Molecular identification of FNDC5 and effect of irisin on the glucose metabolism in common carp (Cyprinus carpio L.)

Liping Yang,Shaoyang Zhi,Guokun Yang,Chaobin Qin,Wenli Zhao,Mingming Niu,Wenlei Zhang,Wenyu Tang,Xiao Yan,Yuru Zhang,Xiaolin Meng,Ronghua Lu,Guoxing Nie

Irisin, encoded by fibronectin type III domain-containing protein 5 (FNDC5) gene, plays a role in energy expenditure and insulin sensitivity in mice. In fish, the function of irisin related to glucose metabolism is less reported. It may increase glucose utilization in fish. The aim of the present study was to characterize the regulatory role of irisin in glucose metabolism in common carp ( Cyprinus carpio L.). In this study, FNDC5a and FNDC5b were isolated from common carp. The cDNA of FNDC5a and FNDC5b were 722 bp and 714 bp, encoding 221 and 207 amino acids, respectively. FNDC5a was abundantly expressed in the brain and gonad. FNDC5b was mainly expressed in brain. Different expression pattern of FNDC5a and FNDC5b under fasting/refeeding and OGTT experiment were identified. The recombinant common carp irisinA and irisinB were prepared by prokaryotic expression system. Glucose concentration was decreased in treatment with irisinA or irisinB in the in vitro and in vivo experiments. The mRNA expression levels of gluconeogenesis-related genes were significantly down-regulated, while the mRNA expression of glycolysis-related genes were significantly up-regulated after treatment with recombinant irisinA or irisinB in liver in vivo and in primary hepatocytes in vitro. Our research shows that irisin inhibits hepatic gluconeogenesis and promotes hepatic glycolysis. Taken together, this study for the first time revealed the two subtypes of FNDC5 and explored the function and mechanisms of irisinA and irisinB in fish glucose homeostasis.

Environment-relevant concentrations of cadmium induces necroptosis and inflammation; baicalein maintains gill homeostasis through suppressing ROS/ER stress signaling in common carps (Cyprinus carpio L.)

Xiaojing Li,Qian Wu,Dan Chen,Yichen Bai,Yuhong Yang,Shiwen Xu

Cadmium (Cd) is a major contaminant in natural environments and exerts adverse effects on aquatic biota at low concentrations. Gill is as vital respiratory organ and may cause pollutants to enter fish during gas exchange. Baicalein (BAI), as a kind of flavonoids, possess antioxidant properties through inactivating free radicals. To confirm the potential effects and approaches of BAI addition in maintaining the gill stability, 90 common carps ( Cyprinus carpio L.) were selected and randomly divided into water environment exposure group (0.22 mg/L Cd) and/or feed added with 0.10 g/kg BAI for 30 days. The analysis of ion content in serum showed that Cd exposure disturbed ion homeostasis , and BAI could reduce serum Cd concentration. The histopathological results of gills showed that Cd exposure caused gill tissue lesions and structural damage, and BAI feeding effectively alleviated this damage. In addition, BAI could enhance antioxidant activity and activate Nrf2/HO-1 axis, thereby reducing oxidative stress and endoplasmic reticulum (ER) stress. Moreover, BAI lightened cytokine imbalance, inflammatory response, and necroptosis. Overall, the results indicated that BAI feeding could maintain gill homeostasis against Cd poisoning via the ROS/ER stress signaling. This trial revealed the properties of BAI resistance to metal Cd in aquaculture and partially elucidated its mechanism.

Taurine Alleviated the Negative Effects of an Oxidized Lipid Diet on Growth Performance, Antioxidant Properties, and Muscle Quality of the Common Carp (Cyprinus carpio L.)

