Effects of nitrification inhibitors DCD and DMPP on maturity, N2O and NH3 emissions during manure composting

Xinyuan Ma,Haobo Bai,Guoxue Li,Lingling Li,Haofeng Meng,Yan Liu,Jing Yuan

In order to reduce N 2 O emissions during composting, the effects of different nitrification inhibitors (NI), dicyandiamide (DCD) and 3,4-dimethylpyrazole phosphate (DMPP), on compost maturity, N 2 O, and NH 3 emissions were studied under continuous incremental addition. This study used pig manure and corn straw as composting materials, based on the total nitrogen (TN) content of the initial mixture, two treatments were set: DCD (2.5% in the early phase and 5.0% in the maturation phase) and DMPP (0.25% in the early phase and 0.75% in the maturation phase) in a composting experiment. The results showed that adding DCD and DMPP did not affect the compost maturity, with the seed germination index (GI) of final compost reaching 80.76%–97.06%. Before the maturity period of compost, ammonia (NH 3 ) emissions accounted for 98.5%–99.4% of total emissions. Compared with the control group (CK), the addition of DCD and DMPP in the early stage reduced NH 3 emissions by 8.85% and 12.83%, respectively, by decreasing the ammonification rate. During the mature stage of composting, N 2 O emissions account for 95.6%–98.9% of the total emissions. The addition of DCD and DMPP delayed N 2 O emissions by 4 and 6 days, respectively, through nitrification inhibition. The DMPP amendment also reduced cumulative N 2 O emissions by 54.50% and increased the nitrogen content of the final compost. Correlation analysis showed that N 2 O was mainly originated from the denitrification of nitrification substrate (NO 2 − -N and NO 3 − -N). This study provides technical support for low-carbon management of agricultural waste.

Application of isotope-based carboxy group derivatization in LC–MS/MS analysis of tissue free-fatty acids for thyroid carcinoma

Jiapeng Leng,Qing Guan,Tuanqi Sun,Yi Wu,Yanjing Cao,Yinlong Guo

Free fatty acids (FFAs) analysis of human tissue is challenging due to the low abundance of tissue lipids and interference from the complex matrix. Based on our previously reported on 2,4-dimethoxy-6-piperazin-1-yl pyrimidine (DMPP) derivatization toward carboxy group, we have further applied DMPP derivatization method in the assay of tissue FFAs in patients with thyroid carcinoma after method optimization including sample pretreatment, and derivatization condition. d 6 -DMPP labeled mixture of FFAs standard was used as the internal standard (IS). The d 0 -DMPP labeled samples (spiked with IS) were subsequently used for lipids profiling by selective reaction monitoring (SRM) detection. The DMPP derivatization combined with SRM not only allowed trace analysis of FFAs in complex matrix due to the extremely high sensitivity, but also minimized the errors in quantitation because of the very similar property between d 0 -DMPP derivatized samples and d 6 -DMPP derivatized IS. Eighteen kinds of FFAs were detected successfully with the increased level in carcinoma tissue compared with the normal tissue, which may be the results of abnormality in the metabolism of fatty acid synthases and lipids. Application of the DMPP based FFAs derivatization is expected to provide a promising tool for the investigation of lipids in vivo , especially for the research of lipid metabolism.

The win-win effects of nitrification inhibitors on soil-crop systems: Decreasing carbendazim residues but promoting soil bacterial community diversities and stabilities and crop yields

Tao Guo,Patrick J. O'Connor,Xinlin Zhao,Tangrong Zhou,Yan Wang,Manyun Zhang

Applying nitrogen (N)-cycling inhibitors is an effective measure to improve N fertilizer utilization efficiency, but the effects of N-cycling inhibitors on fungicide residues in soil-crop systems are unclear. In this study, nitrification inhibitors dicyandiamide (DCD) and 3, 4-dimethylpyrazole phosphate (DMPP) and urease inhibitor N -(n-butyl) thiophosphoric triamide (NBPT) were applied into agricultural soils with fungicide carbendazim applications. The soil abiotic properties, carrot yields, carbendazim residues, bacterial communities and their comprehensive relationships were also quantified. Compared to the control treatment, the DCD and DMPP significantly decreased soil carbendazim residues by 96.2% and 96.0%, and the DMPP and NBPT significantly reduced carrot carbendazim residues by 74.3% and 60.3%, respectively. The nitrification inhibitor applications also generated significant and positive effects on carrot yields and soil bacterial community diversities. The DCD application significantly stimulated soil Bacteroidota and endophytic Myxococcota and modified soil and endophytic bacterial communities. Meanwhile, the DCD and DMPP applications also positively stimulated the co-occurrence network edges of soil bacterial communities by 32.6% and 35.2%, respectively. The linear correlation coefficients between soil carbendazim residues and pH, ETSA and NH 4 + -N contents were − 0.84, − 0.57 and − 0.80, respectively. The nitrification inhibitor applications generated win-win effects on the soil-crop systems by decreasing carbendazim residues but promoting soil bacterial community diversities and stabilities and crop yields.

