Seasonal Variation in Phenolic Compounds and Antioxidant Activity in Leaves of Cyclocarya paliurus (Batal.) Iljinskaja

Yanni Cao,Shengzuo Fang,Xiangxiang Fu,Xulan Shang,Wanxia Yang

Cyclocarya paliurus(Batal.) Iljinskaja is a plant with nutraceutical importance since its leaves have been used historically as folk medicines for hundreds of years. The content of 10 phenolic compounds was determined throughout the growing season by high-performance liquid chromatography (HPLC) with UV detector, while the antioxidant activities ofC. paliurusleaf extracts were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation (ABTS), and ferric reducing antioxidant power (FRAP) methods. Seasonal variations in phenolic concentration and antioxidant activity as well as linkage between the phenolic composition and antioxidant activity were assessed. A significant seasonal variation of phenolic compounds was observed in the leaves and the highest content appeared in May, July, and November. Seventy percent ethanol extract ofC. paliurusleaves possessed a good radical scavenging potency. Meanwhile, a significant correlation between antioxidant activities and contents of phenolics was detected. Results of the relationship between molecular structures and their antioxidant activities showed that both the number and configuration of H-donating hydroxyl groups are the main structural features influencing the antioxidant capacity of phenolics, while glycosylation may reduce the antioxidant capacity. The information provided by this study not only revealed the accumulative dynamics of phenolic compounds, but also established a basis for determining the optimal time for harvesting to improve the content of beneficial compounds in the leaves ofC. paliurusin the future. Keywords:Cyclocarya paliurus;seasonal dynamic;phenolic acids;flavonoids;antioxidant activity;structure-activity relationship

Environmentally-Friendly Extraction of Flavonoids from Cyclocarya paliurus (Batal.) Iljinskaja Leaves with Deep Eutectic Solvents and Evaluation of Their Antioxidant Activities

Xianchao Shang,Jia-Neng Tan,Yongmei Du,Xinmin Liu,Zhongfeng Zhang

Deep eutectic solvents (DESs) are commonly employed as environmentally-friendly solvents in numerous chemical applications owing to their unique physicochemical properties. In this study, a novel and environmentally-friendly extraction method based on ultrasound assisted-deep eutectic solvent extraction (UAE-DES) was investigated for the extraction of flavonoids fromCyclocarya paliurus(Batal.) Iljinskaja (C. paliurus) leaves, and the antioxidant activities of these flavonoids were evaluated. Nine different DES systems based on either two or three components were tested, and the choline chloride/1,4–butanediol system (1:5 molar ratio) was selected as the optimal system for maximizing the flavonoid extraction yields. Other extraction conditions required to achieve the maximum flavonoid extraction yields from the leaves ofC. paliuruswere as follows: DES water content (v/v), 30%; extraction time, 30 min; temperature, 60 °C; and solid-liquid ratio, 20 mg/mL. Liquid chromatography-mass spectrometry allowed the detection of five flavonoids in the extract, namely kaempferol-7-O-α-l-rhamnoside, kaempferol, quercetin, quercetin-3-O-β-d-glucuronide, and kaempferol-3-O-β-d-glucuronide. In vitro antioxidant tests revealed that the flavonoid-containing extract exhibited strong DPPH and ABTS radical-scavenging abilities. Results indicate that UAE-DES is a suitable approach for the selective extraction of flavonoids fromC. paliurusleaves, and DESs can be employed as sustainable extraction media for other bioactive compounds. Keywords:Cyclocarya paliurus;deep eutectic solvent;extraction;flavonoid;antioxidant activity

Untargeted metabolomics analysis based on HS-SPME-GC-MS and UPLC-Q-TOF/MS reveals the contribution of stem to the flavor of Cyclocarya paliurus herbal extract

Lu Liang,Yuchen Liu,Yuxin Liu,Siyi Gan,Xuejin Mao,Yuanxing Wang

The herbal tea made from the leaves and stems of Cyclocarya paliurus is thought to be more flavorful than those made from the leaves only. However, evidence supporting this view is not available yet. 53 VOCs and 54 non-VOCs were identified via HS-SPME-GC-MS and UPLC-Q-TOF/MS, respectively, which were found to vary dramatically in three groups of samples (combination of stems and leaves, separated leaves, and separated stems). 11 VOCs and 18 non-VOCs potential markers are responsible for the change of metabolic effects through the biosynthesis of flavonoids, the synthesis of terpenoids, and the oxidation of fatty acids and carotenoids. In response, the stems were effective to enhance fatty odors, the initial astringency and bitterness and the aftertaste of sweetness of the leaves. Stems are conducive to increasing total phenolic content, total flavonoid content and antioxidant capacity of Cyclocarya paliurus herbal extract because of the increase in polyphenols. In addition, the stems have the potential to be used as raw materials for nerolidol and polyphenols extraction. This study has been the first to determine the contribution of stem to the flavor of Cyclocarya paliurus herbal tea and these results provide the theoretical basis for making Cyclocarya paliurus herbal tea with stems.