Dan Liu,Jiali Mi,Xiao Yan,Chaobin Qin,Junli Wang,Guoxing Nie

In the present study, we conducted a 10-week culture experiment to investigate the effects of taurine on the growth performance, antioxidant properties, and muscle quality of the common carp fed an oxidized lipid diet. There were five experimental groups with three replicates each. Based on the fresh fish oil group (FO), equal amounts of oxidized fish oil (with a thiobarbituric acid-reactive substances value of 49.57 ± 2.34 mg/kg) and 0 g/kg (OFO), 4 g/kg (OT4), 8 g/kg (OT8), and 12 g/kg (OT12) taurine were added to the diet, while the same composition ratio was maintained by changing the microcrystalline cellulose content. Compared to the FO group, the feed conversion ratio, weight gain rate, muscle crude lipid, and n-3/n-6 polyunsaturated fatty acids (PUFA) ratio were significantly lower in the group OFO ( P < 0.05). In addition, compared to the FO group, OFO fish showed an increased content of malondialdehyde and protein carbonylation and decreased hardness, brightness, pH, superoxide dismutase, and catalase levels in the muscle ( P < 0.05). Notably, the growth index significantly improved in the OT4 group compared to that in the OFO group ( P < 0.05). In addition, dietary taurine increased the crude lipid content, n-3/n-6 PUFA, antioxidant capacity, hardness, springiness, brightness, pH, and collagen content in the muscle compared with OFO fish ( P < 0.05). Moreover, taurine supplementation significantly reduced myofiber diameter and increased myofiber density ( P < 0.05) and enhanced the expression levels of paired box 7 ( pax7 ), myogenic factor 5 ( myf5 ), myogenic differentiation antigen ( myod ), and myogenic regulatory factor 4 ( mrf4 ) compared with that of the OFO group ( P < 0.05). Considering growth performance and muscle quality, the optimal supplemental levels of taurine in the oxidized lipid diet were 4 g/kg and 6.84–7.70 g/kg, respectively.

Chlorpyrifos caused necroptosis via MAPK/NF-κB/TNF-α pathway in common carp (Cyprinus carpio L.) gills

Jianqing Chen,Bing Shao,Jinliang Wang,Zhiqiang Shen,Honggui Liu,Shu Li

Chlorpyrifos (CPF) is an organophosphate insecticide and can cause cell death of animals. In the study, the common carp were exposed to CPF at 0 μg/L (the control group), 1.16 μg/L (the low dose group), 11.6 μg/L (the medium dose group), and 116 μg/L (the high dose group), respectively. The carp were euthanized at the 30th day and gills were collected immediately. The ultrastructural and histopathological observations showed obvious necrosis characteristics and inflammatory injury in the CPF-treated groups. CPF exposure activated the MAPK pathway, in which the mRNA and protein expressions of extracellular signal-regulated (ERK), p38 MAP kinase (p38), and c-Jun N-terminal kinase (JNK) were increased; the mRNAs and proteins of NF-κB and TNF-α were activated; and the mRNAs and proteins of necroptosis related genes were changed (the mRNA and protein expression of RIPK1 , RIPK3 , MLKL, and FADD were increased and caspase-8 was decreased) with concentration dependency. Taken together, we concluded that CPF exposure activated the MAPK/NF-κB/TNF-α pathway, promoted inflammatory injure and evoked necroptosis in common carp gills. In addition, CPF-induced inflammation and necroptosis was concentration dependency. The toxic effects of CPF on gills provided data for both aquaculture and toxicological studies.

The Effect of High Levels Carbohydrate on Intestinal Microbiota, Metabolites, and Health of Common Carp (Cyprinus carpio L.)

Jinrui Xiong,Liping Yang,Luming Wang,Shaoyang Zhi,Mengjuan Zhao,Chunchu Xu,Leya Qu,Xiaorui Guo,Xiao Yan,Chaobin Qin,Guoxing Nie

Long-term consumption of high-carbohydrate feed may adversely affect intestinal health of fish; however, the underlying roles remain ambiguous. This study examined the effects of varying carbohydrate levels on the intestinal flora of common carp and assessed how microbial metabolites influence intestinal health. Two hundred seventy common carps were chosen and distributed randomly into three groups that fed diets containing starch at levels of 15% (low-carbohydrate diet [LCD]), 28% (medium-carbohydrate diet [MCD]), and 45% (high-carbohydrate diet [HCD]) for 60 days. A significant increase in final body weight, weight gain rate, and specific growth rate within the MCD group, while feed conversion ratio exhibited a decrease in comparison to the other groups ( p < 0.05). Feeding with a HCD led to decreased activity of catalase and increased malondialdehyde content, which was consistent with reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR) analysis results ( p < 0.05). Specifically, the RT-qPCR results revealed that HCD treatment significantly upregulated il1β , il6 , and il8 transcript levels. Whereas, the il10 messenger RNA (mRNA) was markedly reduced in comparison to the LCD group. Furthermore, the HCD group exhibited an increased abundance of Proteobacteria , accompanied by a reduction in Fusobacteria abundance, and also revealed an upsurge in opportunistic pathogenic bacteria, such as Aeromonas and Shewanella . The correlation analysis demonstrated negative correlations of anti-inflammatory active substances such as fucoxanthin, (S)-reticuline, hecogenin, and uridine with Aeromonas , but positive correlations with Luteolibacter . In summary, dietary carbohydrates might mediate intestinal flora to regulate their metabolites and affect intestinal inflammatory response.

