Integration of Activation by Hypoxia and Inhibition Resistance of Tumor cells to Apoptosis for Precise and Augmented Photodynamic Therapy

Yutong Shao,Miaomiao Chen,Wenlong Chen,Zehui Wang,Mengzhang Sui,Mingyu Tian,Yingnan Wu,Jitao Song,Debin Ji,Fengling Song

Photodynamic therapy (PDT) uses photosensitizers to convert oxygen (O 2 ) to reactive oxygen species (ROS) under irradiation to induce DNA damage and kill cancer cells. However, the effect of PDT is usually alleviated by apoptosis resistance mechanism of tumor living cells. MTH1 enzyme is known to be such an apoptosis-resistance enzyme which is over expressed as a scavenger to repair the damaged DNA. In this work, a hypoxia-activated nanosystem FTPA, which can be degraded to release the encapsulated PDT photosensitizer 4-DCF-MPYM and an inhibitor TH588 is proposed. The inhibitor TH588 can inhibit the DNA repair process by reducing the activity of MTH1 enzyme, and achieve the purpose of amplifying the therapeutic effect of PDT. This work demonstrates that a precise and augmented tumor PDT is achieved by integration of hypoxia-activation and inhibition resistance of tumor cells to apoptosis.

Synergistic effects of sulfur-doped carbon dots/permanganate process for DCF degradation: Mechanism and pathways

Danmei Luo,Heng Zhang,Xingxing An,Jia Zhao,Can Feng,Jialong Yin,Mengfan Luo,Tao Wei,Yang Liu,Yanbiao Shi,Jing Zhang,Bo Lai

Extensive research has explored strategies to enhance permanganate (PM) oxidation capacity, with carbon materials having shown significant promise in treating refractory organic pollutants in water. However, the impacts of carbon quantum dots (CQDs) and heteroatom-doped CQDs on PM oxidation remain unclear, warranting further investigation. In this study, we found that sulfur-doped CQDs (SCQDs) significantly outperformed CQDs, nitrogen-doped CQDs (NCQDs), and boron-doped CQDs (BCQDs) in promoting the oxidation reaction of diclofenac sodium (DCF) by PM. Through quenching experiments, light-shielding tests, and UV-Vis spectroscopy, we revealed that the PM/SCQDs system could rapidly and stably generate Mn(III) and MnO 2 , thereby facilitating the removal of DCF. Characterization analysis showed that SCQDs possess a surface enriched with diverse functional groups, and sulfur doping further enhances their electron cloud density. This structural characteristic enhances the ability of SCQDs to donate electrons to permanganate, thereby promoting its reduction to Mn(III) and MnO 2 . A comprehensive evaluation of key operational parameters governing the PM/SCQDs system performance was conducted. Additionally, the PM/SCQDs system exhibited substantial resistance to interference from common matrix ions and natural organic matter in water. The study provides a new perspective on enhancing permanganate oxidation by SCQDs and offers a promising pathway for efficient water pollutant degradation.

Visible light-based photocatalytic ozonation of DCF over CQDs/FeTiO3 heterostructure in a spinning disk photocatalytic reactor

A. Al-Farraji,Saad H. Ammar,Hanan H. Abed,Zaid H. Jabbar

Photocatalytic ozonation is an advanced oxidation process (AOP) that has recently demonstrated its strong oxidation capabilities for efficient wastewater treatment using sunlight and air as the ozone source. Additionally, the spinning disc photocatalytic reactor (SDPR) has been widely adopted recently as an intensification approach for photocatalytic processes. Herein, an efficient approach has been adopted to enhance the degradation performance of photocatalytic ozonation process over CQDs/FeTiO 3 photocatalyst film in a SDPR for synergistically enhancing the degradation of diclofenac sodium (DCF). The DCF degradations were compared in different treatment systems in the SDPR, including ozonation, catalytic-ozonation, photocatalysis, and photocatalytic ozonation. High photocatalytic ozonation performance (99.3% degradation efficiency, 85.8% mineralization, and high stability after six runs) has been acquired over CQDs/FeTiO 3 photocatalyst film under visible-light exposure. The effective separation of photogenerated charge carriers due to CQDs deposition, and high generation of ORS resulting from the combined systems were responsible for the enhanced photocatalytic performance.