Mengjia Xu,Huixia Feng,Mingquan Guo

Cyclocarya paliurus has been widely applied for therapeutic feeding as a traditional Chinese medicine homologous food with high nutritional and medical value. Nevertheless, the specific bioactive compounds liable for hypoglycemic and hypolipidemic properties and underlying mechanisms remain unexplored. In the present study, the in vitro hypoglycemic and hypolipidemic effects of C. paliurus were assessed by α-glucosidase and pancreatic lipase inhibition assays, and the results indicated that 70% ethanol extract exhibited remarkable α-glucosidase and pancreatic lipase inhibitory activity with the IC 50 value of 3.14 μg/mL and 0.98 μg/mL, respectively. A total of 18 and 15 potential ligand candidates were screened, and the chemical structures were characterized. Furthermore, the docking result showed that chlorogenic acid, quercetin, kaempferol 3- O -rhamnoside, and pterocaryoside A contributed to the underlying hypoglycemic and hypolipidemic effects of C. paliurus , further strengthened the bindings of the enzyme-ligand complex and suppressed the enzyme's activity with the binding energies ranged from −4.00 to −6.31 kcal/mol. The interrelations between the specific bioactive constituents, α-glucosidase or lipase, and hypoglycemic or hypolipidemic activities were also carified. The present study reveals substantial prospects of C. paliurus to be developed as a natural dietotherapy, disease-prevention, and healthcare for diabetes and obesity in the near future.

Protective effects of Cyclocarya paliurus on hyperuricemia and urate-induced inflammation

Li-Hua Zhu,Ying-Yin Xu,Li-ping Zhu,Xian Zheng,Cui-Hua Jiang,Jian-Jing Liu,Jian Zhang,Zhi-Qi Yin

As a new and safe food resource, Cyclocarya paliurus (CP), rich in flavonoids, exhibits hypouricemic, anti-inflammatory and antioxidative properties. However, whether CP aqueous extract (CPA) still possesses the ameliorative effects on hyperuricemia (HUA) and urate-induced renal inflammation remains vacant. In HUA mice, CPA could significantly reduce serum uric acid (UA), alleviate kidney damage and liver steatosis, inhibit xanthine oxidase (XOD) activity, ameliorate renal urate transporters and renal inflammation by inhibiting NLRP3 inflammasome and interleukin-1β (IL-1β). Moreover, the flavonoids quercetin-3- O-β- D-galactopyranoside, quercetin-3- O-β- D-glucopyranoside, and quercetin-3- O-α -L-rhamnopyranoside from CPA could significantly reduce IL-1β and interleukin-6 (IL-6) in MSU-challenged THP-1 cells. Noticeably, quercetin-3- O-β- D-galactopyranoside could inhibit the intracellular XOD activity, reduce intracellular ROS and the expression of pro-IL-1β, IL-1β, and NLRP3 inflammasome. Collectively, CPA shows appreciable therapeutic effects on HUA and evoked renal inflammation. Quercetin-3- O-β -D-galactopyranoside, quercetin-3- O-β -D-glucopyranoside, and quercetin-3- O-α -L-rhamnopyranoside might be the major active compounds of CPA, which obtain the favorable effects through blocking the activation of ROS/NLRP3/IL-1β pathway.

Extraction and properties of Cyclocarya paliurus leaf polysaccharide and its phosphorylated derivatives

Haibin Zhu,Liuming Xie,Weidong Zhang,Qiang Yu,Yi Chen,Jianhua Xie

Chemical modification provides an important idea for advancing and harnessing plant polysaccharides. It is well known that Cyclocarya paliurus (Batalin) Iljinskaja (C. paliurus) , is an important industrial crop resource, and polysaccharide stands as one of the compounds displaying a wide range of bioactivities within the leaves of C. paliurus . The study found that C. paliurus polysaccharides (CPP) were subjected to phosphate modification to obtain two phosphorylated polysaccharides (P-CPPL and P-CPPH) with degrees of substitution (DS) of 0.10 and 0.18, correspondingly. The findings indicated that the phosphorylation changed the chemical composition, relative molecular weight, molar ratio of monosaccharide composition and microstructure of CPP, thus changing the thermal stability and bioactivities of CPP. In vitro bioassay results demonstrated that CPP and its phosphorylated derivatives showed antibacterial activity at high concentrations. In addition, phosphorylation changes the water solubility and spatial configuration of CPP, so that P-CPPL and P-CPPH can bind or interact with enzymes more effectively, thus improving the α-glucosidase inhibitory activity in vitro. In vitro cell experiment results demonstrate that CPP and its phosphorylated derivatives can protect IEC-6 cells from oxidative damage by restoring mitochondrial membrane potential levels and modulating the expression and activation of Caspase-3. In summary, phosphorylation provided an essential way of changing the chemical properties of polysaccharides and enhancing their functionalities.

