We investigated a portable micro-nanochannel biosensor 3D-printed liver microtissues for rapid and sensitive deoxynivalenol (DON) detection. The screen-printed carbon electrode (SPCE) was modified with nanoporous anodic aluminum oxide (AAO), gold nanoparticles (AuNPs), and cytochrome C oxidase (COx) to enhance sensor performance. Gelatin methacrylate hydrogel, combined with hepatocellular carcinoma cells, formed the bioink for 3D printing. Liver microtissues were prepared through standardized and high-throughput techniques via bio-3D printing technology. These microtissues were immobilized onto modified electrodes to fabricate liver microtissue sensors. The peak current of this biosensor was positively correlated with DON concentration, as determined by cyclic voltammetry (CV), within a linear detection range of 2∼40 μg/mL. The standard curve equation is denoted by I CV(μA) = = 18.76956 + 0.03107C DON(μg/mL) , with a correlation coefficient R 2 was 0.99471(n=3). A minimum detection limit of 1.229 μg/mL was calculated from the formula, indicating the successful construction of a portable micro-nanochannel bio-3D printed liver microtissue biosensor. It provides innovative ideas for developing rapid and convenient instrumentation to detect mycotoxin hazards after grain production. It also holds significant potential for application in the prediction and assessment of post-production quality changes in grain.

Deoxynivalenol (DON) contamination in wheat flour induces a number of adverse health effects to consumers and livestock, even at very low concentrations. Direct detection methods for massive screening of DON in wheat flour is still lacking. A new methodology integrating multi-molecular infrared spectroscopy (MM-IR) with two-trace two-dimensional correlation spectroscopy (2T-2DCOS) was developed for in-situ qualitative and quantitative determination of DON in wheat flour as a whole. Typical spectral variation of wheat flour samples with diverse concentration of DON were stepwise characterized by MM-IR and tiny spectral profile differences resulting from concentration variation of DON were visually disclosed by 2T-2DCOS. Based on the obtained key spectral features of DON, 180 of wheat flour samples with DON higher and lower than 1.00 mg/kg were undoubtedly classified by Principal Component Analysis (PCA) and Support Vector Machines (SVM) with an accuracy rate up to 100% (for Second derivative spectra consisted of selected bands, SD-SS). Furthermore, a robust quantitative prediction model was established based on partial least squares (PLS) of SD-SS (Rc: 0.998, RMSEC: 0.135; Rp: 0.968, RMSEP: 0.421), and its excellent predictive capacity of model was validated by both residual prediction deviation (RPD) value of 3.2 and t -test. It was demonstrated that the developed methodology was applicable for screening and quantitative detection of DON in wheat flour based on the novel correlation analysis methods (SD-2DCOS-IR and 2T-2DCOS-IR) with chemometrics tools, which could be utilized both at laboratory and industrial level for quality control purposes of a large wheat flour sample set.

Deoxynivalenol (DON), a mycotoxin primarily produced by Fusarium species, is commonly found in cereal grains and poses risks to human and animal health, as well as global grain trade. This study demonstrates that methyl jasmonate (MeJA), a natural plant hormone, inhibits the growth and conidiation of Fusarium graminearum. Importantly, MeJA significantly reduces DON production by suppressing TRI gene expression and toxisome formation. To explore the molecular mechanism, we identified MeJA-tolerant mutants, including a transcription factor MRT1 and cAMP-PKA pathway-related genes (FgGPA1 and FgSNT1). MeJA treatment reduced PKA activity and intracellular cAMP levels in F. graminearum, suggesting it targets the cAMP-PKA pathway. Notably, the MeJA-resistant mutant FgGPA1R178H enhanced fungal growth, DON production, and cAMP levels in the presence of MeJA. Exogenous cAMP alleviated MeJA’s inhibitory effects on DON production, further supporting this pathway’s involvement. Interestingly, MeJA had no effect on all three MAP kinase pathways (Mgv1, Gpmk1, and FgHog1). Truncated and phospho-mimicking mutations in Mrt1 or FgSnt1 conferred MeJA resistance, suggesting they may act downstream of the cAMP-PKA pathway. In conclusion, MeJA presents a promising approach to control F. graminearum growth and DON production.

Deoxynivalenol (DON), a prevalent mycotoxin contaminating cereal crops globally, poses significant threats to animal and human health through its gastrointestinal toxicity. While DON-induced intestinal damage has been documented in mammals, its metabolic mechanisms in aquatic species remain poorly understood, particularly in adult zebrafish models that offer unique advantages for toxicological studies. Multi-omics analysis revealed 16 key differential metabolites (9 upregulated, 7 downregulated) associated with amino acid metabolism and carbohydrate homeostasis. Pathway enrichment analysis identified significant perturbations in 2-oxocarboxylic acid metabolism and sphingolipid signaling, suggesting mitochondrial dysfunction and epithelial barrier disruption as primary toxicity mechanisms. This study establishes the first adult zebrafish model for DON intestinal toxicity evaluation, demonstrating its utility in revealing conserved metabolic targets across species. The identified pathway-specific biomarkers provide novel insights for developing dietary interventions against mycotoxin exposure.

