miRNA is an effective disease diagnostic biomarker. Developing efficient miRNA in living cells analysis methods, holds significant importance for disease diagnosis and understanding disease progression. Strategies combining DNA nanostructures and enzyme-free amplification have been developed for highly sensitive and rapid imaging of miRNA. However, due to nonspecific amplification of enzyme-free amplification inherent in the probe assembly process and during transfection, these strategies suffer from significant background signals and reduced sensitivity, limiting their further application. To overcome these limitations, we have developed a DNA tetrahedral-based and photocontrolled intramolecular CHA strategy (TCHA-P) to achieve spatiotemporal, highly sensitive, and accurate imaging of miRNA in living cells. In this strategy, the DNA tetrahedron not only serves as a transfection vector but also enhances the local concentration of reactant probes, boosting the reaction kinetics and sensitivity of CHA. Additionally, the light-controlled group (PC-linker) can trigger CHA at selected times and locations, enhancing the spatiotemporal precision of miRNA imaging and ensuring high sensitivity and accuracy by eliminating nonspecific amplification during the assembly and transfection processes. Using miR-10b as a proof-of-concept model, the proposed strategy demonstrated higher signal-to-noise ratio compared to traditional CHA, with a detection limit of 28 pM. Living cell imaging experiments showed that the strategy could effectively differentiate the expression of miR-10b between normal breast cells and breast cancer cells, providing high imaging contrast. The strategy holds promise for broadening the path of nucleic acid amplification-based imaging and is expected to become an effective tool in nucleic acid-based disease diagnosis, treatment, and drug development.

Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) cause severe lung inflammation and damage, compromising respiratory function. Nanomedicine offers hope in controlling this inflammation. DNA nanostructures, as drug carriers, stand out due to their non-toxic, programmable, and precisely controllable traits. However, they face challenges in physiological stability and functionalization as carriers. Here, we introduce TET AG , DNA tetrahedra assembled and functionalized with aminoguanidine without magnesium. Aminoguanidine not only aids tetrahedron assembly but also inhibits the cyclic GMP-AMP synthase and stimulator of interferon (cGAS-STING) pathway, a key inflammatory signal. By integrating STING siRNA into TET AG (TET AG -STING), we achieve synergistic anti-inflammatory effects in ALI/ARDS. Our results show that TET AG offers improved stability and cellular uptake. Importantly, TET AG -STING exhibits potent anti-inflammatory activity in vitro and in a Lipopolysaccharides-induced mouse model following intratracheal administration. This significantly improves lung injury scores and pulmonary functions. These findings underscore aminoguanidine’s therapeutic potential in ALI/ARDS and highlight the promise of DNA nanostructures in medical applications.

Cisplatin is the most widely used anticancer drug, but its side effects limit the maximum systemic dose. To circumvent the side effects, a DNA tetrahedron–affibody nanoparticle was prepared by combination of a DNA chain with cisplatin via interstrand crosslinks or adducts. Each nanocarrier can bind ∼68 molecules of cisplatin. This cisplatin nanoparticle exhibited high selectivity and inhibition for breast cancer HER2 overexpressing cells BT474 and lower toxicity in MCF-7 cells with low HER2 expression. The nano-drug inhibited the growth of BT474 cells by 94.57% at 512 nM (containing 33.3 μM cisplatin), which was higher than that of cisplatin (82.9%, 33.3 μM).

Herein, a bifunctional electrochemical biosensor based on the DNA tetrahedral scaffolds (TDNs) was proposed, OTA@TDNs and AFB1@TDNs were adopted for electrochemical signal output in response to OTA and AFB1 concentration, simultaneously. In order to increase the conductivity of the biosensor, highly porous gold (HPG) was loaded on electrode surface by pulse electrodeposition. Under optimal conditions, the P Fc displayed a linear range with AFB1 concentration between 0.05 ∼ 360 ng·mL −1 with the LOD of 3.5 pg·mL −1 . And the P MB selective and sensitive responses to OTA are achieved with a linear range of 0.05 ∼ 420 ng·mL −1 and a LOD of 2.4 pg·mL −1 . This biosensor has high sensitivity, selectivity and stability for OTA and AFB1 detection in peanut samples. The approach streamlines the experimental procedure, leading to significantly improve the detection efficiency of mycotoxins. Collectively, this method suggest a novel approach for the detection and monitoring of OTA and AFB1 in food sample.

