NIR light-triggered bursting of double-emulsion drop (DED) for micro-droplets generation

Zhaoze Lu,Jian Yu,Kaihua Wang,Wei Cheng,Likai Hou

Micro-droplets have significant applications in microbiologies, pharmaceuticals, cosmetics, and synthetic materials. Here, we present for the first time a near-infrared (NIR) light-triggered double-emulsion drop (DED) bursting method for generating a large number of micro-droplets in several micron. Under the irradiation of NIR light, the inner water phase of the DED containing a trace amount of Prussian blue (PB) rapidly heats up and evaporates rapidly to generate microbubbles due to the photothermal property of PB. By controlling the light intensity, the DED can inflate itself by coalescing these constant microbubbles to burst immediately and tear the middle oil phase to form a large number of micro-droplets. The performance of micro-droplets generated by NIR light-triggered DED bursting is investigated at different oil shell thickness (HO), oil phase viscosity (ηO) and oil type. HO and ηO are both key factors affecting the generation of micro-droplets. DED with lower HO and ηO can generate lower polydispersity and large quantity of micro-droplets by NIR-triggered DEDs bursting. The proportion of micro-droplets below 10 μm can reach up to 95%. Furthermore, camellia oil, as middle oil phase of DEDs, also can be used to generate lower polydispersity and large quantity of micro-droplets in several micron. The presented bursting method has a great potential to generate micro-droplets for micro/nano and biotechnology applications.

S100A9-TLR4 axis aggravates dry eye through the blockage of autophagy

Lihong Liang,Xue Yang,Hao Zeng,Kai Liao,Runze Zhang,Bowen Wang,Jin Yuan

This research focused on how upregulation of S100A9 contributed to the pathogenesis of the dry eye disease (DED) and whether S100A9 served as a promising therapeutic target in DED. Public single-cell RNA sequencing (scRNA-seq) data of a lacrimal gland excision (LGE) murine DED model was analyzed. LGE model was established and expression of protein was measured through immunofluorescence and western blot. DED-related signs were evaluated through tear secretion and fluorescent staining. TUNEL was performed to detect the level of cell death. Briefly, S100A9 was recognized as a highly variable gene in the DED group. LGE model was successfully established, and S100A9 showed a time-dependent increase in the corneal epithelia. Autophagic blockage was predicted by the scRNA-seq data in DED, and further verified by decrease of LC3B-II/LC3B-I and increase of SQSTM1 and p-mTOR/mTOR, while S100A9 inhibitor paquinimod (PAQ) reversed the changes. PAQ also downregulated TLR4, and inhibition of TLR4 also alleviated autophagic blockage in DED. Finally, signs of DED, chronic corneal inflammation and cell death got a remission after either inhibition of S100A9 or TLR4. In general, we deduced a S100A9-TLR4-Autophagic blockage pathway in the pathogenesis of DED.

Molecular mechanism of Wilms tumor 1-associated protein in diabetes-related dry eye disease by mediating m6A methylation modification of lncRNA NEAT1

Chen Guo,Mingyi Yu,Jinghua Liu,Zhe Jia,Hui Liu,Shaozhen Zhao

Dry eye disease (DED) is often secondary to diabetes mellitus (DM). We explored the action of Wilms tumor 1-associated protein (WTAP) in DM-DED via lncRNA NEAT1 m6A methylation. DM-DED mouse models were treated with sh-WTAP/sh-NEAT1 plasmid, followed by assessment of corneal epithelial damage/histopathological changes. HCE-2 cells were exposed to hyperosmotic conditions to establish in vitro DED models and treated with oe-NEAT1/sh-NEAT1/sh-WTAP/nigericin (an NLRP3 inflammasome inducer). Cell viability/apoptosis were evaluated by CCK-8/TUNEL. The levels of WTAP/NEAT1/inflammatory factors/NLRP3 inflammasome- and apoptosis-related markers were determined. m6A modification was examined by MeRIP-qPCR and NEAT1 stability was also detected. DM-DED mice exhibited up-regulated WTAP/NEAT1 expression and severe corneal damage, whereas WTAP/NEAT1 knockdown alleviated inflammation/corneal damage. In hyperosmolarity-induced HCE-2 cells, NEAT1 aggravated inflammation and apoptosis, while NEAT1 knockdown suppressed NLRP3 inflammasome activation and ameliorated cell injury. Hyperosmolarity-induced WTAP expression increased m6A modification and NEAT1 mRNA stability. WTAP mediated m6A methylation of NEAT1 and NLRP3 inflammasome activation in DM-DED mice. Collectively, WTAP upregulates NEAT1 expression and facilitates NLRP3 inflammasome activation by increasing m6A methylation, thereby accelerating DM-DED development. WTAP or lncRNA NEAT1 may become a therapeutic target for DM-DED, which offers a theoretical foundation for the investigation and advancement of nanocarrier-based targeted therapeutic medications for DM-DED.

