The combination of natural antimicrobial peptide BCp12/phenyllatic acid (BCp12/PLA) presents a more efficient antibacterial effect, but its antibacterial mechanism remains unclear. This study studied the synergistic antibacterial mechanism of BCp12 and PLA against S. aureus . The results demonstrated that the BCp12/PLA combination presented a synergistic antibacterial effect against S. aureus , with a fractional inhibitory concentration of 0.05. Furthermore, flow cytometry and scanning electron microscope analysis revealed that BCp12 and PLA synergistically promoted cell membrane disruption compared with the group treated only with one compound, inducing structural cell damage and cytoplasmic leakage. In addition, fluorescence spectroscopy analysis suggested that BCp12 and PLA synergistically influenced genomic DNA. BCp12 and PLA targeted enzymes related to peptidoglycan and DNA synthesis and interacted by hydrogen bonding and hydrophobic interactions with mur enzymes (murC, murD, murE, murF, and murG), dihydrofolate reductase, and DNA gyrase. Additionally, the combined treatment successfully inhibited microbial reproduction in the storage of pasteurized milk, indicating that the combination of BCp12 and PLA can be used as a new preservative strategy in food systems. Overall, this study could provide potential strategies for preventing and controlling foodborne pathogens.

Graphical In this paper, the nanocomposites with dodecahedral structure ZIF-8@ZIF-67 and BBH@ZIF-8@ZIF-67 have been synthesized by one-pot method. Bimetallic MOFs ZIF-8@ZIF-67 exhibited superior antibacterial properties and good biocompatibility. BBH-loaded ZIF-8@ZIF-67 showed enhanced synergistic antibacterial activity. Molecular docking suggests that PBP2a and DHFR may be potential antibacterial targets of BBH. Stimulus-responsive nanocarrier materials have garnered significant attention in biomedical science. Among them, metal-organic frameworks (MOFs) have emerged as promising candidates due to their remarkable antibacterial properties and pH sensitivity. In this study, a pH-responsive bimetallic MOFs carrier, ZIF-8@ZIF-67, was successfully synthesized for the delivery of berberine hydrochloride (BBH). Comprehensive characterization using UV-Vis spectroscopy, FT-IR spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM) confirmed the successful synthesis and structural integrity of ZIF-8@ZIF-67 and BBH@ZIF-8@ZIF-67. Subsequently, the drug release behavior, antibacterial efficacy, and biocompatibility of the composite material were systematically evaluated. Our findings revealed that BBH@ZIF-8@ZIF-67 exhibited well-controlled drug release properties, excellent antibacterial effects and biocompatibility. Furthermore, molecular docking results revealed that penicillin-binding protein 2a and dihydrofolate reductase in S. aureus may serve as two potential antibacterial targets for BBH. This research provides valuable insights into the use of bimetallic MOFs materials for developing highly efficient stimulus-responsive nanocarrier systems in biomedical applications.

Protein immobilization is critical to utilize their unique functions in diverse applications. Herein, we report that orthogonal peptide–protein chemistry enabled multilayer construction can facilitate the incorporation of various folded structural domains, including calmodulin in different states, affibody, and dihydrofolate reductase (DHFR). An extended conformation is found to be the most advantageous for steady film growth. The resulting protein thin films exhibit sensitive and selective responsive behaviors to biosignals, such as Ca2+, trifluoperazine, and nicotinamide adenine dinucleotide phosphate (NADPH), and fully maintain the catalytic activity of DHFR. The approach is applicable to different substrates such as hydrophobic gold and hydrophilic silica microparticles. The DHFR enzyme can be immobilized onto silica microparticles with tunable amounts. The multilayer setup exhibits a synergistic enhancement of DHFR activity with increasing numbers of bilayers and also makes the embedded DHFR more resilient to lyophilization. Therefore, this is a convenient and versatile method for protein immobilization with potential benefits of synergistic enhancement in enzyme performance and resilience.

In this article, three series of dihydrotriazine derivatives bearing a quinoline moiety ( 5a , 5b , 8a – 8c , and 9a – 9m ) have been designed, synthesized, and evaluated as antibacterial agents. Compounds 8a – 8c were found to be the most potent of all of the compounds tested with an MIC value of 1 μg/mL against several Gram-positive ( S. aureus 4220 and MRSA CCARM 3506) and Gram-negative ( E. coli 1924) strains of bacteria. In addition, 3-[4-amino-6-(phenethylamino)-2,5-dihydro-1,3,5-triazin-2-yl)-6-[(3-chlorobenzyl)oxy]quinolin-2-ol ( 8a ) showed potent inhibitory activity (MIC=2 μg/mL) against Pseudomonas aeruginosa 2742, indicating that its antibacterial spectrum is similar to those of the positive controls gatifloxacin and moxifloxacin. Structure-activity relationships (SAR) analyses and docking studies implicated the dihydrotriazine group in increasing the antimicrobial potency of the quinoline compounds. In vitro enzyme study implied that compound 8a also displayed DHFR inhibition. Graphical
