Magnetite (Fe3O4) nanoparticles as drug carriers can achieve precise drug target due to their magnetic property. However, they are easy to aggregate in the physiological environment, which obviously limits their application in drug delivery. The development of the Fe-MIL-88B-derived method to construct the Fe3O4-loaded mesoporous carbon (Fe3O4/carbon) system is a feasible strategy to solve the issue. First, iron atoms evenly distribute in the organic links through coordination bonds in Fe-MIL-88B. After the carbonization of Fe-MIL-88B, mesoporous carbon acts as a barrier to prevent the aggregation of Fe3O4 nanoparticles. Herein, Fe-MIL-88B particles were fabricated by the hydrothermal method and then pyrolyzed to construct Fe3O4/carbon systems. Results showed that Fe3O4 nanoparticles uniformly in situ grew on mesoporous carbon generated by the carbonization of organic components. More encouragingly, the Fe3O4/carbon system loaded with DOX demonstrated pH-responsive DOX release, efficient delivery of DOX into cancer cells, and significant cancer cell killing ability. Therefore, the Fe3O4/carbon systems prepared by the Fe-MIL-88B-derived method might open up a way for targeted and controlled drug delivery.

Herein, we construct a charge - switchable polymer nano micelles poly (2-(hexamethyl eneimino) ethyl methacrylate) - b - poly (ethylene glycol) monomethyl ether methacrylate) - b - poly (diethyl enetriaminepentaacetic acid methacrylate) - b - poly (1-vinyl imidazole) - b - poly (4-vinyl phenylboronic acid) (PC7A-PEG-DTPA-VI-PBA) in different pH solutions. DOX released faster from micelles in a weakly acidic environment (pH 5.0) than at pH 7.4. In order to enhance the anti-tumor effect, the imidazole functional groups in the polymer were used to coordinate CdSeTe quantum dots (QDs) for photodynamic treatment (PDT). In addition, the surfaces of the micelles were further decorated with phenylboronic acidas a targeting group, using DTPA chelating 99mTc for SPECT imaging. It has been successfully demonstrated that the nanoparticles have a good cumulative effect on the tumor site. The structure of the polymer was characterized by 1HNMR. The morphology and particle size of the micelles were characterized by transmission electron microscopy (TEM) and dynamic light scattering (DLS). The drug loading capacity (DLC) and drug loading efficiency (DLE) of the micelles were analyzed by ultraviolet visible spectroscopy. And the pH-sensitive drug release and cytotoxicity of the micelles were verified in vitro. In vitro experiments showed that the nano micelles were noncytotoxic to different cell lines, while DOX@CdSeTe@PC7A-PEG-DTPA-VI-PBA inhibited the proliferation and promoted the apoptosis of B16F10 cells. An in vivo study with C57BL tumor-bearing mice indicated that DOX@CdSeTe@PC7A-PEG-DTPA-VI-PBA nano micelles efficiently inhibited tumor growth. Results showed that the nano micelles had good pH responsibility and biocompatibility, and the loaded DOX could be released in the weak acidic environment of tumor cells, and it was expected to be a good drug delivery system.

Hybrid CaCO 3 microparticles coated by sodium poly(styrene sulfonate) (PSS) and aliphatic poly(urethane-amine) (PUA) were developed as thermal-/pH-responsive drug delivery vehicles via LbL self-assembly technique. The DOX release from the CaCO 3 microparticles was higher than 60% within 36 h, whereas the value of PUA/PSS-coated microparticles was only 20%. The results demonstrated that the PUA/PSS multilayer coating could reduce the drug release rate and significantly assuage the initial burst release of DOX. In addition, the drug release of the hybrid microparticles was found to be thermal-/pH-dual responsive. More interestingly, more than 90% of DOX was released in 36 h at pH 2.1 and 55 °C owing to the combined action of the dissolution of the CaCO 3 core and the shrinkage of aliphatic PUA.

The combination of chemo-photothermal therapy applied to tumor ablation, overcoming the shortcomings of traditional cancer treatment and has become a research hotspot. Tungsten disulfide nanosheets (WS 2 ) are used as the core, and gold nanoparticles is incorporated into the surface layer of WS 2 (WS 2 /Au) by the sodium citrate reduction method, and then gold-doped tungsten disulfide surface is coated by lipids (WS 2 /Au-lipids). The biocompatibility and stability of this functional nanocomposite has been strongly improved under physiological conditions. The excellent photothermal conversion characteristics of WS 2 /Au-lipid has been verified by the photothermal experiment both in vitro and in vivo. Furthermore, the DOX loaded by WS 2 /Au-lipid has a dual response release curve of pH and NIR. The results of in vitro photothermal and chemotherapy combined treatment shows that the survival rate of the cell of the combined treatment group is obviously lower than the single treatment group. Through the investigation of photothermal performance in vivo, it has been found that after modification, the photothermal performance of the compound has been improved, and the drug can be targeted for delivery to the tumor site, making it more concentrated in cancer cells. In addition, in vivo study confirms that the combined treatment group has a more obvious therapeutic effect on tumors than the single treatment group, and no systemic toxicity was shown. It is an excellent carrier in many aspects and has unlimited development and prospects in the field of multiple cancer treatments.

