Nitrogen doped chiral carbonaceous nanotube for ultrasensitive DNA direct electrochemistry, DNA hybridization and damage study

Mengjing Cui,Qiuyue Zhao,Qi Zhang,Mingxuan Fu,Yuexian Liu,Xinyu Fan,Haiyang Wang,Yufan Zhang,Huan Wang

In the interest of developing novel electrocatalyst for high performance DNA biosensing, with distinctive chiral double helix nanostructure , nitrogen doped chiral carbonaceous nanotube (Chiral-CNT) was employed for ultrasensitive label-free DNA biosensing research. Chiral-CNT can quantitative detection of four DNA bases with high sensitivity and selectivity . Without any prehydrolysis and labeling process, direct electrochemistry of single-stranded DNA and double-stranded DNA, qualitative and quantitative detection of DNA hybridization (low detection limit: 0.0268 g L −1 ) were realized. Moreover, sensitive detection of DNA damage induced by fenton reagent was also realized with low detection limit of 0.0350 mg mL −1 and high sensitivity of 7.42 μA mg −1  mL. The high biosensing performance attributes to the unique chiral structure of Chiral-CNT, leads to efficient interreaction between Chiral-CNT and DNA molecule.

A Novel DNA Electrochemical Sensor Based on Grafting of L-Aspartic Acid and Cu2+ Ions on the Terminal of Molecule Beacons

Xia Wang,Xuan Zhang,Cai Gao,Xiaolei Liao,Le Hang,Shulian Jiang,Feng Gao,Lizhang Huang,Qingxiang Wang

A novel electrochemical DNA biosensor based on in-situ labeling of L -Aspartic acid-copper ion (Asp-Cu 2+ ) complex on the terminal of molecule beacons (MBs) was constructed. The MB with bifunctional groups of 3’–SH and 5’–COOH was first anchored on a gold electrode surface through Au-S assembly chemistry, then L -Aspartic acid (Asp) was grafted with the 5’–COOH of MB via succinimide coupling, which was used as a platform molecule for the following coordination assembly of copper ions (Cu 2+ ). Therefore an electroactive MBs probe with the copper complex (Asp-Cu 2+ ) as a signal source was obtained. The hybridization between the loop sequence and the target DNA transferred the hairpin-like MBs to the linear form, and the electroactive moiety of Asp-Cu 2+ was forced to leave away from the electrode surface, resulting in the reduction of the electrochemical response of the biosensor. The hybridization experiment results showed that the oxidation peak currents of developed biosensor attenuated linearly with the logarithmic values of the target DNA concentrations in the kinetic range from 1.0×10 -15 ~1.0×10 -10 M. The detection limit was estimated to be 0.17 fM. Moreover, the biosensor showed excellent selectivity for discriminating the complementary sequences from the mismatched and noncomplementary sequences.

Construction of self-signal DNA electrochemical biosensor employing WS2 nanosheets combined with PIn6COOH

Jimin Yang,Lei Gao,Cheng Peng,Wei Zhang

In this work, a novel self-signal DNA electrochemical biosensor was constructed based on tungsten disulfide (WS2) nanosheets combined with poly(indole-6-carboxylic acid) (PIn6COOH) as the sensing interface. The WS2 nanosheets were synthesized via a simple solvent exfoliation method from bulk WS2, and then PIn6COOH was electropolymerized on the WS2 nanosheet-modified carbon paste electrode to obtain a unique nanocomposite. The electropolymerization efficiency was remarkably improved, ascribed to the physical adsorption between WS2 nanosheets and aromatic In6COOH monomers, resulting in an increase of the electrochemical response of PIn6COOH. Owing to the presence of π–π interactions between the conjugated PIn6COOH/WS2 nanocomposite and DNA bases, the probe ssDNA was noncovalently assembled on the nanocomposite substrate. After the hybridization of the probe ssDNA with the target DNA, the formation of the double-helix structure induced the resulting dsDNA to be released from the surface of the conjugated nanocomposite, accompanied with the self-signal regeneration of the nanocomposite (“signal-on”). The constructed PIn6COOH/WS2 nanocomposite was not only employed as an interface for DNA immobilization but also reflected the signal transduction stemming from DNA immobilization and hybridization without any external indicators or complex labeling processes. A detection limit of 2.3 × 10−18 mol L−1 has been estimated and a dynamic range of 1.0 × 10−17 mol L−1 to 1.0 × 10−11 mol L−1 has been shown for the detection of a PIK3CA gene related to lung cancer. Selectivity of the biosensor has been researched in the presence of noncomplementary and base mismatched DNA sequences.

DNA electrochemical biosensor based on thionine-graphene nanocomposite

Limei Zhu,Liqiang Luo,Zhenxin Wang

A novel protocol for development of DNA electrochemical biosensor based on thionine-graphene nanocomposite modified gold electrode was presented. The thionine-graphene nanocomposite layer with highly conductive property was characterized by scanning electron microscopy, transmission electron microscopy, cyclic voltammetry and electrochemical impedance spectroscopy. An amino-substituted oligonucleotide probe was covalently grafted onto the surface of the thionine-graphene nanocomposite by the cross-linker glutaraldehyde. The hybridization reaction on the modified electrode was monitored by differential pulse voltammetry analysis using an electroactive intercalator daunomycin as the indicator. Under optimum conditions, the proposed biosensor exhibited high sensitivity and low detection limit for detecting complementary oligonucleotide. The complementary oligonucleotide could be quantified in a wide range of 1.0   ×   10 −12 to 1.0   ×   10 −7   M with a good linearity ( R 2   =   0.9976) and a low detection limit of 1.26   ×   10 −13   M (S/N   =   3). In addition, the biosensor was highly selective to discriminate one-base or two-base mismatched sequences.

Fabrication of DNA electrochemical biosensor based on gold nanoparticles, locked nucleic acid modified hairpin DNA and enzymatic signal amplification

Xiaomeng Meng,Minrong Xu,Jianying Zhu,Huanshun Yin,Shiyun Ai

In this paper, a novel sensitive biosensor was fabricated for DNA detection based on gold nanoparticles (AuNPs) locked nucleic acid modified hairpin DNA probe (LNA-m-HpDNA) and enzymatic signal amplification. LNA-m-HpDNA dually labeled with 3′-thiol and 5′-biotin was efficiently assembled on the electrode via the interaction of thiol and gold. The stem-loop structure of LNA-m-HpDNA was unfolded after hybridization with target DNA, which forced biotin away from the electrode surface. Streptavidin-labeled horseradish peroxidase (Streptavidin-HRP) was subsequently immobilized on the electrode surface via the specific conjugation of biotin and streptavidin. Thereafter, the resulting HRP/target DNA–LNA-m-HpDNA (cDNA–LNA)/AuNPs modified Au electrode was successfully assembled for the construction of the DNA biosensor. Cyclic voltammetry and electrochemical impedance spectroscopy were carried out for the characterization of modified electrodes. Electrochemical reduction signal of benzoquinone was investigated in this work to indirectly analyze DNA hybridization via chronoamperometry. Under optimum conditions, the biosensor showed a good linear relationship between the current value and logarithm of the target DNA concentration ranging from 10 to 1000   pM with a low detection limit of 6.0   pM (S/N   =   3). Furthermore, the DNA biosensor exhibited excellent discrimination ability to detect single-base mismatched DNA, three-base mismatched DNA and non-complementary DNA sequence. This strategy could provide a new platform for disease diagnose with high sensitivity.