In situ imaging of microRNA (miRNA) content and distribution is valuable for monitoring tumor progression. However, tumor specific in situ imaging remains a challenge due to low miRNA abundance, lack of biological compatibility, and poor specificity. In this study, we designed a DNA tetrahedral framework complex with hairpins (DTF-HP AP ) consisting of an apurinic/apyrimidinic site (AP site) that could be specifically recognized and cleaved by apurinic/apyrimidinic endonuclease 1 (APE1). Efficient and specific in situ imaging of miR-21 in tumors was thus achieved through catalytic hairpin assembly (CHA) reaction. In this study, DTF-HP AP was successfully constructed to trigger the cumulative amplification of fluorescence signal in situ . The specificity, sensitivity and serum stability of DTF-HP AP were verified in vitro , and DTF-HP AP could be easily taken up by cells, acting as a biosensor to detect tumors in mice. Furthermore, we verified the ability of DTF-HP AP to specifically image miR-21 in tumors, and demonstrated its capability for tumor-specific imaging in clinical samples.

Photodynamic therapy (PDT) holds great promise for treating various types of cancer. However, its clinical advancement is hindered by the intrinsic hypoxic tumor microenvironment, which reduces PDT efficacy, and safety concerns related to the biological specificity of photosensitizers. Here we constructed a MnO 2 nanosheet based DNA tetrahedron-photosensitizer (TMPyP4) nanoplatform (TPM), which integration with tumor targeting (aptamer), gene drugs (DNAzyme) and photosensitizer agents for enhanced PDT. Once the TPM entered cancer cells through aptamer targeting, the MnO 2 nanosheet reacts with endogenous acidic and H 2 O 2 to generate Mn 2+ and oxygen (O 2 ), thereby alleviating the tumor hypoxic. Subsequently, TMPyP4 further converts the generated O 2 into singlet oxygen ( 1 O 2 ) under near-infrared light, enhancing PDT efficacy both in vitro and in vivo. Specifically, the generated Mn 2+ activates the DNAzyme and accelerate tumor target mRNA cleavage, resulting in effective PDT-gene synergistic therapy for tumors. This work offers novel perspectives on advancing precise and highly effective gene-PDT systems.

Doxorubicin (DOX) is a clinical chemotherapeutic drug and patients usually suffer from dose-dependent cytotoxic and side effects during chemotherapy process with DOX. Therefore, developing a reliable strategy for DOX analysis in biological samples for dosage guidance during chemotherapy process is of great significance. Herein, a sensitive and selective electrochemical biosensor for DOX detection was designed based on gold nanoparticles (AuNPs) and DNA tetrahedron (TDN) nanoprobe bifunctional glassy carbon electrode that could detect DOX in human serum and cell lysate samples. AuNPs not only could enhance electron transfer efficiency and detection sensitivity, but also could improve the biocompatibility of electrode. TDN nanoprobes were employed as specific DOX bind sites that could bind abundant DOX through intercalative characteristics to contribute to sensitive and selective detection. Under the optimal conditions, the proposed TDN nanoprobes-based DOX biosensor exhibited a wide linear range that ranged from 1.0 nM to 50 μM and a low detection limit that was 0.3 nM. Moreover, the proposed DOX biosensor displayed nice selectivity, reproducibility and stability, and was successfully applied for DOX detection in human serum and cell lysate samples. These promising results maybe pave a way for DOX dosage guidance and therapeutic efficacy optimization in clinic.

In this work, a rigid DNA tetrahedron (TDN) scaffold was synthesized to precisely control the interenzyme distance by randomly anchoring two pairs of horseradish peroxidase (HRP)/glucose oxidase (GOx) at the vertices of TDN, which could not only avoid the drawbacks of poor controllability and biocompatibility from traditional scleroid skeletons, but also overcome the defect of imprecise regulation for interenzyme distance caused by DNA origami. Impressively, by varying the side length of TDN scaffold, the interenzyme distance was precisely regulated, thus, an optimal TDN scaffold with highest catalytic efficiency was acquired and subsequently applied for constructing an ultrasensitive biosensor for DNA detection with a low detection limit of 3 fM. This strategy paved an avenue for developing new reliable scaffold to precisely regulate the catalytic efficiency of enzyme cascade reaction with ultimate applications in bioanalysis and clinical diagnosis.

Nanopipette as a member of solid-state nanopores has been developed to detect a set of biological molecules. Although the size of the nanopipette can be adjusted artificially, the direct detection of small molecules is still challenging, which is limited by the precision of the nanopipette preparation. In this work, the translocation characteristics of DNA tetrahedrons with varying degrees of the assembly were studied to evaluate their self-assembly process and the advantage of tetrahedral DNA nanostructures as a signal amplification label for biomarkers detection based on current blockages. Self-assembly of different number of single-stranded DNA causes the DNA nanostructure in different shapes. Through the complete assembly of DNA tetrahedron, we demonstrate that the nanopore signal-to-noise ratio is significantly increased and the event signal becomes more stable and significant. Therefore, these attributes allow us to gain more insight into the assembly process of DNA nanostructures and we can use tetrahedral DNA nanostructure to develop an effective assay for identifying small molecules with the nanopipette.

