Pt(IV) prodrug-backboned micelle and DCA loaded nanofibers for enhanced local cancer treatment

Zhiyun Zhang,Yanjuan Wu,Gaizhen Kuang,Shi Liu,Dongfang Zhou,Xuesi Chen,Xiabin Jing,Yubin Huang

Drug-loaded nanocarriers, especially polymeric micelles, have attracted much attention in recent years, but their application is still limited due to their systemic toxicity and unsatisfactory tumor delivery efficiency through intravenous administration. In this study, we demonstrated an implantable prodrug micelle/small drug molecule co-loaded nanofiber strategy for enhanced local cancer treatment. A reduction-responsive Pt(IV) prodrug-backboned micelle, as a prodrug of bioactive Pt(II), and DCA were co-electrospun into poly(vinyl alcohol) (PVA) nanofibers. The prepared Pt(IV) prodrug-backboned micelle/DCA fibers exerted a synergistic effect on cancer cells. In contrast to systemic administration of chemotherapeutic agents, this implantable device exhibited enhanced anti-cancer efficacy with lower systemic toxicity against advanced cervical cancer in vivo. This work provides a facile, efficient and safe prospect of using a prodrug micelle and small molecular drug co-loaded electrospun device for localized and advanced cancer therapy.

Catalytic ignition of the [BMIM]DCA-H2O2 propellant with the Cu(vim)2(DCA)2 complex

Yuqi Zhang,Yuan Yao,Peihao Dou,Qing Cheng,Yanqiang Zhang

An ionic liquid (IL)-H2O2 propellant offers a green alternative to the traditional hydrazine-N2O4 system. Due to the weak oxidizing ability of H2O2, typical ILs cannot be spontaneously ignited. To address this issue, catalytic ignition was proposed, and a series of metal complex catalysts with the formula M(vim)2(DCA)2 (M = Mn, Fe, Co, and Cu) were synthesized. Single-crystal X-ray diffraction analysis shows that the Mn, Fe and Co complexes adopt an octahedral geometry, while the Cu complex exhibits a planar quadrilateral geometry. The physicochemical properties (Td, ΔHc, ΔHf, and Isp) of the fuels (10 wt% M(vim)2(DCA)2 + 90 wt% [BMIM]DCA) were evaluated. Dynamic light scattering and UV-vis spectroscopy analyses confirm that the four complexes disperse well in [BMIM]DCA. Ignition tests reveal that Cu(vim)2(DCA)2 exhibits the best catalytic performance for the ignition of [BMIM]DCA with H2O2, achieving tid as short as 21 ms. EPR studies indicate that Cu(vim)2(DCA)2 catalyzes H2O2 to generate reactive oxygen species (·OH, ·O2−, and 1O2), which subsequently induce the spontaneous ignition of [BMIM]DCA. DFT calculations reveal that the zero-dimensional geometry of Cu(vim)2(DCA)2 features an unsaturated Cu center and electron-rich ligand framework. This typical structure enables efficient electron transfer to activate H2O2, boosting ROS generation and ignition. This work clarifies the crucial role of the metal center in the IL-H2O2 system and provides an important insight into the catalytic ignition of the “double-green” propellants.

A dual-targeting drug co-delivery system for tumor chemo- and gene combined therapy

