Fibronectin-Targeting Dual-Modal MR/NIRF Imaging Contrast Agents for Diagnosis of Gastric Cancer and Peritoneal Metastasis

Ningxin Yang,Yuelin Huang,Xiaoyu Wang,Dengbin Wang,Defan Yao,Gang Ren

The prevalence and fatality rates of gastric cancer (GC) remain elevated, with advanced stages presenting a grim prognosis. Noninvasive diagnosis of GC cancer often proves challenging until the disease has progressed to an advanced stage or metastasized. Initially, the level of fibronectin (FN) in cancer-associated fibroblasts (CAFs) of GC was at least 3.7 times higher than that in normal fibroblasts. Herein, two FN-targeting magnetic resonance/near-infrared fluorescence (MR/NIRF) imaging contrast agents were developed to detect GC and peritoneal metastasis noninvasively. The probes CREKA-Cy7-(Gd-DOTA) and CREKA-Cy7-(Gd-DOTA)3 demonstrated significant FN-targeting capability (with dissociation constants of 1.0 and 2.1 mM) and effective MR imaging performance (with proton relaxivity values of 9.66 and 27.44 mM–1 s–1 at 9.4 T, 37 °C). In vivo imaging revealed a high signal-to-noise ratio and successful visualization of GC metastasis using NIRF imaging as well as successful tumor detection in MR imaging. Therefore, this study highlights the potential of FN-targeting probes for GC diagnosis and aids in the advancement of new diagnostic strategies for the clinical detection of GC.

Photodynamic and Photothermal Co-Induced Efficient Anti-Tumor Immunotherapy

Shanshan Jin,Yongkang Chen,Fahui Li,Ping Yan,Guanhong Guo,Guangzhao Xu,Weiguo Song,Wenda Zhong

Currently, immunotherapy based on photothermal and the application of photodynamic therapy in anti-tumor treatment is showing great potential. Its uniqueness lies in the critical role of small molecule immunomodulators in promoting effective immune responses against tumors, and the use of laser-activated biophysical mechanisms to precisely trigger the swift demise of cancer cells, avoiding damage to surrounding normal tissues. However, the use of photodynamic therapy (PDT) alone is hampered by the tumors’ hypoxic environment, resulting in poor antitumor effects, while photothermal therapy (PTT) alone cannot arouse enough antigen presentation. It is of great significance to design photosensitizers (PSs) that possess both PDT and PTT effects. Herein, a series of PSs with both PDT and PTT efficacy are reported, ultimately selecting Cy7-Naph as the star molecule due to its best overall phototherapeutic effect. Upon reactive oxygen species (ROS) production and thermogenesis in tumor cells, Cy7-Naph induced significant apoptosis and eventually boosted the release of damage-associated molecular patterns (DAMPs) under near-infrared (NIR) light irradiation. By combining Cy7-Naph with the Toll-like receptor agonist Resiquimod (R848), a synergistic treatment for bilateral tumor-bearing mice is achieved. This combination promotes dendritic cell (DC) maturation and increases the infiltration of cytotoxic T lymphocytes (CTLs), leading to significant inhibition of both primary and distant tumors.

Biometabolizable Conjugated Polymers for Photothermal Therapy under the Guidance of Dual-Modal Imaging of Prostate Cancer

Qilai Long,Wei Xi,Yanyun Chen,Lei Xu,Zhibing Xu,Yanjun Zhu,Jianming Guo

As photothermal drugs can convert external light energy into heat energy to kill tumor cells, they have gained avid attention in the field of tumor therapy in recent years. In this study, we designed and prepared a conjugated polymer photothermal agent for prostate cancer by modifying PEG-CY7 on the surface of the conjugated polymer. This study demonstrated that upon excitation by an 808 nm laser, conjugated polymers exhibit good photothermal conversion ability and high thermal stability. Moreover, this conjugated polymer exhibited excellent biocompatibility and high cellular uptake. In in vivo experiments, through dual-modal imaging, we confirmed that the conjugated polymer can effectively accumulate in the tumor tissue and also be eliminated by the normal metabolism of the organism, avoiding toxic side effects. In addition, the conjugated polymer exhibited a good antitumor photothermal and laser-mediated treatment effect in the tumor-bearing mouse model. Thus, our synthesis of conjugated polymers is a promising treatment strategy for prostate cancer.

pH-Responsive nanomicelles for breast cancer near-infrared fluorescence imaging and chemo/photothermal therapy

Jingjing Xu,Xiaojing Yang,Xianjiao Tang,Mei-Xia Zhao

Chemotherapy, despite being the most common and effective cancer treatment, can cause several toxic adverse effects. Multimodal combined antitumor therapy, which accounts for the deficiency of monotherapy, has become an effective strategy for clinical cancer treatment. In this study, we describe the development of pH-sensitive Cy7-PEG-CA-DOX micelles with antitumor activity for controlled drug release, tumor imaging, and their synergy with photothermal therapy (PTT) and chemotherapy. The cyanine dye Cy7 was used as a near infrared (NIR) fluorophore and photosensitizer, and polyethylene glycol (PEG) served as a biocompatible carrier. The anticancer drug doxorubicin (DOX) was loaded onto PEG via aconitic anhydride bonds to increase pH sensitivity. Cy7-PEG-CA-DOX molecules initiated self-assembly to form nanomicelles (∼64.21 ± 0.49 nm) in aqueous solution. In vitro the DOX release rate reached 78% after 48 h at pH 5.5 due to the amide bond between DOX and aconitic anhydride becoming readily broken at low pH. Moreover, intracellular uptake images showed that Cy7-PEG-CA-DOX micelles effectively released DOX in 4T1 tumor cells. In vivo fluorescence imaging revealed that Cy7-PEG-CA-DOX micelles aggregated at tumor sites with optimal tumor imaging properties. More importantly, Cy7-PEG-CA-DOX micelles facilitated the favorable anti-breast cancer properties elicited by PTT and chemotherapy in vivo. This study provides a novel platform for constructing responsive multifunctional anticancer nanoparticles, which have significant potential applicability in controlled drug release and synergistic cancer therapy.

Photothermally driven degradable nanomissile for single-wavelength phototherapy and hypoxia-activated chemotherapy

Feng Ouyang,Li Zhao,Qi Shuai

Viscous tumour tissue seriously affects the effective delivery of nanodrugs and reduces their anti-tumour effect. In order to solve this problem, a nanomotor (Cy7@ZIF-8@TPZ) for penetrating tumour tissue was synthesized. A new amphiphilic molecule Cy7 was designed and synthesized to wrap ZIF-8 loaded with TPZ to obtain the final nanomotor. The synthesized Cy7 can not only realize photodynamic therapy (PDT) and photothermal therapy (PTT) simultaneously under single-wavelength illumination, but also promote the movement of nanoparticles through thermal drive. Through the targeting ability of Cy7 and the enhanced permeability and retention (EPR) effect, the nanomotor can conduct directional deep puncture under illumination. The oxygen continuously consumed by PDT further activates hypoxia-activated chemotherapy, and the combined PTT/PDT/hypoxia-activated CT therapy greatly enhances the anti-tumour efficacy of Cy7@ZIF-8@TPZ. In addition, light-degradable Cy7, acid-degraded ZIF-8 and hypoxia-activated TPZ greatly reduce the side effects of nanomotors.