G-quadruplex plays roles in numerous physiological and pathological processes of organisms. Due to the unique properties of G-quadruplex (e.g., forming G4/hemin complexes with catalytic activity and electron acceptability, binding with metal ions, proteins, fluorescent ligands, and so on), it has been widely applied in biosensing. But the formation process of G-quadruplex is not yet fully understood. Here, a DNA tetrahedron platform with higher reproducibility, regenerative ability, and time-saving building process was coupled with dual polarization interferometry technique for the real-time and label-free investigation of the specific interaction process of guanine-rich singled-stranded DNA (G-rich ssDNA) and Pb2+. The oriented immobilization of probes greatly decreased the spatial hindrance effect and improved the accessibility of the probes to the Pb2+ ions. Through real-time monitoring of the whole formation process of the G-quadruplex, we speculated that the probes on the tetrahedron platform initially stood on the sensing surface with a random coil conformation, then the G-rich ssDNA preliminarily formed unstable G-quartets by H-bonding and cation binding, subsequently forming a completely folded and stable quadruplex structure through relatively slow strand rearrangements. On the basis of these studies, we also developed a novel sensing platform for the specific and sensitive determination of Pb2+ and its chelating agent ethylenediaminetetraacetic acid. This study not only provides a proof-of-concept for conformational dynamics of G-quadruplex-related drugs and pathogenes, but also enriches the biosensor tools by combining nanomaterial with interfaces technique.

DNA-templated silver nanoclusters (DNA-AgNCs) exhibit infinite application foreground for biosensing due to their fluorescence enhancement properties induced by G-quadruplex (G4). Albeit advanced, their synergistic effects of G4 topologies, and spatial proximity between G4 and DNA-AgNCs remain unclear, which will influence the design of DNA-AgNCs based biosensors and their performance. Herein, the fluorescence enhancement of DNA-AgNCs initiated by three distinct G4 topologies (parallel antiparallel, hybrid) and their distance-dependent behaviors were systematically investigated, using a DNA tetrahedron as a mechanically rigid scaffold to precisely regulate the distance between DNA-AgNCs and G4. We found that parallel G4 structures significantly boosted the fluorescence intensity of DNA-AgNCs, and their closer proximity resulted in elevated fluorescence, which were ascribed to a denser surrounding electron environment that facilitates electron transfer to nearby AgNCs, achieving fluorescence enhancement. Consequently, after encoding a specific aptamer into the reconfigurable edge of the DNA tetrahedron, a novel biosensor was fabricated as a proof-of-concept for rapid detection of aflatoxin B1 with a detection limit of 0.068 ng/mL. Our strategy offered some new insights for rational bioassay design and its performance improvement.

Background Assembling framework nucleic acid (FNA) nanoarchitectures and tuning luminescent quantum dots (QDs) for fluorescence assays represent a versatile strategy in analytical territory. Rationally, FNA constructs could offer a preferential orientation to efficiently recognize the target and improve detection sensitivity, meanwhile, regulating size-dependent multicolor emissions of QDs in one analytical setting for ratiometric fluorescence assay would greatly simplify operation procedures. Nonetheless, such FNA/QDs-based ratiometric fluorescence nanoprobes remain rarely explored. Results We designed a sensitive and signal amplification-free fluorescence aptasensor for lead ions (Pb 2+ ) that potentially cause extensive contamination to environment, cosmetic, food and pharmaceuticals. Red and green emission CdTe quantum dots (rQDs and gQDs) were facilely prepared. Moreover, silica nanosphere encapsulating rQDs served as quantitative internal reference and scaffold to anchor a predesigned FNA and DNA sandwich containing Pb 2+ binding aptamer and gQD modified DNA signal reporter. On binding of Pb 2+ , the gQD-DNA signal reporter was set free, resulting in fluorescence quenching at graphene oxide (GO) interface. Owing to the rigid structure of FNA, the fluorescence signal reporter orderly arranged at the silica nanosphere could sensitively respond to Pb 2+ stimulation. The dose-dependent fluorescence signal-off mode enabled ratiometric analysis of Pb 2+ without cumbersome signal amplification. Linear relationship was established between fluorescence intensity ratio ( I 555 / I 720 ) and Pb 2+ concentration from 10 nM to 2 μM, with detection limit of 1.7 nM (0.43 ppb), well addressing the need for Pb 2+ routine monitoring. The designed nanoprobe was applied to detection of Pb 2+ in soil, cosmetic, milk, drug, and serum samples, with the sensitivity comparable to conventional ICP-MS technique. Significance Given the programmable design of FNA and efficient recognition of target, flexible tuning of QDs emission, and signal amplification-free strategy, the present fluorescence nanoprobe could be a technical criterion for other heavy metal ions detection in a straightforward manner.