An emerging fluorescent ratiometric aptasensor based on gold nanoclusters (AuNCs) with aggregation-induced emission (AIE) properties was prepared and studied for deoxynivalenol (DON) detection. The ratiometric aptasensor used red fluorescent AuNCs 620 labelled with DON aptamer (Apt-AuNCs 620 ) as an indicator and green fluorescent AuNCs 519 modified by complementary DNA (cDNA) and magnetic beads (MBs) as internal reference, namely MBs-cDNA-AuNCs 519 . Under the optimal conditions, the aptasensor exhibited two good linear ranges of 0.1–50 and 50–5000 pg/mL for DON detection with coefficient of determination (R 2 ) of 0.9937 and 0.9928, respectively, and the low detection limit (LOD) of 4.09 pg/mL was achieved. Furthermore, this aptasensor was feasible to detect DON in positive wheat samples, and the results were in line with those from HPLC and ELISA, thus providing a promising route to detect DON with high sensitivity in cereals, even for other mycotoxins by replacing the suitable aptamer and cDNA.

A highly sensitive method based on MBs-cDNA@Apt-AuNCs 519 was developed for deoxynivalenol (DON) detection in wheat. The MBs-cDNA@Apt-AuNCs 519 was established using green emission gold nanoclusters (AuNCs 519 ) with aggregation-induced emission properties as signal probes and combining amino-modified DON-aptamer (Apt), biotin-modified DNA strand (the partially complementary to Apt (cDNA)), and streptavidin-modified magnetic beads (MBs). The Apt-AuNCs 519 were well connected with MBs-cDNA without DON but dissociated from MBs-cDNA@Apt-AuNCs 519 with the addition of DON, leading to a noticeable reduction in the fluorescent intensity of the aptasensor. Moreover, this fluorescence aptasensor showed two linear relationships in the concentration range of 0.1–50 ng/mL and 50–5000 ng/mL with a limit of detection of 3.73 pg/mL with good stability, reproducibility and specificity. The results were consistent with high-performance liquid chromatography and enzyme-linked immunosorbent assay methods, further indicating the potential of this method for accurate trace detection of DON in wheat.

In this study, an enzyme-assisted extraction method was used to extract Orchis chusua D. Don (Salep) polysaccharide (SP), which was then modified by sulfation, acetylation, phosphorylation, and carboxymethylation to obtain modified polysaccharides. Furthermore, their degree of substitution, chemical composition, and molecular weight were evaluated. The primary structural features were characterized by UV spectra, FT-IR spectra, Congo-red test, and scanning electron microscope. The phosphorylated polysaccharide (SP-P) was demonstrated the highest scavenging ability on hydroxyl radical and growth-promoting activity on Lactobacillus Bulgaricus . The carboxymethylated polysaccharide (SP-C) was exhibited the strongest DPPH and ABTS radical scavenging effects. The acetylated polysaccharide (SP-A) displayed the best proliferation effects on Bifidobacterium adolescentis , whereas the sulfated polysaccharide (SP-S) maintained moderately stable antioxidant and probiotic ability. These findings indicate that the modified polysaccharides had their potential significance as new antioxidants and probiotics for the food industry. Practical Application This article provides a new source for the development of polysaccharide derivatives as new antioxidants and probiotics for the food industry.

Fusarium graminearum (F. graminearum) is a severe pathogen threatening the safety of agriculture and food. This study aimed to explore the antifungal efficacies of several plant-derived natural compounds (vanillin and its derivatives) against the growth of F. graminearum and investigate the antifungal mechanism of 2-hydroxy-4-methoxybenzaldehyde (HMB), the strongest one. The minimum inhibitory concentration (MIC) of HMB in inhibiting mycelial growth was 200 μg/mL. HMB at MIC damaged cell membranes by increasing the permeability by about 6-fold (p < 0.05) as evidenced by propidium iodide (PI) staining. Meanwhile, the content of malondialdehyde (MDA) and glycerol was increased by 45.91 and 576.19% by HMB treatment at MIC, respectively, indicating that lipid oxidation and osmotic stress occurred in the cell membrane. Furthermore, HMB exerted a strong antitoxigenic role as the content of deoxynivalenol (DON) was remarkably reduced by 93.59% at MIC on 7th day. At last, the antifungal effect of HMB against F. graminearum was also confirmed on wheat grains. These results not only revealed the antifungal mechanism of HMB but also suggested that HMB could be applied as a promising antifungal agent in the preservation of agricultural products.

