Paroxetine, a selective serotonin reuptake inhibitor, is widely used in the clinical treatment of depression. While several antidepressants show promise as geroprotectors, the role of paroxetine in aging remains unclear. In this study, we evaluated the lifespan extension effect of paroxetine in Caenorhabditis elegans ( C. elegans ) and elucidated the underlying mechanisms. The results showed that paroxetine can prolong lifespan concomitant extension of healthspan as indicated by increasing mobility and reducing lipofuscin accumulation, as well as confer protection to nematodes against different abiotic stresses. Paroxetine upregulated ser-7 expression and downregulated dop-4 expression. dop-4 RNA interference (RNAi) mimicked the beneficial effect of paroxetine on lifespan. Conversely, ser-7 RNAi abolished paroxetine-induced lifespan extension and the expression changes of dop-4 and genes related to insulin/insulin-like growth factor 1 signaling (IIS). Moreover, paroxetine exhibited a comparable lifespan extension effect to that observed in daf-2 or age-1 mutants; however, this effect was abolished in daf-16 mutant. Taken together, these results suggest that paroxetine promotes health and longevity in C. elegans through the ser-7 - dop-4 -IIS pathway, underscoring its potential as a geroprotector.

In this study, the kinetics and mechanism of UV/O 3 synergistic oxidative digestion of dissolved organic phosphorus (DOP) were investigated, focusing on the ozone direct oxidation and hydroxyl radical oxidation parts of glufosinate and triphenyl phosphate (TPhP). The p-chlorobenzoic acid (p-CBA) was selected as the probe compound, and two kinds of reaction kinetic models were proposed by competitive kinetic method with R ct according to the different scale of rate constants of hydroxyl radical oxidation. Under the condition of weakly alkaline (pH = 9.0) and weakly acidic environment (pH = 5.0), the second-order rate constants of glufosinate and TPhP was determined indirectly to be k o 3/glufosinate = (2.903 ± 0.247)M −1 s −1 and k o 3/TPhP = (3.307 ± 0.204) M −1 s −1 by ozone direct oxidation, and k ·OH/glufosinate = (1.257 ± 1.031) × 10 9 M −1 s −1 and k ·OH/TPhP = (7.120 × 10 8 ± 0.963) M −1 s −1 by hydroxyl radical oxidation, respectively. The comparison of the contribution levels of the two parts to the digestion process showed that the contribution levels in the digestion of glufosinate and TPhP processes both the contribution of ·OH were higher than those of ozone, 86.3% and 72.6%, respectively.

α -Methyl groups play significant roles in the regulation of water molecules within both small molecular systems and bio-macromolecular systems. Systematically studying the influence of α -methyl on the dynamics of water molecules within hydrogel systems is therefore worthwhile. In this study, we prepared a series of hydrogen-bonded (H-bonded) hydrogels with varying densities of α -methyl groups by copolymerizing methacrylamide (MAm) with its α -methyl-absent analogue, acrylamide (Am). Introducing a small amount of Am (≤6 mol%) into the polymer chain resulted in significant shifts in the viscoelasticity of the hydrogels. The hydrogels exhibit a “time-temperature- α -methyl equivalence”, meaning that introduction of α -methyl-absent monomer has effects similar to elevating temperature and prolonging observation time on the dynamic properties. Based on low-field nuclear magnetic resonance spectroscopy and Raman scattering, a “hydrophilic defects-assisted H-bonds dissociation” mechanism is proposed, depicting that the α -methyl-absent monomer can disturb the rearrangement of water molecules surrounding the polymer chain and accelerate chain dissociation. These findings enabled the copolymer hydrogels with functions such as fast self-healing and tunable interface adhesion.

Purpose Psoriasis is a skin disease characterized by excessive proliferation, inflammation and oxidative stress in keratinocytes. The present study aimed to investigate the therapeutic effects of Dendrobium officinale polysaccharide (DOP) on keratinocyte psoriasis-like models. Methods The HaCaT keratinocyte inflammation models were induced by interleukin (IL)-22 or lipopolysaccharide (LPS), respectively, and oxidative stress damage within cells was elicited by H 2 O 2 and treated using DOP. CCK-8 and EdU were carried out to detect cell proliferation. ELISA, qRT-PCR, and Western blot were conducted to measure the expression of pro-inflammatory cytokines IL17A, IL-23, IL1β, tumor necrosis factor alpha (TNF-α), and IL-6. Reactive oxygen species (ROS) level in keratinocytes was detected by flow cytometry. Cell proliferation-associated proteins (PCNA, Ki67, Cyclin D1) and pathway proteins (p-AKT and AKT), and oxidative stress marker proteins (Nrf-2, CAT, SOD1) were detected by Western blot. Result DOP did not affect the proliferation of normal keratinocytes, but DOP was able to inhibit the proliferative activity of IL-22-induced overproliferating keratinocytes and suppress the expression of proliferation-related factors PCNA, Ki67, and Cyclin D1 as well as the proliferation pathway p-AKT. In addition, DOP treatment was able to inhibit IL-22 and LPS-induced inflammation and H 2 O 2 -induced oxidative stress, including the expression of IL17A, IL-23, IL1β, TNF-α, IL-6, and IL1β, as well as the expression levels of intracellular ROS levels and cellular oxidative stress-related indicators SOD, MDA, CAT, Nrf-2 and SOD1. Conclusion DOP inhibits keratinocyte hyperproliferation, inflammation and oxidative stress to improve the keratinocyte psoriasis-like state.

Poly(methylacrylamide) (PMAM) is a thermo-responsive polymer with an upper critical solution temperature (UCST). Its cloud point ( T cp ) is around 60 °C, unsuitable for certain biomedical and industrial applications. This study brought up a copolymerization strategy to tune the T cp of PMAM with hydrophilic comonomers. Surprisingly, with a small portion of hydrophilic monomer doped, the T cp of the PMAM copolymer can be significantly shifted. For instances, with ≤7 mol% of acrylamide or 1 mol% of oligo(ethylene glycol) methacrylate, the T cp can be shifted in a wide range from ~69 to ~0 °C. Microdifferential scanning calorimetry demonstrated that the enthalpic effect during the phase transition of the solutions is indistinctive, while fluorescence measurement with pyrene as a probe revealed that the hydrogen-bonding within polymer chains is enhanced by the hydrophobic aggregation of methyl groups. Therefore, the doped hydrophilic monomer could remarkably alter the ordering of water-molecules and the extent for the aggregation of methyl groups, leading to the pronounced shifting in the T cp of the copolymers. This work would facilitate the application of PMAM as smart polymer materials and guide the inventions of functional materials based on UCST polymers. Graphical
