In this study, we synthesized DOX-loaded H-ZIF-8@CaCO 3 -PEG nanocarrier, which combined with calcium overload and photothermal therapy (PTT) for synergistic therapy. The nanocarrier comprised a PEG modified calcium carbonate (CaCO 3 ) shell and a hollow imidazolate zeolite skeleton (H-ZIF-8) core, in which the chemotherapy drug doxorubicin hydrochloride (DOX) and photosensitizer indocyanine green (ICG) were simultaneously co-loaded. In vitro cellular experiments revealed that the lethality of DOX&ICG@H-ZIF-8@CaCO 3 -PEG against tumor cells was significantly higher than that of the single chemotherapy owing to the combination with chemotherapy and photothermal therapy as well as calcium overloading. This drug-loaded nanocarrier exhibits notable characteristics such as exceptional stability, elevated drug loading capacity and efficient release of the loaded drug, which provides a promising perspective for efficient synergistic therapy.

Carbon quantum dots (CQDs) are extensively applied for bioimaging due to their unique photoluminescence properties, while less attention has been paid to their inherent bio-functions as targeted drug delivery vehicles. To address this limitation, a novel CQDs-based drug delivery system ( [email protected] ) was constructed using l -arginine as precursor with duplex metal co-doping (Ag&Cu) for further DOX-loading/releasing and synergistic tumor theranostics. Specifically, the functional residues retention ability of CQDs guaranteed the reactive nitrogen species (RNS) replenishment, Ag-doping enhanced the photoluminescence intensity, and Cu 2+ -functionalizing could act as the anchor for further interparticle self-assembly, resulting in intelligent adjustable CQDs size for increasing the drug-loading capacity. After reaching targeted sites, CQDs could act as NO/ONOO − donors for gas-involving therapy, and Cu 2+ could not only generate ROS through chemodynamic therapy (CDT), but also modulate the band structure of as-prepared CQDs for laser-induced photodynamic therapy (PDT) to further replenishing ROS. Simultaneously, with Cu 2+ depletion, the structure of [email protected] was disrupted, which guaranteed in situ DOX-releasing. As such, these orchestrated procedures fully exerted the intrinsic bio-functions of CQDs then act as adjuvants in drug delivery systems, thus representing synergistic effects in suppressing tumor progression and migration with minimal side effects, and provides a paradigm of potent CQDs-based nanotheranostics.

As drug-delivery carriers for cancer chemotherapy, gatekeeper-capped mesoporous silica nanoparticles (MSNs) have been widely studied due to their high drug-loading capability, controlled drug release property and good biocompatibility. However, the currently reported gatekeeper-capped MSNs suffer from complex synthetic procedures, potential toxicity of gatekeepers, unsatisfactory control on drug stimuli-release, etc. In this work, we develop a simple but efficient approach to fabricate PEGylated organosilica-capped mesoporous silica nanoparticles (POMSNs) by employing a disulfide-doped organosilica coating as the gatekeeper formed by the hydrolysis and condensation of a silane coupling agent 3-(mercaptopropyl)trimethoxysilane (MPTMS) to block the mesopores of MSNs. Owing to the glutathione (GSH)-responsive biodegradation behavior of the disulfide-doped organosilica gatekeeper, the DOX-loaded POMSNs exhibit only 20% cell viability towards SMMC-7721 tumor cells, and almost no toxicity towards L-02 cells at a DOX concentration of 50 μg mL−1 was measured, demonstrating their selective cytotoxicity in vitro. More importantly, it is demonstrated that the DOX-loaded POMSNs exhibit a tumor inhibition rate of 71.3% and negligible systematic toxicity. Consequently, the resultant POMSNs show great potential as drug nanocarriers for redox-responsive drug release and passive-targeting tumor chemotherapy.

Thioketal and thioether are moieties used to fabricate reactive oxygen species (ROS)-responsive polymers for drug delivery. In this paper, three amphiphilic copolymers of mPEG-poly(ester-thioether), mPEG-poly(thioketal-ester) and mPEG-poly(thioketal-ester-thioether) were synthesized. The ROS-responsive behaviors of the three copolymers nanoparticles as drug carriers were investigated. The ROS-sensitivity was demonstrated by NMR, DLS, and SEM. mPEG-poly(ester-thioether) nanoparticles exhibited the fastest drug release rate, which possessed the best ROS sensitivity. The in vitro anticancer activity of the DOX-loaded nanoparticles was studied, the results revealed that the mPEG-poly(ester-thioether) nanoparticles showed the most efficient anticancer activity. Notably, all the three ROS-responsive copolymers nanoparticles showed enhanced cellular uptake and anticancer efficacy comparing to the control mPEG- b -PCL nanoparticles.