Fusarium head blight( FHB )caused by Fusarium graminearum species complex (FGSC) is one of the most important diseases around the world. Deoxynivalenol (DON) is a type of mycotoxin produced by FGSC when infecting cereal crops . It is a serious threat to the health of both humans and livestock. Trehalose-6-phosphate phosphatase ( TPP ), a conserved metabolic enzyme found in many plants and pathogens, catalyzes the formation of trehalose. N-(phenylthio) phthalimide (NPP) has been reported to inhibit the normal growth of nematodes by inhibiting the activity of TPP , but this inhibitor of nematodes has not previously been tested against F. graminearum . In this study, we found that TPP in F. graminearum (Fg TPP ) had similar secondary structures and conserved cysteine (Cys356) to nematodes by means of bioinformatics. At the same time, the sensitivity of F. graminearum strains to NPP was determined. NPP exhibited a better inhibitory effect on conidia germination than mycelial growth . In addition, the effects of NPP on DON biosynthesis and trehalose biosynthesis pathway in PH-1 were also determined. We found that NPP decreased DON production, trehalose content, glucose content and TPP enzyme activity but increased trehalose-6-phosphate content and trehalose-6-phosphate synthase ( TPS ) enzyme activity. Moreover, the expression of TR I 1 , TR I 4 , TR I 5 , TR I 6 , and TPP genes were downregulated, on the contrary, the TPS gene was upregulated. Finally, in order to further determine the control ability of NPP on DON production in the field, we conducted a series of field experiments, and found that NPP could effectively reduce the DON content in wheat grain and had a general control effect on FHB. In conclusion, the research in this study will provide important theoretical basis for controlling FHB caused by F. graminearum and reducing DON production in the field.

Glutamine antagonists, such as 6-diazo-5-oxo-L-norleucine (DON), have demonstrated remarkable anti-tumor effects by blocking tumor glutamine metabolism, but their use is frequently causing toxicity. Despite the existence of multiple glutamine antagonist prodrug designs, a tumor-selective prodrug has yet to be developed. Herein, a novel prodrug of DON, Azo-DON, has been developed, which remains stable and inactive in normal tissues with sufficient oxygen levels, while can be selectively reduced to DON by highly expressed azo-reductase in hypoxic tumor environments. This leads to blockade of glutamine metabolism in cancer cells and promotes cell death without affecting T cell proliferation. In a high-hypoxic H22 hepatoma cancer model, Azo-DON showed a 1.8-fold enhancement in glutamine blockade compared to the control group, resulting in a tumor suppression rate (TSR) of 84.2% in vivo with no significant weight loss. In a low-hypoxic CT26 colon cancer model, when combined with vascular disrupting agent nanoparticles (CBP) to induce a hypoxic environment, Azo-DON exhibited a 4.6-fold enhancement in glutamine blockade over the control group, resulted in a remarkable TSR of 96.6% in vivo . This innovative approach represents a promising strategy for the application of broad-spectrum metabolic inhibitors in the field of precision cancer treatment.

Dissolved organic nitrogen (DON) plays a key role in many biogeochemistry and engineering processes such as forming nitrogenous disinfection byproducts . However, detecting aqueous DON at trace levels is challenging currently because conventional DON conversion methods have very high method detection limits (MDL). In addition, DON is measured indirectly by subtracting dissolved inorganic nitrogen (DIN) from total dissolved nitrogen (TDN), which can propagate analytical errors of each analyte. In order to solve these issues, we isolated DON from DIN with electrodialysis before and herein tested vacuum ultraviolet (VUV) in tandem with several oxidants to convert DON completely into nitrate for subsequent N analysis. Results showed that H 2 O 2 was more suitable than chlorine and persulfate because VUV/chlorine or VUV/persulfate is either inefficient to convert DON or subjected to nonnegligible N loss. To verify the efficiency, we evaluated the effects of typical water and operating variables on the conversions of four model DON compounds and their yields of nitrate. Under optimal conditions (pH 10.3 and 500 mg/L H 2 O 2 ), the process converted DON completely into nitrate within just 60 min. Compared to conventional TDN analytical methods, the VUV/H 2 O 2 method features not only better analytical precision but also lower MDL because the formed nitrate can be analyzed at very low MDL by ion chromatography (IC). So, this approach moves one step further to achieve a conceptually new DON analytical method by coupling electrodialysis, VUV/H 2 O 2 , and IC.

Deoxynivalenol (DON) contamination in cereals and their products poses a potential threat to animal and human health, however, physical and chemical detoxification methods deplete nutrients and cannot specifically remove DON. This study aims to identify novel and efficient DON-degrading enzymes, providing practical support for their application in biodegradation. A novel DON-degrading aldo-keto reductase named AKR11A2 was identified from Bacillus velezensis Vel-HNGD-F2 through BlastP comparison. AKR11A2, an enzyme with a molecular mass of 34.8 kDa encoded by a 933 bp gene, exhibited optimal activity at pH 9 and 40°C, while demonstrating remarkable thermal and alkaline stability by retaining over 90% of its activity. UPLC-MS/MS analysis revealed that the m/z of the DON degradation product was 295.1, identified as 3- epi -DON, formed through the direct isomerization of DON. Notably, zebrafish experiments demonstrated that the liver toxicity of the degradation product was significantly lower than that of DON. AKR11A2 effectively degraded 50.69% of the DON in contaminated corn, highlighting its practical application in food safety. These findings indicate that the study achieved the biodegradation of DON and provide a promising theoretical and technological support for the application of DON detoxifying enzymes in food and feed products.