Asymmetries among soil fungicide residues, nitrous oxide emissions and microbiomes regulated by nitrification inhibitor at different moistures

Tangrong Zhou,Zhihong Xu,Shahla Hosseini Bai,Minzhe Zhou,Wenhui Tang,Bin Ma,Manyun Zhang

Carbendazim residue has been widely concerned, and nitrous oxide (N 2 O) is one of the dominant greenhouse gases. Microbial metabolisms are fundamental processes of removing organic pollutant and producing N 2 O. Nitrification inhibitor 3,4-dimethylpyrazole phosphate (DMPP) can change soil abiotic properties and microbial communities and simultaneously affect carbendazim degradation and N 2 O emission. In this study, the comprehensive linkages among carbendazim residue, N 2 O emission and microbial community after the DMPP application were quantified under different soil moistures. Under 90% WHC, the DMPP application significantly reduced carbendazim residue by 54.82% and reduced soil N 2 O emission by 98.68%. The carbendazim residue was negatively related to soil ammonium nitrogen (NH 4 + -N), urease activity, and ratios of Bacteroidetes , Thaumarchaeota and Nitrospirae under 90% WHC, and the N 2 O emission was negatively related to NH 4 + -N content and relative abundance of Acidobacteria under the 60% WHC condition. In the whole (60% and 90% WHC together), the carbendazim residue was negatively related to the abundances of nrf A (correlation coefficient = −0.623) and nrf H (correlation coefficient = −0.468) genes. The hao gene was negatively related to the carbendazim residue but was positively related to the N 2 O emission rate. The DMPP application had the promising potential to simultaneously reduce ecological risks of fungicide residue and N 2 O emission via altering soil abiotic properties, microbial activities and communities and functional genes. Environmental Implication Carbendazim was a high-efficiency fungicide that was widely used in agricultural production. Nitrous oxide (N 2 O) is the third most important greenhouse gas responsible for global warming. The 3, 4-dimethylpyrazole phosphate (DMPP) is an effective nitrification inhibitor widely used in agricultural production. This study indicated that the DMPP application reduced soil carbendazim residues and N 2 O emission. The asymmetric linkages among the carbendazim residue, N 2 O emission, microbial community and functional gene abundance were regulated by the DMPP application and soil moisture. The results could broaden our horizons on the utilizations DMPP in decreasing fungicide risks and N 2 O emission.

A highly sensitive isotope-coded derivatization method and its application for the mass spectrometric analysis of analytes containing the carboxyl group

Jiapeng Leng,Haoyang Wang,Li Zhang,Jing Zhang,Hang Wang,Yinlong Guo

A novel isotope labeling reagent d 0 -/ d 6 -2, 4-dimethoxy-6-piperazin-1-yl pyrimidine (DMPP) has been developed for derivatization toward the carboxyl group based on carbodiimide chemistry for mass spectrometry (MS) analysis. The strengths of this derivatization strategy involve fast labeling (15   s), low chemical background and general access to most carboxylic analytes. This has been demonstrated using a series of compounds containing carboxylic acids, including peptides and proteins. To enhance the MS response of the derivatized analytes, the design of DMPP has been based on integration of the theoretical consideration of high gas-phase hydrogenation capacity and hydrophobicity. In addition, the high abundance product ions at m / z 225 and m / z 231 from d 0 -/ d 6 -DMPP labeled carboxylic acids indicate high efficiency of the gas-phase cleavage induced by the labeling reagent. Quantitative determination of these ions can also be used in single reaction monitoring to achieve extremely high sensitivity toward the target analytes. This has subsequently been used to determine trace free fatty acids in human urine. Furthermore, the DMPP labeled peptides also provide additional sequence information in MALDI–MS/MS because of the formation of sequence-related isotope fragment ions. This DMPP-oriented labeling technique is expected to be a promising tool for the MS detection of many varieties of compounds containing carboxyl groups.

Insight into functional mechanisms of percarbamide and nitrification inhibitors in degrading fungicide residues and shaping microbial communities in soil-plant systems

Yan Wang,Xinlin Zhao,Negar Omidvar,Mengting Liu,Dongsheng Zou,Manyun Zhang

Fungicides and nitrogen (N) fertilizers are essential to maintain plant yield in current intensive agriculture. Percarbamide is a novel type of N fertilizer with strong oxidizing property, and the nitrification inhibitor is widely used in agricultural production. It may be feasible to apply percarbamide and nitrification inhibitor as N management to promote fungicide dissipations in soil-plant system. This study quantified the effects of percarbamide and nitrification inhibitor dicyandiamide (DCD) and 3, 4-dimethylpyrazole phosphate (DMPP) on carbendazim residues, and microbial communities of soil-plant systems, and relationships among carbendazim residues, soil and endophytic microbial communities and plant yields were also comprehensively quantified. Compared with the control, the percarbamide significantly reduced soil carbendazim residues by 29.4% but enhanced the lettuce yield by 28.0%. Soil carbendazim residues were significantly and negatively correlated with the soil total N and NO 3 − -N contents. Soil microbial community structures and co-occurrence networks were more sensitive to N management than their endophytic counterparts. In comparison to the percarbamide alone, the DCD significantly increased the nodes of soil fungal community co-occurrence network which were positively correlated with the plant yield. The DCD outweighed DMPP in increasing the lettuce yield and soil fungal community stability and reshaping soil bacterial community structure. Our study suggested that soil microbial communities were more sensitive to percarbamide and nitrification inhibitor applications than their endophytic counterparts under fungicide pressure and that the DCD outweighed DMPP in reshaping microbial communities. The integrated applications of percarbamide and nitrification inhibitors were promising soil N management strategies to promote fungicide removal and stimulate microbial community in the soil-plant systems.
