maximum yield of diosgenin was 19.15 %. The solid acid reported in this work exhibited higher catalytic activity at a smaller dosage than traditional sulfuric acid and other solid acids. The alcoholysis products were identified by high-performance liquid chromatography-mass spectrometry (HPLC-MS). Alcoholysis catalysed by an as-prepared magnetic solid acid is clearly superior in terms of catalytic activity, dosage, and reusability. It is a promising strategy for the green production of diosgenin.