Pre-existing tumor hypoxia and oxygen consumption by photodynamic therapy (PDT) always generate an inadequate oxygen supply, which often contributes to other therapy resistance, especially for aerobic treatments such as chemotherapy. In order to overcome this problem, we synthesized a chitosan (CS)-capped pH-/NIR-dual responsive and perfluorocarbon-based oxygen self-enriching photodynamic therapy nanosystem which could codeliver oxygen, photosensitizer, and chemotherapeutic drug. Simultaneously, in vitro results revealed that DOX release was stimulated by the acidic pH value with CS as a pH-responsive layer and rising temperature caused by the photothermal effect with an 808 nm laser, which could effectively reduce the nonspecific toxicity caused by premature drug release. Furthermore, due to the higher oxygen-carrying capability of perfluorocarbon, the decreased expression of the hypoxia-inducible factor-1α (HIF-1α) of tumors, as well as the significantly intense photodynamic effect and longer singlet (1O2) lifetime of the loaded photosensitizer, the elevated cytotoxicity was improved. More importantly, both in vitro and in vivo results showed that drug-loaded nanoparticles with oxygen-abundant properties achieved a similar anticancer therapy in both normoxic and hypoxic conditions against the difficulty led by deficient oxygen, as well as a good cellular internalization. Summarily, all these results pointed out that such a pH-/NIR-dual responsive drug carrier could enhance the synergetic anticancer therapy of chemotherapy and PDT by alleviation of tumor hypoxia, which provided a tempting controllable and multifunctional therapeutic application.

The nanoplatform of synergistic chemo-photothermal therapy has superior advantages on antitumor. It is urgently needed to explore novel nanocarrier for improving photothermal performance in drug delivery process. Herein, we synthesized polydopamine doped mesoporous silica-coated reduced graphene oxide (rGO/MSN/PDA) by simply adding dopamine hydrochloride into the oil-water biphasic reaction system as a multifunctional drug carrier for anticancer treatment, which combines chemotherapy and photothermal therapy. The rGO/MSN/PDA showed nearly twice the photothermal conversion efficiency of mesoporous silica-coated graphene oxide (GO/MSN) due to the reduction of GO and doping with PDA. In addition, the rGO/MSN/PDA showed pH-response DOX release abilities, which means higher release of DOX in tumor cells. The cell experiments in vitro proved that rGO/MSN/PDA with better biocompatibility compare to GO/MSN might offer a promising tool for improving the therapeutic effects of hepatocellular carcinoma cells through synergistic chemo-photothermal therapy.

Innovative treatments for peritoneal metastatic cancer have attracted widespread attention from researchers. Here, we propose a drug-loaded nanomotor (PSBMA/ l -Arg/DOX, PLD) based on zwitterionic materials for the treatment of peritoneal metastatic cancer through intraperitoneal injection. Zwitterionic polymer nanocarriers (PSBMA NPs) are obtained by radical polymerization with zwitterionic SBMA as the polymerization monomer and N,N’-Bis(acryloyl)cystamine (BAC) as the cross-linking agent. The zwitterionic substrate of this nanomotor has the ability to resist non-specific protein adsorption in ascites. The loaded l -arginine enables the nanomotor to have the ability to chemotaxis towards high concentrations of ROS/iNOS in tumors and be catalyzed to produce NO, achieving deep penetration into tumor tissue. Furthermore, the disulfide bond (S S) carried by the crosslinking agent used in the preparation of the nanomotor can respond to the high expression of reducing glutathione in the tumor microenvironment and undergo degradation, releasing a large amount of loaded drug DOX. Cell and animal disease model experiments confirme the good therapeutic effect of this drug-loaded nanomotor, providing new therapeutic concepts and strategies for the treatment of peritoneal metastatic cancer.