21.15%-Efficiency and Stable γ -CsPbI3 Perovskite Solar Cells Enabled by an Acyloin Ligand

Jungang Wang,Yuhang Che,Yuwei Duan,Zhike Liu,Shaomin Yang,Dongfang Xu,Zhimin Fang,Xuruo Lei,Yong Li,Shengzhong (Frank) Liu

Cesium lead triiodide (CsPbI 3 ) is a promising light-absorbing material for constructing perovskite solar cells (PSCs) owing to its favorable bandgap and thermal tolerance. However, the high density of defects in the CsPbI 3 film not only act as recombination centers, but also facilitate ion migration, leading to lower PCE and inferior stability compared with the state-of-the-art organic–inorganic hybrid PSC counterpart. Theoretical analyses suggest that the effective suppression of defects in CsPbI 3 film is helpful for improving the device performance. Herein, the stable and efficient γ -CsPbI 3 PSCs are demonstrated by developing an acyloin ligand (1,2-di(thiophen-2-yl)ethane-1,2-dione (DED)) as a phase stabilizer and defect passivator. The experiment and calculation results confirm that carbonyl and thienyl in DED can synergistically interact with CsPbI 3 by forming a chelate to effectively passivate Pb-related defects and further suppress ion migration. Consequently, DED-treated CsPbI 3 PSCs yield a champion PCE of 21.15%, which is one of the highest PCE among all the reported CsPbI 3 PSCs to date. In addition, the unencapsulated DED-CsPbI 3 PSC can retain 94.9% of itsinitial PCE when stored under ambient conditions for 1000 h and 92.8% of its initial PCE under constant illumination for 250 h.

Cytisine eye drops for benzalkonium chloride-induced dry eye: safety and efficacy evaluation

Zijian Wang,Xixi Song,Yanjun Wei,Xianggen Wu,Ying Jie

This experiment aimed to investigate the feasibility of cytisine (CYT) in treating eye diseases with ocular topical application. An in vitro cytotoxicity test, a hen’s egg test-chorioallantoic membrane (HET-CAM), and a mouse eye tolerance test were used to fully reveal the ocular safety profiles of CYT. For the efficacy evaluations, CYT’s effects on cell wound healing, against H2O2-induced oxidative stress damages on cells, and on benzalkonium chloride (BAC)–induced dry eye disease (DED) in mice were evaluated. Results showed that CYT did not show any cytotoxicities at concentrations no higher than 250 μg/ml, while lipoic acid (α-LA) at 250 μg/ml and BAC at 1.25 μg/ml showed significant cytotoxicities within 48 h incubation. The HET-CAM and mouse eye tolerance test confirmed that 0.5% CYT eye drops demonstrated good safety characteristics. Efficacy evaluations showed that CTY significantly promoted cell migration and wound healing. CYT significantly improved cell survival against H2O2-induced oxidative stress damage by reversing the imbalance between the reactive oxygen species (ROS) and antioxidant defense mechanisms. The animal evaluation of the BAC-induced dry eye model revealed that CYT demonstrated a strong treatment effect, including reversing ocular surface damages, recovering corneal sensitivity, and inhibiting neovascularization; HMGB1/NF-κB signaling was involved in this DED treatment by CTY. In conclusion, CYT had strong experimental treatment efficacy against DED with good ocular safety profiles, and it might be a novel and promising drug for DED.