Sepiapterin reductase (Spra and Sprb) regulate carotenoid and pteridine metabolism influencing the koi carp (Cyprinus carpio L.) coloration

X. Tian,Y.S. Shan,N.N. Peng,X. Ma,X. Shi,H. Li,X.J. Li,X.L. Meng

The evolution of teleost coloration is strongly linked with the fish-specific genome duplication (FSGD). Sepiapterin reductase (Spr) is commonly required in pteridine pigment synthesis and two paralogs are found in teleosts. However, the functions of spra and sprb in pigmentation have not been completely elucidated. Here we found that spra was intensively expressed in skin, fins and scales of koi carp, while sprb was ubiquitously expressed in all tissues . Then, the Spr enzyme activity inhibitor, sulfapyridine (SFD), was used to explore the phenotype and genes variation after spr inhibition. Following the administration of SFD, spra was significantly suppressed but sprb was up-regulated in skin. Additionally, the color phenotypes shifted from red to yellow due to a reduction in carotenoid concentration and the number of xanthophores/erythrophores in scales. Furthermore, the genes involved in carotenoid metabolism were influenced. However, pteridine metabolism-related genes rarely exhibited obvious changes. The results indicate that spra inhibition resulted in the dysregulation of carotenoid metabolism and pigment cells reduction, whereas sprb was likely to maintain the conserved functions in pteridine synthesis for metabolic homoeostasis. This study provided evidences for the different functions of spr paralogs in carotenoid and pteridine metabolism and for better understanding of the relationship between genome duplication and color pattern diversification.

Cryoprotective effect of water-tailored trehalose-based natural deep eutectic solvents on frozen-thawed mirror carp (Cyprinus carpio L.) surimi

Xin Du,Baohua Kong,Junjie He,Quanyu Zhang,Geer An,Tingting Zhang,Xiufang Xia

The inhibitory effect of water-tailored natural deep eutectic solvents (NADES) constructed from citric acid and trehalose with different amounts on the quality deterioration and oxidation of frozen-thawed (F-T) mirror carp ( Cyprinus carpio L. ) surimi was studied. NADES was obtained by citric acid to trehalose and the effect of moisture addition (v/v) on the structure, physicochemical, and anti-freezing capacity of NADES was assessed. NADES + 10 % H 2 O has relatively low viscosity (25 %) and strong freezing resistance. However, a 50 % H 2 O addition leads to the disappearance of the hydrogen bond. The addition of NADES effectively inhibits water loss, migration, and mechanical damage on F-T surimi. An inhibitory effect of 4 % (w/w) NADES on oxidation was verified by a decrease in carbonyl contents (17.4 %, 8.63 %) and TBARS (37.9 %, 15.2 %) of surimi compared with control ( P  < 0.05) and sucrose + sorbitol after 5F-T cycles, suggesting the potential of NADES as a cryoprotectant for the food industry.

Environmental cadmium exposure perturbs systemic iron homeostasis via hemolysis and inflammation, leading to hepatic ferroptosis in common carp (Cyprinus carpio L.)

