Patulin (PAT) is an unsaturated lactone mycotoxin primarily produced by Penicillium expansum and Aspergillus clavatus . Given the potential health risks and economic losses associated with PAT, the rapid detection of PAT using fluorescent aptasensors is of significant importance in evaluating food safety. However, it easily increases the cost and complexity caused by signal labeling. We combined TCPP/BDC-NH 2 mixed ligands functionalized Zr metal–organic frameworks (Zr-MOF mix ) and terminated three-stranded DNA gates (ttsDNA gates) to fabricate a label-free fluorescent aptasensor for PAT detection. The Zr-MOF mix system was synthesized via a one-pot strategy and could be used to address the problem of pore size limitation and increase the loading amounts of dyes. TtsDNA gate was integrated into the Zr-MOF mix system to control the release of dyes, exhibiting a high signal-to-background ratio. The single-stranded aptamer region in ttsDNA gate situated away from the surface of the Zr-MOF mix , resulting in a natural release of dyes in the absence of PAT. While binding to PAT resulted in target-induced conformational changes that helped form the hairpin structure of the aptamer. This structure hindered the release of dyes from the pores of Zr-MOF mix , thus reducing the fluorescence signals intensity. The stimuli-responsive DNA-gated material provides a platform for PAT analysis under conditions of a low limit of detection (0.871 pg/mL). Furthermore, the excellent specificity and anti-interference of the fluorescent aptasensor make the system suitable for the analysis of apple juice samples. This label-free strategy is cheaper and simper compared with labeled detection, especially for the development of multi-target-detection.

Novel DNA-gated mesoporous silica nanoparticle (MSN) vehicles functionalized with disulfide-linked acridinamine intercalators are constructed for multi-responsive controlled release. The DNA-gated MSN vehicles release cargo encapsulated in the MSN pores under different stimuli, including disulfide reducing agents, elevated temperature, and deoxyribonuclease I (DNase I), for codelivery of drugs and DNA/genes in different forms. Furthermore, the cascade release of encapsulated and intercalative drugs is controlled by AND logic gates in combination of dual stimuli. The ingeniously designed DNA-gated MSN vehicles integrates multiple responses and AND logic gate operations into a single smart nanodevice not only for codelivery of drugs and DNA/genes but also for cascade release of two drugs and has promising biological applications to meet diverse requirements of controlled release.

This study investigates three metal–organic frameworks (MOFs) with distinct Brunauer–Emmett–Teller (BET) surface areas and pore sizes: MOFZr (1274 m2/g, <0.7 nm), MOFAl (371 m2/g, 1.5 nm), and MOFCr (1917 m2/g, 2 nm). Methylene blue (MB, 1.4 × 0.7 × 0.2 nm3) and triferrocene (tri-FC, 1.2 × 0.9 × 0.3 nm3) were adsorbed onto these MOFs. Specific DNA segments targeting SARS-CoV-2 (PR and PO) were employed to block the pores of the MOFs, leading to the formation of DNA-gated MOFs: MOFX/MB/PR and MOFX/tri-FC/PO. These constructs were subsequently integrated with four-arm poly(ethylene glycol) amine (4-arm-PEG-NH2)-modified gold electrodes to create the corresponding sensors (MOFX/MB/PR+MOFX/tri-FC/PO@4-arm-PEG-NH2@Au NPs@GCE). The DNA-gated MOFAl system exhibited the highest loading capacity and a 3-fold increase in sensing efficiency following the introduction of SARS-CoV-2 target DNA, surpassing the performance of the other systems. This study highlights the enhancement of the DNA-gated MOFs when the three-dimensional structures of the guest molecules closely align with the pore sizes of the MOFs. It emphasizes a critical aspect of the traditional design approach for electrochemical sensors based on MOF encapsulation, which often prioritizes BET surface areas while neglecting the compatibility between the sizes of guest molecules and MOF pore diameters. Moreover, the sensor effectively discriminated between SARS-CoV-2 and SARS-CoV by precisely aligning the SARS-CoV-2 target DNA with PR and PO. In contrast, the specific genetic targets (SR) from SARS-CoV showed complete mismatches with PO and a three-base deviation with PR. This differentiation was achieved in a simple one-step assay, even in 5% serum, across a linear concentration range of 10–9 to 10–14 M. This range signifies an expansion of 2 to 3 orders of magnitude compared to sensors that inaccurately selected MOFs.

In this paper, polyethylenimine (PEI) and Chitosan were simultaneously one-step doped into silicon dioxide (SiO 2 ) nanoparticles to synthesize PEI/Chitosan/SiO 2 composite nanoparticles. The polymer PEI contained a large amount of amino groups, which can realize the amino functionalized SiO 2 nanoparticles. And, the good pore forming effect of Chitosan was introduced into SiO 2 nanoparticles, and the resulting composite nanoparticles also had a porous structure. In pH 7.4 phosphate buffer solution (PBS), the amino groups of PEI had positive charges, and therefore the fluorescein sodium dye molecule can be loaded into the channels of PEI/Chitosan/SiO 2 composite nanoparticles by electrostatic adsorption. Furthermore, utilizing the diversity of DNA molecular conformation, we designed a high sensitive controllable assembly of DNA gated fluorescent sensor based on PEI/Chitosan/SiO 2 composite nanoparticles as loading materials. The factors affecting the sensing performance of the sensor were investigated, and the sensing mechanism was also further studied.

In this paper, a label-free and stimuli-responsive electrochemical biosensing platform was fabricated based on target-driven load release from DNA-gated metal–organic frameworks (MOFs) with cascade amplification. By using MOFs (UiO-66-NH2) as a nanocarrier of electroactive molecules (methylene blue; MB) and the programmably assembled DNA acted as the gatekeeper, the biofunctionalized MOFs (MB@DNA/MOFs) were not only used as an amplified signal label but also worked as three-dimensional tracks for biosensing. In the presence of a target, the nicking endonuclease cleavage process was triggered, leading to the generation of two strands (S1 and S2). Both S1 and S2 act as stimuli to participate in the strand displacement reaction on the MB@DNA/MOFs, which caused the unlocking of the pore to release MB, resulting in the decrease of the signal. Using carcinoembryonic antigen (CEA) as a model target, the cascade-amplified biosensor presented good performance for CEA detection, ranging from 50 fg/mL to 10 ng/mL with a detection limit of 16 fg/mL. The stimuli-responsive DNA-gated MOF-based electrochemical platform exhibited three-dimensional biosensing tracks with rational utilization of the cascade amplification, providing an effective method for cancer biomarker detection.