Fangrong Zhang,Min Li,Yujie Su,Jianping Zhou,Wei Wang

Regulation of gene expression using p53 is a promising strategy for treatment of numerous cancers, and chemotherapeutic drug dichloroacetate (DCA) induces apoptosis and growth inhibition in tumor, without apparent toxicity in normal tissues. Combining DCA and p53 gene could be an effective way to treat tumors. The progress towards broad applications of DCA/p53 combination requires the development of safe and efficient vectors that target to specific cells. In this study, we developed a DSPE-PEG-AA (1,2-distearoryl- sn -glycero-3-phosphoethanolamine-N-[methoxy(polyethyleneglycol-2000)] ammonium salt-anisamide) modified reconstituted high-density lipoprotein-based DCA/p53-loaded nanoparticles (DSPE-PEG-AA/rHDL/DCA-PEI/p53 complexes), which was fabricated as a drug/gene dual-targeting co-delivery system for potential cancer therapy. Here, DCA-PEI was utilized to effectively condense the p53 plasmid, to incorporate the plasmid into rHDL and to act as an antitumor drug to inhibit tumor cell growth. The DSPE-PEG-AA/rHDL/DCA-PEI/p53 complexes exhibited desirable and homogenous particle size, neutral surface charge and low cytotoxicity for normal cells in vitro . The results of confocal laser scanning microscopy (CLSM) and flow cytometry confirmed that the scavenger receptor class B type I (SR-BI) and sigma receptor mediated dual-targeting function of the complexes inducing efficient cytoplasmic drug delivery and gene transfection in human lung adenocarcinoma cell line A549. And in vivo investigation on nude mice bearing A549 tumor xenografts revealed that DSPE-PEG-AA/rHDL/DCA-PEI/p53 complexes possessed specific tumor targeting and strong antitumor activity. The work described here demonstrated that the DSPE-PEG-AA/rHDL/DCA-PEI/p53 complexes might offer a promising tool for effective cancer therapy.

The denitrification characteristics of novel functional biocarriers immobilised by non-dissolved redox mediators

Qing Xu,Jianbo Guo,Chunmei Niu,Jing Lian,Zhenghao Hou,Yankai Guo,Shaoying Li

This study investigated the denitrification characteristics and practical applications of PET–CAs (chloroanthraquinones immobilised on polyethylene terephthalate), which are novel functional biocarriers. The PET–CAs were characterized using Fourier transform infrared spectroscopy (FT-IR) and energy dispersive spectrometry (EDS). Among the PET–CAs, PET-1,8-DCA (PET modified by 1,8-dichloroanthraquinone) was the best functional biocarrier for accelerating the denitrification process with approximately 0.14   mmol anthraquinone. The denitrification process was pseudo-zero order, with C NO3 −   =   175.64–19.28   t and R 2   =   0.9715. The pH of the PET-1,8-DCA system increased and ultimately reached 9.7. The stabilized ORP value with PET-1,8-DCA was approximately 70   mV lower than that of the control with PET . This study explored a novel design concept for the immobilization of quinone.

Efficient cobalt recovery and separation from spent batteries using a novel [TKN][Dca] ionic liquid

Jiateng Shi,Junfeng Wang,Tao Dong,Jie Yan,Danfeng Jiang,Zengxi Li,Haitao Zhang

The ionic liquid tetrakis(decyl)ammonium dicyanamide ([TKN][Dca]) was investigated for the selective extraction of Co(II) from a simulated spent battery leachate, achieving efficient separation of Co(II) from Ni(II) and Li(I). [TKN][Dca] demonstrate a high Co(II) extraction efficiency in a 4 mol/L NaCl solution, without requiring pH adjustment. The separation factor ( β Co/Ni ) for [TKN][Dca] was determined to be 47433. Density functional theory (DFT) calculations and ESI-MS spectra revealed that the predominant anionic species involved in Co(II) extraction were [Co(Dca) 3-x Cl x ] − , with [Co(Dca)Cl 2 ] − and [Co(Dca) 2 Cl] − as the primary complexes. The strong coordination between the dicyanamide anion (Dca − ) and Co(II) enabled selective separation of Co(II) from the leachate. Co(II) was efficiently stripped from the loaded organic phase using Na 2 SO 4 , with the extraction efficiency after stripping comparable to that of fresh [TKN][Dca]. Furthermore, [TKN][Dca] exhibits excellent stability and maintained consistent performance over multiple extraction-stripping cycles, highlighting its potential for sustainable applications in metal separation fields.