Wenli Guo,Jinjin Zhang,Xiaoqian Zhang,Quanzhong Ren,Guangzhe Zheng,Jianxin Zhang,Guoxing Nie

Cadmium (Cd) pollution is considered a pressing challenge to eco-environment and public health worldwide. Although it has been well-documented that Cd exhibits various adverse effects on aquatic animals, it is still largely unknown whether and how Cd at environmentally relevant concentrations affects iron metabolism . Here, we studied the effects of environmental Cd exposure (5 and 50 μg/L) on iron homeostasis and possible mechanisms in common carp. The data revealed that Cd elevated serum iron, transferrin saturation and iron deposition in livers and spleens, leading to the disruption of systemic iron homeostasis. Mechanistic investigations substantiated that Cd drove hemolysis by compromising the osmotic fragility and inducing defective morphology of erythrocytes. Cd concurrently exacerbated hepatic inflammatory responses, resulting in the activation of IL6-Stat3 signaling and subsequent hepcidin transcription. Notably, Cd elicited ferroptosis through increased iron burden and oxidative stress in livers. Taken together, our findings provide evidence and mechanistic insight that environmental Cd exposure could undermine iron homeostasis via erythrotoxicity and hepatotoxicity. Further investigation and ecological risk assessment of Cd and other pollutants on metabolism-related effects is warranted, especially under the realistic exposure scenarios.

Identification of Isthmin-1 in common carp (Cyprinus carpio. L) and the effects on glucose metabolism in vivo and in vitro

Leya Qu,Liping Yang,Yiran Wang,Shaoyang Zhi,Mengjuan Zhao,Jinrui Xiong,Xiao Yan,Chaobin Qin,Guoxing Nie

Isthmin-1 (Ism1) plays roles in glucose uptake in mammals as an adipokine. To investigate its role in the glucose metabolism of common carp ( Cyprinus carpio. L), the Ism1 sequence was cloned, and its expression and distribution in tissues were detected. In addition, we prepared and purified the recombinant Ism1 protein using the E. coli expression system and assessed changes in the expression of key genes related to glucose metabolism through both in vivo injection experiments and primary hepatocyte experiments in vitro. The results revealed that the open reading frame of Ism1 was 1377 bp long, encoding 458 amino acids. Similarity analysis indicated that Ism1 exhibited a close evolutionary relationship with goldfish ( Carassius auratus ), sharing 98.35% amino acid similarity. Ism1 was expressed in all tissues of common carp, with the highest level observed in the heart, followed by the gill, head kidney, and hepatopancreas. Distinct patterns of Ism1 expression were identified during the oral glucose tolerance test and long-term high-carbohydrate and high-fat diet feeding experiments. In vivo studies demonstrated that the serum glucose concentration was reduced on treatment with Ism1, accompanied by a significant upregulation of mRNA levels for gk , hk , and pfk genes in hepatopancreas; conversely pepck and g6pase mRNA levels were significantly downregulated in the hepatopancreas under these conditions as well. Furthermore, our primary hepatocyte experiment confirmed that Ism1 could inhibit pepck and g6pase mRNA expression, while promoting gk , hk , and pfk mRNA expression levels. In conclusion, Ism1, in common carp, could participate in the glucose metabolism, which provides essential information for future studies on the function of Ism1.

Apelin and apj in common carp (Cyprinus carpio L.): Molecular characterization, tissue expression and responses to periprandial changes and fasting–refeeding

Xiao Yan,Chaobin Qin,Dapeng Deng,Guokun Yang,Junchang Feng,Ronghua Lu,Liping Yang,Xiaolin Meng,Xianglin Cao,Guoxing Nie,Guiqin Wang

Apelin and its receptor apj have a variety of biological functions and are involved in appetite and energy regulation. However, the comparative studies of apelin/apj system in fish are still absent. In this study, the open reading frames of apelin and apja and apjb were obtained. Apelin and apj mRNAs were widely distributed in all tissues detected, with the highest level in the brain, followed by the spleen, kidney and foregut. The mRNA expression of apelin and apj s increased rapidly at mealtime and significantly decreased after 30 min of feeding. In addition, there were obvious circadian rhythms (higher at nighttime and lower at daytime) in the expression of apelin and apja and apjb . Furthermore, fasting elevated the mRNA levels of apelin and apj s in the hypothalamus, foregut and hepatopancreas. When the 8-day fasted fish were refed, apelin and apj s mRNA expression significantly decreased. However, there were differences in the response to fasting and refeeding between apja and apjb . The roles of apelin and its receptor apjs in fish are poorly understood. Herein, we employed molecular and physiological studies to characterize their structures, distribution and mRNA expression responses to periprandial changes and fasting–refeeding in Cyprinus carpio L. for the first time.

