Arch-cruciform DNA are self-assembled on AuNPs/VS2scaffold as a highly sensitive and selective electrochemical biosensor for michigan cancer foundation-7 (MCF-7) breast cancer cells. In the construction, arch DNA is formed using two single-strand DNA sequences embedded with the aptamer for MCF-7 cells. In the absence of MCF-7 cells, a cruciform DNA labeled with three terminal biotin is bound to the top of arch DNA, which further combines with streptavidin-labeled horseradish peroxidase (HRP) to catalyze the hydroquinone-H2O2reaction on the electrode surface. The presence of MCF-7 cells can release the cruciform DNA and reduce the amount of immobilized HRP, thus effectively inhibiting enzyme-mediated electrocatalysis. The electrochemical response of the sensor is negatively correlated with the concentration of MCF-7 cells, with a linear range of 10~1 × 105cells/mL, and a limit of detection as low as 5 cells/mL (S/N = 3). Through two-dimensional materials and enzyme-based dual signal amplification, this biosensor may pave new ways for the highly sensitive detection of tumor cells in real samples.Keywords:MCF-7 cells;electrochemistry;2-D materials;signal amplification;DNA assembly

Photothermal therapy (PTT) has received increasing attention for treating tumors. However, a long-standing challenge in PTT is non-uniform distribution of photothermal agents (PAs) in tumor tissues, resulting in limited therapeutic efficiency. Herein, inspired by dandelions blowing away by the wind, we have designed a DNA-assembled visible GRS-DNA-CuS nanodandelion, which can achieve uniform intra-tumor distribution (UITD) of PAs, thus enhancing the photothermal therapeutic efficiency. GRS-DNA-CuS is featured by the formation of hydrogen bond between the core of single-strand DNA-modified Raman nanoprobes (GRS) and the shell of complementary single-strand DNA-modified CuS PAs. Under Raman imaging-guided 1st NIR irradiation, hydrogen bond in GRS-DNA-CuS is explosively broken, resulting in large-sized GRS-DNA-CuS (∼135 nm) be completely dissociated into GRS and ultra-small CuS PAs (∼12 nm) within 1 min. Such an explosive dissociation instantly enhances the local concentration of ultra-small CuS PAs and slightly rises intra-tumor temperature, thus increasing the diffusion coefficient of PAs and promoting their UITD. This UITD of CuS PAs enhances the photothermal anti-tumor effects. Three out of five tumors are completely eliminated under photoacoustic imaging-guided 2nd NIR irradiation. Overall, this study provides one UITD-guided PTT strategy for highly effective tumor treatment by exerting explosive breakage property of hydrogen bond, broadening the application scope of DNA-assembly technique in oncology field.

Here, we designed a catalytic DNA assembly (CDA) amplification strategy to construct a photoelectrochemical (PEC) assay to detect the specific target F3L-gene of Monkeypox Virus (MPXV). Graphdiyne (GDY) and CuSe were synthesized by a surface-assisted deprotection/polymerization method and hydrothermal method, respectively. Then AuNPs and GDY/CuSe nanocomposites were modified on the surface of gold electrode (GE). The hairpin DNA4 (H4) was incubated onto the surface of the electrode by Au-S bond. When the target was present, the CDA was occurred by hairpin DNA1 (H1) and hairpin DNA2 (H2). Then, DNA3 (H3) hybridized with the single stranded region of H1-H2 complex, formed a site that can be hydrolyzed by exonuclease III, led to exonuclease III hydrolysis of DNA and to release the H3 primer (H3P). H3 further opened H4 and triggered a subsequent CDA reaction, ultimately formed a Y-shaped DNA structure on the electrode surface with the help of DNA5 (H5) and DNA6 (H6). The two end sequences of H4 and H6, which constituted the Y-shaped structure, were designed as split H3P fragments. They can be used as primers to trigger CDA reactions for more efficient and characteristic autocatalytic signal amplification. The Y-shaped structure carried H5-ALP, and catalyzed phosphonoxyphenol ( o -PP) to generate benzene-1,2-diol (CA), which could enhance the PEC signal. The linear range is 0.1 fM-10.0 pM with the detection limit of 0.05 fM (3S/N) for MPXV.