Gut microbiota derived DCA enhances FOLFOX efficacy via Ugt1a6b mediated enterohepatic circulation in colon cancer

Qian Fang,Xiaoying Hou,Limei Fan,Yufei Deng,Xiaoxuan Li,Hongyun Zhang,Haiping Wang,Zhengqi Fu,Binlian Sun,Xiji Shu,Hongzhi Du,Yuchen Liu

FOLFOX (5-Fluorouracil, Calcium Folinate combined with Oxaliplatin) is a preferred chemotherapy regimen for colon cancer, but its limited efficacy remains a major challenge, significantly impairs patient outcomes. There is an urgent need to identify strategies to improve its therapeutic effectiveness. Our previous studies have suggested that gut microbiota-derived bile acids may be involved in the anticancer effect of FOLFOX in vitro , however, the underlying mechanism remains unclear. In this study, we investigated the role of bile acids in modulating FOLFOX efficacy and the related mechanisms. We first demonstrated that bile acids depletion (cholestyramine treatment) enhanced FOLFOX efficacy in an orthotopic colon cancer mouse model, suggesting that bile acids play a key role in FOLFOX’s therapeutic effects. Further, based on the system screen of 15 bile acids on FOLFOX efficacy via MTT, colony formation and flow cytometry assay, Deoxycholic Acid (DCA) and Glycodeoxycholic Acid (GDCA) were annotated as potential modulators of FOLFOX efficacy. Among these, DCA was further validated to significantly enhance FOLFOX’s anti-colon cancer effects in vivo . Transcriptomic analysis and subsequent biological experiments revealed that DCA enhanced FOLFOX efficacy via Ugt1a6b. In conclusion, our findings establish that gut microbiota-derived DCA enhances the efficacy of FOLFOX potentially via Ugt1a6b mediated enterohepatic circulation, providing novel insights into a synergistic therapeutic strategy for improving colon cancer treatment.

Development and application of a fluorescent immunochromatographic assay for metabolite G-DCA quantification in ICP

Jianyi Gao,Linxia Shuang,Xiaojin Yang,Ruirui Dong,Gaoying Wang,Rong Wang,Ting Zhang

Intrahepatic cholestasis of pregnancy (ICP) is a pregnancy-specific liver disorder characterized by dysregulated bile acid metabolism, which can lead to severe adverse pregnancy outcomes. Glycine-conjugated deoxycholic acid (G-DCA) is a small-molecule metabolite that has been reported to be elevated in the serum of patients with ICP; however, its clinical application remains limited because conventional chromatographic methods for its quantification are time-consuming, costly, and not well suited for rapid clinical screening. To address this gap, we developed a time-resolved fluorescence immunochromatographic test strip (TRF-ICTS) for the quantitative detection of serum G-DCA. Based on a small-molecule competitive binding design, the assay requires only 10 μL of serum sample and enables rapid detection within 16 min. The TRF-ICTS demonstrated good analytical performance, with a linear range of 0.05–10.0 ng/mL and a detection limit of 0.074 ng/mL, and maintained stable performance over two weeks of storage at both 4 °C and 37 °C. The results showed good quantitative agreement with LC–MS/MS measurements. Evaluation of sensitivity, specificity, and precision demonstrated that all parameters were within acceptable ranges. Clinical validation using serum samples from ICP patients and healthy pregnant women showed that G-DCA levels were elevated across different trimesters and were more pronounced in patients with severe ICP and adverse pregnancy outcomes. Overall, the developed TRF-ICTS provides a rapid and sensitive approach for quantifying G-DCA and supports its use as a complementary biomarker for ICP diagnosis and disease stratification, with potential to provide complementary insights into bile acid changes following UDCA treatment.

Influence of comonomer DCA on thermal, crystallization, and mechanical characteristics of bio-based semi-aromatic PA10T

Zhu Rongli,Pu Zejun,Peng Qiuxia,Wang Xu,Wu Fang,Zheng Pan,Yu Dayang,Chen Keli,Ni Wenlong,Zhong Jiachun