In this report, uniform LaF3:Ln mesoporous spheres have been synthesized by a facile and mild in situ ion-exchange method using yolk-like La(OH)3:Ln mesoporous spheres as templates, which were prepared through a self-produced bubble-template route. It was found that the structures of the final LaF3:Ln can simply be tuned by adding a polyetherimide (PEI) reagent. LaF3:Ln hollow mesoporous spheres (HMSs) and LaF3:Ln flower-like mesoporous spheres (FMSs) were obtained when assisted by PEI and in the absence of PEI. The up-conversion (UC) luminescence results reveal that the doping of Nd3+ ions in LaF3:Ln can markedly influence the UC emissions of the products. It is interesting that an obvious thermal effect is achieved due to the energy back-transfer from Tm3+ to Nd3+ ions under 980 nm near-infrared (NIR) irradiation. The LaF3:Yb/Er/Tm/Nd HMSs show good biocompatibility and sustained doxorubicin (DOX) release properties. In particular, upon 980 nm NIR irradiation, the photothermal effect arising from the Nd3+ doping induces a faster DOX release from the drug release system. Moreover, UC luminescence images of LaF3:Yb/Er/Tm/Nd HMSs uptaken by MCF-7 cells exhibit apparent green emission under 980 nm NIR irradiation. Such a multifunctional carrier combining UC luminescence and hyperthermia with the chemotherapeutic drugs should be of high potential for the simultaneous anti-cancer therapy and cell imaging.

A novel near-infrared (NIR) light-triggered smart nanoplatform has been developed for cancer targeting and imaging-guided combined photothermal-chemo treatment. Notably, Ag 2 S has a dual function of photothermal therapy and fluorescence imaging, which greatly simplifies the structure of the system. It can emit fluorescence at 820 nm under an excitation wavelength of 560 nm. The phase-change molecule of 1-tetradecanol (TD) is introduced as a temperature-sensitive gatekeeper to provide the nanocarrier with controlled release capability of doxorubicin (DOX). The nanocarrier (HAg 2 S@mSiO 2 -TD/DOX) shows a high drug loading capacity of 26.3% and exhibits an apparent NIR-responsive DOX release property. Under NIR irradiation, the photothermal effect of HAg 2 S nanocores facilitated the release of DOX through the melting of TD. The cytotoxicity test shows that the nanocarriers have good biocompatibility. As the same time, the synergistic combination leads to a better cancer inhibition effect than individual therapy alone in vitro. Cell uptake tests indicate that the carriers have excellent fluorescence imaging ability and high cellular uptake for HepG2 cells. This work provides a new strategy for the fabrication of smart nanocarriers with simple structures for fluorescence-mediated combination cancer therapy. Redox-responsive ferrocene (Fc)-grafted supramolecular block copolymers functionalized with hyaluronic acid (HA-g-β-CD/Fc-polymers or HAP) or folic acid (FA-g-β-CD/Fc-polymers or FAP) were synthesized by host–guest interaction. The copolymer structures were characterized by Fourier transform infrared spectroscopy, nuclear magnetic resonance, and gel permeation chromatography. The block copolymers self-assemble into spherical micellar aggregates in aqueous solution, and exhibit reversible redox characteristics in the presence of an oxidant and reductant. The tunable redox response was evaluated by transmission electron microscopy, ultraviolet–visible spectroscopy, cyclic voltammetry and dynamic light scattering. The micelles rapidly release DOX under oxidative and acidic condition in vitro. Furthermore, presence of HA and FA on the micelles enabled effective uptake by the CD44 and FA receptor-overexpressing cancer cells. Taken together, the redox responsive Fc-block copolymer micelles have good biocompatibility, biodegradability, low toxicity, and high selectivity, and are a highly promising drug delivery system for cancer therapy.

The redox homeostasis in tumor cells is an innate defense barrier for reactive oxygen species (ROS)-mediated tumor therapy. Herein, an intratumoral cascade-responsive multifunctional nanodiamond-based platform (NPCD) with a hydrodynamic diameter of 191 ± 5 nm triggering redox dyshomeostasis is fabricated by Cu2+-mediated coordination for polylysinated nanodiamond and doxorubicin (DOX) to achieve cancer-specific diagnosis and therapy. The NPCD nanoplatform enables precise tumor fluorescence localization and chemotherapy through a “turn on” effect due to being specifically activated by GSH to trigger DOX release at a low-pH tumor-specific environment instead of normal cells. Furthermore, NPCD realizes Cu+-based chemodynamic therapy (CDT) and induces GSH depletion by Cu2+ reduction to support CDT. These cause intracellular redox dyshomeostasis, making tumor cells more sensitive to the NPCD nanoparticles. Additionally, NPCD also possesses the intrinsic ability to reverse drug resistance. Of note, NPCD mainly accumulates in the tumor site in 4T1 tumor-bearing mice and enhances the efficacy of therapeutic interventions with negligible side effects. More interestingly, combined with programmed cell death ligand 1 antibody (anti-PD-L1) and imiquimod (R837), NPCD significantly suppresses distant tumor growth, indicating a good immune response. Therefore, this work opens perspectives for the further development of synergistic therapy through a Cu2+-mediated intratumoral cascade reaction.
