Characterization of a d-lyxose isomerase from Bacillus velezensis and its application for the production of d-mannose and l-ribose

Guo Zongren,Long Liangkun,Ding Shaojun

d -Mannose and l -ribose are two important monosaccharides, which have attracted public attention recently because of their great application potentials in food, cosmetic and pharmaceutical industries. Sugar isomerases catalyze the sugar isomerization and therefore can be used as the biocatalysts for production of the high-value sugars from inexpensive sugars. l -arabinose isomerase catalyzes the conversion of l -arabinose to l -ribulose, while d -lyxose isomerase catalyzes l -ribulose and d -fructose to l -ribose and d -mannose, respectively. In this paper, a putative d -LI from Bacillus velezensis (BvLI) was identified, characterized and used to produce d -mannose and l -ribose from d -fructose and l -arabinose, respectively. The recombinant BvLI exhibited a maximum activity at 55 °C and pH 6.5, in the presence of 0.1 mM Co 2+ . Approximately 110.75 g/L d -mannose was obtained from 500 g/L d -fructose in 6 h by the recombinant BvLI, and approximately 105 g/L l -ribose was obtained from 500 g/L l -arabinose in 8 h by the successive biocatalysis of l -arabinose isomerase from Bacillus licheniformis (BlAI) and BvLI.

D-Mannose-Modified M2 Macrophage Membrane-Derived Decoy Nanovesicles for Synergistic Immunotherapy of Asthma

Shukun Zhao,Qiong Jiang,Ling Yu,Quan Liu,Zi Chen,Danting Zhan,Shuyu Chen,Yuqiong Yang,Jiacheng Zhong,Bingxin Guo,Fei Shi,Rongchang Chen,Lingwei Wang,Lang Rao,Junxia Zheng,Qian-Fang Meng,Shanze Chen

Eosinophilic asthma is a chronic inflammatory disease driven by Type 2 immune cells and cytokines. While cytokine-based immunotherapies have shown promising efficacy in asthma treatment, current approaches often fail to simultaneously target multiple cytokines. In this study, we present nanodecoys derived from the cellular membranes of M2 macrophages, which exhibit a high density of cytokine receptors, enabling efficient sequestration of multiple Type 2 cytokines. This mechanism significantly alleviates OVA-induced eosinophilic asthma inflammation in mice. To enhance therapeutic efficacy, D-mannose glycans, a potential immunoregulatory molecule for asthma, were modified onto the surface of the nanovesicles. This modification facilitates the phagocytic clearance of absorbed cytokines by targeting the mannose receptor (CD206). Consequently, the mannose-modified nanovesicles exhibit synergistic immunosuppressive effects on asthmatic inflammation by inhibiting M2 macrophage polarization, CD4+ T cell Th2 polarization, and eosinophil activation, as revealed by single-cell RNA sequencing and in vitro analyses. In summary, this study presents a safe and effective nanotechnology-based immunotherapy for asthma and potentially other Type 2 inflammatory diseases.

Construction of a Concanavalin A electrochemical sensor base on a novel sandwich capture mode

Xionghui Ma,Jianping Li,Yunbin Liu,Yali Yuan,Guobao Xu

Herein, a concanavalin A (ConA) electrochemical sensor was constructed via a sandwich mode. Firstly chitosan functionalized graphene oxide (CS/GO) composite was introduced on the of glassy carbon electrode (GCE) as the substrate. D–mannose (D-man) was immobilized on the surface of CS/GO via Schiff base reaction. Due to the specific affinity between mannose and ConA, the prepared D–man/CS/GO sensing interface could specifically capture ConA which could further react with mannose residues of HRP and form a sandwich configuration. Based on the catalytic effect of horseradish peroxidase (HRP) to H 2 O 2 , a ConA electrochemical sensor with enzyme catalytic amplification was constructed by taking hydroquinone as electrical mediator. The morphology of CS/GO was investigated by transmission electron microscope and Fourier transform infrared spectroscopy. Electrochemical techniques including electrochemical impedance spectroscopy and differential pulse voltammogram (DPV) were used for reveal the characteristics of the sensing platform. Benefit from the high surface area of GO and enzyme catalytic amplification of HRP, the peak current from DPV is linearly dependent on the ConA concentration ranging from 5   ×   10 −9 to 5   ×   10 −7   mol/L with a detection limit of 1.24   ×   10 −9   mol/L. The results demonstrated that the proposed sensor might exploit a new direction for determination of ConA.