In this study, the semi-aromatic poly (decamethylene terephthalamide) acid (PA10T) and an array of PA10T/1010 with various copolymerization content of the third monomer 1,10-decanedioic acid (DCA) were synthesized using a productive one-step melt polycondensation method. Importantly, the addition of DCA’s effect for PA10T on crystallization behaviors, thermal properties, and mechanical performances were extensively investigated. Additionally, the results of DSC demonstrate that all the melting temperature of PA10T/1010 copolyamides are significantly lower than PA10T by introducing the third monomer DCA, and the heat stability has been not significantly affected according to the results of TGA, suggesting the improvement of processing properties. Besides, compared with PA10T, the bending modulus, breaking elongation, tensile strength, and izod impact strength of PA10T/1010-1 all increase obviously, when the feed ratio of DCA and terephthalic acid (PTA) is 10:90. Thus, introducing bio-based DCA for copolymerization is a promising method to enhance both the processing and mechanical properties of PA10T.

Electric double-layer of [emim][DCA] ionic liquid at heterogeneous interface of TiO2/C composite: From simulation to experiment

Jiabao Zhu,Linghong Lu,Lili Shi,Zhongyang Dai,Wei Zhuang,Zhengsong Weng

The hydrophilicity/hydrophobicity of heterogeneous interface of the electrode material is critical for the performance of ionic liquids supercapacitors . However, the behavior of electrolytes at the electrode interface, especially the interface microstructure and properties is not yet clear. We investigated the electric double-layer of [emim][DCA] on slit pore of TiO 2 /C composite with the heterogeneous surface by combining molecular dynamics simulation and experimental measurements. The results of simulation show that coverage ratio of carbon on the surface affect the number density distribution, orientation and diffusion coefficient of [emim][DCA]. Moreover, we calculated the charge distribution of electric double-layer of [emim][DCA] and the capacitance. The results indicate that the capacitance of slit pore with the surface of TiO 2 with 78.7% carbon coverage ratio has the highest capacitance at high surface charge density. We performed an electrochemical experiment using composite of TiO 2 covered with carbon as the electrode material, and obtained the capacitance from cyclic voltammetry (CV). The experimental results also indicate there is an optimal coverage ratio of carbon, which achieves the highest capacitance. Our researches could provide a microscopic perspective of the electric double layer at the heterogeneous interface and the guidance for the design of electrode material.

Parabacteroides distasonis regulates the infectivity and pathogenicity of SVCV at different water temperatures

Zhang Yujun,Gao Yan,Li Chen,Zhang Yong-An,Lu Yuanan,Ye Jing,Liu Xueqin

Background Spring viremia of carp virus (SVCV) infects a wide range of fish species and causes high mortality rates in aquaculture. This viral infection is characterized by seasonal outbreaks that are temperature-dependent. However, the specific mechanism behind temperature-dependent SVCV infectivity and pathogenicity remains unclear. Given the high sensitivity of the composition of intestinal microbiota to temperature changes, it would be interesting to investigate if the intestinal microbiota of fish could play a role in modulating the infectivity of SVCV at different temperatures. Results Our study found that significantly higher infectivity and pathogenicity of SVCV infection in zebrafish occurred at relatively lower temperature. Comparative analysis of the intestinal microbiota in zebrafish exposed to high- and low-temperature conditions revealed that temperature influenced the abundance and diversity of the intestinal microbiota in zebrafish. A significantly higher abundance of Parabacteroides distasonis and its metabolite secondary bile acid (deoxycholic acid, DCA) was detected in the intestine of zebrafish exposed to high temperature. Both colonization of Parabacteroides distasonis and feeding of DCA to zebrafish at low temperature significantly reduced the mortality caused by SVCV. An in vitro assay demonstrated that DCA could inhibit the assembly and release of SVCV. Notably, DCA also showed an inhibitory effect on the infectious hematopoietic necrosis virus, another Rhabdoviridae member known to be more infectious at low temperature. Conclusions This study provides evidence that temperature can be an important factor to influence the composition of intestinal microbiota in zebrafish, consequently impacting the infectivity and pathogenicity of SVCV. The findings highlight the enrichment of Parabacteroides distasonis and its derivative, DCA, in the intestines of zebrafish raised at high temperature, and they possess an important role in preventing the infection of SVCV and other Rhabdoviridae members in host fish. Bile acids modified by the intestinal microbiota promote colorectal cancer growth by suppressing CD8+ T cell effector functions