Extraction, isolation and structural characterization of a novel polysaccharide from Cyclocarya paliurus

Xiaojiang Wang,Xiaoliang Zhao,Youjing Lv,Minghua Hu,Luodi Fan,Qinying Li,Chao Cai,Guoyun Li,Guangli Yu

A water soluble polysaccharide CP-III was extracted and purified from Cyclocarya paliurus . CP-III is identified as a novel pectin-like polysaccharide with molecular weight ( Mw ) of 72.7 kDa. The structural features of CP-III were characterized by methylation and nuclear magnetic resonance (NMR) spectroscopy. Its depolymerized fragments were analyzed by hydrophilic interaction chromatography-Fourier transform mass spectrometry (HILIC-FTMS). The main chain of CP-III is composed of →4)GalA p (α1 → and →2)Rha p (α1 → 4)GalA p (α1→, repeatedly. The residue of →4)Gal p (β1 → and →5)Ara f (α1 → alternately exists on the O -4 of partial →2)Rha p (α1 → residues as side chains. On the O -3 of sectional →5)Ara f (α1 → residues is a secondary branch assembled by →3)Ara f (α1→. Moreover, on the non-reducing terminus of →4)Gal p (β1 → occasionally have an →5)Ara f (α1 → chain. Surprisingly, a sub-branch constructed by →6)Hex p (β1 → with a galacturonate or methyl galacturonate exists on the O -3 of certain →4)Gal p (β1 → residues in the non-reducing terminus. In addition, a terminal Xyl is located on the O -3 of fractional →4)GalA p (β1 → residue. The highly branched polysaccharide CP-III with high water solubility can be used as food supplement and medicinal carrier in the future.

Colchicine-Induced Polyploidization Influences the Morphological, Physiological, and Biochemical Characteristics of Cyclocarya paliurus

Guoliang Bian,Yan Yi,Ziqi Song,Yanmeng Huang,Qianxing Mao,Jian Qin,Xulan Shang

Cyclocarya paliurus(Batal.) Iljinskaja is a rare and multifunctional tree species endemic to China. This study aimed to establish a feasible method for polyploid induction inC. paliuruswith colchicine treatment, and the obtained polyploid plants were identified and characterized. In this study, natural diploid and tetraploidC. paliurusseedlings were treated with different concentrations and durations of colchicine to induce polyploidization. The results indicated that a colchicine concentration of 0.4% for 4 d was the most suitable induction protocol, respectively. Compared with diploid and tetraploid control plants, the induced polyploid plants were shorter and thinner. The number of compound leaves in induced polyploids were fewer, and the compound leaf areas became smaller. The stomatal density of induced polyploids decreased, but the stomata became larger and wider, with an increased number of chloroplasts in the guard cells. The induced polyploids exhibited higher levels of carotenoid, and the contents of soluble sugar, soluble starch, and soluble protein were higher than those of controls. The polyploid plants exhibited an increase in the contents of growth-inhibiting hormones (JA) and a decrease in those of growth-promoting hormones (IAA, GA3, BR). In summary, the induced polyploids with a dwarfing effect would provide superior germplasm resources for leaf-harvesting plantation. AsC. paliurusis endemic to China, our results have a rather local effect. Overall, the established polyploid induction method ofC. paliuruswill provide technical support for creating superiorC. paliurusgermplasm resources and subsequent plant breeding research.

Cholesterol-Lowering Effect of Polysaccharides from Cyclocarya paliurus In Vitro and in Hypercholesterolemia Mice

Yang Zhang,Lei Zeng,Kehui Ouyang,Wenjun Wang

In this study, a new component of Cyclocarya paliurus polysaccharides (CPP20) was precipitated by the gradient ethanol method, and the protective effect of CPP20 on hypercholesterolemia mice was investigated. In vitro, CPP20 had the ability to bind bile salts and inhibit cholesterol micelle solubility, and it could effectively clear free radicals (DPPH•, •OH, and ABTS+). In vivo, CPP20 effectively alleviated hypercholesterolemia and liver damage in mice. After CPP20 intervention, the activity of antioxidant enzymes (SOD, CAT, and GSH-Px) and the level of HDL-C in liver and serum were increased, and the activity of aminotransferase (ALT and AST) and the level of MDA, TC, TG, LDL-C, and TBA were decreased. Molecular experiments showed that CPP20 reduced cholesterol by regulating the mRNA expression of antioxidation-related genes (SOD, GSH-Px, and CAT) and genes related to the cholesterol metabolism (CYP7A1, CYP27A1, SREBP-2, HMGCR, and FXR) in liver. In addition, CPP20 alleviated intestinal microbiota disturbances in mice with hypercholesterolemia and increased levels of SCFAs. Therefore, CPP20 alleviates hypercholesterolemia by alleviating oxidative damage, maintaining cholesterol homeostasis, and regulating gut microbiota.
