Preliminary Study on the Inhibitory Effect and Mechanism of Oleic Acid in Cylindrospermopsis raciborskii

Xiaojia Huang,Yuanyuan Hu,Xiaowei Cheng,Weibin Pan

Cylindrospermopsis raciborskiiis a toxin-producing cyanobacterium that is easy to overlook. It has strong environmental adaptability and is currently spreading around the world and gradually dominating to form a persistent bloom, causing ecological and environmental risks and drinking water safety issues. In this study, we systematically investigated the inhibitory effects of oleic acid onC. raciborskiiand elucidated the underlying mechanisms through morphological observation, physiological assays, and bioinformatics analysis. Our results demonstrated that oleic acid strongly inhibits the growth ofC. raciborskii, with a 72 h half-maximal effective concentration (EC50) of 0.903 mg·L−1. At 1.6 mg·L−1, oleic acid achieved an inhibition rate of 99.5% within 48 h, indicating rapid suppression of cyanobacterial growth. Physiological analyses revealed that oleic acid severely impaired photosynthetic activity, as evidenced by significant reductions in key parameters (rETRmax, α, Fv/Fm, and Fv/Fo) and altered photosynthetic pigment composition, suggesting structural and functional damage to the photosynthetic apparatus. Morphological observations further showed that oleic acid disrupted filament integrity, inducing cell shrinkage, cytoplasmic vacuolation, cell wall detachment, membrane rupture, and eventual cellular disintegration. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis indicated that oleic acid interferes with multiple metabolic processes, including nutrient and cofactor synthesis, membrane transport, and signal transduction, ultimately triggering algal cell death. This study highlights oleic acid as a promising eco-friendly agent for mitigatingC. raciborskiiblooms, offering potential applications in ecological prevention and emergency bloom control.

Influence of linoleic acid on growth, oxidative stress and photosynthesis of the cyanobacterium Cylindrospermopsis raciborskii

S Xu,S-Q Yang,Y-J Yang,J-Z Xu,J-Q Shi,Z-X Wu

In this study, the effects of linoleic acid on the growth, photosynthetic activities and oxidative system of the invasive cyanobacterium Cylindrospermopsis raciborskii were investigated. The results indicated that linoleic acid decreased the growth rate and chlorophyll a concentration in C. raciborskii after 48 h incubation. The maximum photochemical efficiency (Fv/Fm) of photosystem II (PSII) and maximum relative electron transport rate (ETRmax) decreased significantly at concentrations higher than 1 mg L−1. However, non-photochemical quenching (NPQ) significantly increased when cultured at concentrations of 0.5, 1, 2 and 4 mg L−1. PSII electron transfer rate and chlorophyll a fluorescence transients appeared to be affected after the cells were exposed to higher concentrations of linoleic acid; the density of the reaction centres (RCs), quantum energy flux ratios and the performance index on absorption basis decreased, but absorption flux per RC and dissipated energy flux per RC increased. Malondialdehyde content, superoxide dismutase and catalase activities were enhanced when exposed to 2 and 4 mg L−1 of linoleic acid. This suggests that the blockage of electron transport on the donor and acceptor sides of PSII is the mechanism responsible for the toxicity of linoleic acid in PSII of C. raciborskii.

Use of chlorophyll a fluorescence to elucidate the toxicity target of N-phenyl-2-naphthylamine on photosynthetic system of Cylindrospermopsis raciborskii (Cyanobacteria)

Rui Liu,Xiaofei Ran,Fang Bai,Jinzhu Xu,Songqi Yang,Junqiong Shi,Zhongxing Wu

In order to investigate the toxicity targets of the secondary metabolite from aquatic plants, N-phenyl-2-naphthylamine, on Cylindrospermopsis raciborskii photosynthetic system II (PSII), we determined chlorophyll a (Chl a), specific growth rate (μ−d), photosynthetic rapid light curves and polyphasic Chl a fluorescence transients following exposure to N-phenyl-2-naphthylamine. The results showed significant decreases in Chl a following 48 h at high concentration (0.50 mg l−1) exposure to N-phenyl-2-naphthylamine. The PSII electron transfer rate and Chl a fluorescence transients appeared to be affected after the cells were exposed to concentrations of 0.50, 1.00 and 2.00 mg l−1. Exposure to higher concentration of N-phenyl-2-naphthylamine (> 0.50 mg l−1) mainly depressed the density of reaction centers (RC/CS0) and quantum energy flux ratios (ϕP0 and ϕE0) but increased the absorption flux per RC (ABS/RC), dissipated energy flux per RC (DI0/RC) and the average redox state of QA−/QA in the time span from 0 to tFmax (Sm/tFmax). However, exposure to low concentration (≤ 0.5 mg l−1) mainly inhibited the trapping energy fluxes per reaction center (TR0/RC), electron transport energy fluxes per reaction center (ET0/RC) and energy flux ratio (ψ0). This suggested that the donor and the acceptor sides of PSII were targets of N-phenyl-2-naphthylamine in C. raciborskii. Additionally, our results indicated that the toxicity target at low concentrations of N-phenyl-2-naphthylamine was at each step of electron transport between QA− and the Cyt b6/f complex; whereas, the reduction of QA was the potential target at high concentrations. We suggest that N-phenyl-2-naphthylamine may be a potential allelochemical to control harmful cyanobacterial blooms.

The Effect of Bisphenol A on Growth, Morphology, Lipid Peroxidation, Antioxidant Enzyme Activity, and PS II in Cylindrospermopsis raciborskii and Scenedesmus quadricauda

Xiang Rong,Shi Junqiong,Yu Yi,Zhang Hongbo,Dong Congcong,Yang Yanjun,Wu Zhongxing

To investigate the effect of bisphenol A (BPA) on Cylindrospermopsis raciborskii (Cyanobacteria) and Scenedesmus quadricauda (Chlorophyta), we grew the two species at BPA concentrations of 0, 0.1, 1, 2, 5, 10, and 20 mg/L and examined their growth, lipid peroxidation, antioxidant enzyme activity, and chlorophyll a fluorescence. The 96-h EC 50 values (effective concentration causing 50% growth inhibition) for BPA in C. raciborskii and S. quadricauda were 9.663 ± 0.047, and 13.233 ± 0.069 mg/L, respectively. A significant reduction in chlorophyll a concentration was found in C. raciborskii and S. quadricauda when BPA concentrations were greater than 1 and 2 mg/L, respectively. Furthermore, F v / F m , Δ F / F m ′, and qP decreased significantly at 10 mg/L BPA in C. raciborskii but started to decrease at 10 mg/L in S. quadricauda. The changes in chlorophyll fluorescence parameters ( α , rETR max ) that were obtained from the rapid light response curves of both algae species showed similar responses to F v / F m , Δ F / F m ′, and qP under BPA-induced stress. Values for all of the chlorophyll fluorescence parameters in S. quadricauda were higher than in C. raciborskii ; however, the nonphotochemical quenching measured in C. raciborskii was considerably higher than it was in S. quadricauda . In addition, lipid peroxidation (determined as MDA content) and antioxidant enzyme activities (SOD and CAT) increased in both species as the BPA concentration increased. These results suggest that C. raciborskii is more sensitive to the effects of BPA than S. quadricauda and that photosystem II might be a target for the activity of BPA in vivo.
