Magnetic Microsphere/Silica Nanoparticle Composite Structures for Switchable DNA Storage

Jing-Yuan Ma,Kai-Xuan Wang,Ming Ma,Yu Zhang

As an emerging storage medium, DNA is a crucial carrier for future data storage. DNA has a better storage density and a longer storage life than conventional storage media. However, DNA’s key limitations as a storage medium are the precision of information reading and storage stability. Herein, we designed and successfully developed a switchable DNA storage nanocomposite structure (MM-DNA/DNA’-SiO2). The composite structure employs magnetic microspheres (MMs) based on Fe3O4 nanoparticles as the DNA carrier for storing information (MM-DNA) and silicon dioxide nanoparticles modified with complementary strands as the protective outer layer (DNA’-SiO2), and its switchability and DNA sequence reading accuracy were both validated in this study. This method provides a novel nanostructure medium and technical proposal for efficient DNA storage with potential application value.

Magnetic Bead Spherical Nucleic Acid Microstructure for Reliable DNA Preservation and Repeated DNA Reading

Peng Shen,Xiaojun Qu,Qinyu Ge,Ting Huang,Qingjiang Sun,Zuhong Lu

DNA is an attractive medium for long-term data storage because of its density, ease of copying, sustainability, and longevity. Recent advances have focused on the development of new encoding algorithms, automation, and sequencing technologies. Despite progress in these subareas, the most challenging hurdle in the deployment of DNA storage remains the reliability of preservation and the repeatability of reading. Herein, we report the construction of a magnetic bead spherical nucleic acid (MB-SNA) composite microstructure and its use as a cost-effective platform for reliable DNA preservation and repeated reading. MB-SNA has an inner core of silica@γ-Fe2O3@silica microbeads and an outer spherical shell of double-stranded DNA (dsDNA) with a density as high as 34 pmol/cm2. For MB-SNA, each strand of dsDNA stored a piece of data, and the high-density packing of dsDNA achieved high-capacity storage. MB-SNA was advantageous in terms of reliable preservation over free DNA. By accelerated aging tests, the data of MB-SNA is demonstrated to be readable after 0.23 million years of preservation at −18 °C and 50% relative humidity. Moreover, MB-SNA facilitated repeated reading by facile PCR-magnetic separation. After 10 cycles of PCR access, the retention rate of dsDNA for MB-SNA is demonstrated to be as high as 93%, and the accuracy of sequencing is more than 98%. In addition, MB-SNA makes cost-effective DNA storage feasible. By serial dilution, the physical limit for MB-SNA to achieve accurate reading is probed to be as low as two microstructures.

Transformable and stimuli-responsive liquid metal for integrated, sustainable, and biomimetic DNA-based data storage

Yakun Gao,Biao Ma,Gangsheng Chen,Chengtao Xu,Ziyan Kong,Yanjie Chen,Chao Zhao,Duxin Chen,Wenwu Yu,Hong Liu

DNA data storage has emerged as a promising solution to address the challenges of data explosion due to its high storage density and durability. However, traditional storage architectures require substrate replacement and the use of environmentally unfriendly chemicals for multistep operations, hindering seamless and sustainable storage. Here, we report liquid metal (LM)-DNA, an integrated and sustainable DNA storage architecture that leverages a responsive liquid metal as a functionality-adaptive storage medium. LM-DNA allows cascaded storage operations within a single medium, including enzymatic DNA synthesis, hermetic encapsulation through thermal activation, and electrokinetic decapsulation driven by the electrocapillary effect. The green enzymatic DNA data writing and physical data encapsulation/decapsulation avoid toxic chemicals, ensuring a more sustainable route for DNA storage. Furthermore, the fluidity of liquid metal enables biomimetic operations such as file splitting, merging, and reshaping, offering unprecedented flexibility in information management. LM-DNA provides a promising foundation for advancing sustainable and biomimetic information storage technologies.

Engineered Living Memory Microspheroid-Based Archival File System for Random Accessible In Vivo DNA Storage

Hao Luo,Wen Huang,ZhongHui He,Yongcong Fang,Yueming Tian,Zhuo Xiong

Given its exceptional durability and high information density, deoxyribonucleic acid (DNA) has the potential to meet the escalating global demand for data storage if it can be stored efficiently and accessed randomly in exabyte-to-yottabyte-scale databases. Here, this work introduces the Engineered Living Memory Microspheroid (ELMM) as a novel material for DNA data storage, retrieval, and management. This work engineers a plasmid library and devises a random access strategy pairing plasmid function with DNA data in a key-value format. Each DNA segment is integrated with its corresponding plasmid, introduced into bacteria, and encapsulated within matrix material via droplet microfluidics within 5 min. ELMMs can be stored at room temperature following lyophilization and, upon rehydration, each type of ELMM exhibits specific functions expressed by the plasmids, allowing for physical differentiation based on these characteristics. This work demonstrates fluorescent expression as the plasmid function and employs fluorescence-based sorting access image files in a prototype database. By utilizing N optical channels, to retrieve 2 N file types, each with a minimum of 10 copies. ELMM offers a digital-to-biological information solution, ensuring the preservation, access, replication, and management of files within large-scale DNA databases.