Glucose transporter 2 in common carp (Cyprinus carpio L.): molecular cloning, tissue expression, and the responsiveness to glucose, insulin, and glucagon

Deng Dapeng,Yan Xiao,Zhao Wenli,Qin Chaobin,Yang Guokun,Nie Guoxing

Glucose transporter 2 ( glut2 ) has been studied in mammals, aves, and several fish, while the comparative studies of glut2 in common carp are still lacking. In this study, glut2 was firstly isolated and characterized from the liver of common carp. The full-length cDNA of glut2 was 2351 bp with an open reading frame (ORF) of 1512 bp, encoding 503 amino acids. Alignment of glut2 amino acid sequences from different species revealed that common carp glut2 showed higher sequence identity with teleosts, and lower homology with mammals and amphibians. Tissue distribution demonstrated that glut2 mRNA level was mainly expressed in liver, foregut, and midgut. To investigate the actions of glut2 on glucose metabolism, the level of glut2 mRNA was detected after intraperitoneal injection of glucose, human insulin and glucagon (100 ng/g), respectively. Following glucose administration, glut2 gene expression was significantly upregulated at 3 h in the foregut. However, no change was found in hepatic glut2 mRNA level, indicating that glut2 may have a role in intestinal glucose uptake rather than in the liver. Following insulin treatment, the expression of glut2 was markedly downregulated at 3 h and 6 h in the liver, and at 3 h in the foregut, respectively. Furthermore, glut2 mRNA expression was unaffected by glucagon injection in the liver and foregut. These results suggested that the expression of glut2 regulated by pancreatic hormones was different. Taken together, our studies firstly revealed the structure of the glut2 gene and its potential functions in glucose metabolism of common carp.

Dual regulation of bile acids: ameliorating hepatic glycolipid disorders and restoring intestinal health in HFHC-challenged Yellow River carp (Cyprinus carpio L.)

Yan Xiao,Yang Bowen,Mi Jiali,Zhi Shaoyang,Feng Junchang,Cheng Lijiao,Wang Junli,Qin Chaobin,Nie Guoxing

Background This study was designed to investigate how three distinct bile acids (BAs) modulate glycolipid metabolic disorders and hepatointestinal injury induced by excessive intake of lipids and carbohydrates in Yellow River carp ( Cyprinus carpio L.) and elucidate the underlying mechanisms involved. Here, the fish were randomly assigned to five groups: a control group (CON), a high-fat high-carbohydrate diet (HFHC) group, a HFHC + 300 mg/kg chenodeoxycholic acid (CDCA) group, a HFHC + 300 mg/kg ursodeoxycholic acid (UDCA) group and a HFHC + 300 mg/kg hyodeoxycholic acid (HDCA) group. Results The results revealed that the serum triglyceride, glucose, and total cholesterol levels were significantly elevated in HFHC-fed fish, accompanied by increased glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) activities in the serum and hepatopancreas. However, dietary supplementation with bile acids in the HFHC diet significantly improved these negative changes. Analysis of BA-glycolipid metabolism-related gene expression and enzyme activities in the hepatopancreas revealed that CDCA and HDCA inhibited gluconeogenesis (FBPase/PEPCK/G6Pase) and lipogenesis (SREBP-1/FAS), while promoting glycogen accumulation (genes and glycogen levels) and fatty acid β-oxidation (PPARα) via activation of the FXR (farnesoid X receptor) /SHP (small heterodimer partner) pathway. In contrast, dietary UDCA supplementation increased intestinal TGR5 (takeda G protein-coupled receptor 5) expression and suppressed the activities of two key gluconeogenic enzymes, PEPCK and G6Pase. Additionally, dietary BAs supplementation alleviated HFHC diet-induced intestinal inflammation by inhibiting the NF-κB (Nuclear Factor κB) pathway. Bile acids relieved gut dysbiosis, improved microbial alpha diversity and community structure, and enriched beneficial bacteria including Cetobacterium somerae . These microbial changes eventually modulated host substance synthesis and metabolism. HE staining showed that HFHC diet caused hepatopancreatic lesions and intestinal morphological damage in Yellow River carp, which were effectively alleviated by bile acid addition. Conclusions In conclusion, HFHC diets disrupt fish glycolipid metabolism and impair hepato-intestinal health in Yellow River carp, whereas dietary BAs can attenuate these detrimental effects by modulating metabolic pathways and the gut microbiota composition.