Targeting the gut microbiota to investigate the mechanism of Lactiplantibacillus plantarum 1201 in negating colitis aggravated by a high-salt diet

Shufang Chen,Zhongyue Ren,Yalan Huo,Wanyu Yang,Lingling Peng,Huihui Lv,Lijuan Nie,Hua Wei,Cuixiang Wan

High-salt diet (HSD) affects the composition and function of the intestinal microbiota and cause health problems. This study confirmed that HSD aggravates dextran sulphate sodium (DSS)-induced colitis by changing the relative abundance of the gut microbiota, activating the NF-κB pathway, and up-regulating the mRNA levels of inflammatory factors. We explored the effect of L. plantarum 1201 in negating DSS-induced ulcerative colitis, which is aggravated by HSD for the first time. Results show that L. plantarum 1201 rebuilt the balance of intestinal flora by decreasing the ratio of Firmicutes/Bacteroidetes and increasing the relative abundance of Bifidobacterium , Lactobacillus and butyric-producing bacteria. Moreover, L. plantarum 1201 inhibited the up-regulation of inflammatory cytokines (e.g., IL-1β, TNF-α, IL-6, IL-22, and IFN-γ) mRNA levels, increased colonic tight junction protein (ZO-1, ocludin, and claudin-3) expression, and increased serum levels of beneficial metabolites, including alpha-tocopherol (α-T) and D-mannose. By reconstructing an animal model of colitis, we further discovered that α-T and D-mannose inhibited the NF-κB pathway, improved tissue injury, and decreased the expression of pro-inflammatory cytokines (e.g., IL-1β, TNF-α, and IL-6). This study proves for the first time that L. plantarum 1201 attenuates high-salt-aggravated colitis by increasing the serum concentrations of endogenic D-mannose in mice serum and inhibiting the consumption of α-T through intestinal flora. Therefore, regulating the gut microbiota is a potential treatment for high-salt-aggravated colitis.

Mannose-Modificated Polyethylenimine: A Specific and Effective Antibacterial Agent against Escherichia coli

Mei Liu,Jiao Li,Baoxin Li

Polyethylenimine (PEI) has antimicrobial activity against Gram-positive (Staphylococcus aureus, S. aureus) and Gram-negative (Escherichia coli, E. coli), bacteria but is highly cytotoxic, and the selective antimicrobial activity against S. aureus is obviously better than that against E. coli. To reduce the cytotoxicity and improve the antibacterial activity against E. coli, we modified PEI with d-mannose through nucleophilic addition between primary amine and aldehyde groups to get mannose-modified polyethylenimine copolymer particles (Man-PEI CPs). The use of mannose may provide good targeting ability toward E. coli pili. The antibacterial activity of Man-PEI CPs was investigated. Man-PEI CPs shows specific and very strong killing capability against E. coli at a concentration of 10 μg/mL, which is the highest antimicrobial efficiency compared to that of unmodified PEI (220 μg/mL). The antibacterial mechanism demonstrated that the enhancement in antibacterial activity is due to specific recognition of the mannose and destroying the cell wall of the bacteria by PEIs. Importantly, the Man-PEI CPs show less cytotoxicity and excellent biocompatibility. The results indicate that Man-PEI CPs have great potential as novel antimicrobial materials to prevent bacterial infections and provide specific applications for killing E. coli.