Jingjing Cong,Pianpian Liu,Zili Han,Wei Ying,Chaoliang Li,Yifei Yang,Shuling Wang,Jianbo Yang,Fei Cao,Juntao Shen,Yu Zeng,Yu Bai,Congzhao Zhou,Lilin Ye,Rongbin Zhou,Chunjun Guo,Chunlei Cang,Dennis L. Kasper,Xinyang Song,Lei Dai,Linfeng Sun,Wen Pan,Shu Zhu

Concentrations of the secondary bile acid, deoxycholic acid (DCA), are aberrantly elevated in colorectal cancer (CRC) patients, but the consequences remain poorly understood. Here, we screened a library of gut microbiota-derived metabolites and identified DCA as a negative regulator for CD8 + T cell effector function. Mechanistically, DCA suppressed CD8 + T cell responses by targeting plasma membrane Ca 2+ ATPase (PMCA) to inhibit Ca 2+ -nuclear factor of activated T cells (NFAT)2 signaling. In CRC patients, CD8 + T cell effector function negatively correlated with both DCA concentration and expression of a bacterial DCA biosynthetic gene. Bacteria harboring DCA biosynthetic genes suppressed CD8 + T cells effector function and promoted tumor growth in mice. This effect was abolished by disrupting bile acid metabolism via bile acid chelation, genetic ablation of bacterial DCA biosynthetic pathway, or specific bacteriophage. Our study demonstrated causation between microbial DCA metabolism and anti-tumor CD8 + T cell response in CRC, suggesting potential directions for anti-tumor therapy.

A novel selenium analog of HDACi-based twin drug induces apoptosis and cell cycle arrest via CDC25A to improve prostate cancer therapy

Zhiyong Shi,Miaomiao Liu,Xiaowen Zhang,Jingyang Wang,Junwei Zhang,Zeyan Peng,Li Meng,Ruijing Wang,Lihong Guo,Qiang Zhang,Jing Li,Liang Yang,Jie Liu,Yang Xu,Jie Yan,Jianlin Cui,Shan Ren,Yang Gao,Yanming Wang,Zhi Qi

Cancer therapy has moved from single agents to more mechanism-based targeted approaches. In recent years, the combination of HDAC inhibitors and other anticancer chemicals has produced exciting progress in cancer treatment. Herein, we developed a novel prodrug via the ligation of dichloroacetate to selenium-containing potent HDAC inhibitors. The effect and mechanism of this compound in the treatment of prostate cancer were also studied. Methods: The concerned prodrug SeSA-DCA was designed and synthesized under mild conditions. This compound's preclinical studies, including the pharmacokinetics, cell toxicity, and anti-tumor effect on prostate cancer cell lines, were thoroughly investigated, and its possible synergistic mechanism was also explored and discussed. Results: SeSA-DCA showed good stability in physiological conditions and could be rapidly decomposed into DCA and selenium analog of SAHA (SeSAHA) in the tumor microenvironment. CCK-8 experiments identified that SeSA-DCA could effectively inhibit the proliferation of a variety of tumor cell lines, especially in prostate cancer. In further studies, we found that SeSA-DCA could also inhibit the metastasis of prostate cancer cell lines and promote cell apoptosis. At the animal level, oral administration of SeSA-DCA led to significant tumor regression without obvious toxicity. Moreover, as a bimolecular coupling compound, SeSA-DCA exhibited vastly superior efficacy than the mixture with equimolar SeSAHA and DCA both in vitro and in vivo . Our findings provide an important theoretical basis for clinical prostate cancer treatment. Conclusions: Our in vivo and in vitro results showed that SeSA-DCA is a highly effective anti-tumor compound for PCa. It can effectively induce cell cycle arrest and growth suppression and inhibit the migration and metastasis of PCa cell lines compared with monotherapy. SeSA-DCA's ability to decrease the growth of xenografts is a little better than that of docetaxel without any apparent signs of toxicity. Our findings provide an important theoretical basis for clinical prostate cancer treatment.