DNA adsorption and recognition are selective for carbon nanotubes (CNTs). In this work, we reported a novel method for control of electrochemiluminescence (ECL) enhancement through selective assembly of CNTs with DNA on indium tin oxide (ITO) electrode. The successful assembly of CNTs with well-preserved electrochemical conductivity was confirmed by scanning electron microscope and electrochemical impedance spectroscopy measurements. In comparison with the bare ITO electrode, the presence of CNTs catalyzes electrochemical reaction of an ECL coreactant, tri- n -propylamine, present in the solution, producing significantly enhanced ECL signal and ensuring substantial signal amplification. It is demonstrated that the efficiency of ECL enhancement is inversely correlated with the DNA recognition reaction, offering a promising ECL sensing platform for label-free and universal bioassay. The proposed strategy definitely differs from those works previously reported the introduction of CNTs for ECL enhancement, allowing the rise of ECL enhancement to be designed based on a biological recognition process. Just instead of the capture probe sequences, it is confirmed that the assembly of DNA with CNTs enables quantitative analysis of the different types of targets, including nucleic acid, Hg 2+ and thrombin.

Upconversion nanoparticles (UCNPs) have attracted increasing interest owing to their excellent properties in chem/bio sensing. And DNA is the most frequently used biomolecule to functionalize the nanoparticles. However, there is still an urgent need for an effective and convenient strategy to assemble DNA strands on UCNPs. Herein, we utilized polydopamine (PDA) to coat hydrophobic UCNPs by a water-in-oil microemulsion method, which were then assembled with DNA through hydrogen bonding and π–π stacking interactions between the nucleotides of DNA and the aromatic groups of the PDA shell on the surface of the UCNPs. This strategy exhibited several merits such as decreasing the random coordination between phosphates of the DNA backbone and lanthanide ions of the UCNP surface and retaining the intrinsic DNA biorecognition functions for targets. A proof-of-concept biosensor for carcinoembryonic antigen (CEA) based on luminescence resonance energy transfer (LRET) was constructed using CEA aptamer-stabilized silver nanoparticles (Apt-AgNPs) as the energy acceptor. Due to the strong affinity between CEA and its aptamer, the upconversion luminescence was recovered in a [CEA]-dependent manner. This facile assembly strategy can be extended to various targets and sensors by altering the aptamer.

The target-triggered DNA assembling probe is presented for highly selective protein detection. Target-triggered DNA assembling is used in an amplification strategy based on affinity binding for identification and determination of proteins in general. Specifically, it was applied to the platelet derived growth factor-BB (PDGF-BB). A hairpin DNA (H-DNA) probe was designed containing (a) an aptamer domain for protein recognition and (b) a blocked DNAzyme domain for DNAzyme cleavage. An assistant DNA (A-DNA) probe containing aptamer and complementary domains was also employed to recognize protein and to induce DNA assembly. Once H-DNA and A-DNA recognize the same protein, H-DNA and A-DNA are in close proximity to each other. This induces DNA assembling for protein-triggered complex (Protein-Complex) with free DNAzyme domains. The free DNAzymes trigger the circular cleavage of molecular beacons for amplified signals. The assay is performed by fluorometry at an excitation wavelength of 980 nm and by collecting fluorescence at 545 nm. The platelet derived growth factor-BB (PDGF-BB) was accurately identified and selectively determined by this assay with a 22 pM detection limit (using the 3 σ criterion). The responses for PDGF-BB is nearly 6-fold higher than for PDGF-AB, and 16-fold higher than PDGF-AA. This upconversion assay avoids any interference by the autofluorescence of biological fluids.
Schematic representation of the principle of the target-triggered DNA assembling probes mediated amplification strategy based on affinity binding for PDGF-BB. The UCNP probe is used for the quantitation of PDGF-BB with high selectivity.

Silicon, as the second richest element in the earth, is of great importance to the current nanoscience. Functionalization of silica nanomaterials with DNA molecules can significantly broaden their biological application. However, current assembly methods usually involve complex and time-consuming steps or detrimental reagents, limiting their real application. In this work, we report a metal ion-assistant DNA assembly method to simply, rapidly and robustly immobilize short DNA molecules on the surface of silica nanoparticles (SiNPs) and use it to perform different tasks. By fully screening the metal ions, metal ions such as Zn 2+ , Al 3+ , Y 3+ and Zr 4+ , are found to be effective on the mediation of the interaction between DNA and SiNPs. The DNA density can be finely tuned by adjusting the added concentration of DNA or metal ions, and the adsorbed DNA is highly stable in the metal ion-containing buffer. Through simply adjusting the kind of metal ions and DNA, different biological applications such as biosensing, purification, and gene regulation can be achieved. This work not only offers fundamental insights into the biointerface science of SiNPs with DNA, but holds great potential in the area of biomolecule purification, nucleic acid delivery, biosensing, and bioimaging.