D-mannose-modified nanoliposomes enhance the targeted delivery of ovalbumin to improve its anti-inflammatory, antioxidant, and macrophage polarization effects

Jiahao Liao,Jianyu Huang,Xiaoyi Chi,Xiaoqiong Li,Jian Kuang,Jianqiang Li,Xiangyu Bian,Jinzhu Pang,Jinjun Li,Weiwei Wang

In this study, D-mannose-modified nanoliposomes (MLipo@OVA) were developed to facilitate the targeted delivery of ovalbumin (OVA) to macrophages. The subsequent anti-inflammatory, antioxidant, and macrophage-polarizing effects of OVA were investigated. The nanoliposomes were designed with DSPE-PEG and DPPC to enhance the bioavailability and stability of OVA. In vitro analysis showed that MLipo@OVA significantly increased macrophage uptake, with D-mannose further enhancing this effect. MLipo@OVA reduced pro-inflammatory cytokines (TNF-α, IL-1β) and increased anti-inflammatory IL-10 levels in LPS-induced macrophages, primarily via the inhibition of the TLR4/NF-κB pathway. Furthermore, ROS levels were decreased, SOD and GSH activity was enhanced, the Nrf2/HO-1 pathway was activated, and the compound exhibited strong antioxidant activity. MLipo@OVA also promoted M2 macrophage polarization by upregulating CD206 and Arg-1 and downregulating iNOS. Overall, these results indicate that MLipo@OVA could be used as a nanomedicine to facilitate targeted anti-inflammatory and antioxidant therapies.

Detecting N-Phenyl-2-Naphthylamine, L-Arabinose, D-Mannose, L-Phenylalanine, L-Methionine, and D-Trehalose via Photocurrent Measurement

Feng Li,Ruoxi Yang,Jian Xu,Guohai Xu,Ye Wu

The concentration of small molecules reflects the normality of physiological processes in the human body, making the development of simple and efficient detection equipment essential. In this work, inspired by a facile strategy in point-of-care detection, two devices were fabricated to detect small molecules via photocurrent measurement. A linear response of the photocurrent against the concentration of the small molecules was found. In the first device, metal ions were introduced into gel substrates made by xanthan gum to enhance photocurrent response. N-phenyl-2-naphthylamine was measured when iron or manganese ions were used. L-Phenylalanine was measured when the gel was modified by samarium, iron, cerium, or ytterbium ions. L-(+)-Arabinose was detected via the gels modified by iron or holmium ions. D-(+)-Mannose was detected when the gel was modified by ytterbium, manganese, chromium, or sodium ions. L-Methionine was detected in the gels modified by samarium, zinc, or chromium ions. The second device was based on a paper sheet. A sugar-like molecule was first synthesized, which was then used to modify the paper. The detection was possible since the photocurrent showed a linear trend against the concentration of D-Trehalose. A linear fit was conducted to derive the sensitivity, whose value was found to be 5542.4. This work offers a novel, simple, and environmentally sustainable platform that is potentially useful in remote areas lacking medical devices.

MicroRNA-195 liposomes for therapy of Alzheimer's disease

Dan Su,Zhong Chen,Xiaobin An,Junkai Yang,Jinan Yang,Xuqiao Wang,Yang Qu,Chen Gong,Yani Chai,Xiaoying Liu,Wei Cheng,Dongyang Wang,Yan Wu,Jing Ma,Xinyue Zhao,Qin Wang,Yun Xu,Haisheng Peng,Jing Ai