Study on the Removal Characteristics of IBP and DCF in Wastewater by CW-MFC with Different Co-Substrates

Ge Qin,Hongyu Feng,Rendong Yu,Fuchao Zheng,Xufei Jiang,Lu Xia,Shuqing An

Pharmaceuticals and Personal Care Products (PPCPs) constitute a group of compounds that are challenging to break down and potentially pose risks to both ecosystems and human health when they accumulate in water bodies. This study established and operated small-scale constructed wetland–microbial fuel cells (CW-MFCs) continuously for 60 days, maintaining a hydraulic retention time (HRT) of 3 days. The research aimed to assess the treatment efficiency of wastewater containing Ibuprofen (IBP) and Diclofenac (DCF) using different co-substrates (glucose, sucrose, and sodium acetate) and to analyze the impact of these co-substrates on the composition of bacterial communities within the CW-MFC. After 60 days of operation, CW-MFC achieved removal rates of 89.29% for IBP and 84.10% for DCF. The elimination of IBP was primarily dependent on co-metabolic degradation processes occurring in both the anode and cathode, while DCF removal relied on anodic co-metabolism. Additionally, various co-substrates have an influence on the bacterial community diversity of the anode and cathodes. The possible bacterial groups involved in PPCP degradation were identified. In summary, Glu was identified as a more suitable co-substrate for CW-MFC in the removal of IBP and DCF, while SA as a co-substrate favored the induction and enrichment of EAB in the anodes. These findings offer valuable insights into the potential of CW-MFC for mitigating emerging contaminants. Keywords:constructed wetlands;constructed wetland–microbial fuel cells;PPCPs;co-metabolic;Ibuprofen;diclofenac

Based on FRET to construct color-tunable ultralong lifetime room temperature phosphorescent carbon dots in aqueous solution

Feng Lu,Xinhuan Xu,Xingdong Zhu,Linxin Shen,Weizheng Wan,Min Hu

Room temperature phosphorescent (RTP) Carbon Dots have been capturing increasing attention in recent years, while building a general method to adjust the emission color of RTP carbon dots is still a big challenge. Herein we report a simple method that combine the carbon nanodots and dyes (R6G and DCF) in SiO 2 nanosphere to get a series of multicolor RTP nanodots (CD@SiO 2 @dye) with long lifetime in aqueous solution. Leverage on chitosan quaternary ammonium as matrix and diethylenetriamine as N-doping resource to form a cross-linked skeleton as a luminescent center (namely CD), and a rigid network is formed by silica encapsulation (CD@SiO 2 ) to restrict the non-radiative transition process to generate the phosphorescence. The CD-based composites, with 1.10 s green (503 nm) phosphorescence emission, serve as activator to stimulate the corresponding luminescence of organic dyes. Then, based on Förster resonance energy transfer (FRET) process from CDs (as donor) to organic dyes (as acceptor) under UV excitation, the CD@SiO 2 @R6G emit ultra-long lifetime (1.13 s) orange-yellow (570 nm) afterglow, and CD@SiO 2 @DCF emit ultra-long lifetime (1.20 s) yellow-green afterglow (530 nm). Furthermore, it also achieves RTP colors control when the ratio of CDs and the dyes changes, the ratio of green emission and dye’s emission activated by CDs will gradually change as well. These kinds of materials keep the inherent advantages of low toxicity and luminous stability, and achieve adjustable RTP color in aqueous solution. Our research provides a strategy to synthesize water-soluble long-life RTP CDs with adjustable color and lifetime.