A novel and enzyme-free electrochemical sensor was designed for multiplexed DNA detection based on a surface-confined DNA assembly enabled target recycling amplification. In this designed system, two hairpin DNA probes (hairpin 1 and hairpin 2) were immobilized on the surface of gold nanoparticles/graphene/nafion modified glassy carbon electrode (AuNPs/Gr/Nf/GCE). In the presence of two DNA targets, the hairpin structures of hairpin 1 and hairpin 2 opened through the complementary hybridization, resulting in the formation of hairpin 1: target DNA 1 and hairpin 2: target DNA 2. When they met methylene blue (MB)-labeled hairpin 3 (hairpin 3-MB) and ferrocene (Fc)-labeled hairpin 4 (hairpin 4-Fc), DNA cyclic amplification was activated, and the DNA targets were released and could be reused for the next recycling. Meanwhile, coupled with the utilization of positively charged gold nanoparticles ((+)AuNPs), multiplexed DNA detection was realized in the concentration range from 10 fM to 1.5 pM with the detection limits of 3.6 fM and 2.5 fM for target DNA 1 and target DNA 2, respectively. Moreover, satisfactory results were obtained when detecting DNA targets in human serum samples, suggesting that this proposed sensing platform is promising for the applications in complex biological samples.

Kanamycin (KAN) and tobramycin (TOB), as aminoglycoside antibiotics, can induce irreversible ototoxicity and dose-dependent nephrotoxicity risks to human health through residual contaminations. Consequently, sensitive monitoring of TOB and KAN residues in animal-derived foods is of critical importance; however, methods that can enable simultaneous detection of these two antibiotics have been rarely reported. This work demonstrates an ultrasensitive and multiplexed electrochemical aptasensor for simultaneous sensing of KAN and TOB via catalytic gold nanoparticles (AuNPs)/reduced graphene (rGO) nanocomposites and cascaded DNA assembly signal amplifications. Upon binding with their corresponding aptamers in duplex probes, KAN and TOB trigger initiator strands liberation to trigger cascade hybridization DNA assemblies, which enrich numerous auxiliary probes labeled with distinct electroactive reporters, methylene blue (MB) and ferrocene (Fc), on AuNPs/rGO-modified electrode surface. Subsequent electro-catalytic oxidation of MB and Fc yields markedly amplified and well-resolved redox currents, leading to highly sensitive aptamer-based assay of KAN and TOB in dynamic range of 0.1–300 nM with detection limits of 42.14 pM (KAN) and 58.42 pM (TOB). Such multiplexed sensor also enables the monitoring of low-level antibiotic residues in milk matrices, thereby providing promising potentials for the construction of sensitive aptasensors for convenient and simultaneous detection of various antibiotic residues.

Breast cancer is the most common malignant disease among women worldwide. Nowadays, combined therapy against several therapeutic targets is becoming a promising treatment to enhance the survival rate of the patients with some lethal subtypes, and also proposes high demand on the discrimination of the co-existing targets in breast cancer. In this work, we designed in situ automatous DNA assembly reaction and applied it for the simultaneous identification of dual therapeutic targets using electrochemical techniques . Taking triple-negative breast cancer cell MDA-MB-231 as a model, chained strand displacement reactions were initiated after the capture probes recognized the surface biomarkers, epidermal growth factor receptor and intercellular adhesion molecule-1, respectively. Then, an increased electrochemical signaling was created to reveal the co-expression of the two targets using quantum dots as electrochemical labeling. Electrochemical results demonstrated high sensitivity and specificity of our method toward the identification of the coexisted therapeutic targets even in the serum samples, which also allowed to monitor the enhanced efficiency of combined therapy. Therefore, our method suggested a potential use in the accurate identification of therapeutic targets in breast cancer that might provide more information to facilitate the combined therapy in clinic.

Dolastatin derivatives possess excellent anticancer activity and have been translated into clinical trials for cancer therapy. Drug delivery systems enable dolastatin derivatives to break the limitation of instability during blood circulation and ineffective cell internalization in the application. Nevertheless, their potential has not been thoroughly established because of the limited loading efficacy and complicated chemical modification. Herein, we rationally propose a rolling circle amplification-based polymer–DNA assembled nanoflower for targeted and efficient delivery of dolastatin-derived drugs to achieve efficient anticancer therapy. The polymer–DNA assembled nanoflower with targeted aptamer conjugate is widely applicable for loading dolastatin-derived drugs with high encapsulation efficiency. The developed monomethyl auristatin E (MMAE) loaded PN@M exhibited increased cellular uptake and enhanced inhibitory effect, especially in multidrug-resistant tumor cells. The results of in vivo anticancer effects indicate that nanoflower as a dolastatin derivatives delivery system holds considerable potential for the treatment of malignant cancer.