The complex etiologies and mechanisms of Alzheimer's disease (AD) underscore the importance for devising multitarget drugs to achieve effective therapy. MicroRNAs (miRNAs) are capable of concurrently regulating the expression of multiple proteins by selectively targeting disease- associated genes in a sequence-specific fashion. Nonetheless, as RNA-based drugs, their stability in the circulation and capacity of traversing the blood–brain barrier (BBB) is largely compromised, thereby limiting their potential clinical applications. In this study, we formulated the nanoliposomes encapsulating polyethyleneimine (PEI)/ miR-195 complex (DPMT@PEI/ miR-195 ) that was engineered through dual modifications to contain P -aminophenyl-alpha-d-mannopyranoside (MAN) and cationic cell-penetrating peptide (TAT). DPMT@PEI/ miR-195 exhibited the enhanced BBB- and cell membrane penetrating capability. As expected, we observed that DPMT@PEI/ miR-195 administered through intravenous tail injection of produced greater effectiveness than donepezil and the same range of effect as aducanumab in alleviating the cognitive decline in 7-month-old APP/PS1 mice. Moreover, the combination treatment with DPMT@PEI/ miR-195 and donepezil effectively ameliorated the deterioration of cognition in 16-month-old APP/PS1 mice, with enhanced effects than either DPMT@PEI/ miR-195 or donepezil alone. Furthermore, DPMT@PEI/ miR-195 effectively attenuated the positive signals of Aβ, AT8, and CD68 in APP/PS1 mice without notable side effects. Our findings indicate DPMT@PEI/ miR-195 as a promising potentially new agent or approach for the prophylaxis and treatment of early and advanced stages of Alzheimer's disease.

Antioxidative and Cytoprotective Effects of Rosa Roxburghii and Metabolite Changes in Oxidative Stress-Induced HepG2 Cells Following Rosa Roxburghii Intervention

Yangchen Mao,Ruyi Sha,Yuhao Sun,Zhenzhen Wang,Jun Huang

Rosa Roxburghii(RR), a traditional Chinese medicinal fruit, is rich in bioactive substances that make it a potential natural antioxidant resource. This research aimed to study the antioxidant properties of RR by in vitro experiments and through intracellular assessment in H2O2-induced HepG2 cells. A non-targeted metabolic analysis was conducted to indicate changes in intracellular and extracellular metabolites. Differential metabolites and metabolic pathways were explored using PCA, PLS-DA, and KEGG pathway analysis. The results showed that RR rich in bioactive substances exhibited a significant antioxidative property in vitro and intracellularly. This property may be achieved by scavenging free radicals, increasing the activity of catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and the levels of bicinchoninic acid (BCA) while reducing the reactive oxygen species (ROS) generation. This study identified 13 differential metabolites intracellularly and 7 extracellularly, among which the key differential metabolites included D-glucopyranose, D-mannose, fructose, citric acid, malic acid, cholesterol, and cholestenone. These key metabolites primarily regulated glucose-related metabolism, the citrate cycle, and the primary bile acid biosynthesis pathway in H2O2-induced HepG2 cells. These findings provide potential application evidence of RR in the development of natural resources for functional foods.

Construction of mannose-modified polyethyleneimine-block-polycaprolactone cationic polymer micelles and its application in acute lung injury

Su Meiling,Hu Heping,Zhao Xuan,Huang Chengyuan,Yang Bowen,Yin Zongning

This study evaluated the D-mannose modified polyethyleneimine-block-polycaprolactone biomacromolecule copolymer micelles (PCL-PEI-mannose) as a targeted delivery of the glucocorticoid dexamethasone (DXM) to lung inflammation tissues and enhances the vehicle for its anti-inflammatory effects. Dexamethasone was encapsulated in the hydrophobic core of cationic polymer micelles by solvent evaporation. The polymeric micelles exhibited sustained-release within 48 h, good blood compatibility, and colloidal stability in vitro. The cellular uptake of mannose-modified micelles was higher compared with the non-modified micelles. And drug-loaded targeted micelles could inhibit the production of inflammatory factors in activated RAW264.7 cells. The distribution results indicated that drug-loaded targeted micelles highly improved the lung targeting ability, reduced the wet/dry ratio of injured lung tissue, and relieved the lung inflammation, accompanied by the decrease of inflammatory cell infiltration, myeloperoxidase activity, and inflammatory mediator levels in bronchoalveolar lavage fluid. These findings suggested that PCL-PEI-mannose delivery system could facilitate the lung-specific delivery and inhibit the inflammatory response. Collectively, PCL-PEI-mannose polymer micelles could be used as a potential delivery system for the treatment of acute lung injury (ALI). Graphical abstract
